Introduction. Alzheimer’s disease (AD) is one of the most common neurodegenerative diseases, which is a serious health problem for societies that live longer. Spontaneous dominant mutations and polymorphisms of selected genes play an important role in development of AD. Aim. Several polymorphisms in selected genes strongly associated with development of Alzheimer’s disease were highlighted in this review: APOE, CYP46, APP, PSEN1, PSEN2, UBQLN1, BACE1, PRND, APBB2, TOMM 40. These gene polymorphisms have a significant role in the development of Alzheimer’s disease and they have potential to be biomarkers. Researchers combine efforts to find significant polymorphisms that would ensure that a person is predisposed to the occurrence of disease symptoms. This topic is often taken up by scientists seeking to develop effective genetic tests for diagnosing AD. Material and methods. Analysis of literature from web of knowledge: Web of Science (all database), NCBI and PubMed. Results. We reviewed the selected important genes and polymorphisms which are most often associated with development of AD. Conclusion. It should be noted that nowadays scientists strive not to focus on only one polymorphism in the gene but on several polymorphisms in different genes concomitantly and above all on interactions between them to the diagnosis of this disease. Only this approach to AD will contribute to the creation of appropriate identification methods. Moreover, we should use the new generation tools - the platform for collecting data and personalized medicine.

The aging population is a significant social, medical and economic problem due to increasing prevalence of chronic diseases in elderly population. Alzheimer's disease (AD) is the most common form of dementia and the most common neurodegenerative disease. It is characterized by a progressive deterioration of memory and cognitive function. So far, there is neither an effective prevention nor cure for dementia, so more and more attention is paid to the prevention of this group of diseases, particularly to the appropriate diet. Preventive intervention gives the best results if introduced before the first symptoms of dementia, i.e., around the age of 50. This is when the nutritional status, number of synapses, cognition, and neuropathological changes in the nervous system compensate each other, which increases the chances of staying healthy for a longer period of time. It has been proven that dietary habits, which lead to the development of cardiovascular and metabolic diseases, significantly increase the risk of dementia. On the other hand, a Mediterranean diet rich in antioxidants, fiber and omega-3 polyunsaturated fatty acids may have a protective effect on the neurodegenerative process. The beneficial effect of many nutrients on the course of AD has been demonstrated. These include: glutathione, polyphenols, curcumin, coenzyme Q10, vitamins B6, B12, folic acid, unsaturated fatty acids, lecithin, UA, caffeine and some probiotic bacteria. A diet rich in saturated fatty acids and branched-chain amino acids (BCAA) promotes the progression of dementia. Dietary intervention should be introduced as early as possible to minimize the risk of developing dementia. The Mediterranean and DASH diets have been documented to protect against AD. However, the MIND diet is reported to be much more effective in preventing cognitive decline/dementia than either the Mediterranean or DASH diets alone.

Alzheimer’s disease (AD) is an incurable neurodegenerative disease, which is accompanied by chronic inflammation. The immune system has an important role in the process of the disease. The deposition of amyloid β (Aβ) protein is a key pathological feature in Alzheimer’s disease. This article reviews immunotherapeutic strategies against AD. In murine models of AD, both active and passive immunization against Aβ induces a marked reduction in an amyloid brain burden and an improvement in cognitive functions. The findings from murine studies lead to clinical studies. One Phase II clinical trial of active immunization against Aβ was discontinued after 18 patients developed meningoencephalitis. After this lesson learned, new immunotherapeutic strategies, including both active and passive immunization, are investigated in clinical centers.
Choroba Alzheimera (AD) jest nieuleczalną chorobą neurodegeneracyjną, której towarzyszy przewlekły proces zapalny. Układ immunologiczny może mieć istotny wpływ na przebieg procesu chorobowego. Głównym patologicznym wyznacznikiem choroby Alzheimera jest gromadzenie w obrębie mózgu złogów β-amyloidu. W obecnym artykule przedstawiono informacje na temat możliwości zastosowania immunoterapii w leczeniu choroby Alzheimera. Metody immunoterapeutyczne, mające usuwać amyloid β z chorych mózgów, dały bardzo pozytywne rezultaty w badaniach na zwierzętach. Zarówno metody aktywnej, jak i biernej immunizacji powodowały wyraźne zmniejszenie zawartości amyloidu w mózgach myszy transgenicznych oraz poprawę ich funkcji poznawczych. Bardzo dobre wyniki na modelach zwierzęcych pozwoliły przeprowadzić wstępne badania kliniczne. Ich wyniki są również obiecujące, choć obarczone ryzykiem zapalenia mózgu. Jedyne dotychczas badanie II Fazy z zastosowaniem szczepionki przeciwko Aβ zostało przerwane z powodu rozwoju u 18 pacjentów zapalenia opon mózgowych i mózgu. Obecnie ośrodkach badawczych pojawiają się nowe rodzaje technik immunoterapeutycznych.

The most common form of dementia in the elderly population is Alzheimer’s disease (AD). World Health Organization (WHO) thus defines Alzheimer’s disease: “a set of neurodegenerative brain symptoms, resulting in progressive impairment of memory, thinking, cognition, counting, language, ability to learn and assessing situations, which disturb everyday life”. It usually develops in 65+-yearolds, and the risk of AD in 85+-year-olds is as high as 50%. It has become a considerable threat to the society, given the longer life expectancy and an increase in the retirement-aged population. AD prevention and treatment methods described in this paper are still being developed and perfected. Studies on genesis of the disease aim at comprehensive understanding of its causes. New, alternative treatment methods are still sough. Researchers develop and test biomarkers that could facilitate early diagnosis.

Dementia, the plague of the aging populations, is highly prevalent in East Asia, with currently over 9,8 million dementia patients. Alzheimer’s disease (AD), the most common cause of dementia, awaits for more detailed research in terms of understanding patomechanism of the disease, but more importantly, in supporting the treatment of this incurable disease in modern medicine. Therefore, alternative therapies assisting traditional therapeutic options become more popular, with the comeback of traditional medications, such as Traditional Japnese Medicine (TJM) - Kampo. The aim of this study was to present the use and effectiveness of TJM in treatment of symptoms of AD. Among articles in PubMed Medline database in years 2008-2018, 8 articles were selected for further analysis. Most of the research (38%) consisted of observational studies using different neuropsychiatric scales and methods of assessment of dementia. Studies presented significant improvements of behavioral symptoms with additional therapy with Yokukansan, Orengedoku-to, Ninjin’yoeito and other Kampo formulas in AD patients. Reviewed research presents effectiveness of Kampo treatment in supporting of cognitive and behavioral symptoms reduction, as well as protection of beta-amyloid induced toxicity. However, more research involving bigger cohorts is needed in order to confirm results on the larger scale, in order to introduce the beneficial aspects of Kampo in AD populations worldwide.

Lately, different therapy strategies for treating or slowing the progression of Alzheimer's disease are being analyzed. Moreover, the last two decades have seen a considerable research effort directed towards discovering the causes of Alzheimer's disease with the ultimate hope of developing safe and effective pharmacological treatments. In addition to the therapeutic strategies based on targeted drugs, the regimens will require the simultaneous application of neuroprotective drugs. Therefore, although there is currently no "cure" for Alzheimer's disease, a large number of potential therapeutic strategies emerged lately. In this small mini-review we will selectively describe some of the compounds derived from plants that could have a great potential in the treatment of various diseases, including Alzheimer's disease. In this way, there are many plant species that have been traditionally used for memory disorders. The differentiated results and powerful activity of these extracts are making these neuroprotective strategies to be somehow plausible for the treatment of Alzheimer's disease. In addition, these plants can be examined in order to isolate and identify their active ingredients and this can serve as a starting point to find safer and more effective agents for therapeutic use. On thing is certain: as the effective treatment options are limited, there is a demand for new drugs. Thus, plant extracts or vegetal compounds could represent an important part in this equation.

Alzheimer’s disease is the most common form of dementia that can cause a brain neurological disorder with progressive memory loss as a result of brain cell damage. Prevention and treatment of disease is a key challenge in today’s aging society. Accurate diagnosis of Alzheimer’s disease plays an important role in patient management, especially in the early stages of the disease, because awareness of risk allows patients to undergo preventive measures even before irreversible brain damage occurs. Over the years, techniques such as statistical modeling or machine learning algorithms have been used to improve understanding of this condition. The objective of the work is the study of the methods of detection and progression of Alzheimer’s disease through artificial intelligence techniques that have been proposed in the last three years. The methodology used was based on the search, selection, review, and analysis of the state of the art and the most current articles published on the subject. The most representative works were analyzed, which allowed proposing a taxonomic classification of the studied methods and on this basis a possible solution strategy was proposed within the framework of the project developed by the Cuban Center for Neurosciences based on the conditions more convenient in terms of cost and effectiveness and the most current trends based on the use of artificial intelligence techniques.

Alzheimer’s disease (AD) is the most common cause of dementia and is responsible for up to 75% of the nearly 47 million dementia cases worldwide. AD is the clinical manifestation of toxic β-amyloid and neurofibrillary tangles accumulation caused by altered proteostasis. Due to not fully understood molecular mechanisms of AD only symptomatic treatment is available. Considering that cognitive decline is associated with insulin resistance, metabolic alterations may be the primary causes of the AD and insulin therapy is proposed as a novel approach in AD treatment. Intranasal administration of insulin appears to be a profitable solution for drug delivery to the brain due to blood-brain barrier bypassing, higher bioavailability and the lack of systemic side effects. When applying this method, insulin is sprayed into the nasal cavity, enters the mucosa and is transported along the axon bundles to the brain. Clinical trials of AD treatment with intranasal insulin revealed improvement in cognition, verbal memory and functional status. Notably, enhancement in memory and cognition were observed both in healthy adults and in AD patients and were ApoE ε4-related. The use of cell-penetrating peptides (CPPs) improve cellular uptake of insulin, enhance bioavailability and increase the direct insulin transport into the deeper regions of the brain such as the olfactory bulb and hippocampus. Animal studies revealed beneficial effects on AD symptoms of other molecules applied in connection with CPPs and most of them approve CPPs formulations for use in clinical trials. This review article summarize clinical trials results of intranasal insulin administration in AD treatment and propose the use of CPPs as an additive to conventional and experimental therapies for AD.
