Opis:
The objective of our study was to investigate the effect of stimulation of the cAMP-dependent pathway on the expression of an orphan nuclear receptor, SF-1/Ad4BP in mouse adrenal tumour, Y-1 cells in culture. We evaluated the temporal pattern of the effects of corticotropin (ACTH) and the adenylyl cyclase activator forskolin on the level of SF-1 mRNA, and compared the time course of induction of SF-1 with that of CYP11A1. Forskolin, corticotropin and 8-Br-cAMP significantly elevated the level of the SF-1 transcript, after 1.5 h of incubation, with a concomitant increase of SF-1 protein level, observed after 6 h. The CYP11A1 transcript increased gradually over the incubation period, and reached the maximal level after 12 to 24 h. The steady-state level of the SF-1 transcript was unaffected by forskolin when the cells were incubated with actinomycin D, indicating that stimulation of the cAMP pathway results in enhanced transcription of the gene. The effect of forskolin was augmented by cycloheximide, suggesting that an inhibitory protein, whose synthesis was inhibited by cycloheximide, could be involved in negative regulation of SF-1 expression. It is concluded that SF-1 expression is positively regulated by the cAMP pathway at the transcriptional level, and can represent the primary event in cAMP-mediated induction of steroid hormone synthesis in Y-1 cells.

In present study, Ho₂O₃ and Dy₂O₃ doped Bi₂O₃ composite materials for intermediate-temperature solid oxide fuel cells (IT-SOFCs) were investigated. $(Bi_2 O_3)_{1-x-y}(Ho_2 O_3)_x(Dy_2 O_3)_y$ ternary systems (x=0.11, 0.13, 0.15 and y=0.01, 0.03, 0.05, 0.07) were fabricated using conventional solid-state synthesis techniques. The samples were characterized by means of X-ray powder diffraction, scanning electron microscopy, energy dispersive X-ray spectroscopy, differential thermal analysis/thermal gravimeter, and the four-point probe technique. X-ray powder diffraction measurements indicated that all samples have the stable fluorite type face centered cubic (fcc) δ-Bi₂O₃ phase. Scanning electron microscopy micrographs of all of the samples showed that grain size distribution was uniform. Four-point probe technique measurements showed that the conductivity of the samples increase with increase of temperature. Additionally, it has been found that the maximum conductivity values of all samples fall in a range $8.44×10^{-2}-4.60×10^{-1} Scm^{-1}$ and their conductivity values corresponding to the intermediate-temperature region vary in the range $1.65×10^{-3}-2.30×10^{-1} Scm^{-1}$. The activation energy values of the samples were calculated from łogσ graphics versus 1000/T using the Arrhenius equation. It was found that there is a good agreement between the activation energy values and conductivity values.

In a series of studies it was shown that 1,4-dihydropyridine derivatives (1,4-DHP) show antimutagenic and anticlastogenic properties and accelerate repair of oxidant and ionising radiation generated DNA damage. Here, effects of one of 1,4-DHP compounds (sodium 3,5-bis-ethoxycarbonyl-2,6-dimethyl-1,4-dihydropyridine-4-carboxylate denoted as DHP) in X-irradiated L5178Y cells (murine lymphoma sublines, LY-R and LY-S) are reported. DHP treatment 1 h before, during and after X-irradiation gave a radioprotective effect in double strand break (DSB) repair competent LY-R cells: there was an increase in post-irradiation proliferation and cell viability as well as a slight acceleration of break rejoining as measured by the neutral comet assay. In the radiosensitive LY-S cells with impaired non-homologous end-joining system, the radioprotective effect was seen as enhanced growth and viability. There was, however, no effect on the DSB repair rate. Notably, there was no dependence of the biological effects on DHP concentration in the range of concentrations studied (1 nM - 100 mM), suggesting an all-or-none effect, as in cellular signaling induction observed in radioadaptation or bystander effect. We assume that DHP acts by decreasing fixation of radiation inflicted DNA damage, among others, by increasing the rate of DNA repair and enhancing the efficiency of checkpoint control. Direct confirmation of this assumption is necessary.

Objectives: This study is a preliminary study to investigate the effects of carvacrol (CRV) obtained from thyme on the apoptosis process in neuroblastoma cells. Methods: In this study, seven groups were designed as control, CRV, CRV + AP-18, CRV + melatonin, CRV + melatonin + AP-18, melatonin and melatonin + AP-18. All groups were stimulated using CNM (cinnamaldehyde) which is TRPA1 channel stimulator. Levels of Reactive oxygen species (ROS), caspase-3, caspase-9, mitochondrial depolarization, apoptosis, intracellular free calcium and 3- (4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide (MTT) were measured. Data were evaluated using one way ANOVA analysis. Results: Levels of ROS, mitochondrial depolarisation, caspase-3 and -9 and apoptosis were significantly higher in all groups treated with CRV compared to control (P <0.05). On the other hand, in melatonin-treated group and melatonin + AP-18 treated group, ROS and caspase-3 were significantly lower than control (P <0,05). MTT levels were significantly decreased in all groups treated with CRV compared to control (P <0.05). On the other hand, in melatonin-treated group and melatonin + AP-18 treated group, MTT was higher than control(P <0,05). Conclusion: It has also been shown that CRV can also exert its effects through TRPA1 channels in neuroblastoma cells, which may accelerate the apoptosis process by acting on these channels, increasing ROS and caspase-3 levels. Changes in MTT levels support this result. However, in order to better evaluate the effects of CRV on the apoptosis process, it would be useful to investigate changes in caspase-9, mitochondrial depolarization, and calcium channels.
