Investigations are presented in the preparation of composite dressing material based on two biopolymers -chitosan and sodium alginate with the addition of sulfanilamide as medication designed for the healing of bedsores. The dressing was prepared in the form of film. The biopolymers used in the construction of the film make the dressing biodegradable and resorbable in the wound’s environment. Mechanical properties of the film were tested: thickness, extension strength, tenacity and elongation at maximum stress The ability of the material to match the wound was examined, too, as well as the transmission of water vapor. Sulfanilamide as bacteriostatic agent was added to the prepared composites. Mechanical and sorption properties of the composite dressings with addition of the active substance depend largely on their composition. The sorption properties were tested before and after addition of the medication .The release of the medication is intricate and proceeds according to kinetics of first order. Susceptibility of the composite materials to hydrolytic and enzymatic degradation was assessed.

Opis:
The synthetic zeolite-sodalite obtained by the hydrothermal conversion of fly ash with aqueous sodium hydroxide was used in the experiments. Its adsorption properties in relation to lithium ions were examined. The effects of: solution pH, presence of polymeric substance – poly(acrylic acid) and order of individual adsorbates addition were determined. To specify the binding mechanism of lithium ions on the sodalite surface, besides adsorption experiments, the measurements leading to the solid surface charge density and zeta potential determination, were performed. As a result, the structure of mixed adsorption layer composed of polymer+metal complexes was characterized. The presented study concerns two important issues: management of environmentally harmful wastes such as coal combustion products as well as searching for new sources of lithium and effective methods of its acquisition.

The aim of presented work was to identify of PCR amplifiedDNAfragments differentiating aluminium tolerant and sensitive forms of rye and to locate the markers on rye chromosomes. For identification of markers, the PCR system with semi-specific primers targeting intron-exon sequences of plant genes was applied. The modified method of bulked segregant analysis was used. The pooled DNAs of two or three F2 segregating populations were screened together with DNA of their parental inbred lines. Potential marker of tolerance gene was located on rye chromosomes using wheat/rye (Chinese Spring/Blanco) additional lines. The specific probes obtained from DNA fragments differentiating sensitive and tolerant forms of rye were hybridized to PCR amplified DNA fragments of sensitive and tolerant forms of rye and the set of wheat/rye addition lines. Independently of the method of digoxygenin labelling (primer extension or Taq polymerase reaction), the probes obtained showed similar hybridization patterns. The results of hybridisation of 21 probes prepared from 12 DNA fragments confirmed connection of selected DNA fragments with to aluminium tolerance or sensitivity. Most of these DNA fragments originated from tolerant forms of rye. Using this method it was possible to locate eight DNA fragments on rye chromosomes. Three DNA fragments hybridised to chromosome 4R, two DNA fragments to chromosome 6R and single DNA fragments to chromosomes 1R, 2R and 3R. Four DNA fragments indicating clear relationship with character studied were not located on particular chromosomes using this set of wheat/rye addition lines. Hybridisation of probes prepared from four DNA fragments revealed length polymorphism. Probes prepared from two DNA fragments were characterised as dominant markers. In other cases the type of marker (dominant/codominat) was not fully documented.

Twelve DNA fragments amplified on templates of Triticum urartu, Aegilops speltoides and Aegilops squarrosa were separated, reamplified and used as digoxigenin-labelled probes. The species and genome specificity of probes was evaluated by Southern dot-blot hybridisation to Triticum durum, the donors of wheat genomes, rye and rye with introgressed wheat chromosome fragments. In comparison to labelled genomic DNA of T. durum, the probes obtained by labelling PCR-amplified fragments exhibited higher specificity to wheat genomes. Under the applied hybridisation conditions all probes showed different degree of cross-hybridisation to rye DNA. Some probes indicated quantitative difference in their specificity to wheat genomes, but generally the intensity of hybridisation to the genomes A, S and D was independent of the origin of an amplified fragment. Selected probes, used in dot-blot hybridisation system together with genomic DNA of T. durum, may increase the sensitivity of screening wheat chromosomal fragments introgressed to rye.

Opis:
The influence of a kind of support electrolyte on the ionic polyamino acids adsorption at the chromium (III) oxide – polymer solution interface was investigated. The NaCl and CaCl2 were used as the background electrolytes. In order to determine the effect of the electrolyte, the same value of ionic strength of the test solutions were taken. It was proved that formation of intermolecular and intramolecular complexes in the presence of divalent calcium ions is responsible for essential changes in polymer adsorption. Related to the ionic character of polyamino acid two different adsorption behaviours can be observed. The increase of the ASP adsorption amount in the presence of calcium ions may be explained by formation of complexes between the dissociated carboxylic groups and Ca2+ ions. The opposite situation takes place in the case of polylysine – the application of CaCl2 results in the dramatic decrease in the polymer adsorption caused by blocking the active sites available for LYS macromolecules. In order to make a comprehensive analysis, the zeta potential and surface charge density measurements were performed taking into account the kind of the background electrolyte. The above-mentioned tests were carried out in the absence and presence of the polyamino acid at two different concentrations – 10 and 100 ppm respectively.

Objective: To assess the prevalence of UGT1A1*28 and UGT1A1*60 polymorphisms of UGT1A1 gene and their association with hyperbilirubinemia. Study design: The study was performed at a single centre - at the Department of Obstetrics of the Medical University of Gdansk in Poland. DNA was isolated from Guthrie cards of 171 infants. Only full term newborns (gestational age 38-42 weeks) were included in the study. Fluorescent molecular probes were used for UGT1A1 promoter variation analysis. The presence of UGT1A1*28 polymorphism was detected with a dual-probe system, and UGT1A1*60 with a SimpleProbe™. Result: Homozygous UGT1A1*28 and UGT1A1*60 genotypes were detected in 14.6% and 20.5% of the newborns, respectively. Homozygous (G/G) genotypes of UGT1A1*60 polymorphism were found in all of the UGT1A1*28 (i.e. (TA)7/(TA)7) homozygotes. More than 80% (55/66) of the children with "wild" type UGT1A1*28 genotype (where no polymorphism was detected) (i.e. (TA)6/(TA)6) carried the "wild" (T/T) genotype of UGT1A1*60 as well. The UGT1A1*28 polymorphism was detected more often among neonates with elevated bilirubin. Hyperbilirubinemia was diagnosed more frequently in boys. Conclusion: Polymorphisms of the UGT1A1 gene frequently co-exist in neonates. The presence of UGT1A1*28 polymorphism and male gender seem to predispose to neonatal hyperbilirubinemia.

Oligochitosans obtained through degradation of macromolecules of chitosan with a high degree of deacetylation turned out to be biologically active, contributing to an increase of nitric oxide levels in Ehrlich ascites tumor (EAT) cells through inducing expression of the isoform of inducible nitric oxide synthase (iNOS) gene. An increase of NO levels in EAT cells in the presence of the investigated oligochitosans might contribute to nitrosylation of L-cysteine – an allosteric effector of the M2 isoenzyme of pyruvate kinase (PK), which switches the PK kinase activity, responsible for ATP synthesis, to the histone kinase activity that may participate in histone H1 phosphorylation. Lack of the histone activity of the PK M2 isoenzyme may contribute to decreased histone H1 phosphorylation and thus inhibit EAT cells proliferation.

Isoenzyme M2 pyruvate kinase, which is a marker of cancer transformation, can take both tetramer (cytosol) and dimer (nucleus) forms. The former is responsible for ATP synthesis, and the latter demonstrates histone H1 kinase activity. Regulation of the expression of pyruvate kinase through which Akt controls the expression of genes involved in Ehrlich ascites tumour (EAT) cell proliferation, migration and death, also involves cross-talk with the other signalling pathways, transcription factors and co-regulatory proteins such as β-catenin and c-Myc. Treatment of EAT cells with chitosans significantly reduced their proliferation (by 45-60%), expression of nuclear β-catenin, c-Myc as well as cell migration. After 48–72 hours of treatment of the cell with oligochitosans, lower levels of p-Akt were detected. Simultaneously, decreased expression of isoenzyme M2 PK protein levels was observed. The dimeric form (nucleus) can participate in H1 histone phosphorylation, which contributes to increased EAT cell proliferation.

Opis:
Preliminary studies of proliferation of Ehrlich ascites tumor (EAT) cells and normal mammary gland epithelial cells have demonstrated the process to be inhibited by degradation products of microcrystalline chitosan, i.e. oligomers. Inhibition of proliferation has been also accompanied by a decreased activity of the M2 pyruvate kinase (PK) isoenzyme in nucleoplasm, what may indicate the role of this enzyme in regulation of tumor cell proliferation. Determinations of nitrogen oxide in tumor and normal cells point to a higher level of this endogenous effector in normal cells. An increase of nitrogen oxide levels in Ehrlich ascites tumor cells effected by chitosan oligomers may indicate increased nitrosylation, and particularly an increased amount of compounds containing sulfhydryl groups and their participation in regulation of nucleoplasm M2 PK isoenzyme activity. Chitosan oligomers have smaller molecules as compared to microcrystalline chitosan and for this reason appear to be more effective than the latter in acting upon the negatively charged cell membrane surfaces, thus contributing to proliferation inhibition.
