The aim of this study was to evaluate the usefulness of quantitative real-time PCR (RQ-PCR) for the monitoring of molecular remission in follicular lymphoma (FL) patients during long-term follow-up. RQ-PCR by the use of TaqMan® detection system is a sensitive tool to monitor minimal residual disease (MRD) in FL through amplification of the t(14;18) fusion gene during and post-therapy. In most cases the breakpoint region occurs within the major breakpoint region (MBR). Among 75 patients diagnosed with FL, cells harboring the fusion gene BCL2/JH were found in peripheral blood of 31 patients (41%). We further monitored 30 of these patients in a period varying from 6 months to 5 years by RQ-PCR. In our study the level indicating the possibility of the presence of MRD was established at more than five t(14;18)-positive cells in the background of 83000 normal cells. The results of this work also confirmed that the presence of MRD detected by RQ-PCR is an indication for careful observation of patients because of a higher risk of disease recurrence.

Opis:
Hemagglutinin (HA), as a major surface antigen of influenza virus, is widely used as a target for production of neutralizing antibodies. Monoclonal antibody, mAb6-9-1, directed against HA of highly pathogenic avian influenza virus A/swan/Poland/305-135V08/2006(H5N1) was purified from mouse hybridoma cells culture and characterized. The antigenic specificity of mAb6-9-1 was verified by testing its cross-reactivity with several variants of HA. The mimotopes recognized by mAb6-9-1 were selected from two types of phage display peptide libraries. The comparative structural model of the HA variant used for antibody generation was developed to further facilitate epitope mapping. Based on the sequences of the affinity- selected polypeptides and the structural model of HA the epitope was located to the region near the receptor binding site (RBS). Such localization of the epitope recognized by mAb6-9-1 is in concordance with its moderate hemagglutination inhibiting activity and its antigenic specificity. Additionally, total RNA isolated from the hybridoma cell line secreting mAb6-9-1 was used for obtaining two variants of cDNA encoding recombinant single-chain variable fragment (scFv) antibody. To ensure high production level and solubility in bacterial expression system, the scFv fragments were produced as chimeric proteins in fusion with thioredoxin or displayed on a phage surface after cloning into the phagemid vector. Specificity and affinity of the recombinant soluble and phage-bound scFv were assayed by suitable variants of ELISA test. The observed differences in specificity were discussed.

The Church Songbook by Rev. Fr. Jan Siedlecki is not a homogenous publication in terms of its 41 editions over 140 years. From the beginning, it contained both the old and the new songs. The author did not pay too much attention to the documentation of their origins and neither did Rev. Fr. Michał Marcin Mioduszewski CM from whose collection Rev. Fr. Siedlecki borrowed the old songs. The situation changed along with the new editor-in-chief, Rev. Fr. Wendelin Świerczek CM. It was him who raised the bar to a new high in the 1928 anniversary edition of the critical edition of the songbook, which he oversaw until the end of his life (he passed away in 1974). The shape of various editions was influenced by changing editors. In the 41st edition of Rev. Fr. Siedlecki’s songbook from 2015, there are 243 songs for which the source are the previous editions. In addition, for 170 songs, the source are Fr. Rev. Mioduszewski’s songbooks. 102 songs in this edition of the songbook have been printed for the first time. The other ones originate from other sources. From this simple analysis, it follows that for 515 songs out of 840 in the 41st edition, the source is the songbook of Fr. Rev. Siedlecki or the songbook of his predecessor, Fr. Rev. Mioduszewski. Many church songs sung in Poland owe their functioning to the songbook of Fr. Rev. Jan Siedlecki (his individual editions). Thus, the decision of the Conference of the Polish Episcopate, by which the 41st edition was approved as the Polish national liturgical songbook, has to be recognised as historical justice or, if you will, a natural consequence.

The purpose of the article. The study aims to analyze how these special rules of financing during the pandemic affected the financial situation of hospitals in Poland. Methodology. We analyzed a sample of 86 Polish hospitals from 2019 to 2021. The financial condition is assessed based on financial ratios and the synthetic measure M1. We employ the Kruskall-Wallis test to confirm the differences between the values of financial ratios in the analyzed period. Results of the research. We confirm that due to the special rules and financing, combined with underperformance, the financial condition of Polish hospitals has slightly improved – especially in the case of operating margin, EAT to debt ratio and ROA.
