Matrix attachment regions (MARs) are thought to participate in the organization and segregation of independent chromosomal loop domains. Although there are several reports on the action of natural MARs in the context of heterologous genes in transgenic plants, in our study we tested a synthetic MAR (sMAR) with the special property of unpairing when under superhelical strain, for its effect on reporter gene expression in tobacco plants. The synthetic MAR was a multimer of a short sequence from the MAR 3' end of the immunoglobulin heavy chain (IgH) enhancer. This sMAR sequence was used to flank the β-glucuronidase (GUS) reporter gene within the T-DNA of the binary vector pBI121. Vectors with or without the sMARs were then used to transform tobacco plants by Agrobacterium tumefaciens. Transgenic plants containing the sMAR sequences flanking the GUS gene exhibited higher levels of transgene expression compared with transgenic plants which lacked the sMARs. This effect was observed independently of the position of the sMAR at the 5' side of the reporter gene. However, variation of the detected transgene expression was significant in all transformed plant populations, irrespective of the construct used. Most genes whose expression has been studied in transgenic plants are generally expressed in appropriate patterns. However, transgene expression can vary within an extremely wide range, often showing only a very low level [1, 2]. Variation in transgene expression is frequently attributed to corresponding variation in the transcription potential of different chromosomal insertion sites. DNA sequences called scaffold/matrix attachment regions (S/MARs) are involved in the structural and functional organization of all eukaryotic genomes. Evolutionarily, the structures of these sequences seem to be conserved. Typically, S/MARs are located every 5 to 200 kb of sequence and are known to bind specifically to components of the nuclear scaffold, therefore suggesting that they are responsible for loop domain base formation [3, 4].Of the MAR elements reported, many do not display extensive sequence homology. It is therefore reasonable to assume that the scaffold probably recognizes some structural features of the MAR DNA rather than a specific sequence [5].MARs appear to be functionally conserved, since animal MARs can bind to plant nuclear scaffolds and vice versa [6, 7].Most MARs have been generally characterized as AT-rich sequences. However, AT-richness per se is not a sufficient criterion for specific sequence recognition of MARs by specific binding proteins [7]. Their capacity to bind to the nuclear matrix is determined by the specific structure of DNA. A prominent structural characteristic of different MARs is their strong potential for extensive unpairing when subjected to superhelical strain [8, 9]. The ability to assume a stably unpaired conformation has been described for several MARs. For example, within the MAR 3' end of immunoglobulin heavy chain (IgH) enhancer there is an AATATATTT motif that is a nucleation site for DNA unwinding [10]. Concatamerized oligonucleotides containing seven repeats of this sequence exhibited a strong affinity for the nuclear scaffold and increased SV40 promoter activity in stably transformed mouse cells [11].In this paper we present results of our studies that concern the effect of a synthetic MAR on transgene expression in tobacco plants. The synthetic MAR sequences were used to flank the β-glucuronidase (GUS) gene whose transcription was under control of the 35S CaMV promoter in the binary vector pBI121. This construct was introduced into tobacco plants and the GUS reporter gene expression was monitored in stably transformed plants.

This position forces him to establish new foundations of human solidarity and morality as it is no longer possible to refer to a universal concept of human nature. Critics stress the fact that questioning the ontic foundations inevitably destroys all the things that rest upon them such as morality, community and culture. Thus, Rorty's proposal to distinguish between private and public languages does not seem a satisfactory solution to the problem. For A. Bielik-Robson the very concept of irony contains "foundations" of human solidarity. In her view, irony exposes one to the experience of de-differentiation and creates a new sense of human affiliation based on the observation that however different from one another we might be, we nevertheless remain equally vulnerable. Such a claim coincides with Rorty's views much closer than his own opinions. It is through the language of irony that we are able to save our identity, the identity of our culture as well as of our moral sensitivity.

Human colon adenocarcinoma LS180 parental cell line and selected variants, characterized by different metastatic capacity were used to examine, whether a correlation exists between β-actin expression, its subcellular distribution and metastatic potential of these cells. Cytosolic fraction (supernatant 105000 × g), isolated from the tumor cells was used as a source for actin quantification. The higher level of β-actin was observed in the cytosol of three selected sublines to compare with LS180 parental line. Statistically significant increase of β-actin level in highly motile EB3 cells variant should be underlined to compare with the other sublines. Distinct differences in the phenotype of adenocarcinoma cell variants were found, such as the changes in cells shape, cells spreading and ability to attach to the surface of culture dish. Actin cytoskeleton was visualized with fluorescence microscopy application and microfilaments rhodamine-conjugated phalloidin staining. β-actin subcellular localization was done by immunofluorescence staining with monoclonal anti-β actin antibodies. In the elongated cells (LS180, 3LNLN), this isoactin is dispersed in the whole cell body and concentrates in pseudopods and at the leading edges, when in the rounded variant (EB3) β-actin dominates mainly in cortical ring under cellular membrane and it is also seen in the subtle protrusions. Summary of our former (Nowak et al., 2002, Acta Biochim. Polon., 49: 823) and current data lead to the conclusion that there is a distinct correlation between metastatic capacity of examined human colon adenocarcinoma cells, the state of actin polymerization, actin cytoskeleton organization and β-actin expression.

Modem economic and social system requires that certain activities are undertaken by local government in order to create favourable conditions for the society and economy. Municipal infrastructure is one of the main stimulants of local growth. It is a keystone integrating the functional and spatial structure of the city. In the beginning of the nineties the municipal infrastructure in Łódź was a developmental barrier for the city. Both roads and housing required new investments. Reactivation of the territorial self-government changed the approach of local authorities towards infrastructure The local government was also given financial recourses to realize its assignments. The aim of this article is to present the municipal investments in roads and housing in Łódź between 1990 and 2002. The authors present also the impact of the investments on spatial structure of the city and its developmental opportunities.

Therefore, organization itself, as well as its management system has to be evaluated in terms of effectiveness which is a term wider than only the ability of achieving goals. A commune too is an organization which has to be evaluated in order to function properly, that is to satisfy the needs of the local community. As far as the private sector is concerned, the literature widely discusses the problem of measuring effectiveness. Thus the aim of the paper is to transfer the major theoretical approaches to the concept onto the field of managing communes.

The goal of the article is to highlight the links between Tadeusz Nowak’s novel A jak królem, a jak katem będziesz (When You Are a King, When You Are a Hangman), with the trend known as magical realism. Pointing out to the connections with this trend, which is very much alive in the contemporary world literature, allows one to survey the writings of the Polish author from a new, broader perspective. One is able to perceive the whole richness of the author’s prose, where realism and the miraculous seem to grow out of reality and are equivalent to one another. The elements of magical realism  distinguished by the authoress (the element of magic in the realistically constructed depicted world, the permeation of two worlds and two separate realities, the refl ection of the magical mentality etc.) are also present in Nowak’s other works, and particularly in his short-stories dating back to different periods, such as Przebudzenia (Awakenings), W puchu alleluja, In the fluff of the hallelujah? or Półbaśnie (Half-Fairy Tales).

In the seasons 2004 and 2006, interdisciplinary researches at the settlement of the early phase of Eastern Linear Pottery Culture at Moravany were continued. Archaeological excavations have limited spatial dimension compared to previous seasons, as a priority in the years 2004-2006 was the completion of the palaeoeconomic, paleoenvironmental and chronological analyses. I.a. 14 new radiocarbon dates were obtained. In conjunction with the absolute dates obtained previously and with typological evidence it gives rise to dating of the settlement for a period between about 5500 and 5300/5200 BC.

The aim of the present study was to examine the eff ect of the central serotoninergic lesion on the antinociceptive eff ects of morphine (5.0 mg/kg sc), nefopam (20 mg/kg ip), indomethacin (5.0 mg/kg ip) and imipramine (10 mg/kg ip) in the model of exteroceptive sensation - formalin test (chemical stimulus), and interoceptive sensation - writhing test (model of visceral pain). Furthermore monoamine synthesis rate after examined analgetics in the frontal cortex, thalamus, brain stem and spinal cord was assayed. MATERIAL AND METHODS On the 3 rd day of postnatal life male rats were administered with 5.7- DHT (70 μg/10 μl in 0.1% ascorbic acid solution ICV; 35 μg/5 μl per side); control animals received vehicle (70 μg/10 μl of 0.1% ascorbic acid solution ICV). Rats continued to be housed until 8-10 weeks, for further experimentation. RESULTS It was shown that the central serotoninergic lesion slightly modifi ed analgesia evoked by nefopam and indomethacin in formalin test but much more profound reduction in analgesia produced by morphine and indomethacin injection was observed in visceral model of nociception. In biochemical study it was shown that analgetics employed in this study (with exception of indomethacin) altered synthesis rate of serotonin (5-HT) and dopamine (DA) in the examined parts of the rats brain. The above indicates that besides serotoninergic pathway other monoaminergic systems (noradrenergic and dopaminergic) participate in the central mechanism of action of these drugs. CONCLUSION It is likely that similar nociception abnormalities may occur in patients with serotoninergic system dysfunction, so that it points out on the requirement of analgetics dosage adjustment.
WSTĘP Celem pracy była ocena wpływu lezji (zniszczenia) ośrodkowego układu serotoninergicznego na przeciwbólowe działanie morfi ny (5.0 mg/kg sc), nefopamu (20 mg/kg ip), indometacyny (5.0 mg/kg ip) i imipraminy (10 mg/kg ip) w testach czucia bólu eksteroceptywnego, tj formalinowym (bodziec chemiczny) a ponadto czucia interoceptywnego, tj. bólu trzewnego w teście wicia. Ponadto zbadano wpływ stosowanych analgetyków na procesy syntezy monoamin w wybranych częściach mózgu (kora przedczołowa, wzgórze, most, rdzeń kręgowy) u szczurów kontrolnych oraz z lezją. MATERIAŁ I METODY Zniszczenie ośrodkowego układu serotoninergicznego wykonano u 3 dniowych noworodków szczurzych stosując dokomorowo 5.7-DHT w dawce 70 μg/10 μl w 0.1% roztworze kwasu askorbinowego. Właściwe testy behawioralne i biochemiczne wykonano po osiągnięciu przez zwierzęta 8 tygodnia życia. WYNIKI Wykazano, że chemiczna lezja ośrodkowego układu serotoninergicznego tylko w niewielkim stopniu modyfi kuje przeciwbólowe działanie nefopamu i indometacyny w modelu bólu zapalnego natomiast wyraźnie osłabia przeciwbólowe działanie morfi ny i indometacyny w modelu bólu trzewnego u szczurów. W badaniach biochemicznych wykazano, że stosowane leki (za wyjątkiem indometacyny) zmieniają szybkość syntezy serotoniny (5-HT) oraz dopaminy (DA) w badanych strukturach mózgu. Powyższe wskazuje, że w ośrodkowych mechanizmach analgetycznego działania stosowanych leków poza układem serotoninergicznym uczestniczą w różnym stopniu inne układy monoaminergiczne, tj. noradrenergiczny i dopaminergiczny. WNIOSKI Uzyskane wyniki przemawiają za tym, że trwała dysfunkcja ośrodkowego układu serotoninergicznego może prowadzić do zmienionej reakcji biologicznej na leki przeciwbólowe.

BACKGROUND: We investigated the reactivity of 5-HT1B receptor in adult rats administered as neonates with noradrenergic neurotoxin N-(-2-chloroethyl)-N-ethyl-2- bromobenzylamine (DSP-4; 50 mg/kg sc x2, P1 and P3). MATERIAL AND METHODS: At 8-10 weeks (after lesion), control and DSP-4 rats were divided into two subgroups injected either with saline or 5-hydroxytryptamine (serotonin; 5-HT) synthesis inhibitor (p-chlorophenylalanine; p-CPA; 100 mg/kg ip x 2 every 24h). On the following day food intake was examined after 5-HT1B receptor ligands administration; agonist CP 94,253, 4.0 mg/kg ip; and antagonist, SB 216641, 4.0 mg/kg ip respectively. Additionally, the level of monoamines, their metabolites and 5-hydroxytrptophane (5- HTP) as an indicator of 5-HT synthesis rate were analyzed by HPLC/ED methods. RESULTS AND CONCLUSIONS: CP 94,253 in a dose of 4.0 mg/kg ip signifi cantly decreased food intake in control and DSP-4 rats in comparison to respective control (saline). 5-HT1B antagonist (SB 216641) alone did not alter the examined parameter but injected 30 min before CP 94,253 markedly abolished its eff ect in both tested groups. Similar results were obtained in p-CPA pretreated animals. The level of amines and their metabolites was not diff erent between control and DSP-4 treated rats. Administration of p-CPA (2 x 100 mg/kg ip) reduced the concentration of 5-HT and 5-hydroxyindoleacetic acid (5-HIAA) in the hypothalamus of control rats by 94.9 % and 96.7 % respectively. In DSP-4 rats the percentage of 5-HT displacement was 92.5 % (5-HT) and 95.7 % (5-HIAA). As it was presented CP 94,253 and SB 216641 alike, administered in doses of 4.0 mg/kg ip, did not alter biogenic amines and their metabolites content in the hypothalamus of both examined groups (control and DSP-4). There were also no diff erences in the 5-HT synthesis rate in the hypothalamus between control and DSP-4 rats after 5-HT1B receptor ligands injection. We concluded that in contrast to our previous studies, DSP-4 being without eff ect on hypothalamus noradrenalin (NA) level and 5-HT synthesis rate did not also aff ect reactivity of terminal 5-HT1B autoreceptors in this brain area. On the basis of this experiments, it should be noted that in contrast to our previous studies which have shown that the consequence of DSP-4 administration is dramatic reduction in the NA contents in the frontal cortex, hippocampus, striatum, etc., with concomitant desensitization of 5-HT1A and 5-HT1B receptors (evaluated by behavioral and biochemical methods), in this experiment, there were no such eff ects. This proves that the noradrenergic fi bers in the hypothalamus are much more resistant to the neurotoxic action of DSP-4, furthermore, for the development of 5-HT1B receptors desensitization in the specifi c brain structure, decrease in NA contents in this part of the brain is required. WSTĘP: W niniejszej pracy zbadano reaktywność ośrodkowego receptora serotoninergicznego 5-HT1B u dorosłych szczurów, które w okresie noworodkowym otrzymały neurotoksynę N-(-2-chloroethyl)- N-ethyl-2-bromobenzylamine (DSP-4; 50 mg/kg sc x 2, 1-go i 3-go dnia). MATERIAŁ I METODY: W 8-10-tym tygodniu życia zwierzęta kontrolne i z lezją podzielono na dwie podgrupy, jedna otrzymała sól fi zjologiczną, druga inhibitor syntezy serotoniny (5-hydroksytryptaminy; 5-HT) - p-chlorofenyloalaninę (p-CPA). Następnie po podaniu ligandów receptora 5-HT1B (agonisty; CP 94253 4.0 mg/kg ip i/lub antagonisty; SB 216641 4.0 mg/kg ip) oceniono ilość spożywanej karmy. Dodatkowo zbadano również zawartość amin biogennych po podaniu ligandów receptora 5-HT1B oraz oceniono szybkość syntezy serotoniny w podwzgórzu. WYNIKI I WNIOSKI: CP 94253 stosowany zarówno u szczurów kontrolnych jak i DSP-4 zmniejszał ilość spożywanej karmy w porównaniu do odpowiednich grup kontrolnych (po podaniu soli fi zjologicznej). Podobne efekty zaobserwowano u zwierząt, które wcześniej otrzymały p-CPA. Nie stwierdzono zmian zawartości amin biogennych po podaniu DSP-4. Również CP 94253 oraz SB 216641 nie wpływały na stężenie amin biogennych i ich metabolitów w badanej strukturze mózgu. Po podaniu p-CPA (2 x 100 mg/kg ip) obserwowano spadek zawartości 5-HT oraz 5-HIAA w podwzgórzu u kontroli odpowiednio o 94.9% i 96.7%, natomiast w grupie DSP-4 o 92.5 % (5-HT) i 95.7 % (5-HIAA). Szybkość syntezy 5-HT nie różniła się pomiędzy kontrolą a grupą DSP-4, również CP 94253 4.0 mg/kg ip i/lub SB 216641 4.0 mg/kg ip pozostawały bez wpływu na badany parametr. Na podstawie przeprowadzonych eksperymentów stwierdzić należy, że w przeciwieństwie do naszych poprzednich badań, w których wykazaliśmy, że konsekwencją podania DSP-4 jest spadek zawartości NA w korze czołowej, hipokampie i prążkowiu, które wywołuje zjawisko desensytyzacji receptorów 5-HT1A i 5-HT1B (oceniane metodami behawioralnymi i biochemicznymi), w niniejszym doświadczeniu nie stwierdzono takich efektów. Powyższe przemawia za tym, iż zakończenia noradrenergiczne w podwzgórzu są znacznie bardziej oporne na neurotoksyczne działanie DSP-4 oraz, że dla rozwoju desensytyzacji receptorów 5-HT1B w określonej strukturze mózgu konieczny jest spadek zawartości NA w tej części mózgu. Innymi słowy głęboka dysfunkcja układu noradrenergicznego pod postacią zniszczenia zakończeń noradrenergicznych w korze czołowej, prążkowiu oraz hipokampie pozostaje bez wpływu na „status” receptorów 5-HT1B w podwzgórzu.
