The aim of the work was analyzing of genomic DNA of Malassezia pachydermatis isolates from clinical cases otitis externa from dogs using RAPD method with arbitrary primers Eric1R, Eric2, BG2 and FM1. Materials and methods. 47 strains of M. pachydermatis isolates from clinical cases otitis externa from dogs were tested. Isolation of genomic DNA was provided according with MasterPureTM Yeast DNA Purification Kit EPICENTRE procedure. The quality of isolated genomic DNA was determined electrophoreticaly. For differentiation the following primers were used: Eric1R, Eric2, BG2 and FM1. Primers Eric 1R and Eric 2 were used together in one reaction or amplificated separately. Obtained products were analyzed electrophoreticaly in 1.5% agarose gel. For determination of phylogenic tree Quantity one VersaDoc (BioRad) and Statgraphics plus 4.1 programs were used. Results. High degree of heterogeneity of DNA among investigated isolates of M. pachydermatis was shown using FM1 primer. Dendrograms were prepared by calculation euclid's distance of different parameters (size and count of RAPD products) by nearest neighbor method. Basing on phylogenic tree four main types (phylogenic groups) of M. pachydermatis isolates were shown. The other five groups non-count was shown also.

Opis:
Veterinary medical mycology often differs from the human counterpart by the clinical aspects, the variety of fungi involved, and the antimycotic drugs available. Since the 1980s, there has been a significant increase in the number of humans affected by mycoses, and the spectrum of fungal species defined as pathogenic has broadened due to the increasing immunosuppression in the population. In veterinary mycology, this phenomenon has not been observed or is still insufficiently investigated. Only in the last few years the interest and awareness of the importance of animal mycoses and their treatment have increased. Convincing data present identification of previously human pathogens e.g. Cryptococcus gattii or Sporothrix brasiliensis, in animals. There are also revisions in taxonomy of some groups of fungi. The development of new antifungal drugs with better pharmacological properties, a broader spectrum of activity and fewer adverse effects was largely stimulated by the aforementioned increase of cases of fungal infections and the variety of causative agents. Some of these newly developed antifungal drugs are very expensive and, as is well known, economic considerations have a much greater influence on the decision to treat animals than it is in humans. Unfortunately, it is still often believed that the value of an animal is calculated in relation to the cost of therapy. This article reviews the literature on the treatment of fungal and yeast infections from the veterinary perspective of antifungal therapy. Particular attention was paid to the characteristics of disseminated mould mycoses in dogs, as there are numerous reports in the scientific literature in this field.

Yeast infections such as otitis externa and seborrheic dermatitis in dogs and cats are frequently associated with Malassezia pachydermatis secondary infection. It is part of the normal cutaneous microflora of most warm-blooded vertebrates, however, under certain conditions, it can become a causative agent of infection that needs to be treated pharmacologically. Azole derivatives are the drugs of the first choice. An interesting trend in developing resistance is the use of natural substances, which include manuka honey with confirmed antimicrobial properties. The main intention of this research was to evaluate the mutual effect of manuka honey in combination with four conventional azole antifungals – clotrimazole, fluconazole, itraconazole, and miconazole – on 14 Malassezia pachydermatis isolates obtained from dogs and 1 reference strain. A slightly modified M27–A3 method (CLSI 2008) and the checkerboard test (Nikolić et al. 2017) were used for this purpose. Our results show an additive effect of all 4 antifungals with manuka honey concurrent use. Based on the determined values of fractional inhibitory concentration index (FICI – 0.74±0.03 when manuka honey combined with clotrimazole, 0.96±0.08 with fluconazole, 1.0±0 with miconazole and 1.16±0.26 with itraconazole), it was found in all cases that the effect of substances used is more pronounced in mutual combination than when used separately.

The present study investigated the effectiveness of three different disinfectants: preparation H1 (two-component preparation based on hydrogen peroxide); Pedox (multi-component preparation based on peroxyacetic acid) and Savo hypochlorite preparation) against Malassezia pachydermatis. The antifungal activity of disinfectants was tested by quantitative suspension method according to STN EN 1650. The results confirmed 100% effectiveness of these disinfectants at all concentrations and exposure times tested.

A total of 100 Malassezia pachydermatis strains recovered from skin and mucosal membranes of dogs were evaluated for their adhesive properties. Two types of growth, related to colony morphology on Sabouraud agar, were observed (type I and II). The mean number of fungal cells attaching to canine buccal epithelial cells was found to be 17. The number of adhered cells was greater (statistically significant at the level of p < 0.01) in strains belonging to the type I.
