This report presents purification and characterization of the extracellular domain of rat Fas protein, called FIP (FasL interfering protein), expressed as inclusion bodies in Escherichia coli. FIP was extracted from the inclusion bodies, solubilized with 8 M urea, purified by a single-step immobilized metal ion (Ni2+) affinity chromatography and refolded. SDS/PAGE and mass spectrometry analysis of the purified protein verified its purity. Fluorescence spectrum analysis showed that the refolding procedure caused structural changes which presumably might have led to oligomerization. The purified FIP has biological activities: it binds specifically soluble Fas ligand and protects human Jurkat lymphocytes against FasL-dependent apoptosis. This efficient procedure of FIP expression in E. coli and renaturation may be useful for production of therapeutically important proteins.

Neuron-astrocyte interactions are critical for signalling, energy metabolism, extracellular ion and glutamate homeostasis, volume regulation and neuroprotection in the central nervous system. Glutamate uptake by astrocytes may prevent excitotoxic glutamate elevation and determine neuronal survival. However, activation of astrogliosis and brain inflammation during cerebral ischemia exacerbates primary brain damage, resulting in upregulation of cytotoxic/infammatory cytokines and mediators, that alter blood flow and increase vascular permeability, thus leading to secondary damage and accumulation of immune cells in the brain. Although, astrocytes are more resistant than neurons to ischemic injury, astrocyte death have been demonstrated in animal models of brain ischemia. Exposure of cultured cortical astrocytes to glutamate induces apoptotic cell death and similar events can be detected in ischemic brain. FK506, an inhibitor of calcineurin and immunosuppressive drug, is neuroprotective in animal models of neurologic diseases, including focal and global ischemia. FK506 exerts a complex action on many processes ongoing in pathological brain: it directly protects neurons from glutamate-induced neuronal cell death, reduces gliosis and brain inflammation in animal models of stroke, and inhibits Glu-induced apoptosis of astrocytes in vitro and in ischemic brain. Altogether, recent findings suggest that modulation of astrocyte functions and astrocytic survival/cell death in neurodegeneration may be a novel therapeutic strategy in neurological disorders.

In recent years, the development of IT systems for fleet monitoring was observed. Tire pressure monitoring systems are constantly improved. Decreased values in tire pressure can cause deformation of tires. Monitoring of tire pressure is an important function in oversized transport trucks. Tire pressure and rolling resistant influence fuel consumption. The purpose of this paper was to determine the impact of tire pressure on fuel consumption in a fleet of trucks with tire pressure monitoring system installed and to determine the impact of other factors that may affect fuel consumption, such as the vehicle weight, brake usage and cruise control usage. The results of the research were developed using a multiple regression model describing the above dependence.

Opis:
Introduction. Breast cancer is the most common cancer among women and is the second cancer frequently occurring worldwide of newly-diagnosed cancers. There is much evidence showing the influence of life style and environmental factors on the development of mammary gland cancer (high-fat diet, alcohol consumption, lack of physical exercise), the elimination of which (primary prevention) may contribute to a decrease in morbidity and mortality. Secondary prevention, comprising diagnostic tests (e.g. mammography, ultrasonography, magnetic resonance imaging, breast self-examination, as well as modern and more precise imaging methods) help the early detection of tumours or lesions predisposing to tumours. Objective. The aim of this study paper is to review current knowledge and reports regarding primary and secondary prevention of breast cancer. State of knowledge. It is estimated that nearly 70% of malign tumours are caused by environmental factors, whereas in breast cancer this percentage reaches 90–95%. There are national programmes established in many countries to fight cancer, where both types of prevention are stressed as serving to decrease morbidity and mortality due to cancers. Conclusions. Cancer prevention is currently playing a key role in the fight against the disease. Behaviour modification, as well as greater awareness among women regarding breast cancer, may significantly contribute towards reducing the incidence of this cancer. Another important aspect is the number of women undergoing diagnostic tests, which still remains at an unsatisfactory level.

Opis:
Transforming growth factor beta (TGF-β) is a multifunctional cytokine involved in the regulation of cell proliferation, differentiation and survival/or apoptosis of many cells. Knock-out experiments in mice for the three isoforms of TGF-β have demonstrated their importance in regulating inflammation and tissue repair. TGF-β is implicated in the pathogenesis of human diseases, including tissue fibrosis and carcinogenesis. TGF-β receptors act through multiple intracellular pathways. Upon binding of TGF-β with its receptor, receptor-regulated Smad2/3 proteins become phosphorylated and associate with Smad4. Such complex translocates to the nucleus, binds to DNA and regulates transcription of specific genes. Negative regulation of TGF-β/Smad signalling may occur through the inhibitory Smad6/7. Furthermore, TGF-β-activated kinase-1 (TAK1) is a component of TGF-β signalling and activates stress-activated kinases: p38 through MKK6 or MKK3 and c-Jun N-terminal kinases (JNKs) via MKK4. In the brain TGF-β, normally expressed at the very low level, increases dramatically after injury. Increased mRNA levels of the three TGF-β isoforms correlate with the degree of malignancy of human gliomas. TGF-βs are secreted as latent precursors requiring activation into the mature form. TGF-β may contribute to tumour pathogenesis by direct support of tumour growth and influence on local microenvironment, resulting in immunosuppression, induction of angiogenesis, and modification of the extracellular matrix. TGF-β1,2 may stimulate production of vascular endothelial growth factor (VEGF) as well as plasminogen activator inhibitor (PAI-I), that are involved in vascular remodelling occurring during angiogenesis. Blocking of TGF-β action inhibits tumour viability, migration, metastases in mammary cancer, melanoma and prostate cancer model. Reduction of TGF-β production and activity may be a promising target of therapeutic strategies to control tumour growth.

Control of eukaryotic transcription involves combinatorial interactions between transcription factors and regulatory sequences in the genome. In addition, chromatin structure and modification states play key roles in the complex control of transcription. The maturation of methods for detecting in vivo protein-DNA interactions on a genome-wide scale, detailed maps of chromatin features and transcription factor localization over entire genomes of eukaryotic cells increases our understanding of the properties and nature of transcriptional regulatory mechanisms. The knowledge gained from large-scale investigations has challenged some conventional concepts of promoter structure and function, and provided valuable insights into the complex gene regulatory mechanisms. Here, we review recent experimental and computational advances that have enabled identification and analysis of regulatory regions on an unprecedented scale, allowing for systematic determination of the transcriptional regulatory networks in eukaryotic cells. Analysis of high-throughput gene expression data together with genomic sequence(s) holds promise of revealing features of cis-regulatory sequences responsible for co-regulation of many genes. This general approach has proved successful in simpler organisms, and recent studies in Metazoa reported features over-represented in groups of genes with particular expression patterns revealing gene regulatory mechanisms.
