Genetic diversity caused by transposable element movement can play an important role in plant adaptation to local environments. Regarding genes, transposon-induced alleles were mostly related to gene bodies and a few of them to promoter regions. In this study, promoter regions of 9 stress-related genes were searched for transposable element insertions in 12 natural accessions of Arabidopsis thaliana. The promoter screening was performed via PCR amplification with primers designed to flank transposable element insertions in the promoter regions of the reference accession Col-0. Transposable element-associated insertion/deletion (indel) polymorphisms were identified in 7 of the 12 promoter loci across studied accessions that can be developed further as molecular markers. The transposable element absence in the promoter regions of orthologous genes in A. lyrata indicated that the insertion of these transposable elements in A. thaliana lineage had occurred after its divergence from A. lyrata. Sequence analysis of the promoter regions of CML41 (Calmodulin-like protein 41) and CHAP (chaperone protein dnaJ-related) confirmed the indel polymorphic sites in four accessions - Col-0, Wassilewskija, Shahdara, and Pirin. The observed indel polymorphism of the CHAP promoter region was associated with specific gene expression profiles in the different accessions grown at a normal and elevated temperature in a plant growth chamber. The collected data can be a starting point for gene expression profiling studies under conditions resembling the natural habitats of accessions.

Bisphenol A (BPA) is a widely used chemical, that can potentially be toxic to plants. In this study we examined the toxicity of 5-50 mg/l of BPA on Arabidopsis thaliana. Additionally, the effects of 0.5-5 mg/l of BPA were examined after four weeks of development. BPA had no effect on the germination rate and the chlorophyll a/b ratio. The chlorophyll a and carotenoid content was significantly elevated in seedlings treated with 5 mg/l of BPA. In 4-week-old plants there was no change in the chlorophyll and carotenoid content and photosynthetic parameters (Fv/Fm, Fv/F0 and PI) were unaffected, which suggests no photoinhibition. No oxidative stress symptoms were observed. BPA significantly decreased leaf protein content. A low concentration of BPA seems to have no significant effect on A. thaliana flowering, but further investigation is needed. The results obtained indicate that a low concentration of BPA has no negative effect on the growth and development of A. thaliana.

Abscisic acid (ABA) and phototropins act antagonistically to control stomatal movements. Here, we investigated the role of ABA in phototropin-directed chloroplast movements in mesophyll cells of Arabidopsis thaliana. We analyzed the expression of phototropins at mRNA and protein level under the influence of ABA. PHOT1 mRNA level was decreased by ABA in the dark while it was insensitive to ABA in light. PHOT2 mRNA level was independent of the hormone treatment. The levels of phototropin proteins were down-regulated by ABA, both in darkness and light. No impact of exogenous ABA on amplitudes and kinetics of chloroplast movements was detected. Chloroplast responses in wild type Arabidopsis and three mutants, abi4, abi2 (abscisic acid insensitive4, 2) and aba1 (abscisic acid1), were measured to account for endogenous ABA signaling. The chloroplast responses were slightly reduced in abi2 and aba1 mutants in strong light. To further investigate the effect, abi2 and aba1 mutants were supplemented with exogenous ABA. In the aba1 mutant, the reaction was rescued but in abi2 it was unaffected. Our results show that ABA is not directly involved in phototropin-controlled chloroplast responses in mature leaves of Arabidopsis. However, the disturbance of ABA biosynthesis and signaling in mutants affects some elements of the chloroplast movement mechanism. In line with its role as a stress hormone, ABA appears to enhance plant sensitivity to light and promote the chloroplast avoidance response.

Arabidopsis microRNA162 (miRNA162) level regulation was studied under abiotic stresses, such as drought and salinity. The TaqMan® microRNA assay proved that A. thaliana miRNA162 level was elevated under these stresses, confirming its salt and drought responsiveness. The promoter region analyses of A. thaliana miRNA162a and b genes (MIR162a and MIR162b) identified numerous salinity and drought responsive elements. However, our results indicated that Arabidopsis MIR162a was presumably the main locus responsible for the mature ath-miRNA162 accumulation under the stresses tested, and the MIR162b was generally rather weakly expressed, both in control and under the stress conditions. The MIR162a structure was confirmed to be complex and the pri-miRNA162a hairpin structure was shown to span an alternative exon and an intron. The MIR162a transcription generated a few pri-miRNA162a splicing isoforms that could be functional and non-functional. Upon drought and salinity stresses, the regulation of the pri-miRNA162a alternative splicing pattern revealed an increase of a functional pri-miR162a isoform and a preferential distal polyA site selection under the stress conditions. Apart from the potential transcriptional regulation of the miRNA genes (MIRs) expression, the data obtained point to an essential role of posttranscriptional regulation of Arabidopsis microRNA162 level.

In eukaryotes, ribosomal 5S rRNAs are products of multigene families organized within clusters of tandemly repeated units. Accumulation of genomic data obtained from a variety of organisms demonstrated that the potential 5S rRNA coding sequences show a large number of variants, often incompatible with folding into a correct secondary structure. Here, we present results of an analysis of a large set of short RNA sequences generated by the next generation sequencing techniques, to address the problem of heterogeneity of the 5S rRNA transcripts in Arabidopsis and identification of potentially functional rRNA-derived fragments.

Fatty alcohols and derivatives are important for proper deposition of a functional pollen wall. Mutations in specific genes encoding fatty acid reductases (FAR) responsible for fatty alcohol production cause abnormal development of pollen. A disrupted AtFAR2 (MS2) gene in Arabidopsis thaliana results in pollen developing an abnormal exine layer and a reduced fertility phenotype. AtFAR2 has been shown to be targeted to chloroplasts and in a purified form to be specific for acyl-ACP substrates. Here, we present data on the in vitro and in planta characterizations of AtFAR2 from A. thaliana and show that this enzyme has the ability to use both, C16:0-ACP and C16:0-CoA, as substrates to produce C16:0-alcohol. Our results further show that AtFAR2 is highly similar in properties and substrate specificity to AtFAR6 for which in vitro data has been published, and which is also a chloroplast localized enzyme. This suggests that although AtFAR2 is the major enzyme responsible for exine layer functionality, AtFAR6 might provide functional redundancy to AtFAR2.

In this article, the morphology of a brown alga is revisited and compared to the phyllotaxis of land plants. The alga, Sargassum muticum (Yendo) Fensholt has a highly organized thallus with a stipe, the stem-like main axis, and hierarchically organized lateral branches of successive orders. Around each of these axes, the lateral organs: blades, side-branches, and receptacles grow in a spiral disposition. As in land plants, this organization is related to an apical mode of growth. Measurements performed along the mature differentiated axes as well as in their meristematic regions confirm the similarity of the large-scale organization of this brown alga with that of the land plants. In particular, the divergence angle between successive elements has similar values and it results from the existence around the meristem of parastichies having the same Fibonacci ordering. This is remarkable in view of the fact that brown algae (Phaeophyceae) and land plants (Embryophyta) are two clades that diverged approximately 1800 million years ago when they were both unicellular organisms. We argue that the observed similarity results from a morphogenetic convergence. This is in strong support of the genericity and robustness of self-organization models in which similar structures, here Fibonacci related spirals, can be obtained in various situations in which the genetic and physiological implementation of development can be of a different nature.

The aim of the study was investogated the effect of selected types of soot on Arabidopsis thaliana in vitro culture. In experiments different type the soot were used: (1) formed in the presence of a commercial fuel additive containing significant quantities of sodium chloride and copper(II) sulfate(VI) (2) prepared by adding of the newly developed additive without NaCl and with triiron tetraoxide instead of copper and (3) prepared without additive. The soot were added to the culture media (to the root) or applied on the leaves. The parameters such as root length, fresh weight, photosynthetic efficiency (Fv / Fm), the concentration of photosynthetic pigments, index of tolerance (IT) were analyzed. The fresh weights of the investigated plants were comparable or slightly lower than the fresh weights of control plants. The values of the photosynthetic efficiency were comparable to the control when the soot were added to culture medium (to the root) or slightly lower when were applied on the leaves. However, content of photosynthetic pigments were higher than in control plants. The highest indexes of tolerance (even above 100%) were observed for plants growing on media containing the soot formed in the presence of additive with iron.
Celem pracy było zbadanie wpływu wybranych rodzajów sadzy na Arabidopsis thaliana w hodowlach in vitro. Stosowano sadzę powstałą: (1) w obecności komercyjnego dodatku paliwowego zawierającego chlorek sodu i siarczan(VI) miedzi(II), (2) dodatku nowoopracowanego zawierającego tetratlenek triżelaza i (3) bez dodatku. Sadze dodawano do pożywek hodowlanych (dokorzeniowo) lub nanoszono na blaszki liściowe (dolistnie) i analizowano parametry takie jak: długość korzenia, świeża masa, wydajność kwantowa fotosyntezy (Fv/Fm), stężenie barwników fotosyntetycznych, indeks tolerancji (IT). Świeża masa roślin była porównywalna lub nieco niższa niż roślin kontrolnych. Wartości wydajności kwantowej fotosyntezy były porównywalne z kontrolą gdy sadza była podawana dokorzeniowo lub nieco niższe, gdy stosowano ją dolistnie. Zawartość barwników fotosyntetycznych była wyższa niż w roślinach kontrolnych. Najwyższy indeks tolerancji (nawet powyżej 100%) charakteryzował rośliny rosnące na podłożach z dodatkiem sadzy powstałej w obecności katalizatora zawierającego żelazo.

Biostimulants are compounds of diverse formulations, that improve quantity and quality of yield. Their positive effect on plants is more evident in stressful conditions. Biostimulants increase a plant’s tolerance to stresses and enable the repair of damage caused by unfavourable conditions. In this study the effect of Asahi SL on Arabidopsis thaliana L. grown in the presence of cadmium (Cd2+) was evaluated.Cd2+ negatively affected examined parameters and processes, leading to disturbances in plant growth and performance. A. thaliana treated with Asahi SL were higher and advanced in development. Biomass accumulation was stimulated by Asahi SL due to a higher efficiency of the photosynthetic apparatus, manifested by higher: (i) leaf area, (ii) chlorophyll content and (iii) intensity of photosynthesis. Despite higher intensity of transpiration and lower stomatal resistance, the relative water content was unchanged in Asahi SL – treated plants due to the stimulation of root development. Therefore, it can be concluded that Asahi SL protects plants against Cd2+ stress.
Biostymulatory poprawiają ilość i jakość plonu. Preparaty te pozytywnie wpływają na procesy życiowe rośliny, zazwyczaj wyraźniej w warunkach stresu. Stosowanie biostymulatorów zwiększa tolerancję roślin na stresy i przyśpiesza procesy naprawcze uszkodzeń wywołanych przez niekorzystne warunki. W pracy zbadano wpływ Asahi SL na rośliny Arabidopsisthaliana L. rosnące w obecności kadmu (Cd2+). Cd2+ negatywnie wpływał na mierzone parametry i procesy, prowadząc do zaburzeń wzrostu i osłabienia kondycji roślin. Rośliny A. thaliana traktowane Asahi SL były wyższe, a ich rozwój był przyśpieszony. Akumulacja biomasy zwiększyła się u roślin rosnących w obecności Asahi SL w wyniku lepszej wydajności aparatu fotosyntetycznego, wyrażonej zwiększaną (i) powierzchnią liści, (ii) zawartością chlorofilu i (iii) intensywnością fotosyntezy. Pomimo wyższej transpiracji i obniżonych oporów aparatów szparkowych, względna zawartość wody roślin traktowanych Asahi SL nie zmieniała się, co związane było z stymulacją rozwoju systemu korzeniowego. Można zatem wnioskować, że Asahi SL chroni rośliny rosnące w warunkach stresu wywoływanego Cd2+ .

Several studies have used A. thaliana as a model to identify the physiological and molecular mechanisms underlying iron deficiency tolerance in plants. Here, Arabidopsis thaliana and Thellungiella salsuginea were used to investigate the differential responses to iron deficiency of these two species. Plants were cultivated in hydroponic medium containing 5 or 0 μM Fe, for 10 days. Results showed that rosette biomass was more reduced in T. salsuginea than in A. thaliana when grown on Fe-deficient medium. As a marker for iron deficiency tolerance, the induction of ferric chelate reductase (FCR) and phosphoenolpyruvate carboxylase (PEPC) activities was observed only in A. thaliana roots. In addition, we found that the accumulation of phenolic acids in roots of N1438 ecotype of A. thaliana was stimulated by Fe deficiency. Furthermore, an increase of flavonoids content in the root and exudates was observed under Fe-deficiency in this ecotype. Unlike other abiotic stresses, it appears that iron deficiency effects were more pronounced in Thellungiella than in Arabidopsis. The higher tolerance of the Arabidopsis plant to iron deficiency may be due to the metabolic changes occurring in the roots.

Most flowering plants, including important crops, require double fertilization to form an embryo and endosperm, which nourishes it. Independence from fertilization is a feature of apomictic plants that produce seeds, from which the plants that are clones of the mother plant arise. The phenomenon of apomixis occurs in some sexual plants under specific circumstances. Since the launch of a fertilization-independent mechanism is considered a useful tool for plant breeding, there have been efforts to artificially induce apomixis. We have been able to produce fertilization-independent endosperm in vitro in Arabidopsis over the last few years. This paper demonstrates the methods of improving the quality of the endosperm obtained using plant and mammalian steroid hormones. Additionally, it shows the study on the autonomous endosperm (AE) formation mechanism in vitro. This paper examines the effect of exogenous steroid hormones on unfertilized egg and central cell divisions in culture of unpollinated pistils of Arabidopsis Col-0 wild-type andfie-1 mutant. All media with hormones used (estrone, androsterone, progesterone, and epibrassinolide) stimulated central cell divisions and fertilization-independent endosperm development. The stages of AE development followed the pattern of Arabidopsis thaliana wild type after fertilization. Subsequent stages of AE were observed from 2-nuclear up to cellular with the most advanced occurring on medium with 24-epibrassinolide and progesterone. The significant influence of mammalian sex hormones on speed of AE development and differentiation was noticed. Using restriction analysis, the changes in methylation of FIE gene was established under in vitro condition. The authors of this paper showed that Arabidopsis thaliana has a high potency to fertilization-independent development.
