Aeromonas hydrophila is a valuable indicator of the quality of water polluted by sewage and pathogens that pose a risk for humans and cold-blooded animals, including fi sh. The main aim of this research was to evaluate anthropogenic pollution of river water based on genetic diversity of 82 A. hydrophila strains by means of RAPD, semi-random AP-PCR (ISJ) and the rep-BOX conservative repeats test. Genetic diversity of A. hydrophila was HT = 0.28 (SD = 0.02) for all DNA markers (RAPD, semi random and rep-BOX). None of the analyzed electrophoretic patterns was identical, implying that there were many sources of strain transmission. The presence of genes for aerolysin (aerA), hemolysin (ahh1) and the cytotoxic enzyme complex (AHCYTOGEN) was verifi ed for all tested strains, and drug resistance patterns for tetracycline, enrofl oxacin and erythromycin were determined. The most diverse A. hydrophila strains isolated from river water were susceptible to enrofl oxacine (HS = 0.27), whereas less diverse strains were susceptible to erythromycin (HS = 0.24). The presence of the multidrug resistance marker (ISJ4-25; 1100 bp locus) in the examined strains (resistant to three analyzed drugs) indicates that intensive fi sh cultivation affects the microbiological quality of river water.
Aeromonas hydrophila jest cennym wskaźnikiem jakości wody w przypadku zanieczyszczeń ściekami oraz mikroorganizmami względnie patogennymi dla człowieka i zwierząt zimnokrwistych, w tym ryb. Celem niniejszych badań była ocena zanieczyszczenia antropogenicznego na podstawie zróżnicowania genetycznego 82 szczepów A. hydrophila poprzez analizy RAPD, pół-przypadkowo amplifi kowanej klasy AP-PCR (ISJ) i konserwatywnego powtórzenia rep-BOX. Zróżnicowanie genetyczne A. hydrophila wyniosło HT = 0,28 (SD = 0,02) dla wszystkich markerów DNA (RAPD, pół-przypadkowe i rep-BOX). Wszystkie szczepy dla wszystkich markerów ujawniły indywidualny wzór elektroforetyczny, nie ujawniono jednego źródła rozprzestrzeniania się szczepów. U szczepów potwierdzono obecność genów aerolizyny (aerA), hemolizyny (ahh1) i kompleksu enzymów cytotoksycznych (AHCYTOGEN), jak również określono wzorzec oporności na tetracyklinę, enrofl oksacynę i erytromycynę. Najbardziej zróżnicowane okazały się szczepy A. hydrophila wrażliwe na enrofl oksacynę (HS = 0,27) a najmniej zróżnicowane były szczepy wrażliwe na erytromycynę (HS = 0,24). Wyselekcjonowany marker wielolekooporności (locus ISJ4-25, 1100 pz) obecny u szczepów (opornych na 3 rozpatrywane leki) świadczy o wpływie intensywnej hodowli ryb na jakość mikrobiologiczną wody rzecznej.

Out of the twenty-one A. hydrophila complex isolates obtained during a routine examination of human diarrhoeal faeces, two A. hydrophila subsp. dhakensis isolates (P1097 = CCM 7329 and P1165) were successfully identified by ribotyping. The correct taxonomic position of the A. hydrophila subsp. dhakensis CCM 7329 was verified by cpn60 sequencing (GeneBank accession number HM536193). The remaining A. hydrophila complex isolates were identified as A. hydrophila subsp. hydrophila. The ability of biochemical tests and fatty acid methyl ester analysis to reliably discern both A. hydrophila subsp. dhakensis and A. hydrophila subsp. hydrophila was limited. In contrast to the A. hydrophila subsp. hydrophila, the faecal isolates of A. hydrophila subsp. dhakensis did not produce acid from arbutin. When compared in a two-dimensional plot, the A. hydrophila subsp. dhakensis faecal isolates contained higher amounts of the two minor fatty acids C13:0 and C17:1 ω8c than the A. hydrophila subsp. hydrophila reference strain. This is the first detected occurrence of the less frequent A. hydrophila subsp. dhakensis in our region and ribotyping was proved as a suitable method for the identification of A. hydrophila subsp. dhakensis.

The purpose of this study was to distinguish histopathological changes in carp fish (Carassius auratus) infected by Aeromonas hydrophila with different densities. Five treatments were used in the survey for this research method. The treatments used were bacterial infusion of A. hydrophila in the test fish through intramuscular injection with various densities, namely: A. without infection, B: 105 CFU/mL, C: 106 CFU/mL, D: 107 CFU/mL, and E: 108 CFU/mL. The parameters observed were histopathological changes in gill organs, kidneys and hearts of carp fish (gold fish). Data from histopathological observations were analyzed descriptively. The results present gill organs, liver and kidneys of carp fish infected with the bacteria A. hydrophila at various densities experiencing histopathological changes in the form of hemorrhages, necrosis and congestion and hyperplasia of the renal tubules. Other changes that occur were in the kidney organ experiencing a melanomacrophage center (MC). Tubules were coated with connective tissue and lymphocyte infiltration. The most severe organ damage that occurs in carp fish infected with the bacteria A. hydrophila at a density of 108 CFU/mL (treatment E) is shown by a low survival of 5%.

This research aims to determine the best currents speed and utilization of venturi to increasing survival, growth of Cyprinus carpio L. and reducing the infection rate of A. hydrophila bacteria. fingerlings. This research used an experimental method completely randomized design (CRD) with four treatments and four replications. The treatments used were the use of currents and venturi on the maintenance media with currents velocities of 0 m/s (A), 0.06 m/s (B), 0.1 m/s (C), and 0.14 m/s (D). Maintenance of the test fish for 40 days then followed by a challenge test for 14 days. Koi fish fingerlings tested to use A. hydrophila with a density of 108 CF /ml. The parameters observed included white blood cells, red blood cells, clinical symptoms, survival, daily growth rate, feed conversion ratio and water quality. The results showed that the use of currents and venturi with a flow rate of 0.14 m/s was effective in improving the performance of the koi fish's immune system and reducing the infection rate of A. hydrophila bacteria. During the flow administration during the 40 days maintenance period, the white blood cell count increased by 20.51% from a value of 9.19 × 104 cells/mm3 to 11.56 × 104 cells/mm3. Red blood cells increased by 40.65% from 1.60 × 106 cells/mm3 to 2.69 × 106 cells/mm3. Currents treatment with a speed of 0.14 m/s produced the highest survival rate after challenged with a value of 86.7%. Also this treatment was effective in preventing infection of A. hydrophila bacteria, it can be seen from the macroscopic clinical symptoms that appear less than the treatment current. The best results of daily growth rate and feed conversion ratio provided in currents treatment with a speed 0.1 m/s.

This study aims to examine the growth characteristics of B. cereus and B. zhangzhouensis and to see their potential in preventing the growth of Aeromonas hydrophila in vitro. This research was conducted in June 2020 - May 2021 at the Laboratory of Microbiology and Biotechnology, Faculty of Fisheries and Marine Sciences, Padjadjaran University. The method used is the exploratory method and the data obtained were analyzed descriptively. The growth curve was performed using the turbidimetric method. The inhibition test used the disc diffusion method with 5 treatments, chloramphenicol antibiotics (4000 ppm), distilled water, bacterial culture, 100% supernatant and 50% supernatant. The curve results showed that the growth of B. cereus and B. zhangzhouensis was slow. The stationary phase of B. cereus occurred at 40-72 hours, while in B. zhangzhouensis the stationary phase occurred at 48-72 hours. The results of the inhibition test showed that the antimicrobial activity of B. cereus and B. zhangzhouensis was weak with a value range of 0.71 - 1.6mm, besides that the results of the inhibition zone produced by these two bacteria have different inhibitory abilities in each treatment.

Many tropical plants have interesting biological activities with potential therapeutic applications. Garcinia mangostana Linn. (GML) belongs to the family of Guttiferae and is named “the queen of fruits”. It is cultivated in the tropical rainforest of some Southeast Asian nations like Indonesia, Malaysia, Vietnam, Philippines, and Thailand. This research aims to determine the concentration of solution mangosteen rind (Garciana mangostana) which is the most effective to healing of black tilapia juvenile (Oreochromis niloticus Bleeker) when infected by A. hydrophila bacteria to produce the highest survival of black tilapia juvenile. This research used experimental method and experimental design by including a complete randomized design (CRD) with five treatments and three replications. The used treatment was soaking black tilapia juvenile for 24 hours in mangosteen rind. The sample infected by A. hydrophila bacteria as much as 0.1 mL/fish at concentration of 108 cfu/mL with intramuscular injection method was used. The concentrations of mangosteen rind solution were 0, 500, 600, 700, and 800 ppm. The parameters observed were clinical symtoms, fish response to feed, fish response to shock, recovery process, and water quality. Data of the clinical symtoms, response fish to feed, response fish to shock, recovery process and water quality were analysed descriptively, while the survival rate data of black tilapia juvenile were analysed using F test at 95% confidence level and continued with Duncan test. The results showed, the use of mangosteen rind solution with 600 ppm concentration was effective to treat tilapia juvenile which infected by A. hydrophila bacteria resulted in the fastest recovery on the third day and the highest survival rate of 91.76.

This research was conducted from October 2018 until April 2019 in the Faculty of Fisheries and Marine Sciences, Universitas Padjadjaran. This study aims to determine the optimum potassium diformate dosage which was added to feed to increase the immune status of Pangasianodon hypophthalmus (Sauvage, 1878). The research method used is the experimental Complete Random Design (CRD) with 4 treatments and 3 replications. The treatments are A (control), B (0.3% KDF), C (0.5% KDF), and D (0.7% KDF). Observed parameters are survival rate, gross clinical signs, intestine pH levels, total leukocyte count (white blood cells), total erythrocyte count (red blood cells), and water quality. The results showed that the 0.5% KDF addition in feed is the most effective to improve immune status. This treatment also is the most effective to improve Pangasius immune status after being challenged with A. hydrophila with survival rate of 93.33%, leukocyte count 2.98×106 cells/mm3 and erythrocyte count 1.02×106 cells/mm3. Increased leukocyte count was 65.26% and erythrocyte count - 58.23%.

This research aims to determine the effective dosage of potassium diformate (KDF) in feed to enhance sangkuriang catfish immune performances. The research was conducted on November - January 2019, at Aquaculture Laboratory Building No. 4 Faculty of Fisheries and Marine Sciences Universitas Padjadjaran. This study used an experimental method of Completely Randomized Design (CRD), with four treatments and three replications. The treatments are the addition of potassium diformate into feed with doses A (0%), B (0.5%), C (0.7%) and D (0.9%). Observed parameters are clinical symptoms, total leukocyte count, total erythrocyte count, specific growth rate, survival rate and water quality. The results showed that 0.5% KDF feed produced the highest survival rate of 85% and the highest specific growth rate of 3.59% after 40 days. Furthermore, due to treatment, total erythrocyte count increased from 2.09×106 cells/mm3, to 2.10×106 cells/mm3 (8.1%) and total leukocyte count increased from 7.1×104 cells/mm3, to 10.7×104 cells/mm3 (42.81%). The addition of 0.5% KDF in feed is effective in improving the immune performances of sangkuriang catfish challenged by A. hydrophila, with the highest survival rate of 97%. Moreover, recovery of total leukocyte count and total erythrocyte count was faster than other treatments as of day 7th, with a value 1.97×106 sel/mm3and 12.60×104 sel/mm3. It also resulted in the lowest gross clinical signs and the fastest recovering.

This study aims to determine the best density of Bacillus sp. which is given through feed to increase the body resistance of carp that is infected by pathogenic bacteria, Aeromonas hydrophila. The research was conducted at the Aquaculture Laboratory and Laboratory of Microbiology and Biotechnology at FPIK UNPAD. This study used an experimental method with a completely randomized design consisting of 4 treatments and 3 replications, namely treatment A (control), treatment B (Bacillus sp. 106 cfu/mL feed), treatment C (Bacillus sp. 108 cfu/mL feed) and treatment D (Bacillus sp. 1010 cfu/mL feed) for two weeks later in Aeromonas hydrophila infection with an intramuscular density of 108 cfu/mL for seven days. Observation parameters were clinical symptoms, fish response to feed, fish response to shock and survival. The results of this study showed that treatment C (Bacillus sp. 108 cfu/mL feed) resulted in 89% survival and reduced clinical symptoms.

This study aims to see the condition of hematocrit levels and differential leukosit after immersion in LAB isolated and after in vivo, using Aeromonas hydrophila bacterium. LAB isolated were obtained from the organs of the common carp intestine. The study used a Completely Randomized Design (RAL) consisting of four treatments and three replications. The treatment used was immersion of test fish for 24 hours in several isolates of LAB with a density of 108, namely A: without LAB isolate (control), B: isolate CcB7, C: isolate CcB8, D: isolate CcB15. Immersion was done three times, with a frequency of seven days. Parameters observed included hematocrit levels, leukocyte differentiation and survival rate. The results showed that immersion in LAB isolate CcB15 will enhance the differential leukocytes and hematocrit levels that serve as indicators of carp body resistance, which is shown by high levels of white and red blood cells.
