In this study, milk samples were taken in sterile 20 ml containers from 90 cows with clinical mastitis in various enterprises around Aydın province between May-September 2017. The presence of Aspergillus flavus M1 toxin in milk samples was investigated by ELISA. Aflatoxin M1 ELISA results were compared with positive controls with 0 ppt, 5 ppt, 15 ppt, 30 ppt, 60 ppt and 100 ppt. For the 88 samples examined; 5 ppt in 7 (8,0 %), 15 ppt in 12 (13,6 %), 100 ppt in 32 (36,4 %) and 0 ppt in 37 (42,0 %) aflatoxin M1 was detected. According to these results, 32 (36,4 %) AFM1 positivity was detected in the European Union countries over the limit (50 ppt) accepted for raw milk. In our study, DNA obtained from 88 mastitis milk samples examined with Aflatoxin M1 ELISA kit was subjected to Aspergillus flavus species specific PCR process. As a result of PCR; Aflatoxin M1 at a level of 0 ppt in 7 (36,8 %), 5 ppt in 1 (% 5,2), 15 ppt in 2 (10,5 %) and 100 ppt in 9 (47,5 %) ELISA kit toxin presence was determined. As a result, 32 samples of 100 ppt AFM1 positivity were detected by ELISA and 9 samples 100 ppt positivity was detected by PCR in our study. The results of the ELISA test revealed that more positive results were formed due to cross-reactions, and the PCR method with FLA gene primers was more reliable for diagnosis.

Aspergillus flavus is a saprophytic fungus which can grow on corn and produce aflatoxins which render it unsafe for consumption as food and feed. In this study, aflatoxin and non-aflatoxin producing isolates of A. flavus were grown separately on wet (20% water added), sterile or non-sterile cracked corn. Wet and dry cracked corn controls were included as needed. Secondary metabolic volatiles were identified and aflatoxin concentrations determined over a 12-day period. Volatiles unique to the toxigenic A. flavus isolates were determined by comparison with volatiles produced by the respective corn controls and the non-toxigenic A. flavus isolate. The number and identity of the volatiles produced by these A. flavus isolates varied by isolate, whether sterile or non-sterile corn was the substrate, and the sampling day. Overall, most of the volatiles were produced before day 8 after inoculation. Aflatoxin production was 10-fold lower on the sterile corn, compared to the nonsterile corn. Volatiles unique to the aflatoxin producing isolates were identified on both substrates after comparison with those produced by the non-aflatoxin producing isolate, as well as the corn control samples. Results indicate that several factors (substrate, fungal isolate, culture age) affect volatile and aflatoxin production by A. flavus.

Opis:
Intensive poultry production is associated with extreme densities of birds crammed into industrial-scale hen houses. Consequently, the bird keepers are exposed to elevated concentrations of dust. Exposure to dust, containing pathogenic mycological agents, may cause exacerbate asthma, allergic alveolitis, as well as organic dust toxic syndrome. Geographic Information System (GIS) allows to view, understand, question, interpret, and visualize biological data, among them mycological threats. The aim of the presently reported study was to create the mycological database from the chicken farms and their visualization on the map of Lower Silesia, Poland. Field data for Aspergillus fumigatus, A. flavus, Candida sp. were collected in Wrocław laboratory in 2008. GIS database was formed using ArcView 9.2. computer software. The visualization data on the digital maps were analyzed for the Health and Safety Executive.

Fungal antagonist, Talaromyces flavus, is one of the most important biological agents of soil-borne fungal diseases including Verticillium and Fusarium wilt. In this study, to increase the effectiveness of T. flavus isolates obtained from greenhouse cucumbers and field grown tomatoes five chemical stabilizers were evaluated. Based on the results of previous studies, the most effective substrate for the growth, sporulation and stability of T. flavus isolates related to the above-mentioned plants was a mix of rice bran and peat-moss. Different chemical stabilizers were mixed with the above-mentioned substrate containing spore suspensions of various T. flavus isolates. For each plant, a completely randomized experiment was conducted under greenhouse conditions with seven treatments and three replications. The results of this study indicated that treatments containing sodium nitrate and D-cycloserine were more effective than those containing other stabilizers. The overall results of this study suggest that the use of some chemical stabilizers may enhance the biocontrol potential of fungal antagonists in controlling different plant diseases including Verticillium and Fusarium wilt.

Timer decay is caused by primarily enzymatic activities of microorganisms. For the first time fungal diversity of timber degrading fungi was studied in Gujarat, India. Timber Degrading Fungi belonging to Aphyllophorales are economically important as many of these cause serious damage in sawmills of Gujarat. To find out the association of the timber degrading fungi and timber decay problems in sawmills a survey was conducted during 2007 to 2011 in different sawmills of 5 districts of Gujarat i.e. Vadodara, Ahmedabad, Bharuch, Rajkot and Jamnagar. In the present study teak wood present in sawmills was infected with 14 types of fungi in which Lenzites sterioides and Trametes versicolor damaged the wood severely was reported for the first time. In all 94 sawmills were surveyed, the 28 sawmills were from Vadodara, 29 from Ahmedabad, 12 from Bharuch, 21 from Rajkot and 4 from Jamnagar. Out of 94 sawmills survyed, 84 sawmills were having timber rotting fungi associated with wood. Maximum fifteen and thirteen fungal species were observed in saw mills of Chhani road, followed by 11 in Station road, 7 in Dhabhoi road and 6 in Harni, Vadodara. Fours woods uninfected are Beyo, Marinty, Ash, and Arjun. Fourteen different types of fungi were found associated with teak, followed by seven in pinus, madhuca, Acacia nilotica, six in babul, neem, four in tamarind, Pithacoelobium and three in mango, Eucalyptus, African Mahagoni, Kapoor, Peltophoram rouxburghii, Derris pinnata wood respectively. The commonly observed timber decaying fungi were Schizophyllum commune, Flavodon flavus and Ganoderma lucidum belonging to Basidiomycota. Ascomycota members included was Daldinia concentrica and Xylaria polymorpha.

The aim of this study was to assess the total aflatoxin (TAT) contents of roasted, dehulled and hulled groundnuts (Arachis hypogaea L.) and of roasted cashew nuts (Anacardium occidentale), sold and consumed in some public areas of Gboko metropolis of Benue State, Nigeria. The items were sampled from 5 different locations based on human traffic densities and sellers of the product and then analysed for TAT contents using direct competitive Enzyme-linked Immunosorbent Assay (ELISA) method. In the roasted cashew nuts, TAT was only detected in samples collected at two of the sampling sites; Mechanic Site/Adekaa (>20.00 µg/kg) and Gboko Main Market (0.30 µg/kg) but not detected in samples collected at the three others (Gboko motor park, Abagu and Tipper Garage). Nevertheless, the detected levels at Gboko Main Market were below the maximum tolerable limits (MTL) of 4 µg/kg set by EU and 10.0 µg/kg set by CAC and NAFDAC, Nigeria for ‘ready to eat’ food substances unlike the situation at the Mechanic Site/Adekaa sampling location. In the roasted groundnuts, levels of TAT higher than the indicated MTL were documented in samples collected at Tipper Garage (12.20 µg/kg) and Abagu (5.70 µg/kg) only exceeded the EU MTL, while the results at Gboko motor parks (2.00 µg/kg), Gboko main market (1.30 µg/kg) and Mechanic site/Adekaa (0.50 µg/kg) were below the EU MTL. Samples of hulled groundnuts collected at Gboko main market recorded TAT levels of 0.10 µg/kg while at Mechanic site/Adekaa it was not detected. Dehulled groundnuts sampled at Gboko Main market (0.40 µg/kg) and Mechanic site/Adekaa (0.20 µg/kg) recorded TAT levels within CAC and NAFDAC MTLs. The dehulled groundnuts samples collected at the Tipper Garage sample location, which recorded a moisture level of 4.20 %; yet among the rest of all the other samples had moisture levels below the recommended maximum tolerable limits of 5.80 % for stored groundnuts and cashew nuts. The results of microbial counts revealed that total mesophilic bacteria ranged between < 1.0 × 101 to >1.0 × 102 CFU/L, fungi varied between < 1.0 ×101 to > 1.0 × 101 CFU/L while E.coli counts varied between 0.00 to 0.32 CFU/L. The microbial contagion was generally low. Nevertheless, prolonged feasting on the aflatoxin-contaminated nuts is possible in the study area due to the rising food insecurity of majority of the masses. The chronic and acute exposures therefore, presents health concern of aflatoxicosis.

Microbial conversions are gaining importance in the synthesis of important drug metabolites and their intermediates as they are good alternative to chemical synthesis since they are enantio-selective and regio-selective and even can be carried out at ambient temperature and atmospheric pressure. Till date, biocatalytic reduction of acetophenone and its derivatives has been widely reported. In the present study, we have made an attempt to carry out the microbial bioreduction of o-hydroxyacetophenone by screening some of the selected microorganisms which were obtained from culture collection centre as well as those which are isolated in our Microbiology lab. The selected microorganisms include Aspergillus ochraceous, Aspergillus flavus, Aspergillus tubingenesis, Aspergillus niger, Rhizopus stolanifer MTCC 162, Rhizopus stolanifer MTCC 2591 and Baker’s yeast.Among the seven microorganisms screened for the bioreduction of o-hydroxyacetophenone, Baker’s yeast and Aspergillus tubingenesis showed significant bioconversion where as Aspergillus ochraceous exhibited the least bioconversion.In our earlier study it was found that Aspergillus flavus has the required bioreductase enzyme, which showed the maximum conversion of p-chloroacetophenone to p-chlorophenylethanol. Hence optimization of culture conditions to get maximum enzyme expression and hence maximum conversion was thought off. The parameters considered for the study include effect of various Carbon sources, Nitrogen source, Metal ions, incubation Temperature and media pH on enzyme expression. The optimized culture a condition at which maximum bioconversion was achieved was maltose among various carbon sources. Tryptone was found to have maximum effect among the nitrogen sources. Media pH 7.6 and incubation temperature of 35 °C was found to be favourable for maximum enzyme activity. Among various divalent metal salts, addition of magnesium sulphate to the media significantly increased the enzyme activity.
