STAT proteins belong to the transcriptional factors family, and each of them performs a unique function in extracellular signal transduction and in direct regulation of transcription. Their function is based on controlling genes expression, which is involved in cell survival, proliferation, chemoresistance and angiogenesis. Phosphorylated STAT3 is observed in 70% of human cancers. STAT3 as an oncogenic protein is constitutively activated in many primary human cancers by different cytokines as: IL-6 IL-7, IL-10, IL-20, leptin, granulocyte colony-stimulating factor (G-CSF), epidermal growth factor (EGF), platelet-derived growth factor (PDGF), and oncogenic proteins such as Src and Ras. Moreover, STAT3 can be activated by receptor and nonreceptor tyrosine kinases such as: epidermal growth factor receptor kinase (EGFR), activated Janus kinase (JAK) or kinase regulating extracellular signals (ERK). An important role of STAT3 is the regulation of cancer cells autonomous properties. The blocking of STAT3 expression in human cancer cells inhibits proliferation in vitro and cancer progression in vivo. To inhibit gene expression of STAT3, antisense oligonucleotides, rybozimes and DNAzymes can be used. The STAT3 protein can be blocked by tyrosine kinase inhibitors, negative dominants for the STAT3 protein, complementary to small nonpeptide particle drugs. Among the newest methods of gene expression regulation is the RNA – RNAi method of interference.
Białka STAT (signal transducer and activator of transcription – przekaźnik sygnału i aktywator transkrypcji) to rodzina czynników transkrypcyjnych, z których każdy pełni unikalną funkcję w przekazywaniu sygnałów zewnątrzko-mórkowych oraz bezpośrednim regulowaniu transkrypcji. Ich funkcja polega na kontroli ekspresji genów, które zaangażowane są w przeżycie komórek, proliferację, chemiooporność oraz angiogenezę. Ufosforylowany STAT3 obserwuje się w blisko 70% ludzkich nowotworów. Pełniąc rolę białka onkogennego ulega on konstytutywnej aktywacji w wielu pierwotnych nowotworach u ludzi, będąc aktywowanym przez wiele różnych cytokin, takich jak IL-6 IL-7, IL-10, IL-20, leptyna, czynnik stymulujący wzrost kolonii granulocytów (granulocyte colony stimulating factor – G-CSF), epidermalny czynnik wzrostu (epidermal growth factor – EGF), płytkowy czynnik wzrostu (platelet-derived growth factor – PDGF), a także białka onkogenne, m.in. Src i Ras. Ponadto STAT3 może być aktywowany poprzez receptorowe i niereceptorowe kinazy tyrozynowe, takie jak: kinaza receptora epidermalnego czynnika wzrostu (kinase of epidermal growth factor receptor – EGFR), aktywowana kinaza Janus (activated Janus kinase – JAK), kinazy regulujące sygnały zewnątrzkomórkowe (kinases regulating extracellular signals – ERK). Jego istotną funkcją jest regulacja autonomicznych właściwości komórek nowotworowych. Blokowanie ekspresji STAT3 w ludzkich komórkach nowotworowych hamuje proliferację in vitro oraz progresję nowotworów in vivo. W celu wyciszenia ekspresji genów STAT3 można wykorzystać oligonukleotydy antysensowe, rybozymy i DNAzymy. Samo białko STAT3 można zablokować wykorzystując inhibitory kinazy tyrozynowej, dominanty negatywne wobec białka STAT3, komplementarne wobec leków małe niepeptydowe cząsteczki. Wśród najnowszych metod regulacji ekspresji genów znajduje się metoda wykorzystująca proces interferencji RNA – RNAi.

The power of teaching by using computer programs is well known. But some of commercial programs are very expensive and out of a range of many universities. In this paper we propose a original computer program STAT_STUD, which is almost free, very functional and easy to use. The program includes the following modules: descriptive and mathematical statistics, calculator and generator. In the paper we discuss the mentioned modules and present the merits of the program.

Opis:
Interferons (IFNs) induce gene expression by phosphorylating latent transcription factors belonging to the signal transducer and activator of transcription (STAT) family, mediated by janus kinases (Jaks). STAT dimers directly activate genes containing the IFNγ activation site (GAS) DNA element, with different STAT proteins displaying slightly different intrinsic DNA binding specificities. The combinatorial association of STATs with the additional DNA binding adaptor protein interferon regulatory factor (IRF)9 expands the range of enhancer elements that can be targeted by the JAK-STAT pathway to interferon-stimulated response element (ISRE) and IRF response element (IRE). Based on the amino-acid sequence similarity within the IRF family and functional overlap with the STAT family, in this paper we hypothesize that other IRF members could serve as adapter proteins for the STATs during IFN responses to redirect them to subsets of ISRE, GAS and/or IRE-containing IFN-stimulated genes (ISGs). In addition, the fact that STAT2 homodimers are not capable of binding consensus GAS sites leaves the possibility for a novel type of DNA-binding site bound by STAT2 homodimers and potentially other STAT complexes.

Opis:
In this study the recharge by precipitation of the upper Narew catchment was evaluated. Data from the MARS-STAT database and from the ground net of the rain gages belonging to the Institute of Meteorology and Water Management (IMGW) were acquired for the period 1978–2002. Analysis has indicated average 5% underestimation of the mean annual precipitation estimated with the MARS- -STAT data in comparison to the IMGW data. Evaluation of monthly data brought significantly bigger differences. Presented results indicate need of precise MARS-STAT data verifi cation before this data source could be applied in hydrological modeling.

Out of the two forms of genitive plural of the Ukrainian noun stat't'a ‘article', namely stattej and statej, the former has been assumed to be purely Ukrainian, whereas the latter a Russified one. The paper attempts to demonstrate that the relationship is not necessarily as simple, moreover, that such an interpretation does not altogether answer the question of why only the form of genitive plural, and why of this very word, would have become the object of a stronger Russification.

Interleukin-6 is a potent inducer of acute-phase response gene transcription. The intracellular signal transduction mechanisms by which this and other biological effects of the cytokine are achieved include activation of the JAK-STAT signaling pathway. More specifically, activation of the signal transducers and activators of transcription STAT1, 3, and 5 in response to IL-6 has been described. We examined the relative potency of these three STAT factors for the activation of acute-phase gene promoters in HepG2 cells in a reporter gene-based assay, where specific STAT factors could be activated via recombinant receptor constructs bearing different STAT-recruiting modules. These experiments indicate that amongst the STAT factors known to be activated by IL-6 STAT3 is the most potent activator of acute-phase gene transcription.

Okra (Abelmoschus esculentus L. Moench) yield is characterized by low productivity but may be increased through the adoption of new technology or the efficient allocation of existing resources. In addition, improving the level of resource allocation is important for improving productivity. This study investigated resource use efficiency in small-scale okra production. A multi-stage sampling procedure was used to select 100 respondents. The primary data generated through the survey of 100 okra farmers were analyzed using descriptive statistics, the ordinary least squares regression model, and marginal value analysis. The average age of the respondents was 45 years, with 7 members in the household, and 2.8ha being farmed. The ordinary least squares regression estimate indicated age, labor, farm size, and herbicide and insecticide use that influenced okra production. Returns to scale was 2.09; resource use efficiency indicated the following values: farm size 0.343, herbicide application 0.857, insecticide application 0.75, and labor 4.80. The high cost of inputs (1.93), low numbers of extension contact with farmers (1.82), high incidence of pests and disease (1.49), non-access to credit (1.39), difficulty in obtaining labor (1.23), drought (1.15), and marketing problems (1.09) are constraints negatively affecting okra production. Although the positive and statistically significant effect of pesticides, farm size, and labor indicate these inputs can effectively improve okra production, small-scale okra production is inefficient because farmers use too many pesticides and farmland and less labor than is needed. The marginal product of inputs is not enough to offset the corresponding marginal cost. Therefore, small-scale farmers should operate on smaller farms than they currently do, apply pesticides at recommended rates, and spend more on labor. So, training should be organized through extension workers and agricultural research stations to improve scientific knowledge of farmers for efficient use of productive resources.

Monitoring toxic metal concentrations in fish is very important for human health because fish, which are consumed by humans, can accumulate toxic metals from water up to hazardous levels. The aim of this study has been to monitor the levels of Cd, Pb, Cu, Cr and Ni in three deep-sea fish species as well as to make a risk assessment of their consumption. The samples were digested in a microwave oven and the determinations were made by atomic absorption spectrophotometry (AAS) and inductively coupled plasma-atomic emission spectrophotometry (ICP-AES). The mean lead concentrations in the Salmo salar and Sarda sarda species were found to be 955 and 948 μg kg-1, respectively, and these levels are three-fold higher than the risk threshold level of 300 μg kg-1. Cd concentrations in only six samples were slightly higher than the maximum allowable concentrations (MACs) of 100 μg kg-1. Mean Cr and Ni concentrations in Salmo salar were found to be 866 and 472 μg kg-1, respectively, and these results were higher than in Sarda sarda (mean 388 μg Cr kg-1 and 356 μg Ni kg-1) and Merlangius merlangus (mean 303 μg Cr kg-1 and 336 μg Ni kg-1). The measured Pb concentrations in all muscles of Salmo salar and Sarda sarda were found to be significantly higher than the MAC values, while Cd in all studied samples was around or lower than the MACs. However, there is no carcinogenic risk for humans, and the risk of developing cancer over an average human lifespan is between 2.5 and 13 in 1,000,000.

Breakdown of the Soviet Union has clearly revealed insufficiency and in many directions even unsuitability of the existing for many years system of economic and statistical information for assessment of radical changes taking place in economy and social life. Serious reconstruction was needed of the existing fundamentals of primary accounting and statistics by means of reforming common methodological and organizational basis of statistics itself, as well as adjusting it in accordance with international rules and standards. First of all, a serious step in the movement “towards the west” was the support proposed by The European Union in the field of transmission of “know-how” through the TACIS Program - Program of technical assistance to the CIS countries. The Program has covered a wide range of directions: reforming of the system of government administration; restructuring of public enterprises and development of private sector, finances and banking sector, development of transport and communication infrastructure, energy sector, creation of efficient production system, etc. But statistics were not forgotten. In the total TACIS budget its share is rather low, not more than 0.2%. However, statistical projects play a special role in the creation of a new information infrastructure and promotion of the efficient exchange of data. Cooperation with the EU within the framework of the TACIS Program has played and important role in the development of statistics in the CIS countries. However, it should be emphasized that for the development of the statistical potential of these countries it is also very important to interact with statistical services of other leading international organizations, first of all, with UNECE, OECD, IMF, the World Bank, ILO and other. The solution of the tasks faced by statisticians of the Commonwealth countries was promoted and will be promoted by the discussion of urgent problems at the Conference of European Statisticians, active cooperation with statistical organizations of the region. Big role in building up national statistics was played by the Interstate Statistical Committee of the Commonwealth of Independent States, which at the first stage of CIS existence CIS-STAT became the main moderator of international standards introduction. Today CIS-STAT faces new challenges. First of all, this is methodological unification of different large-scale statistical works. All the measures taken have enabled the creation and development of the interstate statistical system of the Commonwealth, although not everything took place in favorable conditions. Many national statistical services face serious financial difficulties constantly and for that reason they lag behind in technical equipment and other supplies, can not keep qualified personnel and have to reduce statistical activities. Some national statistical services are forced to give up conduct of a number of important statistical works.
