In transgenic plant development, the low transformation efficiency of Agrobacterium with exogenous DNA is the major constraint, and hence, methods to improve its transformation efficiency are needed. Recently, nanoparticlemediated gene transfer has evolved as a key transformational tool in genetic transformation. Since silver nanoparticles (AgNPs) can induce pores on the cell membrane, their efficacy in the improvement of conventional calcium chloride freeze-thaw technique of transformation of Agrobacterium was explored in this study. Agrobacterium cells in the exponential growth phase were exposed to different concentrations of AgNPs (0.01, 1, 5, 10, and 20 mg/l), and the half-maximal effective concentration (EC50) was determined via Probit analysis using the SPSS software. Transformation efficiency of AgNPs alone and in combination with calcium chloride was compared with that of the conventional calcium chloride freeze-thaw technique. AgNPs at a concentration of 0.01 mg/l in combination with calcium chloride (20 mM) showed a ten fold increase in the transformation efficiency (3.33 log CFU (colony-forming unit/microgram of DNA) of Agrobacterium tumefaciens strain EHA 105 with plasmid vector pART27 compared with the conventional technique (2.31 log CFU/μg of DNA). This study indicates that AgNPs of size 100 nm can eliminate the freeze-thaw stage in the conventional Agrobacterium transformation technique, with a 44% improvement in efficiency. The use of AgNPs (0.01 mg/l) along with 20 mM calcium chloride was found to be an economically viable method to improve the transformation of Agrobacterium with exogenous plasmid DNA.

Opis:
Purpose: This paper aims to investigate the possibilities of using 3D printing by fused deposition modelling (FDM) technology for developing micro-fluidic devices by printing a benchmark test part. A low-cost desktop printer is evaluated to compare the minimum possible diameter size, and accuracy in the microchannel body. Design/methodology/approach: The parts were designed using SolidWorks 2016 CAD software and printed using a low-cost desktop FDM printer and Polylactic acid (PLA) filament. Findings: Desktop 3D printers are capable of printing open microchannels with minimum dimensions of 300 μm width and 200 μm depth. Research limitations/implications: Future works should focus on developing new materials and optimizing the process parameters of the FDM technique and evaluating other 3D printing technologies and different printers. Originality/value: The paper shows the possibility of desktop 3D printers in printing microfluidic devices and provides a design of a benchmark part for testing and evaluating printing resolution and accuracy.

Opis:
Alzheimer’s disease (AD) is a brain disorder and the main reason for dementia. In this regard, there is a need to understand the alterations that occur during aging to develop treatment strategies to mitigate or prevent neurodegenerative consequences. Onion and garlic root extracts contain natural polyphenols with high antioxidant capacity; therefore, the present study aimed to investigate the protective effect of these extracts free from mycotoxin contamination on a rat model of AD. Antifungal and antibacterial assays were performed for onion and garlic extracts. Several groups of AD-induced rats were administered 1, 2, and 3 mg/kg onion or garlic extract through intragastric intubation for 30 days. After treatment, histopathological analysis, expression of apoptosis-related genes, and analyses of DNA damage and reactive oxygen species (ROS) generation were conducted in the brain tissues. The results indicate that treatment of AD-induced rats with several doses of onion and garlic root extracts decreased histopathological lesions, the expression levels of apoptotic genes, and the rate of DNA damage and inhibited intracellular ROS generation in the brain tissues. The results suggest that the protective role of onion root extract could be attributed to its content of flavonoids and flavonoid compounds through the improvement of antioxidant capacity and regulation of gene expression patterns. The higher activity levels of free radical scavenging of azino-bis (3-ethylbenzothiazoline-6-sulphonic acid (ABTS) and antioxidant ferric reducing antioxidant power (FRAP) levels found in garlic root extract are most probably responsible for its protective effect against neurodegenerative damage.

Wprowadzenie do hodowli szczepu St. maltophilia KB2 nanocząstek tlenku niklu nie zahamowało procesu biodegradacji fenolu, a reakcja populacji bakterii była uzależniona od stężenia nanocząstek i użytego surfaktantu. Opracowano metodykę przygotowania stabilnej dyspersji badanych nanocząstek oraz oceniono wpływ wybranych surfaktantów na wzrost komórek szczepu St. maltophilia KB2.
The introduction of nickel oxide nanoparticles into the St. maltophilia KB2 cultures did not inhibit the phenol biodegradation process, and the reactions of the bacterial population were depended on the nanoparticles concentrations and the surfactant used. The methodology for the preparation of the tested nanoparticles stable dispersion was developed and the influence of selected surfactants on the growth of St. maltophilia KB2 cells was assessed.

Most studies of wetlands in Uganda use for water and wastewater treatment have focused on natural undisturbed pristine wetland and constructed wetlands. There exists a gap in literature surrounding treatment abilities of restored wetland or wetlands on a path to recovery. Water quality parameters were sampled covering wet and dry seasons in a restored wetland. Soil properties were also assessed and compared with a referenced natural site. The objectives of the study were to (i) determine the effects of wetland restoration on nutrient retention and release, (ii) analyze temporal dynamics of water quality parameters in the restored wetland, and (iii) describe soil properties in depths of the restored wetland in relation to a reference natural site. Values of pH, electrical conductivity (EC), temperature, total dissolved solids (TDS), TN and NO3-N showed significant differences across seasons. The wetland demonstrated little water quality benefits but the overall nutrient removal efficiency when compared to literature values was found to be not impressive. Rates for P and N removal were between 3.5 - 19% and -10.8 – 32.4% respectively. These results could be attributed to flooding, low residence time of the water, small size of the wetland, low influent load, extra nutrient inputs from fertilizers and pollution sources as well as release from soil. We recommended that management activities like harvesting; redesigning the channels for water distribution throughout the system and removal of invasive plant species be carried out. This study will form a baseline in understanding the ecology of restored wetlands in Uganda.

Interkosmos - the international space research program - is multilayered; it covers a variety of aspects: political, scientific, economic, propaganda, technical, etc. In the article, the authors provide insight into technical support for the Interkosmos program carried out at the Dnepropetrovsk Space and Rocket Center. The engineering background for the conversion of the Center-developed military rocketry for use in space exploration is studied. The authors, relying on the recollections of the participants, reveal the little-known aspects of the creation of a powerful satellite development facility at the Dnepropetrovsk Space and Rocket Center. The article brings into the foreground the importance of KB Yuzhnoye’s idea of developing a ‘unified spacecraft’, meaning a space platform where it was possible to install miscellaneous devices for pursuing a wide range of scientific issues. Using the example of Poland and India, some results obtained during launches of unmanned and manned spacecraft under the Interkosmos program are studied. The participation of the mentioned countries in the creation process of many scientific instruments for implementation of the Interkosmos program and the importance of this research and design direction are accentuated.
Międzynarodowy projekt badań kosmicznych Interkosmos składał się z wielu elementów; w jego funkcjonowaniu można wyodrębnić różne aspekty: polityczny, naukowy, ekonomiczny, propagandowy, techniczny etc. W artykule autorzy przedstawiają techniczny wykład Dniepropietrowskiego Ośrodka Kosmiczno-Rakietowego w program Interkosmos. Artykuł omawia techniczne zagadnienia przekształcenia produkowanych w Ośrodku rakiet wojskowych do wykorzystania w eksploracji kosmosu. Autorzy, opierając się na wspomnieniach uczestników tych wydarzeń, opisują mało znane aspekty utworzenia prężnych zakładów produkujących satelity w Dniepropietrowskim Ośrodku Kosmiczno-Rakietowym. Artykuł podkreśla znaczenie rozwijanej w Biurze Projektowym Jużnoje idei stworzenia „zunifikowanego statku kosmicznego” - podstawy konstrukcyjnej, na której można by instalować różne instrumenty do rozwiązywania szerokiego wachlarza problemów badawczych. Na przykładzie Polski i Indii zostały przedstawione wyniki, które uzyskano podczas bezzałogowych i załogowych misji kosmicznych w ramach programu Interkosmos z zaznaczeniem udziału tych krajów w opracowaniu instrumentów naukowych służących programowi oraz znaczenie tego kierunku badań i rozwoju.

In the paper, a back swept impeller of centrifugal compressor is experimentally studied and numerically validated and modified to increase its pressure ratio and improve efficiency, as well as to analyse the effect of splitter blade location between two main blades. The back swept multi splitter blade impeller was designed with a big splitter positioned close to the main blade suction surface and a smaller splitter close to the pressure surface. Adding this multi splitter improves the overall performance of the modified impeller due to less intensive flow separation and smaller pressure loss. In particular, the total pressure ratio was observed to increase from 4.1 to 4.4, with one percent increase in efficiency.

Indoneesiella echioides (L.) Nees is a well-known herb used in ethnomedicine. Flavonoids and alkaloids are major constituents of Indoneesiella echioides (L.) Nees. In this study, we investigated the anticancer activity of KB and normal HGF-1 cells. We demonstrated the effects of ethanolic extract of Indoneesiella echioides (L.) Nees on the cell growth and apoptosis in KB and normal HGF-1 cells were analyzed by the generation of reactive oxygen species (ROS), the level of mitochondrial membrane potential (∆ψM) and apoptotic morphological changes were analyzed by AO/EtBr, AO and Hoechst staining. Our results indicated that the ethanolic extracts of Indoneesiella echioides (L.) Nees shows better anticancer activity through it’s induces increased cell death, ROS generation, alteration of mitochondria membrane potential and apoptosis.

Objectives Welding processes that generate fumes containing toxic metals, such as hexavalent chromium (Cr(VI)), manganese, and nickel (Ni), have been implicated in lung injury, inflammation, and lung tumor promotion in animal models. Bronchiolar epithelium Clara cells/club cells, coordinate these inflammatory responses. Clara cells secretory protein (CC16) with ant-inflammatory role. Material and Methods The pulmonary toxicity of welding dust (WD) was assessed for Wistar rats exposed to 60 mg/m³ of respirable-size welding dust (mean diameter 1.17 μm for 1 and 2 weeks (6 h/day, 5 days/week)) or the aerosols of soluble form (SWD) in the nose-only exposure chambers. Additionally the effect of antiinflammatory betaine supplementation was assessed. Clara cells secretory protein, differential cell counts, total protein concentrations and cellular enzyme (lactate dehydrogenase – LDH) activities were determined in bronchoalveolar lavage fluid, and corticosterone and thiobarbituric acid reactive substances (TBARS) and prolactin concentrations were assessed in serum. Histopathology examination of lung, brain, liver, kidney, spleen was done. Additionally slices of brain and lung were exanimated in laser ablation inductively coupled plasma mass spectrometry. Results Both WD and SWD exposure evoked large bronchiolar infiltration shoved in histopathology examination. In this study, TBARS inversely correlated with a significant decrease of CC16 concentration that occurred after instillation of both WD and SWD indicating decreased anti- inflammatory potential in the lung. In WD exposed rats prolactin correlated with nuclear factor-kappa B (NF-κB), LDH, TBARS and serum levels Cr, Ni and inversely with c-Jun. In SWD exposed rats prolactin correlated with CC16 indicated effect of prolactin on the population of epithelial cells. Conclusions In the current study, deleterious effects of repeated inhalation stainless steel welding dust form on club (Clara) cell secretory protein (CC16) were demonstrated. Clara cells secretory protein relation with prolactin in exposed rats to welding dust were shown and explored whether the NF-κB and c-Jun/activator protein 1 related pathway was involved. The cometabolic biodegradation of 4-Chlorophenol (4-CP) by the Stenotrophomonas maltophilia KB2 strain in the presence of phenol (P) was studied. In order to determine the kinetics of biodegradation of both substrates, present alone and in cometabolic systems, a series of tests was carried out in a batch reactor changing, in a wide range, the initial concentration of both substrates. The growth of the tested strain on phenol alone was described by Haldane kinetic model (mm = 0:9 1/h, Ksg = 48:97 gg/m3, KIg = 256:12 gg/m3, Yxg = 0:5715). The rate of 4-CP transformation by resting cells of KB2 strain was also described by Haldane equation and the estimated parameters of the model were: kc = 0:229 gc=gxh, Ksc = 0:696 gc=m3, KIc = 43:82 gc=m3. Cometabolic degradation of 4-CP in the presence of phenol was investigated for a wide range of initial 4-CP and phenol concentrations (22–66 gc/m3 and 67–280 gg/m3 respectively). The experimental database was exploited to verify the two kinetic models: CIModel taking only the competitive inhibition into consideration and a more universal CNIModel considering both competitive and non-competitive inhibition. CNIModel approximated experimental data better than CIModel.
