Opis:
Diclofenac sodium is often used drug with a strong anti-inflammatory, analgesic and antipyretic effect, belonging to NSAID. Diclofenac sodium was detected on plates precoated with silica gel 60F254. Nineteen new visualizing (known as dyes) namely: brilliant cresyl blue, malachite green, alkaline blue, methylene blue, methylene violet, Janus blue, bromophenol blue, brilliant green, methyl green, methyl red, bromocresol green, bromothymol green, gentian violet, crystalline violet, aniline blue, neutral red, titan yellow, thymol blue, bromothymol blue were used to detected diclofenac sodium. Rhodamine B and fuchsine were used as comparative visualizing reagents for detection of diclofenac sodium. Among all studied new visualizing reagents, brilliant-cresol blue, malachite green and titan yellow are the best to detect diclofenac sodium. Similar detection and quantitative limit values of diclofenac sodium were obtained without using visualizing reagent and after detection using the following reagents: fuchsin, alkaline blue, Janus blue, methylene blue, aniline blue and crystal violet. The coloured spots and spectrodensitograms of diclofenac sodium obtained after the use of individual visualizing reagents can be an additional parameter in studying its identity.

Lipid was extracted from the muscle of Cuttle fish by solvent extraction method and then purified by suitable conventional method. Various physical constants e.g. refractive index, viscosity, specific gravity, crude fat, crude fibre and ash contents and chemical parameters e.g. saponification value, saponification equivalent value, acid value, iodine value, acetyl value, peroxide value, thiocyanogen value, Reichert-Meissl value, Polenske value, Henher value, cholesterol content etc. of the lipid sample have been determined and compared with those of different standard oils. Fatty acids composition of the sample was investigated by Thin Layer Chromatography (TLC) method. The muscle lipid of Cuttle fish was found to contain palmitic acid, stearic acid, linolenic acid and erucic acid respectively with some other unknown fatty acids. Antimicrobial activities of the lipid were tested by standard method and found moderate to potential antibiotic and antifungal property in this lipid. The lipid containing muscle of Cuttle fish was analyzed quantitatively for the determination of percentages of protein and minerals (N, P, K, Ca) contents by modified Kjeldahl method.

Opis:
Fucoxanthin, diadinoxanthin and diatoxanthin are carotenoids found in brown algae and most other heterokonts. These pigments are involved in photosynthetic and photoprotective reactions, and they have many potential health benefits. They can be extracted from diatom Phaeodactylum tricornutum by sonication, extraction with chloroform : methanol and preparative thin layer chromatography. We assessed the utility of an additional column chromatography step in purification of these pigments. This novel addition to the isolation protocol increased the purity of fucoxanthin and allowed for concentration of diadinoxanthin and diatoxanthin before HPLC separation. The enhanced protocol is useful for obtaining high purity pigments for biochemical studies.

Opis:
The trans fatty acids in milk fat from the indoor feeding ( March, April) and the pasture feeding period (May) were determined. An argentation thin layer chromatography (Ag-TLC) and gas liquid chromatography (GLC) on a polar capillary column (CP Sil 88) were used.The content of trans fatty acids in milk fat showed considerable seasonal variations. The trans 18:1 content for the pasture period was 5.73 g/100 g milk fat (MF) and only 1.83 g/100 g MF for the indoor feeding period. About one-half of trans 18:1 isomers in the samples from the indoor feeding period and more than 60% in the samples from May was trans-11 18:1 acid. The level of two trans 18:2 isomers (cis-trans and trans-cis) was about 4 times higher (0.81 g/100 g MF) in the pasture feeding period than in the indoor feeding period (0.18 g/100 g MF).
Przedmiotem badań byla zawartość kwasów tłuszczowych trans w tłuszczu mleka z okresu żywienia oborowego (marzec, kwiecień) i okresu żywienia pastwiskowego (maj). Oznaczenia przeprowadzano metodą chromatografii gazowej (GLC) na polarnej kolumnie kapilarnej (CP Sil 88) w połączeniu z metodą chromatografii cienkowarstwowej z azotanem srebra (Ag-TLC). Zawartość izomerów trans w tłuszczu mleka wykazywała znaczne wahania sezonowe. W okresie żywienia oborowego łączna zawartość izomerów trans 18:1 wynosiła 1,83 g/100 g tłuszczu, a w okresie żywienia pastwiskowego 5,73 g/100 g (tab. 1). Około 50% izomerów trans w tłuszczu z okresu żywienia oborowego i ponad 60% w tłuszczu mleka uzyskanego w maju stanowił izomer trans-11 18:1 (tab. 2). Zawartość izomerów trans kwasu 18:2 (cis-trans i trans-cis) była ponad czterokrotnie wyższa w tłuszczu mleka z okresu żywienia pastwiskowego (0,81 g/100 g tłuszczu) w porównaniu z tłuszczem mleka z okresu żywienia oborowego (tab. 4).

We carried out histochemical studies to find the localization of ginsenosides in roots of Panax quinquefolium cultivated in Poland. We performed an anatomical study on the structure and localization of secretory canals on the cross section of 4-year-old American ginseng roots. We observed the occurrence of large secretory canals, mainly in the middle part of the secondary cortex and less in the phloem layer. In our studies, moreover, we demonstrated the production of secretory canals within the periderm layer. After the anatomical study, the 4-year-old ginseng root was divided into periderm, cortex and xylem, and the ginsenosides were extracted from each part of the root. The TLC separation of ginsenosides was performed on silica gel Si60 glass plates with chloroform-methanol-ethyl acetate-water-hexane, 20+22+60+8+4 (v/v) as mobile phase. Quantitative analysis of ginsenosides was performed by using the TLC-densitometric method. Concerning the distribution of ginsenosides in the different anatomical parts of the root of Panax quinquefolium, they were contained in the periderm layer at the highest level.
W celu okreoelenia lokalizacji ginsenozydów w korzeniach żeń-szenia pięciolistnego (Panax quinquefolium L.) uprawianego w Polsce, przeprowadzono badania histochemiczne. Badania anatomiczne dotyczące budowy i rozmieszczenia kanałów wydzielniczych wykonano na przekrojach poprzecznych 4-letnich korzeni żeń-szenia. Z obserwacji mikroskopowych wynika, że największe kanały wydzielnicze są zlokalizowane w środkowej i zewnętrznej części kory wtórnej. Kanały o mniejszej średnicy położone były w obrębie floemu. Wykazano ponadto występowanie kana- łów wydzielniczych w tkankach perydermy. Do badań analitycznych korzenie żeńszenia podzielono na trzy części (perydermę, korę i floem), a następnie wyekstrahowano z nich frakcje ginsenozydów. Analizę ilościową ginsenozydów prowadzono metodą TLC na płytkach szklanych pokrytych żelem krzemionkowym Si60 przy użyciu fazy ruchomej: chloroform metanol octan etylu woda heksan, 20+22+60+8+4 (v/v). Rozwinięte płytki poddawano analizie densytometrycznej. Analiza ilościowa ginsenozydów w różnych tkankach korzeni Panax quinquefolium wykazała największą zawartość tych związków w perydermie.

Carotenoids (neoxanthin, violaxanthin, lutein and β-carotene) present in plant cell play a role in photoprotection. Industrial pollution causes oxidative stress in plants, while carotenoids react with free radicals and dissipate the excess of excitation energy. In this way carotenoids prevent the negative influence of free radicals on metabolism and can even restore some of the damages. This is confirmed by results of our analysis of the level of xanthophylls in 16 and 17-year-old trees of Scots pine (Pinus sylvestris L.). Developed from seed collected in north-eastern, the trees grow in a relatively unpolluted control site, and in a polluted site located 2 km for away from the Phosphorus Fertilizer Works. In the polluted site the environment is contaminated with SO2, NOx and F, Al, F, Pb, Cu. The needles analysed in this study were visually undamaged. Material was collected in experimental plot from 6 trees in October'98 and April'99, between 12.00 and 13.00 hours, at full sunlight. The pigments were analysed quantitatively and qualitatively by thin-layer chromatography. The paper presents results of content and distribution of neoxanthin, violaxanthin, lutein and β-carotene of Scots pine needles from a healthy control tree and a stressed tree. Marked differences in pigment levels depended on the stage of needle development and level of pollution.

Background. Mangrove forests have long been known as a source of phytochemical compounds producing various secondary metabolites. Despite the ubiquitous diversity of polyisoprenoids in the plant kingdom, few studies have focused on the distribution of polyisoprenoids in mangrove plants. The present study describes the distribution and occurrence of a new class of prenyl derivates – polyprenyl acetone as well as other polyisoprenoids in fourteen species of Indonesian mangroves, with an emphasis on chemotaxonomic importance. Material and methods. The leaves and roots of fourteen North Sumatran mangroves were analyzed using two-dimensional thin layer chromatography and electrospray ionization mass spectrometry. Results. In the leaves, the distribution of several types of polyprenyl acetones, polyprenols, and dolichols was detected and classified into types: type-I, having a predominance of dolichols over polyprenols (more than nine-fold), was observed in Acrostichum aureum (younger leaves), Avicennia alba, Av. lanata, Av. officinalis, Bruguiera parviflora, Ceriops tagal, Nypa fruticans, and Rhizophora mucronata; type-II, having the presence of both polyprenols and dolichols, was observed in Acanthus ilicifolius, Acr. aureum, B. cylindrica, and R. apiculata; type-III having a predominance of polyprenols over dolichols (more than nine-fold), was not observed in any North Sumatran mangroves; type-IV, having the presence of both polyprenyl acetones and dolichols, was observed in Aegiceras corniculatum; type-V, having the presence of polyprenyl acetones, polyprenols, and dolichols, was observed in Sonneratia caseolaris and Xylocarpus granatum. In the roots, type-I distribution was observed in Ae. corniculatum, Av. alba, Av. lanata, Av. officinalis, B. parviflora, C. tagal, N. fruticans, R. apiculata, R. mucronata, S. caseolaris, and X. granatum. Type-II distribution was observed in Ac. ilicifolius, Acr. aureum, and B. cylindrica. Type-III, -IV, and -V distributions were not observed in mangrove roots. Cluster analysis demonstrated that polyisoprenoid patterns in the leaves and roots form distinct separation into appropriate genera and tribe, suggesting that mangrove polyisoprenoids are chemotaxonomically significant. Conclusions. The major polyisoprenoid alcohols in Indonesian mangroves were found to be dolichols rather than polyprenols. The diversity of polyisoprenoids in both leaves and roots of mangroves may provide chemotaxonomic marker. The discovery of a new class of polyprenyl acetone is the first report from mangrove plants.
