Opis:
The discovery of conifer somatic embryogenesis (SE) and the subsequent development of SE protocols for a range of genera and species have opened new research opportunities to forest biotechnologists and a means towards mass clonal propagation for the forest industry. This paper provides a general description of the conifer SE process, followed by a review of protocols developed specifically for several conifer species in the Pinaceae family for which production of somatic trees has been demonstrated. Additionally, future research needs, including approaches for developing markers to optimize SE process and the production of high quality embryos, are discussed.

Opis:
Somatic embryogenesis (SE) of conifers for clonal propagation has emerged from its earliest beginnings in 1980 to become an integral component of tree improvement strategies. With its capacity for long-term germplasm preservation and scale-up technologies,it is seen to be the preferred avenue to accelerate selection and operational deployment of value-added genotypes. At present,programs for numerous species from the Pinaceae family are underway worldwide,with activities ranging from selection trials to pilot production for plantation forestry. This paper will provide a review of current efforts in conifer SE including cryopreservation,commercialization and deployment strategies,and transgenics. A discussion of challenges and issues is directed at genetic fidelity, intellectual property and future needs.

Opis:
Somatic embryogenesis is the latest, and potentially the most efficient, method for the vegetative micropropagation of plants. Over the past three decades, numerous laboratory studies have investigated somatic embryogenesis of forest trees, yielding positive results for a number of economically important tree species. The first test trials were run and plantations were planted with interior spruce in the 90s by CellFor Inc. (Canada). However, at the beginning of the XXI century, the program to produce spruce and Douglas fir somatic seedlings was stopped for economic reasons. Thus, currently no operational program is ongoing except on a small scale in New Brunswick. In order to integrate somatic embryogenesis technology into operational reforestation programs, the production costs of forest tree somatic seedlings needs to be reduced, and the awareness of foresters and forest landowners that the material obtained through somatic embryogenesis is valuable needs to be increased. This awareness would enable implementation of this technology on a large scale for production and forest management throughout Europe including Poland. In this review, the importance of somatic embryogenesis in scientific research and in global and European forestry is presented. Our main aims are to provide basic information on the challenges in researching somatic embryogenesis of forest trees and to raise interest in this tree propagation technique in both scientists and foresters.

Many chrysanthemum mutants are chimeras built from tissues of a varied genetic composition. Regeneration in vitro of somatic embryos from the whole mutated ligulate florets or only from their small mutated fragments can lead to the separation of chimera components and, as a result, producing a new original cultivar. There was determined the effect of growth regulators (2,4-D; KIN or BAP) and explant type (whole or transversely-cut-into-half ligulate florets) on the efficiency of somatic embryogenesis of Chrysanthemum × grandiflorum (Ramat.) Kitam. ‘Cool Time’. For the induction of the somatic embryogenesis ant the regeneration of somatic embryos the MS medium [Murashige and Skoog 1962] with 18.08 μM 2,4-D as well as with this auxin and 4.44; 8.88; 22.20 μM BAP or 4.65; 9.30; 23.25 μM KIN was used. The best results were obtained when transversely-cut-into-half ligulate florets were inoculated onto the medium with 4.65 μM KIN and 18.08 μM 2,4-D. This results may increase the probability of success in separation of chimera components in chrysanthemum breeding.
Wiele mutantów chryzantem jest chimerami zbudowanymi z tkanek o różnym składzie genetycznym. Regeneracja in vitro zarodków somatycznych z całych objętych mutacją kwiatów języczkowatych lub tylko z ich niewielkich zmutowanych fragmentów może doprowadzić do separacji komponentów składowych chimery i w rezultacie do uzyskania nowej, oryginalnej odmiany. W badaniach określono wpływ regulatorów wzrostu (2,4-D; KIN lub BAP) oraz rodzaju eksplantatu (cały lub przecięty poprzecznie na pół kwiat języczkowaty) na wydajność embriogenezy somatycznej u Chrysanthemum × grandiflorum (Ramat.) Kitam. ‘Cool Time’. Do indukcji embriogenezy somatycznej oraz do regeneracji zarodków somatycznych zastosowano pożywkę MS [Murashige and Skoog 1962] uzupełnioną jedynie 18,08 μM 2,4-D, a także tą auksyną oraz 4,44; 8,88; 22,20 μM BAP lub 4,65; 9,30; 23,25 μM KIN. Najlepsze rezultaty uzyskano, inokulując przecięte poprzecznie na pół kwiaty języczkowate na pożywkę z 4,65 μM KIN i 18,08 μM 2,4-D. Uzyskane wyniki mogą przyczynić się do zwiększenia prawdopodobieństwa separacji komponentów chimer w hodowli chryzantem.

Objective. The aim of the research was to verify whether intellectually disabled people are responsive to motor stimulation, and whether the specific physical exercises, besides traditional rehabilitation and occupational therapy, would result in physical fitness enhancement. Materials and methods. The research was carried out on 259 persons with moderate and significant intellectual disability, participants of occupational therapy workshops. They were divided into two groups: a control group that underwent traditional rehabilitation treatment and occupational therapy, and an experimental group that additionally performed feasible physical exercises for 10 months. Participants’ body mass and height were measured to calculate their body mass index (BMI). Physical fitness was assessed with the Eurofit Special test and additional balance tests, at the beginning and at the end of the experiment. Results. The results showed that the body mass of both men and women increased in the control group, and was reduced in the experimental group. The results of the physical fitness tests were more varied, in which the control group obtained similar results in the repeated measurements, and the experimental group significantly improved the initial results after 10 months of performing the feasible physical exercises. Conclusions. The applied physical exercises performed in the experimental group were effective because they caused body mass loss and significant improvement in physical fitness.

Embryogenic cultures of plants are exposed to various stress factors both in vitro and during cryostorage. In order to safely include the plant material obtained by somatic embryogenesis in combination with cryopreservation for breeding programs, it is necessary to monitor its genetic stability. The aim of the present study was the assessment of somaclonal variation in plant material obtained from embryogenic cultures of Picea abies (L.) Karst. and P. omorika (Pančić) Purk. maintained in vitro or stored in liquid nitrogen by the pregrowth-dehydration method. The analysis of genetic conformity with using microsatellite markers was performed on cotyledonary somatic embryos (CSE), germinating somatic embryos (GSE) and somatic seedlings (SS), obtained from tissues maintained in vitro or from recovered embryogenic tissues (ETc) and CSE obtained after cryopreservation. The analysis revealed changes in the DNA of somatic embryogenesis-derived plant material of both Picea spp. They were found in plant material from 8 out of 10 tested embryogenic lines of P. abies and in 10 out of 19 embryogenic lines of P. omorika after in vitro culture. Changes were also detected in plant material obtained after cryopreservation. Somaclonal variation was observed in ETc and CSE of P. omorika and at ETv stage of P. abies. However, most of the changes were induced at the stage of somatic embryogenesis initiation. These results confirm the need for monitoring the genetic stability of plants obtained by somatic embryogenesis and after cryopreservation for both spruce species.

Mutational witches’ broom is a part of the tree crown with abnormally dense branching and slow shoot growth, which putatively originates from a cell in a bud apical meristem and can be visually differentiated from the normal crown. The witches’ broom forms a large branching system which competes sufficiently with other parts of the tree crown. However, the competitive relationship and the interaction between the mutant and normal crown parts have not yet been studied. We investigated the patterns and dynamics of the competitive relationship between witches’ broom and normal branching systems of the same tree of five Pinaceae species by tree ring analysis. Three saw-cuts were made in each tree: on the lateral branch with the witches’ broom, on the proximal part of the axial branch in front of the branch with the witches’ broom, on the distal part of the axial branch right after the branch with the witches’ broom. An- nual radial growth was measured to the nearest 0.01 mm, and annual ring areas from three saw-cuts were analysed to compare the growth dynamics before and after witches’ broom appearance. The growth of the lateral branch with the witches’ broom sharply increased in the year of witches’ broom appearance by 2–10 times. The growth of the distal part of the axial branch sharply decreased in the same year, which could be explained only by the witches’ broom appearance. All branches with witches’ brooms gradually became thicker than the distal part of the branch, 3–150 times at the peak of growth, and occasionally outgrew even the proximal part of the branch. Thus, witches’ broom is drastically morphologically and physiologically different from the rest of the crown. All witches’ brooms had their own vertically oriented axis of symmetry and represented autonomous branching systems that were not subordinate to any other branching systems. They break the normal donor-acceptor relationship in a tree and eagerly consume resources without being fully involved in their production. The mutation clearly affects the hormonal regulation of growth and morphogenesis and turns a witches’ broom into a new metabolic sink.

The potential of immature zygotic embryos to produce embryogenic tissue was tested using culture media differed in inorganic salt composition as well as plant growth regulator content. Explants cultured on DCR as well as MLV media gave the highest initiation percentages with maximum values of 8.8% (MLV) to 10.4% (DCR). Plant growth regulators content (standard or reduced) had no profound effect on embryogenic tissue initiation. Somatic embryo maturation as well as germination was dependent on the cell line and salt composition of the medium. Plantlet regeneration occurred in three cell lines out of five tested. During the maturation process profound changes occurred in the internal organisation of somatic embryos such differentiation of root meristem in precotyledonary somatic embryos and formation of provascular strands in cotyledonary somatic embryos.

Mitochondrial DNA polymerase gamma (POLG) is the only DNA polymerase involved in maintaining the mitochondrial genome. Recent studies demonstrated an association of CAG repeat polymorphism in the second exon of POLG gene with the risk of cancer. We investigated the CAG repeat variability in the POLG gene in tumor and non-tumor tissues from colorectal cancer patients and in DNA samples isolated from blood obtained from age-matched healthy persons. Somatically occuring CAG-repeat alterations in cancer tissues have been observed in 10% of patients, but no association has been found between the CAG repeat variants in the POLG gene and colorectal cancer risk.
