One may hear that over time languages tend to simplify their grammar and notably their morphological system. This intuition, probably based on linguists’ knowledge of the rich inflectional systems of older Indo-European languages, has been challenged, particularly by sociolinguistic typologists (e.g. Trudgill 2011; Braunmuller 1984, 2003; Nichols 1992). They hypothesise that languages spoken by small and isolated communities with a dense network may complexify their grammar (Trudgill 2011: 146-147). The present article investigates the nominal inflection systems of 14 varieties of German in order to survey whether there is any such diachronic tendency towards simplification and whether instances of complexification can be observed, too. The varieties under analysis include present-day Standard German, Old High German and Middle High German (two older stages of German) and eleven present-day non-standard varieties which make part of the Alemannic dialect group. First, it will be shown that there is a diachronic tendency towards simplification if we consider the total complexity of nominal inflection. Second, however, we can identify instances of diachronic complexification too if we take a closer look at single categories. Interestingly, diachronic complexification appears only in the non-standard varieties, not so in the standard variety. This may support the hypothesis that isolated varieties are more complex than non-isolated ones.

Bone material is one of the most difficult biological samples used for extraction of DNA. The primary objective of molecular tests is to determine the concentration and quality of DNA, and these results are of significant importance in further analyses. The aim of the study was to evaluate the usefulness of a method for DNA isolation from bone material of selected animal species from Cervidae family using a commercially available GeneMatrix Bone DNA Purification Kit manufactured by Eurx. Quantitative evaluation was performed with a spectrophotometer. Pure DNA isolates were obtained from 21 out of 24 bone fragments. The efficiency of the applied isolation method varied. The difference between the lowest and highest DNA concentration was more than hundredfold. Qualitative analysis was carried out by means of electrophoresis in 1% agarose gel. No high molecular mass DNA was obtained. The genetic material was present in small quantity and it was fragmented. It was concluded that the Bone DNA Purification Kit manufactured by Eurx may be effective only in isolation of DNA from bones stored in good conditions.

Samples of crude mucin were incubated at room temperature for 48 and 96 h in a sodium azide containing buffer, pH 7.0. Then each sample was purified, reduced and alkylated with iodo[^14C]acetamide. Electrophoretic analysis demonstrated that radioactivity was incorporated into the mucin subunits and proteins of 100 and 140 kDa. The results of our experiments suggest that the released proteins can be a part of mucin molecule, cleaved by proteolysis and reduction of disulfide bridges.

In this paper a cross-shaped isolator consisting of cuboidal magnets and a cylindrical isolator are compared by resonance frequency to volume ratio and shape. Both isolators are capable of obtaining a low resonance frequency, i.e. 0.15 Hz and 0.01 Hz for the cross and cylinder, respectively. The volume of both isolators is comparable, only the shape is different, resulting in a tall structure with a small footprint for the cross and a flat with a large diameter cylindrical structure. A sensitivity analysis shows that due to the large amount of magnets, the cross-shaped isolator is less sensitive to manufacturing tolerances.

The impreciseness and conjectural character of the notions of island and continent have been the subject of many debates. Most researchers agree that unique features of an island include isolation and significant linkages between the economy and life of the inhabitants with the sea. Some characteristic features are related to spatial development and a concentration of towns and the transport network near the sea. However, Madagascar, which is regarded as an island in all kinds of classifications, fulfils only some of the above criteria. As far as the natural life is concerned, some consequences of spatial isolation can be observed, particularly the predominance of endemic and relict species in the fauna and flora, whereas in the economic and social sphere, Madagascar reveals features which are characteristic of continents. Is therefore Madagascar rightly considered an island?

The object of this study are chitinolytic enzymes produced by bacterium Clostridium paraputrificum J4 isolated from the gastrointestinal tract of a healthy human. In particular, we focus on the development of purification protocols, determination of properties of the enzymes and their activity profiles. The process of bacteria cultivation and isolation of chitinolytic complex of enzymes showing specific activities of endo-, exo-chitinase and N-acetyl-β-glucosaminidase was optimized. A range of various purification procedures were used such as ultrafiltration, precipitation, chromatographic separations (ion-exchange, size exclusion, chromatofocusing) in altered combinations. The optimal purification protocol comprises two or three steps. Individual samples were analyzed by SDS/PAGE electrophoresis and after renaturation their activity could be detected using zymograms. Mass spectroscopy peptide fragment analysis and MALDI analysis of the purest samples indicate presence of endochitinase B (molecular mass about 85 kDa) and of 60-kDa endo- and exochitinases.

The need for cost optimization in a broad sense constitutes the basis of operation of every enterprise. In the case of IT structure (which is present in almost every field of activity these days), one of the most commonly applied technologies leading to good cost-to-profit adjustment is virtualization. This allows the location of several operational systems with IT systems on a single server. In order for such an optimization to be carried out correctly, it has to be strictly controlled by means of allocating access to resources (which is known as performance isolation). Modern virtualizers allow us to set up this allocation in quantitative terms (the number of processors, size of RAM, or disc space). It appears, however, that in qualitative terms (processor time, RAM, or hard disc bandwidth), the actual allocation of resources does not always correspond with this configuration. This paper provides an experimental presentation of the achievable quality level of performance isolation of the Hyper-V virtualizer.

The paper is concerned with qualitative analysis of a non-linear model describing vibration of a vibration-isolated hand grinder. A discontinuous description of grinding forces is introduced, which accounts for the possible separation of the grinding wheel from the object during the process. Eight non-linear ordinary differential equations are obtained which describe dynamics of the system. Numerical analysis is done using methods of numerical integration and the Fast Fourier Transform. The influence of selected parameters on the character of vibration is studied and some measures are calculated which characterize the quality of the vibration isolation system.
Praca dotyczy analizy jakościowej nieliniowego modelu, opisujący drgania wibroizolowanej ręcznej szlifierki. Model opisano układem ośmiu równań różniczkowych zwyczajnych drugiego rzędu. Wprowadzono nieciągły opis sił skrawania, uwzględniający możliwość chwilowego oderwania się ściernicy od obrabianego przedmiotu. Do analizy wykorzystano procedury matematycznego całkowania skojarzone z algorytmami szybkiej transformaty Fouriera. Zbadano wpływ parametrów na charakter drgań oraz wyznaczono pewne wskaźniki jakości działania zastosowanego układu wibroizolacji.

The main element of genomic molecular analysis is isolation of DNA from biological material. DNA isolation is an important process of PCR methods. Efficency of PCR processes depends of purness and concentration of DNA. Many methods allows to izolate good quality DNA. There are many Purification Kits and methods of extraction genomic material. We show available techniques and methods of increasing efficiency of the process in our studies. Previous data shows that operating with kits needs optimization, whilst protocols do not have all information about isolation process. We concentrate on Epicentre MasterPure DNA Purification Kit, which is cheap, fast and allows to purification nucleic acids from various materials. We show detailed advantages and disadvantages of this set. Our main aim is to develop protocol to get optimal concentration (50-100 ng/μL) and pureness (A 260/280; 1.6-1.9) of DNA. We also should optimize the process of DNA isolation to obtain efficiency of about 100%. Optimized methodology of DNA isolation, with using MasterPure DNA Purification Kit allows to get good quality material in laboratory without specialistic equipment. This survey contains our assumptions of ways how we can influent on purification process according to previous data.

Naegleria belongs to the free-living amoeba family and is well-known as a human pathogen. It is recognized as etiological agent of primary amoebic meningoencephalitis involving central nervous system which always leads to death. To date, there is not a single report demonstrating Naegleria isolation and identification from environmental sources of Rawlakot, Azad Jammu and Kashmir Pakistan, and thus the aim of this study. Naegleria was isolated on non-nutrient agar plates seeded with heat killed E. coli and confirmed by morphological properties of the both stages of cyst or trophozoites. Furthermore, PCR was conducted along with direct sequencing of the PCR product for molecular identification. PCR and sequencing data verified the amplification of Naegleria sp. (07) and Vahlkampfia sp. (01) from both water and soil samples. Interestingly two species were successfully isolated and cultured on both 30 and 45°C. To the best of our knowledge this is the first report demonstrating the Naegleria isolation and molecular characterization from environmental sources of Rawlakot, Azad Jammu and Kashmir, Pakistan. The author is anxious for further evaluation of the pathogenic potential of the identified species and explores drinking water across Pakistan to investigate its quality and frequency of FLA, which might be a possible human hazard in future.

The capability of conifers for interspecific hybridization is well known. Five-needle pines from the section Quinquefoliae of the subgenus Strobus play an important role in ecosystems and have great economic significance. Interspecific hybridization that occurred in nature or under controlled pollination suggests that reproductive isolation is relative, being expressed to different degrees. Controlled pollination allows us to gather knowledge about reproductive compatibility and provides valuable material for breeding work. Artifi- cial crosses of five-needle pines began with a purely practical purpose to increase blister rust resistance and it was subsequently suggested that species crossability should be taken into account in their classification. We carried out a number of controlled pollinations using Pinus sibirica, P. koraiensis and natural P. sibirica × P. pum- ila hybrid clones as maternal trees along with pollen of 11 species and four hybrids of the five-needle pines. For the first time, seeds were obtained from the interspecific crosses P. koraiensis × P. armandii, P. koraiensis × P. monticola, P. koraiensis × P. sibirica, P. sibirica × P. ayacahuite, P. sibirica × P. armandii, and P. sibirica × P. wallichiana, as well as from some combinations with hybrids. Based on the results and available information on the crossability of the species, we concluded that complete reproductive isolation does not exist among five-needle pines, and confirmed that interspecific gene flow is common in this section.

The aim of the article is verification of possibility of use of UNEP isolation indicator for measuring isolation of tropical islands for tourism purposes. 30 tropical islands were included in the study. The correlation between intensity of tourism movement and isolation indicator by Spearman rank correlation was -0,46. More isolated islands have smaller intensity of tourism movement and less isolated have higher intensity of tourism movement ratio. But there are also exceptions from this rule - territories which are more isolated and with higher intensity of tourism movement such as Maldives and less isolated with the smaller intensity of tourism movement such as Sao Tome and Principe. UNEP isolation indicator is not ideal measure for tourism purposes.
