Opis:
Pseudomonas syringae pv. syringae (Pss) constitutes a diverse group of bacterial strains that cause canker of stone fruits, blight of cereals and red streak of sugarcane. The purpose of this study was to determine how diverse Iranian strains of Pss are when they come from different hosts. We compared a total of 32 Pss strains isolated from stone fruits, barley, wheat and sugarcane from different geographical regions of Iran based on their phenotypic and molecular properties. Strains showed some variation regarding carbon and nitrogen utilization. Pss strains were similar in their protein banding patterns. Additional bands were found in sugarcane strains. Most strains showed one indigenous plasmid DNA and a few had two and some none. The genes of syrB and syrD encoding syringomycin synthesis and secretion, respectively, were amplified using specific primers in polymerase chain reaction. Syringomycin, producing strains amplified two DNA fragments of 752 and 446 bp representing syrB and syrD genes, respectively. Primer specificity was shown for Pss using various genera. Based on the results of this study, it is suggested that Pss strains from different hosts and geographical regions show diversity in phenotypic and molecular characters. It is thought that phenotypic variation is due to adaptation to specific hosts and niches for survival and pathogenicity.

Tomato (Solanum lycopersicum L.) is attacked by Pseudomonas syringae pv. tomato causing heavy damage to the crops. The present study focused on the application of aqueous fruit extracts of neem (Azadirachta indica L.) on a single node of aseptically raised tomato plants. Observations were done, and the changes in the activity and isoenzyme profile of polyphenol oxidase (PPO) and lysozyme, both at the site of treatment as well as away from it, were noted. The results demonstrate that neem extract could significantly induce the activities of both the enzymes as well as upregulate the de novo expression of additional PPO isoenzymes. Induction of systemic acquired resistance (SAR) by natural plant extracts is a potent eco-friendly crop protection method.

Opis:
Biocontrol of plant diseases has emerged as an eco-friendly measure of plant protection and has experienced a lot of devotion in the last two decades. Biocontrol agents include application of microbial agents, their secretion products and natural extracts from different parts of several plants. The present study, therefore, aimed at evaluating the potency of aqueous extracts of Tagetes erecta L. (marigold) in controlling bacterial speck disease in tomato plants. The experimental design consisted of two groups of 50 plants each: group 1 – sprayed with sterile water (control); and group 2 – sprayed with marigold extract. Spraying was performed under aseptic conditions at the third node from the base of each plant. Challenge inoculation with the bacterial speck pathogen Pseudomonas syringae pv. tomato was performed to analyze the disease severity on the test plants. The parameters of study were analysis of alteration in the activity and gene expression of peroxidase (POX), phenyl ammonia lyase (PAL), and polyphenol oxidase (PPO), as well as isoform expression of POX and PPO. The results demonstrated strong inductive effects of the extract on the activity and genes of POX, PAL and PPO. De novo expression of POX and PPO isoforms following marigold extract treatment was also observed. The observations indicate that marigold extract could be a promising biopesticide.

The objective of the present study was to evaluate the antibacterial properties of a plant secondary metabolite - caffeine. Caffeine is present in over 100 plant species. Antibacterial activity of caffeine was examined against the following plant-pathogenic bacteria: Ralstonia solanacearum (Rsol), Clavibacter michiganesis subsp. sepedonicus (Cms), Dickeya solani (Dsol), Pectobacterium atrosepticum (Pba), Pectobacterium carotovorum subsp. carotovorum (Pcc), Pseudomonas syringae pv. tomato (Pst), and Xanthomonas campestris subsp. campestris (Xcc). MIC and MBC values ranged from 5 to 20 mM and from 43 to 100 mM, respectively. Caffeine increased the bacterial generation time of all tested species and caused changes in cell morphology. The influence of caffeine on the synthesis of DNA, RNA and proteins was investigated in cultures of plant pathogenic bacteria with labelled precursors: [3H]thymidine, [3H]uridine or 14C leucine, respectively. RNA biosynthesis was more affected than DNA or protein biosynthesis in bacterial cells treated with caffeine. Treatment of Pba with caffeine for 336 h did not induce resistance to this compound. Caffeine application reduced disease symptoms caused by Dsol on chicory leaves, potato slices, and whole potato tubers. The data presented indicate caffeine as a potential tool for the control of diseases caused by plant-pathogenic bacteria, especially under storage conditions.

Opis:
The biotechnical preparations: Biosept Active (based on a grapefruit extract) and BioZell (based on thyme oil) as well as Hypericum perforatum extract, streptomycin solution and fungicide Champion 50WP (active ingredient substance – e.i. 50% copper hydroxide) were investigated for antimicrobial effects against plant pathogenic bacteria: Agrobacterium tumefaciens, Pseudomonas syringae pv. syringae and Xanthomonas arboricola pv. corylina. The screening was carried out in vitro on three media: Nutrient Agar (NA Difco), Pseudomonas Agar F (Merck) – analogue of King B and 523. In the experiments, the agar plate method was applied. There were no statistically significant differences in the effect of streptomycin and Champion 50WP on the growth inhibition of three bacteria strains for medium 523 and Nutrient Agar and of P. syringae pv. syringae and X. arboricola pv. corylina for medium King B. It was determined that the antibacterial activity of Biosept Active and BioZell biopreparations and H. perforatum extract against Agrobacterium tumefaciens (strain C58), Pseudomonas syringae pv. syringae (strain 760) and Xanthomonas arboricola pv. corylina (strain RIPF-x13) were dependent on the strain of pathogen as well as the growth medium used. According to the research results obtained, the Biosept Active preparation and H. perforatum extract demonstrated high bacteriostatic activity against three bacterial strains grown on the Nutrient Agar medium.
