Enterocytozoon bieneusi is the most commonly identified Microsporidia in humans and has also been detected worldwide in a large group of wild living and domestic animals. The identification of E. bieneusi in wildlife has raised the question of the importance of animal reservoirs in the epidemiology of microsporidiosis and the implications of the infection with this pathogen in hosts. This review summarizes the available molecular data on the variety of E. bieneusi genotypes, both potentially zoonotic and host-specific isolated from wild living mammals and birds. In contrast to microsporidial infections of humans and domestic animals, wildlife deserves attention as a source of significant environmental reservoir of E. bieneusi.

Modern molecular techniques have greatly increased our knowledge concerning phylogenetic and functional diversity of microorganisms inhabiting the soil environment. Soil ecosys-tem is relatively complex with a high level of microbiologically diversity. The application of traditional culture-based techniques dose not reflect the total diversity of microbial community in-habiting in soil environment. On the other hand commonly used molecular methods allow for quick and accurate identification and evaluation of the genetic diversity of microorganisms in-habiting this environment. Free-living bacteria belonging to the genus Azotobacter commonly occurring in soil. Azotobacter spp. are the subject of many studies conducted both in Poland and in the world. The interest in these bacteria is largely related to their properties very useful for agriculture. Owing to their capability of fixing atmospheric nitrogen and making it available to plants and production of plant growth promotion and fungicidal substances, they are used in the production of soil bacterial inoculants. In addition, these bacteria are an excellent indicator of soil fertil-ity, which is why they are often used as test microorganisms in many studies. The paper presents an overview of molecular mi-crobiological techniques used to identify and evaluate the genetic diversity of Azotobacter spp. in studies conducted both in Poland and across the world. The ITS PCR, PCR-RFLP methods and 16S rRNA gene amplification are used to identify bacteria of the ge-nus Azotobacter, and PCR MP, RAPD PCR and ARDRA are used to assess the genetic diversity of these microorganisms.

Cultivation-based assays represent the gold standard for the assessment of virus infectivity; however, they are time-consuming and not suitable for every virus type. Pre-treatment with platinum (Pt) compounds followed by real-time PCR has been shown to discriminate between infectious and non-infectious RNA viruses. This study examined the effect of Pt and palladium (Pd) compounds on enveloped DNA viruses, paying attention to two significant pathogens of livestock – bovine herpesvirus-1 (BoHV-1) and African swine fever virus (ASFV). Native or heat-treated BoHV-1 suspension was incubated with the spectrum of Pt/Pd compounds. Bis(benzonitrile)palladium(II) dichloride (BB-PdCl 2) and dichloro(1,5-cyclooctadiene) palladium(II) (PdCl 2-COD) produced the highest differences found between native and heat- -treated viruses. Optimized pre-treatment conditions (1 mM of Pd compound, 15 min, 4°C) were applied on both virus genera and the heat inactivation profiles were assessed. A significant decrease in the detected quantity of BoHV-1 DNA and ASFV DNA after heat-treatment (60°C and 95°C) and consequent incubation with Pd compounds was observed. BB-PdCl 2 and PdCl 2-COD could help to distinguish between infectious and non-infectious enveloped DNA viruses such as BoHV-1 or ASFV.

Opis:
The structure of native α1-antitrypsin, the most abundant protease inhibitor in human plasma, is characterised primarily by a reactive loop containing the centre of proteinase inhibition, and a β-sheet composed of five strands. Mobility of the reactive loop is confined as a result of electrostatic interactions between side chains of Glu342 and Lys290, both located at the junction of the reactive loop and the β structure. The most common mutation in the protein, resulting in its inactivation, is Glu342→Lys, named the Z mutation. The main goal of this work was to investigate the influence of the Z mutation on the structure of α1-antitrypsin. Commonly used molecular modelling methods have been applied in a comparative study of two protein models: the wild type and the Z mutant. The results indicate that the Z mutation introduces local instabilities in the region of the reactive loop. Moreover, even parts of the protein located far apart from the mutation region are affected. The Z mutation causes a relative change in the total energy of about 3%. Relatively small root mean square differences between the optimised structures of the wild type and the Z mutant, together with detailed analysis of 'conformational searching' process, lead to the hypothesis that the Z mutation principally induces a change in the dynamics of α1-antitrypsin.

Conformational analysis of two tachykinin family peptides: Scyliorhinin I (ScyI) and Scyliorhinin II (ScyII) was carried out by 1D- and 2D-NMR (DQF-COSY, TOCSY, HMQC, HMBC, NOESY and ROESY) and molecular dynamics calculation methods in water and DMSO. Scyliorhinin I is an equipotent agonist of NK-1 and NK-2 tachykinin receptors and Scyliorhinin II is a selective agonist of the NK-3 tachykinin receptor. In DMSO, two groups of conformations (major and minor) were obtained for both peptides based on the experimental data. The conformations proposed for ScyI represent a folded structure, which show certain similarities to the structures reported for other NK-1 and NK-2 tachykinin agonists. In water ScyII displays a flexible, extended structure, whereas in DMSO the structure is more compact and in the fragment from the centre to the C-terminus several beta -turns may be present.

Opis:
A significant number of DNA-based techniques has been introduced into the field of microorganisms' characterization and taxonomy. These genomic fingerprinting methods were developed to detect DNA sequence polymorphisms by using general principles, such as restriction endonuclease analysis, molecular hybridization, and PCR amplification. In recent years, some alternative techniques based on ligation of oligonucleotide adapters before DNA amplification by PCR, known as Ligation-Mediated PCR methods (LM PCR), have been successfully applied for the typing of microorganisms below the species level. These molecular methods include: Amplified Fragment Length Polymorphism (AFLP), Amplification of DNA fragments Surrounding Rare Restriction Sites (ADSRRS), PCR Melting Profiles (PCR MP), Ligation Mediated PCR/Shifter (LM PCR/Shifter), Infrequent-Restriction-Site Amplification (IRS PCR), double digestion Ligation Mediated Suppression PCR (ddLMS PCR). These techniques are now applied more and more often because they involve less time, are comparably inexpensive, and require only standard lab equipment. Here, we present a general review of this group of methods showing their possibilities and limitations. We also identify questions and propose solutions which may be helpful in choosing a particular LM PCR method for the achievement of the required goal.

Introduction and objective. Conventional, culture-based methods of bacterial identification and drug-susceptibility testing are considered the gold standard in medical microbiology. In recent years, classical microbiological methods have been supplemented with modern analytical and molecular methods. The aim of the review was to discusses the methods which have been permanently adapted to bacteriological microbiological diagnostics. Abbreviated description of the state of knowledge. Currently, PCR, as well as other nucleic acid amplification tests and sequencing techniques, are part of the standard repertoire of microbiological diagnostics. With regard to the quality and speed of pathogen identification, the introduction of mass spectrometry techniques into routine microbiological diagnostics work-up has been revolutionary. Within a short time in many laboratories, Matrix-Assisted Laser Desorption/Ionisation – Time of Flight Mass Spectrometry (MALDI TOF MS) systems have almost completely replaced conventional biochemical pathogen identification. Conclusions. Microbiological diagnostics is an indispensable element of a targeted therapy. The techniques used in the laboratory depend primarily on the laboratory’s apparatus, the costs of the analysis, as well as the sensitivity and specificity of a method. However, regardless of the culture-based methods universality, advanced techniques have permanently established themselves in diagnostics. Confident information about the detected organism and treatment possibilities in a combination with the clinical context are conducive to successful therapy. Although modern methods still require validation and close collaboration between clinicians, microbiologists and bioinformaticians, these methods, once deemed to be the future, have already arrived.

In this paper we present our approaches and results concerning application of molecular modelling techniques in the design of new chemotherapeutic agents for the control of eukariotic systems, comprising compounds for the treatment of systemic fungal infections and tumor deseases. In the case of anti-tumor agents we focused our attention on molecular properties of natural and synthetic anthraquinones. In the area of antifungal compounds we adopted two approaches. In one of them we examine molecular nature of undesirable properties of polyene macrolide antifungal antibiotic - amphotericin B using molecular modelling techniques. Another approach was aimed at the development of selective inactivator of glucosamine synthase, a novel target for antifungal compounds. In this problem we have used computational chemistry methods to identify structural features responsible for the selective inactivation of the target enzyme.

Opis:
The nuclear magnetic resonance relaxation times of solvent water nuclei are known to decrease upon addition of diamagnetic solute protein. For this reason NMR relaxation methods are able to provide information on molecular dynamics changes of water protons and their interaction with macromolecules' surfaces. We present results of measurements of relaxation rates $R_1$ = 1/$T_1$, $R_2$ = 1/$T_2$ and $R_{1ρ}$ = 1/$T_{1ρ}$ in the rotating frame for three proteins: chicken egg white lysozyme, egg white albumin, and bovine serum albumin, obtained at proton resonant frequency of 60 MHz. Besides the relaxation rates dependences on concentration in the 4-23% (g/100 g solution) range, the analysis of the Carr-Purcell-Meiboom-Gill CPMG multi-echo $T_2$ experiments with variable pulse rate τ was performed. The dependences of relaxation rates on protein concentration are linear at low concentration. When protein concentration increases the slope of the straight line rapidly changes at so-called "critical" concentration which depends on MW of the diluted protein. Investigated dispersion of $T_2$, obtained using the CPMG method with a variable pulse rate, for concentrations higher and lower than the "critical" one, exhibits unequal behavior. At high concentration one-exponential curves and at low concentration two-exponential curves correspond closely with experimental data. The obtained parameters of exponents allow an estimation of the ratio of the amount of water with the determined motion freedom, that is free and bounded water, in solution. We showed that the CPMG dispersion method applied to aqueous protein solutions may widen the current understanding of the nature of molecular dynamics of hydrated water protons in non-perturbed environment.
