The aim of the present study was: – to compare methods for concentration and isolation of Legionella DNA from water; – to examine the efficacy of various modifications of PCR test (PCR, semi-nested PCR, and real-time PCR) for the detection of known numbers of Legionella pneumophila in water samples artificially contaminated with the strain of this bacterium and in randomly selected samples of environmental water, in parallel with examination by culture. It was found that filtration is much more effective than centrifugation for the concentration of DNA in water samples, and that the Qiamp DNA Mini-Kit is the most efficient for isolation of Legionella DNA from water. The semi-nested PCR and real-time PCR proved to be the most sensitive methods for detection of Legionella DNA in water samples. Both PCR modifications showed a high correlation with recovery of Legionella by culture (p<0.01), while no correlation occurred between the results of one-stage PCR and culture (p>0.1).

A total of 123 water samples were examined in parallel by culture and semi-nested PCR for the presence of Legionella. They comprised: 35 samples of hot water distributed by the urban municipal water supply system (MWSS) taken in institutions, 45 samples of hot water distributed by urban MWSS taken in dwellings, 27 samples of cold water distributed by rural MWSS taken in dwellings, and 16 samples of cold well water taken in rural areas. The greatest frequency of the isolation of Legionella by culture (88.6%) was recorded in the samples of hot water from the urban institutions, having been greater compared to all other sources (p<0.001). The frequency of Legionella isolation from hot water in urban dwellings (28.9%) was significantly greater compared to the combined value (2.3%) for cold water from rural MWSS and wells (p<0.001). Strains belonging to Legionella pneumophila serogroups 2-14 predominated in the examined samples, while strains of L. pneumophila serogroup 1 and strains of Legionella spp. (other than L. pneumophila) were 3-fold less numerous. The rates of positive findings in the semi-nested PCR (stage 2) were greater than culture isolations in all kinds of samples, except for urban institutions. The correlation between the culture and PCR results was positive for samples of hot water from urban MWSS (p<0.01), but not for samples of cold water from rural MWSS and wells (p>0.5). A significant correlation was found between rates of PCRpositive results and numbers of Legionella pneumophila serogroups 2-14 strains, but not for other Legionella serogroups or species. In conclusion, our results support the opinion that though PCR cannot be a substitute for the isolation of Legionella by culture, it could be regarded as an useful complementary method.

A total of 190 Dermacentor reticulatus ticks (80 males, 110 females) collected on the territory of Ostrów Lubelski, Suchawa, Zalutyń and Kazimierz Dolny (Lublin Province, eastern Poland) were examined by reverse transcription PCR and nested PCR methods for the presence of hantavirus RNA. None of the examined Dermacentor reticulatus specimens showed the presence of the hantavirus-specific RNA in spite of using two pairs of primers and the clearly positive results obtained with the positive control. Thus, the hypothesis about the possible participation of ticks in the transmission of hantaviruses was not confirmed.

The main aim of the study was to determine the risk of two selected zoonosis infections caused by Giardia duodenalis and Cryptosporidium spp. among an occupational group of veterinarians in Poland. Two hundred and ninety-seven samples of stool were tested for the presence of Giardia cysts and Cryptosporidium oocysts using Direct Fluorescent Assay (DFA). There were no positive results for Cryptosporidium. The presence of Giardia cysts was found in two samples of faeces (0.67%). The risk with regard to the parasites Giardia duodenalis and Cryptosporidium spp. seems to be low among the group of veterinarians.

Chorobami odkleszczowymi nazywamy choroby zakaźne przenoszone za pośrednictwem drobnych pajęczaków, zwanych kleszczami (lxodida). W Polsce są one wywoływane przez co najmniej 5 gatunków (lub grup gatunków) mikroorganizmów (wirusów, bakterii, pierwotniaków). Spośród nich największe znaczenie ma krętek Borrelia burgdorferi wywołujący boreliozę z Lyme, który jest częstą przyczyną chorób zawodowych u leśników i rolników. Główne kierunki profilaktyki chorób odkleszczowych obejmują: szczepienia ochronne przeciwko kleszczowemu zapaleniu mózgu i opon mózgowo-rdzeniowych, unikanie spożywania surowego mleka, stosowanie odpowiedniej odzieży ochronnej podczas pracy w lesie lub na obrzeżach lasu, przegląd ciała i wzięcie natrysku po powrocie z pracy w lesie, szybkie i umiejętne usuwanie kleszczy przyssanych do ciała, stosowanie repelentów, edukację zdrowotną, opiekę medyczną nad zagrożonymi pracownikami leśnictwa i rolnictwa, działania prewencyjne ze strony pionu bhp lasów państwowych, redukcję liczebności kleszczy w środowisku naturalnym.
Tick-borne diseases are infectious diseases transmitted by small arachnids called ticks (lxodida). In Poland, these diseases are caused by at least five species (or groups of species) of microorganisms (viruses, bacteria. protozoa). Of which, the most important is the spirochete Borrelia burgdorferi causing Lyme borreliosis, which is the frequent cause of occupational diseases in the forestry and agricultural workers. The main directions of the prevention of tick-borne diseases include: vaccination against tick-borne encephalitis, avoiding of the consumption of raw milk, use of the proper clothing during work in the forest, inspection of the body and taking shower after coming from forest. quick and proper removing of ticks adhered to body, use of repelltents, health education, providing medical care for endangered forestry and agricultural workers, preventive actions pursued by the management of the forest service, reduction of the tick number of natural environment.

Opis:
Objective. Determination of the concentration and species composition of filamentous fungi in root vegetables (carrots and red beetroots), and the adjacent rhizosphere soil, collected on organic and conventional farms in Eastern Poland. Materials and method. During the period 2015–2016, a total number of 80 samples of vegetables and 40 samples of soil were examined. From each type of farm, 20 samples of vegetables and 20 samples of the adjacent soil were examined. In addition, the study included 20 samples of vegetables from organic farms and 20 samples of vegetables from conventional farms purchased on the markets in the city of Lublin in Eastern Poland. In order to determine the concentration and species composition of filamentous fungi, both in vegetables and soil, the method of plate dilutions on Malt Agar medium (Difco) with chloramphenicol was used. The study was conducted in two parallel repetitions. Inoculated media were incubated at the temperature of 30° C for 72 hours, then at room temperature for 72 hours. The species composition of fungal flora was determined using macroscopic and microscopic methods, with the help of keys and atlases. Results. It was found that the mean concentration of fungi was higher in vegetables and soil from conventional farms than in those from organic farms. In the case of carrots and soil from conventional farms, this concentration was 4.93 and 5.10 log10 CFU g-1, respectively, whereas from organic farms – respectively, 3.81 and 4.20 log10 CFU g-1. In the case of beetroots and soil from conventional farms, the mean concentrations were also higher compared to organic farms – 5.09 vs. 3.93 and 4.95 vs. 4.23 log10 CFU g-1. In the examined vegetables and rhizosphere soil, 61 species of filamentous fungi were found, of which 12 belonged to the genus Penicillium, 4 to the genus Fusarium, and 2 species each to the genera: Aspergillus, Cladosporium and Trichoderma. As many as 28 (45.9%) fungal species that occurred in vegetables and soil are regarded as pathogenic for humans.

Introduction. Mortality due to various causes, despite continuous efforts to improve the quality of medical services, is a serious problem for modern healthcare systems. Ischaemic heart disease and stroke are the cause of over 15 million deaths annually, and are therefore known as the world’s number one killer. Objective. The aim of this study is to characterise the missions and the most common reasons for dispatching Helicopter Emergency Medical Service (HEMS) crews, with special emphasis on the differences between urban and rural areas. Materials and method. The study was conducted using a retrospective analysis of HEMS missions, including flights to accidents and diseases carried out by HEMS crews in Poland from January 2014 – December 2018. The final analysis included 35,213 cases of HEMS missions. Results. The study group consisted mainly of male patients (66.40%), aged 50–64 (22.06%), mean age of the entire analysed group – 47.71 (SD: 25.96). The main reason for HEMS missions were strokes (21.63%). Analysis of patients’ clinical status revealed that the clinical status of patients treated in rural areas was more severe, which was indicated by the Glasgow Coma Scale – GCS (12.03 vs 12.35) and the Revised Trauma Scale – RTS (10.14 vs 10.60) scores. When assessed by the National Advisory Committee for Aeronautics (NACA) score, body injuries and fatal diseases were observed more often in patients in rural areas (NACA 7 6.12% vs 3.46%) (p<0.05). Conclusions. Multi-organ injuries, head traumas, sudden cardiac arrest, traumatic brain injuries, collapse and epilepsy, were more frequent reasons for HEMS missions in rural areas than in urban areas.
