African swine fever is an acute, febrile and highly virulent porcine disease causing serious economic losses worldwide. The pK205R protein of the African swine fever virus (ASFV) is largely expressed in the early stages of infection, which has given the K205R gene extensive attention. In this study, the ASFV K205R was cloned and expressed in Escherichia coli BL21 (DE3). Expression of histidine-tagged pK205R with a molecular mass of 44 kDa was determined by 12% sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot analysis. Optimisation of culture conditions allowed induction of the recombinant protein with 0.4 mM Isopropyl β-D-thiogalactoside (IPTG) at 37oC for 2 h. The protein existed in cellular supernatant and was purified using a Ni-NTA resin column. The purified protein was used to immunize rabbits four times to enable the production of polyclonal antibodies, and the antiserum titre was detected by ELISA. The results showed that the purified pK205R can react with ASFV positive serum specifically by Western blotting. The pK205R had high antigenicity, which indicated that pK205R could be used as an antigen for detection of ASFV-specific antibodies in ELISA testing, and the recombinant protein could contribute to further research of the action and structure of pK205R.

Opis:
For one kind of high current diodes composed of a ceramic-metal welding vacuum interface, the electrical design was presented. For compactness, a radial type insulator and a cone-column anode crust were adopted. The shielding methods around cathode and anode region were applied to mitigate the influence of welding solder to vacuum flashover. Finite Element Analysis (FEA) simulation results indicated that by adjusting the anode outline and shielding shape, the electric fields along the ceramic were well distributed. High voltage test was conducted on a long-pulse accelerator and experimental results confirm the theoretic design: the diode can stably hold on 400 kV and 200 ns voltage pulse.

Opis:
Jarosite residues, which are generated in a zinc production plant by a hydrometallurgical process, contain a large amount of valuable metal components. In this study, a method was proposed for the recovery of lead, zinc and iron from the residues and simultaneous sulfur fixation through direct reduction followed by magnetic separation. The influences of the roasting temperature, roasting time and the concentration of SO2 gas in the direct reduction process were researched in detail. Results showed that the volatilization rates of lead, zinc and sulfur were 96.97%, 99.89% and 1.09%, respectively, and the iron metallization rate was 91.97% under optimal reduction conditions; roasting temperature 1523 K for 60 min. The magnetic concentrate with the iron content of 90.59% and recovery rate of 50.87% was obtained from the optimal reduction product by grinding and magnetic separation. The optimum fineness for separation 96.56% less than 37 μm accounted with magnetic field strength 24 kA/m. The theoretical analysis was carried out by thermodynamics, X-ray powder diffraction, gas analysis and scanning electron microscopy.
