Introduction and objective. The aim of the study was to compare the effectiveness of selected oocysts concentration methods, DNA extraction protocols and PCR assays targeting the B1 gene, for the development of procedures which would be effective and useful in laboratory practice for the detection of T. gondii in faecal samples from cats. Materials and method. In order to compare the influence of the flotation fluids on microscopy and PCR detection of T. gondii, saturated solutions of saccharose, MgSO4, ZnSO4 and NaNO3 were used. To determine the sensitivity of PCR tests used: Real time PCR (RT) and nested PCR, water samples spiked with T. gondii tachyzoites and oocysts were tested. DNA was extracted using DNeasy Blood and Tissue Kit (Qiagen) (K1). The same PCR tests were used to assess the efficacy of T. gondii DNA detection in samples of cat faeces spiked with oocysts, using DNA extraction by a K1 set and a K2 set (QIamp DNA Stool Mini Kit, (Qiagen). Results. The initial results showed that NaNO3 was most useful as a flotation fluid due to the lack negative effect on the oocysts and amplification efficacy in PCR. The level of detection for water samples (100 μl) was determined as 100 tachyzoites and 1–50 oocysts in RT, and 2–20 oocysts in nested PCR. The limit of detection (LD) for stool samples (250 mg) spiked with oocysts, where the K1 set was used, determined as 250 and 5 oocysts in RT and nested PCR, respectively. For samples extracted with the K2 set, LD in RT was determined as 1–50 oocysts (depending on the variant) and 50 oocysts in nested PCR. Conclusions. The most effective methods for detection of T. gondii in cat faeces seem to be centrifugal flotation with NaNO3, followed by DNA extraction with removing of inhibitors (K2 set) and Real Time PCR targeting B1 gene.

A total of 190 Dermacentor reticulatus ticks (80 males, 110 females) collected on the territory of Ostrów Lubelski, Suchawa, Zalutyń and Kazimierz Dolny (Lublin Province, eastern Poland) were examined by reverse transcription PCR and nested PCR methods for the presence of hantavirus RNA. None of the examined Dermacentor reticulatus specimens showed the presence of the hantavirus-specific RNA in spite of using two pairs of primers and the clearly positive results obtained with the positive control. Thus, the hypothesis about the possible participation of ticks in the transmission of hantaviruses was not confirmed.

The main aim of the study was to determine the risk of two selected zoonosis infections caused by Giardia duodenalis and Cryptosporidium spp. among an occupational group of veterinarians in Poland. Two hundred and ninety-seven samples of stool were tested for the presence of Giardia cysts and Cryptosporidium oocysts using Direct Fluorescent Assay (DFA). There were no positive results for Cryptosporidium. The presence of Giardia cysts was found in two samples of faeces (0.67%). The risk with regard to the parasites Giardia duodenalis and Cryptosporidium spp. seems to be low among the group of veterinarians.

A total of 123 water samples were examined in parallel by culture and semi-nested PCR for the presence of Legionella. They comprised: 35 samples of hot water distributed by the urban municipal water supply system (MWSS) taken in institutions, 45 samples of hot water distributed by urban MWSS taken in dwellings, 27 samples of cold water distributed by rural MWSS taken in dwellings, and 16 samples of cold well water taken in rural areas. The greatest frequency of the isolation of Legionella by culture (88.6%) was recorded in the samples of hot water from the urban institutions, having been greater compared to all other sources (p<0.001). The frequency of Legionella isolation from hot water in urban dwellings (28.9%) was significantly greater compared to the combined value (2.3%) for cold water from rural MWSS and wells (p<0.001). Strains belonging to Legionella pneumophila serogroups 2-14 predominated in the examined samples, while strains of L. pneumophila serogroup 1 and strains of Legionella spp. (other than L. pneumophila) were 3-fold less numerous. The rates of positive findings in the semi-nested PCR (stage 2) were greater than culture isolations in all kinds of samples, except for urban institutions. The correlation between the culture and PCR results was positive for samples of hot water from urban MWSS (p<0.01), but not for samples of cold water from rural MWSS and wells (p>0.5). A significant correlation was found between rates of PCRpositive results and numbers of Legionella pneumophila serogroups 2-14 strains, but not for other Legionella serogroups or species. In conclusion, our results support the opinion that though PCR cannot be a substitute for the isolation of Legionella by culture, it could be regarded as an useful complementary method.

The aim of the present study was: – to compare methods for concentration and isolation of Legionella DNA from water; – to examine the efficacy of various modifications of PCR test (PCR, semi-nested PCR, and real-time PCR) for the detection of known numbers of Legionella pneumophila in water samples artificially contaminated with the strain of this bacterium and in randomly selected samples of environmental water, in parallel with examination by culture. It was found that filtration is much more effective than centrifugation for the concentration of DNA in water samples, and that the Qiamp DNA Mini-Kit is the most efficient for isolation of Legionella DNA from water. The semi-nested PCR and real-time PCR proved to be the most sensitive methods for detection of Legionella DNA in water samples. Both PCR modifications showed a high correlation with recovery of Legionella by culture (p<0.01), while no correlation occurred between the results of one-stage PCR and culture (p>0.1).

Opis:
Objective. An assessment of the prevalence of Toxoplasma gondii infection in cats from southwestern Poland using serology, coproscopy and PCR methods. Materials and method. In total, 208 cats (139 females and 68 males), aged 0.5–12 years (mean=2.6) from 25 localities in southwestern Poland were examined by indirect immunofluorescence assay (IFAT) to estimate the T. gondii serological status. Faecal samples of 41 cats were examined for the presence of oocysts/DNA T. gondii by microscopy and Real-time/nested PCR. After flotation (with NaNO3), pellets from faecal samples were disrupted by 10 cycles of freezing (liquid nitrogen) and warming. DNA was extracted using QIamp DNA Stool Mini Kit (Qiagen), according to the manufacturer’s instruction. Results. The positive results in IFAT for anti-T. gondii IgG and IgM antibodies were found in 143 of 208 tested cats (68.8%). Among positive results, 14.5%, 34.1% and 51.4% were detected in titre ranges 128–512, 1,000–2,000 and ≥ 4,000, respectively. In 23.1% of cat sera anti- T. gondii IgM antibodies were found. The prevalence of anti-Toxoplasma antibodies was significantly greater in older cats (>1 year) (83.5%) than in younger cats (48.3%) (P<0.05), in females (74.1%) than in males (58.8%) (P<0.05), and in cats kept outdoors than indoors (69.7% vs. 16.7%) (P<0.01). Among the 41 faecal samples examined, the presence of structures resembling T. gondii oocysts was found in 2 samples (4.9%), and for one of these samples (2.4% of the total) the result was also confirmed by PCR. Conclusions. T. gondii infection in domestic cats is highly prevalent in southwestern Poland. Information on the prevalence of infection in cats can be useful for assessing T. gondii environmental contamination and the risk for public health.

Opis:
A total of 684 Ixodes ricinus ticks (321 nymphs, 184 males, and 179 females) were collected by fl agging lower vegetation in 6 forest districts located on the territory of Lublin province (eastern Poland). Ticks were examined by polymerase chain reaction (PCR) method for the presence of Anaplasma phagocytophilum DNA with two pairs of primers: EHR521/EHR747 for detecting 16S rRNA gene, and LA6/LA1 for detecting ankA gene. To study the relationship between infection in ticks and people occupationally exposed to tick bite, blood serum samples of 261 forestry workers employed in the same forest districts were examined by immunofl uorescence method for the presence of specifi c antibodies against A. phagocytophilum. A total of 70 ticks out of 684 examined (10.2%) showed the presence of A. phagocytophilum 16S rRNA gene. The prevalence of infection was signifi cantly dependent on tick’s stage (χ2=49.2, p<0.00001) and geographical locality (χ2=34.4, p<0.00001). The percentage of I. ricinus females infected with A. phagocytophilum (24.6%) was signifi cantly greater compared to males (6.5%) and nymphs (4.4%) (p<0.00001). Only 19 ticks out of 684 examined (2.8%) showed the presence of A. phagocytophilum ankA gene, signifi cantly less compared to 16S rRNA gene (p<0.00001). The prevalence of infection demonstrated by the presence of ankA gene was also signifi cantly dependent on tick’s stage (χ2=23.6, p<0.00001) but not on locality (χ2=9.8, p=0.082). A signifi cant correlation was found between the presence of A. phagocytophilum 16S rRNA gene in I. ricinus female ticks from the particular forest districts and the serologic response to A. phagocytophilum of forestry workers employed in the same districts (p<0.05). No signifi cant correlation was found between the presence of A. phagocytophilum ankA gene in I. ricinus ticks and serologic response of exposed workers. In conclusion, detection of A. phagocytophilum infection in ticks by PCR with the use of EHR521/EHR747 primers detecting 16S rRNA gene is signifi cantly more sensitive compared to LA6/LA1 primers detecting ankA gene.
