Nowadays nanostructures are more and more often designed as carriers for drug delivery, especially to improve the drug pharmacokinetics and pharmaco-dynamics. Numerous kinds of nanostructures are considered a good prospect for medical applications thanks to their small size, acceptable biocompatibility and toxicity. Due to the fact that nanotechnology is a new field of science, every nano-scale product must be thoroughly examined regarding its toxicity to the human body. This study provides new insights into effects of exposing endothelial cells to the selected nanostructures. Dendrimers of the fourth generation (PAMAMs), multi-walled carbon nanotubes (MWCNTs) and silver nanoparticles (SNPs) were used to evaluate nanostructures influence on endothelial cells in vitro. The nanostructures were evaluated via transmission electron microscopy and dynamic light scattering technique. The cells previously exposed to the nanostructures were observed and analyzed via the atomic force microscopy and scanning electron microscopy to obtain a quantitative evaluation of the cells morphology. The presence of multi-walled carbon nanotubes and silver nanoparticles on the cells surface was confirmed by the scanning electron microscopy. Our results confirm that the surface association and/or uptake of nanostructures by the cells resulting from physicochemical and biological processes, affect the cells morphology. Morphological changes can be induced by the membrane proteins interaction with nanomaterials, which trigger a sequence of intracel-lular biological processes.

The cracking of materials and fracture surface is of great practical and academic importance. Over the last few years the development of the fractography of crystalline alloys resulted in a useful tool for the prediction or failure analysis. Many attempts have been made to observe cracks using optical microscopy, X-ray topography and transmission electron microscopy (TEM). Of these techniques, the resolution of optical microscopy and X-ray topography is too poor. By contrast, the resolution of TEM is high enough for detailed information to be obtained. However, in order to apply TEM observations, a thin foil specimen must be prepared, and it is usually extremely difficult to prepare such a specimen from a pre-selected region containing a crack. In the present work, deformation mechanisms fracture surfaces of Ni-based metallic glass samples have been studied by specially designed experiments. In order to study the deformation mechanisms and fracture the Ni-based metallic glasses have been investigated in the tensile test. The structure and fracture surfaces after the decohesion process in tensile tests were observed using transmission electron microscope (TEM) and scanning electron microscope (SEM), respectively. The studies of structure were performed on thin foils. Moreover the investigated tape was subjected to a banding test. Then, the tape was straightened and the thin foil from the area of maximum strain was prepared. This thin foil sample was deformed before the TEM investigation to obtain local tears.

Mg60 Zn35 Ca5 amorphous powder alloys were synthesized by mechanical alloying (MA) technique. The results of the influence of high-energy ball-milling time on amorphization of the Mg60 Zn35 Ca5 elemental blend (intended for biomedical application) were presented in the study. The amorphization process was investigated by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM). Initial elemental powders were mechanically alloyed in a Spex 8000 high-energy ball mill at different milling times (from 3 to 24 h). Observation of the powder morphology after various stages of milling leads to the conclusion that with the increase of the milling time the size of the powder particles as well as the degree of aggregation change. The partially amorphous powders were obtained in the Mg60 Zn35 Ca5 alloy after milling for 13-18h. The results indicate that this technique is a powerful process for preparing Mg60 Zn35 Ca5 alloys with amorphous and nanocrystalline structure.

Mucopolysaccharidoses (MPS) are severe inherited metabolic disorders caused by storage of glycosaminoglycans (GAGs). The level of accumulated GAGs is an important parameter in assessment of the severity of the disease and the efficacy of treatment; unfortunately, biochemical methods are often unreliable and only semi-quantitative. Therefore, finding other methods for estimation of GAG levels and/or assessment of the efficacy of applied therapy is very important. Although monitoring of GAG levels during therapy is crucial, in this work it is proposed that analysis of the ultrastructure of MPS cells by electron microscopic techniques can be considered as an alternative and reliable method for assessment of lysosomal storage. The number of complex lysosomal structures was found to be significantly higher in MPS cells relative to controls, while it decreased significantly in response to either enzyme replacement therapy or substrate reduction therapy. Thus, this parameter, easily assessed by electron microscopy, appears to correspond to the physiological state of MPS cells.

Two coal samples from the Sośnica coal mine, Poland, were analysed in this study. One sample was the natural char collected at the contact of magmatic intrusion, and another sample was the raw coal that was pyrolised in a laboratory furnace. Temperature of pyrolysis was similar to that calculated for the intrusion. The obtained char was analysed to compare its features with those characterizing a natural char sample. Optical microscopy, transmission electron microscopy and Raman spectroscopy studies show that the char from unaltered coal is characterized by the least developed structure, texture and microtexture compared to the natural char. Hence, it can be concluded that geological pressure generated by both the intrusion and the overburden, strongly affects the process of molecular ordering that took place during the heating of coal. The textural, structural or microtextural parameters of coal cannot be used as a geo-thermometer, because they are strongly dependent not only on the temperature but also on other factors.

Detailed studies on the effects of pulsed laser interference heating on surface characteristics and subsurface microstructure of amorphous Fe80Si11B9 alloy are reported. Laser interference heating, with relatively low pulsed laser energy (90 and 120 mJ), but with a variable number (from 50-500) of consecutive laser pulses permitted to get energy accumulation in heated areas. Such treatment allowed to form two- Dimensional micro-islands of laser-affected material periodically distributed in amorphous matrix. The crystallization process of amorphous FeSiB ribbons was studied by means of scanning and transmission electron microscopy. Detailed microstructural examination showed that the use of laser beam, resulted in development of nanostructure in the heated areas of the amorphous ribbon. The generation of nanocrystalline seed islands created by pulsed laser interference was observed. This key result may evidently give new knowledge concerning the differences in microstructure formed during the conventional and lased induced crystallization the amorphous alloys. Further experiments are needed to clarify the effect of pulsed laser interference crystallization on magnetic properties of these alloys.

The structure near the interface of bimetallic strips strongly influences their properties. In this work, the interfacial layers of explosively welded aluminium and copper plates were investigated by means of a scanning electron microscope (SEM), equipped with a high resolution system for local orientation measurements (SEMFEG/EBSD), and a transmission electron microscope (TEM), equipped with energy dispersive spectrometry (EDX) for the analysis of chemical composition changes. The SEMFEG/EBSD-based local orientation measurements in the areas close to the interface, in both sheets, revealed fine-grained layers characterized by the clearly marked tendency of the copper-type rolling texture formation. The texture was described by an increased density of the orientations near the f112g<111>, f123g<634>and f110g<112>positions. The internal microstructure of the intermetallic inclusion is mostly composed of dendrites. The electron diffractions and the TEM/EDX chemical composition measurements in the intermetallic inclusions revealed only crystalline phases, both equilibrium and 'metastable'. Additionally, no significant regularity in the phase distribution with respect to the parent sheets was observed.
Mikrostruktura w pobliżu strefy połączenia silnie wpływa na własności układów bimetalowych. W niniejszej pracy analizie poddano układy warstwowe Al/Cu wytworzone technologia zgrzewania wybuchowego. Badania prowadzono z wykorzystaniem skaningowej mikroskopii elektronowej wyposażonej w wysokorozdzielczy system pomiaru orientacji lokalnych oraz transmisyjnej mikroskopii elektronowej wyposażonej w analizator składu chemicznego. Systematyczne pomiary orientacji lokalnych w pobliżu strefy połączenia, w obydwu łączonych płytach, ujawniają formowanie się strefy silnie rozdrobnionych ziaren. Strefa ta scharakteryzowana jest przez formowanie się tekstury walcowania typu miedzi z dobrze uwidocznionymi składowymi zbliżonymi do położeń: f112g<111>, f123g<634> oraz f110g<112>. Wewnętrzna struktura 'intermetalicznych inkluzji' zbudowana jest z dendrytów. Badania z wykorzystaniem elektronowej mikroskopii transmisyjnej dokumentują, ze wewnątrz strefy przetopień następuje formowanie się faz krystalicznych, zarówno równowagowych jak i 'metastabilnych'. Skład chemiczny formujących się faz nie wykazywał związku z położeniem ziaren względem płyt bazowych.

The present study deals with the application of High Resolution Transmission Electron Microscopy (HRTEM) to dark layers, occurring in the speleothems of Domica Cave (Slovakia). Chemical pre-treatment was necessary for sample purification and the effective extraction of carbon soot. For purposes of comparison, soot aggregates obtained from laboratory experiments on the combustion of beech wood and collected from a diesel engine also were studied. HRTEM analyses of combustion products permit a distinction to be made between soot aggregates that originated in different combustion processes. The diameter of spherical, primary particles depends on the conditions of combustion, notably temperature. Burning in diesel engines produces soot with relatively small, primary particles (diameter dp = 34 ± 4 nm). Primary, spherical particles of soot aggregates, obtained from the combustion of beech wood, were larger (diameter dp = 42 ± 5 nm). The diameters of primary particles of soot separated from Domica flowstones (samples DOM1 and DOM2) were similar to the wood samples (dp = 50 ± 9 nm). Another type of carbonaceous particle, obtained in the combustion process, had a spherical shape, but the diameter of about 50–500 nm was significantly larger than that of soot. Analyses performed on two samples (DOM S1 and DOM S2) confirmed that the black laminae owed their colour to particles, formed during wood combustion and later retained in the speleothems.

Carbon micro/nanofibers prepared by catalytic chemical vapor deposition have been characterized in the form of powders and in the form of filaments, intercorporated in the matrix of ZrO2. Scanning electron microscopy, transmission electron microscopy, high resolution electron microscopy, electron spectroscopy for chemical analysis and Raman spectroscopy have been used. The outer diameter of the fibers varied from 50 nm to 600 nm with an average diameter of 120 nm, length from several micrometers to several tens of micrometers and inner diameters from 20 nm to 230 nm. Two types of fibers have been identified; cylindrical which consists of a distinct graphite layers parallel to the fiber axes and bamboo - shaped fibers with walls which are built from domains with different orientations of graphite layers. The fibers contain 99.05 at.% carbon and 0.95 at.% oxygen with a binding energy of O (1s) electrons of 532.7 e V which corresponds to carbon in C-O bonds. In the first-order Raman spectra, the position of the band G was found at 1600 cm-1 and D at 1282 cm-1. The CNFs in ZrO2+ CNFs composite have been relatively well dispersed, however clusters of CNFs together with porosity are present as a result of the difficulty of dispersing, too. TEM and HREM revealed that the CNFs are usually located at the grain boundaries of ZrO2 in the form of undamaged nanofibers or disordered graphite.
Włókna węglowe przygotowane metodą osadzania z par (tzw. Chemical Vapour Deposition), były badane bezpośrednio po wytworzeniu oraz po wprowadzeniu do osnowy ceramicznej metodami elektronowej mikroskopii skaningowej, transmisyjnej oraz spektroskopii ramanowskiej. Średnica włókien wahała się pomiędzy 50 nm, a 600 nm przy sredniej wartości ok.120 nm, podczas gdy ich długość wynosiła od kilku do kilkudziesięciu mikrometrów. Identyfikowano dwa rodzaje budowy włókien, tj. cylindryczne oraz z wewnętrznymi przekładkami typu „łodygi bambusa“. Włókna węglowe zawierały do 0.95 at. % tlenu o energi wiazania O (1s) ok.532.7 e V, co odpowiada wiązaniu C-O. Na pierwszym widmie Ramana pasmo G identyfikowano dla 1600 cm-1, a pasmo D dla 1282 cm-1. Badania kompozytu wykazałay równomierny rozkład włókien w ceramicznej osnowie. W sąsiedztwie większych aglomeratów stwierdzono tendencje do występowania pustek. W czasie spiekania przeważająca część włókien zachowała swoją rurkową strukturę, natomiast pozostały materiał węglowy uległ przemianie do słabo uprządkowanych struktur grafitu lub nawet uległ amorfizacji.

Changes in the microstructure of nanostructured bainitic steel induced by quasi-static and dynamic deformation have been shown in the article. The method of deformation and strain rate have important impact on the microstructure changes especially due to strain localization. Microstructure of nanostructured steel Fe-0.6%C-1.9Mn-1.8Si-1.3Cr-0.7Mo consists of nanometer size carbide-free bainite laths and 20-30% volume fraction of retained austenite. Quasi-static and dynamic (strain rate up to 2×102 s-1) compression tests were realized using Gleeble simulator. Dynamic deformation at the strain rate up to 9×103 s-1 was realized by the Split Hopkinson Pressure Bar method (SHPB). Moreover high energy firing tests of plates made of the nanostructured bainitic steel were carried out to produce dynamically deformed material for investigation. Adiabatic shear bands were found as a result of localization of deformation in dynamic compression tests and in firing tests. Microstructure of the bands was examined and hardness changes in the vicinity of the bands were determined. The TEM examination of the ASBs showed the change from the internal shear band structure to the matrix structure to be gradual. This study clearly resolved that the interior (core) of the band has an extremely fine grained structure with grain diameter ranging from 100 nm to 200 nm. Martensitic twins were found within the grains. No austenite and carbide reflections were detected in the diffraction patterns taken from the core of the band. Hardness of the core of the ASBs for examined variants of isothermal heat treatment was higher about 300 HV referring to steel matrix hardness.

Opis:
Various transmission electron microscopy techniques were used for recognition of different kinds of bainitic structures in 100CrMnSi6-4 bearing steel. Upper and lower bainite are morphologically different, so it is possible to distinguish between them without problem. For new nanobainitic structure, there is still controversy. In studied bearing steel the bainitic ferrite surrounding the retained austenite ribbon has a high density of dislocations. Significant fragmentations of these phases occur, bainitic ferrite is divided to subgrains and austenitic ribbons are curved due to stress accommodation.
W celu rozpoznania i scharakteryzowania poszczególnych morfologii bainitycznych w stali łożyskowej 100CrMnSi6-4 zastosowano różne techniki transmisyjnej mikroskopii elektronowej. Rozpoznawanie bainitu górnego i dolnego nie nastręcza problemu, ze względu na ich zróżnicowane morfologie. W przypadku bezwęglikowego nanobainitu, ciagle jeszcze wiele jest kontrowersji. W badanej stali łożyskowej ferryt bainityczny otaczający wstęgi austenitu szczątkowego cechuje się dużą gęstością dyslokacji. Obydwie fazy wykazują znaczną fragmentacje; ferryt bainityczny podzielony jest na podziarna a wstążki austenitu ulegaja wygięciu w wyniku akomodacji naprężeń.

Opis:
New five ciprofloxacin (CIP) complexes of dioxouranium(II), oxozirconium(II), zirconium(IV), oxovanadium(II) and vanadium(IV) in the proportion 1:2 have been prepared using CIP as a drug chelate with UO2(NO3)2. 6H2O, ZrOCl2. 8H2O, ZrC4, VOSO4. xH2O and V2O5 respectively. The CIP complexes have been characterized based on the elemental analysis, molar conductance, magnetic, (FTIR & 1HNMR) spectral and thermal studies. The molar conductance studies of the synthesized complexes in DMSO solvent with concentration of 10–3 M indicate their non-electrolytic properties. At room temperature, the magnetic moment measurements revealed a diamagnetic behavior for all CIP prepared complexes. The different formulas of the new complexes can be represented as [UO2(CIP)2(NO3)2] (I), [VO(CIP)2(SO4)(H2O)] (II), [V2(O)(O2)2(CIP)2] (III), [Zr(O)(CIP)2(Cl)2] (IV), and [Zr(CIP)2(Cl)4] (V). The thermal analysis data of the complexes indicates the absence of coordinated water molecules except for vanadyl(II) complex (II). The CIP chelate is a uni-dentate ligand coordinated to the mentioned metal ion through terminal piperazinyl nitrogen. The transmission electron microscopy (TEM) investigation confirms the nano-structured form of the complexes.
