Using in vitro androgenesis serves as a unique opportunity to produce doubled haploid (DH) plants in many species. More benefits of this biological phenomenon have kept these methods in the focus of fundamental research and crop breeding for decades. In common wheat (Triticum aestivum L.), in vitro anther culture is one of the most frequently applied DH plant production methods. The efficiency of in vitro wheat anther culture is influenced by many factors, such as the genotype, growing conditions, collection time, pre-treatments, and compositions of media and culture conditions. According to some critical review, the genotype dependency, low efficiency and albinism are mentioned as limitations of application of the anther culture method. However, some research groups have made significant efforts to diminish the effects of these bottlenecks. Due to the improvements, a well-established in vitro anther culture method can be an efficient tool in modern wheat breeding programs.

This paper briefly cites the various procedures for the production of doubled haploids in barley and wheat. Various associated terms are defined and the factor involved in haploid production are outlined. Isolated microspore cultures offer some advantages over anther culture. Our current procedures for isolated microspore culture of barley and wheat are presented and compared.

A large population of anther culture-derived barley regenerants and their progeny was tested for allele segregation at 1 isozyme and 8 morphological marker loci. The segregation of genetic markers was examined separately for haploid, diploid and polyploid regenerants. All the 9 analysed genes except al (albino lemma) on chromosome 3 segregated according to the expected 1:1 ratio in the microspore-derived barley population. There was no difference in allele distribution between haploid and diploid regenerants. Among the limited number of 34 analysed tetraploids a significant excess of the dominant allele at locus о (orange lemma) of chromosome 6 was also observed. The recombination frequency between linked genes (n - lk2 on chromosome 1 and r - s on chromosome 7) estimated in the DH population did not differ significantly from recombination rates calculated in F₂ progeny or presented in barley chromosome maps. The phenomenon of gametic selection is discussed in relation to the genotype dependency of anther culture response and procedures used for DH production in barley.

The influences of genetic and environmental factors on the anther culture responses of wheat were investigated. Significant differences for callus induction, plant regeneration, and green plant percentages were observed when the nucleus of Triticum acstivum L. cv. Selkirk was transferred to ten alien cytoplasms by substitution backcrosses. In most cases, the alien cytoplasms decreased anther culture responses, but sometimes they were as good as or better than the T. aestivum cytoplasm. Significant within-genotype variation for anther culture responses were observed for wheat varieties Chris, Yecora Rojo, WA7176 and Edwall, indicating genetic heterogeneity in the present commercial cultivars, and potential for improving anther culture responses by in vitro prescreening. When five genotypes (Chris, Pavon 76, Butte 86, WA6916, and Edwall) were cultured across three (potato-4 liquid, 100 g L⁻¹ ficoll-supplemented, and 6 g L⁻¹ agar-solidified) induction media, the liquid and ficoll-containing media were 10 to 15 times more productive than the agar-solidified medium. Whereas, the ficoll medium was not significantly different from the liquid medium. Several low concentration starch media appeared promising to replace current induction media. The starch media sustained the highcallus-induction properties of the liquid medium, while improving callus aeration similar to that observed on solid media, resulting in markedly higher plant regeneration and green plant percentages.

The effect of C17 and W14 induction media on the formation of embryo-like structures (ELS) from F3 generation of nine hexaploid oat hybrids was investigated in the study. In all genotypes, the highest number of ELS (0.6 - 12.1/100 anthers) was obtained on C17 medium. The efficiency of plant regeneration on medium 190-2 was tested, in relation to different ELS culture conditions. The highest rate of green plants per 100 ELS (3.3 - 42.4) was produced by incubation at 22oC in the dark for the first two weeks. Among 36 green regenerants, 28 (77.8%) were haploid and 8 (22.2%) were spontaneous doubled haploids, fully fertile. After colchicine treatment of haploid plants, 19 were partially fertile and set from 1 to 15 seed per panicle.

Several aspects of in vitro culture have potential for cereal improvement. This paper focuses on evaluation of somaclonal variation (SV) from immature embryo callus culture, and doubled haploid (DH) production via anther culture in barley. Genetically stable SV was observed for several seedling morphological traits such as albino, yellow, light green and lethal. SV occurred at approximately half the frequency of azide-induced mutagenesis. The potential for widespread application of anther culture-mediated DH production in barley breeding and genetic studies was increased through culture procedure improvements and understanding the inheritance of anther culture response. Methodology improvements included substitution of inexpensive gelrite for expensive ficoll or agarose, ability to grow anther donor plants under field as well as growth chamber conditions and flexibility in cold pretreatment/storage of anther donor spikes for 4-6 weeks prior to anther plating. From diallel analysis, inheritance of anther culture response was complex with additive and dominance effects for embryoid formation, total plant regeneration and green plant regeneration and reciprocal effects (maternal) for green plant regeneration. High x low responder crosses generated F₁’s that were intermediate in response and low x low crosses sometimes produced F₁ heterosis for green plant regeneration. Therefore, some recalcitrant types appear to be usable in anther culture DH production systems within a breeding program.

Gametoclonal variation among anther culture-derived plants of three barley genotypes was estimated on the basis of cytological analysis (DH1, DH2 generation), observation of morphological variants (DH2, DH3) and chlorophyll mutation test (DH2, DH3). Individual head rows were grown in the field to detect possible chimeric structure of regenerants and to assess the number of variants and mutations in each line. Spontaneously doubled plants were the most frequent class (70%) among regenerants and almost 90% of them were completely fertile. There was a difference in proportion of haploids produced by different genotypes, but the highest frequency observed did not exceed 21%. The remaining regenerants were tetraploid, and contained chromosomal mutations or chimeras. In total, there were about 15% of polyploids and plants carrying chromosomal aberrations (translocations, inversions) among DH1 individuals. The changes in chromosome number and structure were the main source of observed variation. The level of gene mutation induced in vitro was relatively low. No more than 1% of microspore-derived plants expressed visible morphological changes in DH2 progeny. Only two morphological variants derived from the Bruce cultivar proved to be homozygous mutants (dwarf type) stable up the to third generation. The frequency of DH plants carrying chlorophyll mutation was 5.8%, but most of them (82%) were chimeric and had only a small mutation sector. The level of gametoclonal variation depended on the donor plant genotype. The highest proportion of variants and mutations was observed among DH plants derived from the Bruce cultivar, while the lowest was recorded among plants regenerated from anther culture of the doubled haploid line H930-36. Mechanisms leading to the observed variation and implications resulting from the presented experiments concerning implementation of anther culture in barley breeding were discussed. It was concluded that this method resulted in a high frequency of spontaneous doubling, a low frequency of genetic changes, and being less time and effort-consuming than the ’Bulbosum’ technique, can be applied to most barley breeding programs.

F₁, F₂ populations as well as F₃ and F₄ wheat lines coming from more than 300 cross combinations have been used for androgenic wheat studies and more than 5000 haploid plants were produced. Promising doubled haploid lines were tested together with lines produced in the conventional way in yield trials. The two best doubled haploid lines overyielded the standard. On the basis of results related to the general combining ability it is advisible to cross parents with a good combining ability for characters such as high percentage of responding anthers and embryo/callus induction with parents indicating a good combining ability for green plant production. In this way ideal lines can be obtained for doubled haploid production.
