Antibiotic resistance has become a global public health concern in the last few years. Given the widespread rate of recurrence, increasing attention is being turned toward environmental pathways that potentially contribute to antibiotic resistance genes (ARGs) dissemination outside the clinical realm. In this study, a metagenome analysis of intestinal virus-like particle fraction (VLPs) from a wild coyote (Canis latrans) revealed for the first time, multiple ARGs, such as B-lactamases and multidrug efflux pumps. Description of ARGs presence in natural environments is critical to understand the emergence of resistant strains.

Introduction and objective. Salmonella is a foodborne pathogen which causes gastrointestinal illness in consumers, and exhibits resistance to antimicrobials of veterinary and clinical significance. The aim of this study is to detect the prevalence and antimicrobial resistance of Salmonella isolates from meat in Latvia. Materials and method. A total of 3,152 samples of raw and ready-to-eat (RTE) meats were collected during the official control and in-house control procedures in 2015. Samples were tested in accordance with ISO 6579:2002. All S. Typhimurium, S. Enteritidis and other isolates recovered from the official control samples (S. Derby, S. Give) were tested for antimicrobial resistance. The minimum inhibitory concentration (MIC) values were investigated in line with the requirements of the European Committee on Antimicrobial Susceptibility Testing (EUCAST). Results. The prevalence of Salmonella was 0.8% (25/3152). The highest prevalence (1.5%) of Salmonella was found in minced meat and meat preparations (7/481), while the lowest (0%) in frozen meat and meat preparations (0/349) and RTE meats (0/364). The most common serovars were S. Typhimurium (36%, 9/25) and S. Derby (32%, 8/25). In total, 62% (13/21) of Salmonella isolates were resistant to at least one antimicrobial agent. Altogether, 40% (8/20) of isolates were resistant to sulfamethoxazole, 25% (5/20) to nalidixic acid, ciprofloxacin, ampicillin and 20% (4/20) to tetracycline. All isolates were susceptible to ceftazidime, cefotaxime, meropenem, azithromycin and tigecycline. S. Typhimurium exhibited antimicrobial resistance more often (87.5%) than other serovars. Conclusion. The study shows that the presence of Salmonella in meat, together with the high prevalence of resistant strains, is a significant public health related issue in Latvia.

This review summarizes current data on resistance among Salmonella spp. isolates of food origin from countries in different regions of the world. The mechanisms of resistance to different groups of antimicrobial compounds are also considered. Among strains resistant to quinolones and/or fluoroquinolones the most prevalent mechanism is amino acid substitutions in quinolone resistance-determining region (QRDR) of genes gyrA, parC but mechanism of growing importance is plasmid-mediated quinolone resistance (PMQR) associated with genes qnrA, qnrB, qnrC, qnrD, qnrS but frequency of their detection is different. Resistance to sulfonamides is mostly associated with genes sul1 and sul2, while resistance to trimethoprim is associated with various variants of dhfr ( dfr) genes. Taking into account Salmonella spp. strains isolated from food, resistance to β-lactams is commonly associated with β-lactamases encoding by blaTEM genes. However strains ESBL and AmpC – positive are also detected. Resistance to aminoglicosides is commonly result of enzymatic inactivation. Three types of aminoglycoside modifying enzyme are: acetyltransferases (AAC), adenyltransferases (ANT) and phosphotransferases (APH). Resistance to tetracyclines among Salmonella spp. isolated from food is most commonly associated with active efflux. Among numerous genetic determinants encoding efflux pumps tetA, tetB, tetC, tetD, tetE and tetG are reported predominatingly. One of the most common mechanisms of resistance against chloramphenicol is its inactivation by chloramphenicol acetyltrasferases (CATs), but resistance to this compound can be also mediated by chloramphenicol efflux pumps encoded by the genes cmlA and floR. It is important to monitor resistance of Salmonella isolated from food, because the globalization of trade, leading to the long-distance movement of goods, animals and food products, encourages the spread of resistant pathogens around the world.
W artykule przedstawiono aktualne dane na temat mechaniznów lekooporności pałeczek Salmonella spp. pochodzących z żywności. Wśród szczepów opornych na chinolony i/lub fluorochinolony najczęściej identyfikowanym mechanizmem są substytucje aminokwasów w obrębie regionów determinujących oporność na chinolony (QRDR-quinolone resistancedetermining region) w genach gyrA i parC, jednak coraz częściej identyfikowane są geny qnr (qnrA, qnrB, qnrC, qnrD, qnrS) związane z plazmidami (PMQR - plasmid-mediated quinolone resistance). Oporność na sulfonamidy jest najczęściej związana z genami sul1 i sul2, natomiast różne warianty genów dhfr ( dfr) warunkują oporność na trimetoprim. Biorąc pod uwagę szczepy Salmonella spp. pochodzące z żywności, oporność na antybiotyki β-laktamowe związana jest zazwyczaj z produkcją β-laktamaz kodowanych przez geny blaTEM. Jednakże coraz powszechniej identyfikowane są szczepy produkujące β-laktamazy o rozszerzonym spektrum substratowym (ESBL) oraz cefalosporynazy AmpC. Oporność na aminoglikozydy najczęściej wynika z wytwarzania enzymów modyfikujących cząsteczki leku: acetylotransferaz (AAC), adenylotransferaz (ANT) oraz fosfotransferaz (APH). Oporność wobec tetracyklin wśród pałeczek Salmonella spp. izolowanych z żywności najczęściej związana jest z mechanizmem aktywnego usuwania leku za pomocą pomp (efflux) kodowanych, najczęściej przez geny tetA, tetB, tetC, tetD, tetE i tetG. Jednym a najczęściej wykrywanych mechanizmów oporności na chloramfenikol jest jego inaktywacja w wyniku działania acetylotransferazy chloramfenikolowej (CAT). Oporność na chloramfenikol może być również związana ze zjawiskiem aktywnego wypompowywania leku. Pompy efflux są kodowane przez geny floR (warunkujące oporność także na florfenikol) lub cml. Istotne znaczenie ma monitoring lekooporności wśród szczepów Salmonella spp. pochodzących z żywności, ponieważ transport środków spożywczych oraz zwierząt do i z krajów całego świata ułatwia rozprzestrzenianie się szczepów lekoopornych.

Objectives Assessment of microbial air quality and surface contamination in ambulances and administration offices as a control place without occupational exposure to biological agents; based on quantitative and qualitative analysis of bacteria, yeasts and filamentous fungi found in collected samples. Material and Methods The sampling was done by wet cyclone technology using the Coriolis recon apparatus, imprint and swab methods, respectively. In total, 280 samples from 28 ambulances and 10 offices in Warszawa were tested. Data was analyzed using Shapiro-Wilk normality test, Kruskal-Wallis test with α = 0.05. P value ≤ 0.05 was considered as significant. Results The levels of air contamination were from 0 to 2.3×10¹ colony-forming unit (CFU)/m³ for bacteria and for yeast and filamentous fungi were from 0 to 1.8×10¹ CFU/m³. The assessment of office space air samples has shown the following numbers of microorganisms: bacteria from 3.0×10¹ to 4.2×10¹ CFU/m³ and yeast and filamentous fungi from 0 to 1.9×10¹ CFU/m³. For surface contamination the mean bacterial count in ambulances has been between 1.0×10¹ and 1.3×102 CFU/25 cm² and in offices – between 1.1×10¹ and 8.5×10¹ CFU/25 cm². Mean fungal count has reached the level from 2.8×10⁰ to 4.2×10¹ CFU/25 cm² in ambulances and 1.3×10¹ to 5.8×10¹ CFU/25 cm² in offices. The qualitative analysis has revealed the presence of Acinetobacter spp. (surfaces), coagulase – negative Staphylococci (air and surfaces), Aspergillus and Penicillium genera (air and surfaces). Conclusions The study has revealed a satisfactory microbiological quantity of analyzed air and surface samples in both study and control environments. However, the presence of potentially pathogenic microorganisms in the air and on surfaces in ambulances may endanger the medical emergency staff and patients with infection. Disinfection and cleaning techniques therefore should be constantly developed and implemented. Int J Occup Med Environ Health 2017;30(4):617–627

Escherichia coli producing extended-spectrum beta-lactamase (ESBL) are antimicrobial-resistant Enterobacteriaceae important in the livestock production sector, especially dairy cows because these antimicrobial-resistant bacteria can be transferred to consumers via consumption. If antimicrobials are continually used in dairy farms, this may result in antimicrobial resistance. Therefore, investigation of antimicrobial resistance and finding new alternative methods for inhibiting ESBL-producing E. coli is essential. Hence, the aim of this study was to examine the ability of selected antimicrobials to inhibit E. coli ATCC 25922, control bacteria and ESBL-producing E. coli isolated from dairy farms. We also investigated the capacity of polyphenol extract from 10 varieties of medicinal plants to inhibit ESBL-producing E. coli using a broth microdilution method. It was found that control bacteria were susceptible to all antimicrobial agents, i.e., ampicillin, cefotaxime, ciprofloxacin, chloramphenicol, gentamycin, imipenem, nalidixic acid, tetracycline, and sulfamethoxazole/ trimethoprim. However, ESBL-producing E. coli exhibited both susceptibility and resistance to selected antimicrobials. The polyphenol extracted from Psidium guajava Linn at the lowest concentration was 4.5 mg/mL, which could inhibit control bacteria, but at the same concentration could not inhibit ESBL-producing E. coli. These phenomena indicated that ESBL-producing E. coli had both susceptibility and resistance to antimicrobials. Polyphenol, which could inhibit non-resistant E. coli, could not inhibit ESBL-producing E. coli.

This study is aimed to investigate culturable airborne bacteria concentrations and the composition of methicillin-resistant staphylococci in eleven different locations on the basis of specific activities conducted within different parts of the European side of Istanbul. The highest bacterial levels were observed at the Bakirkoy station (1 100 CFU/m3) while the second highest levels were found at the Bahcelievler station (1 040 CFU/m3) in October; the lowest levels (10 CFU/m3) were measured at other different stations (Atakoy, Yesilkoy). Fifteen methicillin-resistant isolates [Staphylococcus hominis (n=11), S. cohnii spp. cohnii (n=2), S. sciuri (n=1), S. capitis spp. capitis (n=1)] were identified. The disc diffusion method was used to identify the antimicrobial resistance of these isolates, it was observed that the most common resistance was to penicillin (P) (n=11), doxycycline (DO) (n=4) and tetracycline (T) (n=5). None of the isolates was resistant to imipenem, amoxicillin/clavulanic acid, vancomycin (IPM, AMC, VA). However, multiple antimicrobial resistance was found to be 26.7%. The results of this study revealed the importance of isolated methicillin-resistant staphylococci in the stations with densely active human population and traffic, for public health. As a result, the importance of resting along known shorelines, where culturable airborne bacteria concentrations are much lower, and its importance for human health have been emphasized.

Enterokoki są dobrze przystosowane do funkcjonowania w środowisku sera i wraz z innymi bakteriami mlekowymi stanowią grupę niestarterowych bakterii mlekowych. Ich aktywność proteolityczna i lipolityczna przyczynia się do rozwoju typowych właściwości sensorycznych serów. Dzięki wytwarzaniu bakteriocyn enterokoki wpływają na mikrobiotę sera, z jednej strony ograniczając ewentualny rozwój drobnoustrojów patogennych i powodujących psucie a z drugiej strony stymulują procesy autolityczne innych bakterii mlekowych. Dzięki tym cechom drobnoustroje te mają potencjał do stosowania jako kultury dodatkowe lub ochronne do serów. Niektóre gatunki Enterococcus wykazują cechy wirulencji i antybiotykooporność i z tego powodu rodzaj ten nie posiada statusu QPS (qualified presumption of safety).
Enterococci are well adapted to function in the cheese environment and, along with other lactic acid bacteria, constitute a group of non-starter lactic bacteria. Their proteolytic and lipolytic activity contributes to the development of typical sensory properties of cheeses. By producing bacteriocins, enterococci influence the cheese microbiota, on the one hand, limiting the possible development of pathogenic and spoilage microorganisms, and, on the other hand, stimulating the autolytic processes of other lactic bacteria. Due to these characteristics, enterococci have the potential to be used as adjunct or protective cultures for cheeses. Some Enterococcus species show virulence and antibiotic resistance and therefore the genus does not have qualified presumption of safety status (QPS).

Campylobacter species are among the most frequently identified bacterial causes of human gastroenteritis. Because Campylobacter spp. harbored by cattle can be transmitted to humans, in this study we investigated antimicrobial resistance of thermophilic Campylobacter isolated from cows. Our study included 150 strains of Campylobacter (143 strains of C. jejuni and 7 strains of C. coli) isolated from cows in South-Western Poland. The minimal inhibitory concentration (MIC) to ciprofloxacin, erythromycin, gentamicin and tetracycline were determined using the agar dilution methodology. All strains of C. coli were susceptible to all four drugs studied. The most frequently detected resistance of C. jejuni was to ciprofloxacin (26 strains 18.2%). Resistance to tetracycline was observed in 5 strains (3.5%). All strains of C. jejuni were susceptible to erythromycin and gentamicin.

Penicillin, the first antibiotic introduced into clinical practice opened a new era in medicine. The ‘golden age’ of antibiotic discoveries in the 1950s, 60s and 70s significantly helped our fight against bacterial infections. In parallel with the introduction of new drugs, resistance strains were identified. This was, however, neglected because of the belief that pharmaceutical companies would continuously supply us with new products. In contrary, a pipeline of new antibiotics slowly dried out and in the 1980s we realized that the proportion of resistant bacteria was increasing faster than the supply of new antibiotics. New mechanisms of resistance emerged and multidrug and pandrug resistant bacterial strains started to spread globally. Antimicrobial resistance is recognized now as one of the greatest threats to public health worldwide. The WHO and EU as well as national agencies are calling for actions which should be immediately undertaken if we do not want to lose the battle.

These are members of the expanding class of substances known as Contaminants of Emerging Concern (CECs). Antibiotic resistance in the environment and garbage generated by PPCP endanger life. The World Health Organisation (WHO) now recognises antibiotic resistance as a significant global health problem due to the expected increase in mortality caused by it. In the past ten years, mounting data has led experts to believe that the environment has a significant impact on the development of resistance. For human diseases, the external environment serves as a source of resistance genes. It also serves as a major pathway for the spread of resistant bacteria among various habitats and human populations. Large-scale DNA sequencing methods are employed in this thesis to better comprehend the dangers posed by environmental antibiotic resistance. The quantification of the number is an important step in this process. Metagenomic measurement of the number of antibiotic resistance genes in various contexts is a crucial step in this process. However, it’s also crucial to put this data into a broader context by integrating things like taxonomic information, antibiotic concentrations, and the genomic locations of found resistance genes.
