Two series of benzimidazole derivatives were sythesised. The first one was based on 5,6-dinitrobenzimidazole, the second one comprises 2-thioalkyl- and thioaryl-substituted modified benzimidazoles. Antibacterial and antiprotozoal activity of the newly obtained compounds was studied. Some thioalkyl derivatives showed remarkable activity against nosocomial strains of Stenotrophomonas malthophilia, and an activity comparable to that of metronidazole against Gram-positive and Gram-negative bacteria. Of the tested compounds, 5,6-dichloro-2-(4-nitrobenzylthio)-benzimidazole showed the most distinct antiprotozoal activity.

Silver nanoparticles (NPs) have at least one dimension of a particle smaller than 100 nm and contain 20-15,000 silver atoms. Due to its antibacterial activity nanosilver (NS) is used for medical purposes. NS particles can be obtained by various methods. Potentially, the best method of the NS synthesis for medical purposes is based on a brief flow of electric current between two silver electrodes placed in deionized water. It is accepted that the major antibacterial effect of silver is its partial oxidation and releasing silver ions, which interact with thiol groups of peptidoglicans of bacterial cell wall, and proteins of the cell membrane causing cell lysis. Silver ions can also bind to bacterial DNA preventing its replication and stopping synthesis of bacterial proteins. The rise in exposure to silver NPs has spurred interest into their toxicology. NS undergoes a set of biochemical transformations including accelerated oxidative dissolution in gastric acid, binding to thiol groups of serum and tissue proteins, exchange between thiol groups, sulfides and selenides, binding to selenoproroteins and photoreduction in skin to zerovalent metallic silver. Animal studies have shown that exposure to NS may lead to liver and spleen damage. NS can also stimulate an increased secretion of proinflammatory cytokines by monocytes. As a spectrum of NS applications is still growing, the complex evaluation of a safety of its use becomes an important task. This requires an elucidation of not only the influence of NS on human cells and organism, but also its biotransformation in organism and in environment.

The sol-gel derived silica spheres with surfaces modified by silver nanoparticles were used to enhance the photodynamic effect. The silica nanoparticles were prepared by modified Stöber synthesis. The diameter of obtained silica spheres was ca. 100 nm. These silica spheres we used as a carrier for noble nanoparticles. It was shown that Ag-Au nanoparticles have an antibacterial activity against Escherichia coli. This effect depends on the nanoparticles concentration and it is stronger for higher concentrations. Laser irradiation enhances this effect, and starting from certain concentration it is possible to kill Escherichia coli, totally, when using laser light.

In this study, a solution of dibutyrylchitin (DBC)/polylactide (PLA) blend micro and nanofibres were successfully fabricated using blends of 2,2,2-trifluoroethanol (TFE) as solvents. Fibres were produced from the solutions by electrospinning. The DBC/PLA blend solutions in various ratios were studied for electrospinning into micro/nanofibres. The morphology of the micro and nanofibres was observed by scanning electron microscope (SEM). The biggest diameters of DBC/PLA fibres were obtained for the blended microfibres in ratios of 10/90 and 25/75. The smallest diameter was observed for pure polymers. The antibacterial properties were examined for materials obtained by electrospinning. In the experiments, materials with antibacterial properties were made. It is likely that the electrospun micro and nanofibres will be used in the native extracellular matrix for tissue engineering.

InWO3-InMoO3 loaded-TiO2 nanomaterial was synthesised by co-precipitation method and sonication technique. The nanomaterial was characterized by High-resolution scanning electron microscopy (HR-SEM) with elementary dispersive X-ray (EDX), High-resolution transmission electron microscopy (HR-TEM), photoluminescence spectroscopy (PL) and UV-Vis diffuse reflectance spectroscopy (DRS). The photocatalytic activity of InWO3-InMoO3 loaded-TiO2 nanomaterial was studied from the photodegradation of methyl green (MEG) and methylene blue (MB) under UV-light irradiation at 365 nm. The hydroxyl radical formation in the mechanism was confirmed by fluorescence quenching technique. The mineralization confirmed by chemical oxygen demand measurements. A flaxisible mechanism is proposed for higher catalytic activity of InWO3-InMoO3-TiO2 under UV-light for 365 nm. This catalyst was found to be stable and reusable without appreciable loss of catalytic activity up to five cycles. The InWO3- InMoO3 loaded-TiO2 nanomaterial on MEG was high photocatalytic activity that of MB so high photocatalytic activity has been discussed by first order kinetics, rate constant value and high quantum yield. This material was showed increased hydrophobicity that of TiO2 electrochemical study and antibacterial activity of the analysis was also studied.

Because of side effects and increasing resistance to antibiotics, considerable attention hasbeen given to extract sand biologically active compounds isolated from medicinal plants. In the present study, phytochemical screening and antibacterial activities of different concentrations of various extracts of Onosma chlorotricum were evaluated and compared with those of vancomycin and amikacin. Total phenolic and flavonoid contents and the antibacterial activity of methanol, n-hexane, and aqueous extracts (5 mg/ml to 0.156 mg/ml final concentration) of O. chlorotricum against Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli were evaluated using disc diffusion and microdilution methods. Total phenolic and flavonoid contents were determined according to the Folin-Ciocalteu and aluminum chloride colorimetric assays, respectively. The results showed that the total phenolic and flavonoid contents of these extracts ranged from 56.10 ± 0.13 to 74.12 ± 0.05 mg GAE/g dry extract and from 19.3 ± 0.6 to 23.20 ± 0.41 mg QE/g dry extract, respectively. The methanol extract with the highest phenolic and flavonoid content showed the highest antibacterial activity against all the tested bacterial strains, with the highest inhibition zone of 21 ± 0.7 mm and the lowest MIC and MBC values of 78.12 μg/ml for S. aureus. The antibacterial effects and the total phenolic content of O. chlorotricum were remarkable. The results suggest that the effects of methanol extract on wound healing could be tested in an animal model.

The objective of this study was to evaluate the chemical composition CH3OH-CH2Cl2 (1:1) extract and biological activities of various extracts derived from the aerial parts of the brown marine alga Stoechospermum marginatum (C. Agardh). Gas chromatography (GC) and gas chromatography-mass spectroscopy (GC-MS) were used to analyze the composition of the essential oil. Total phenolics assay demonstrated a high value in hexane extract (HE), with a lower value for chloroform extract (CE), and the lowest value for methanol extract (ME). DPPH (2,2-diphenyl-1-picrylhydrazyl) assay showed that extracts of S. marginatum possess radical scavenging activity (RSA). Tests of the antioxidant property of the extracts revealed both electron and hydrogen transfer mechanisms. The antibacterial activity of the ME, CE, and HE as well as an ethanol extract was estimated against seven Gram-positive and Gram-negative bacteria. The ethanol extract showed the highest antibacterial activity, and the HE showed the lowest.

Three Schiff bases derived from thiosemicarbazide and ketones (Vanillin, Benzophenone and Acetophenone) were used to study their antibacterial activities against some pathogenic bacteria by disc diffusion method. Of these, benzophenone thiosemicarbazone showed significant antibacterial activity as compared with that of Kanamycin. All these three compounds were found to possess cytotoxic effect. Minimum inhibitory concentration of these compounds was also determined.

Background. During the recent years, appropriate attention has been paid to the oxidative stress which damages the body’s cells, proteins, and DNA. Therefore, the need of antioxidants becomes a therapeutic and preventive priority. In addition, microbial infections also constitute a public health problem. Objective. To find efficient, reliable and safe alternatives sources to synthetic antioxidants, antibiotics and antifungals drugs. Materials and methods. Extract and essential oil of Psidium guajava were screened for their antioxidant and antimicrobial activities against gram positive bacteria (Enterococcus faecalis and Staphylococcus aureus), gram negative bacteria (Citrobacter freundii, Escherichia coli and Pseudomonas sp) and fungi (Candida albicans,Candida tropicalis and Cryptococcus neoformans), as well as to determine the functional groups of phytochemicals present in the essential oil by Fourier transform infrared spectroscopy (FTIR). Results. The results indicate that P. guajava leaves extract demonstrated very high antioxidant activity and P.guajava essential oil showed the highest polyphenols content. The antioxidant capacity showed a significant negative linear correlation to total polyphenolic content (TPC) with Pearson’s correlation coefficients. P. guajava essential oil shows high antibacterial and antifungal activity against all the studied bacteria and fungi.The FTIR analysis of P. guajava essential oil showed the presence of several functional groups (ethers, esters, ketones, terpenes, alkanes, aldehydes, aromatic hydrocarbons, alcohols, and phenols). The relationship between the chemical composition and antimicrobial activity of P.guajava essential oil suggests that the attribution of its antimicrobial activity to a particular compound or a synergistic effects between its different constituents remains difficult. Conclusions. The present study demonstrated that Psidium guajava is a valuable source of active compounds with anti- oxidant and antimicrobial activities. This finding suggests the new use of the fruits and the leaves extracts of this plant in the treatment of bacterial and fungal infections, as well as for the extraction of new antioxidants. Therefore, it is necessary to be carried out in another study to identify the active(s) compound(s) in P.guajava essential oil with respect to their mechanisms and synergistic actions.

The in vitro antibacterial activity of pure caffeine powder and its interaction with first line antibiotic against bacterial isolates were investigated with the macrobroth dilution and the checkerboard assay methods. This study showed that caffeine and the antibiotics exhibited various degrees of antibacterial activities. While caffeine had MICs ranging between 67.19 and 268.75 µg/ml, chloramphenicol was characterized by MICs between 0.98 and 31.25 µg/ml, kanamycin - 15.63-62.5 µg/ml, nalidixic acid - 0.49-250 µg/ml, erythromycin - 0.49-62.5 µg/ml, tetracycline - 1.99-62.5 µg/ml and metronidazole - 15.63-31.25 µg/ml. Combining ½ MICs and MICs of caffeine with the antibiotics as well as direct combination of caffeine and the antibiotics resulted in significant reduction of antibiotics' effectiveness. The fractional inhibitory concentration index (FICI) for the combination of ½ MICs of caffeine with different antibiotics showed antagonistic interactions with the antibiotics except kanamycin which had additive and indifferent interactions with caffeine. The FICI of the MICs of caffeine combined with antibiotics showed a reduction in the number of antagonistic interactions as chloramphenicol, nalidixic acid and erythromycin showed some indifferent interactions while kanamycin was the only antibiotic that showed indifferent interaction against all the bacterial isolates. The direct combination of caffeine and the antibiotics resulted in significant antagonistic interactions higher than in the case when caffeine, at the ½ MICs and MICs, was combined with the antibiotics. Although caffeine demonstrated significant antibacterial activity against the selected bacterial isolates, its combination with the selected antibiotics resulted in significant antagonistic interactions. Caffeine should not be combined with antibiotics as this could result in serious therapeutic failure and, possibly, drug toxicity in vivo.

Celem pracy była ocena aktywności przeciwbakteryjnej modyfikowanego lizozymu w stosunku do bakterii Gram-ujemnych Pseudomonas fluorescens i Proteus mirabilis. Porównano antybakteryjne działanie monomeru lizozymu i jego preparatu uzyskanego w wyniku modyfikacji termiczno-chemicznej. W otrzymanym preparacie lizozymu oznaczono udział form oligomerycznych, aktywność hydrolityczną i hydrofobowość. Stwierdzono wzrost inhibitującego działania lizozymu po modyfikacji w porównaniu z monomerem lizozymu i próbą bez dodatku enzymu w stosunku do badanych bakterii. Wykazano, że modyfikowany preparat lizozymu wykazujący aktywność przeciwbakteryjną wobec bakterii gramujemnych, charakteryzuje się wyższym udziałem form oligomerycznych jak również wyższą hydrofobowością powierzchniową.
The aim of the study was to assess the antibacterial activity of modified lysozyme in relation to Gram-negative bacteria Pseudomonas fluorescens and Proteus mirabilis. Antibacterial action of lysozyme monomer and its preparation produced as a result of thermo-chemical modification were compared. In the obtained lysozyme preparation the proportion of oligomeric forms, hydrolytic activity and hydrophobicity were determined. An increase was found in the inhibitory action of lysozyme after modification in comparison with lysozyme monomer and the sample without an addition of the enzyme in relation to the analysed bacteria. It was shown that the modified lysozyme preparation exhibiting antibacterial activity against Gram-negative bacteria is characterised by a higher proportion of oligomeric forms, as well as higher surface hydrophobicity.

Silver nanoparticles (AgNPs) have been synthesized in the presence of Strawberry fruit extract (SBFE) at room temperature. The synthesized AgNPs was characterized by UV-vis spectroscopy, SEM, EDS, XRD, TEM and FTIR. The UV-vis spectra of the AgNPs show SPR band at 450 nm. TEM results indicate that AgNPs are spherical in shape and size range between 7–65 nm. Antibacterial activity of the synthesized AgNPs has been assessed against Pseudomonas aeruginosa and Bacillus licheniformis. The results show that AgNPs exhibit inhibitory effect and effect is a function of AgNPs concentration. The antibacterial activity of the prepared AgNPs has been compared with two antibiotics, amoxicillin and ciprofloxacin. It is found that the antibiotics perform better than AgNPs.

Opis:
Rothmannia wittii, which is found in Binh Chau-Phuoc Buu Nature Reserve, Bung Rieng ward, Xuyen Moc District, Ba Ria-Vung Tau Province, Vietnam, is a rare species of Rubiaceae family. The present study aimed to determine the phytochemical composition and antimicrobial activity of the ethanolic extract of the R. wittii trunk. A total of 10 compounds were identified by liquid chromatography-mass spectrophotometry (LC-MS), including 1) benzene, 1-(1,5-dimethyl-4- hexenyl)-4-methyl, 2) diphenylsulfone, 3) 6β-hydroxy-10-O-acetylgenipin, 4) cinnamyl-cinnamate, 5) genipin, 6) megastigmatrienone, 7) ethyl p-methoxy-trans-cinnamate, 8) rothalazepane, 9) 1,3,6-trimethylpyrano[2,3-d ]imidazole-2,5(1H,3H)-dione, and 10) shanzhiol. The extract also exhibited an antibacterial activity against six bacterial strains: Bacillus cereus, Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Salmonella typhimurium and Salmonella enteritidis, as determined via disk diffusion agar method.
