In growing maize, an increase in the content of phenolic compounds and selected phenolic acids in soil was found after the incorporation of white mustard, buckwheat, spring barley, oats and rye mulches into the soil. The highest content of phenolic compounds in soil was found after oats mulch incorporation (20% more than in the control soil). The highest content of selected phenolic acids was found for the soil with the oats and rye mulch. Among the phenolic acids investigated, ferulic acid was most commonly found in the soil with the plant mulches. However, two phenolic acids: the protocatechuic and chlorogenic acid, were not detected in any soil samples (neither in the control soil nor in the mulched soil). At the same time, a decrease in the primary weed infestation level in maize was found in the plots with all the applied plant mulches, especially on the plots with oats, barley and mustard. The plant mulches were more inhibitory against monocotyledonous weeds than dicotyledonous ones. During high precipitation events and wet weather, a rapid decrease in the content of phenolic compounds in soil and an increase in the primary weed infestation level in maize were observed.
W uprawie kukurydzy stwierdzono wzrost zawartości związków fenolowych i wybranych kwasów fenolowych w glebie po wprowadzeniu do niej mulczu z: gorczycy białej, gryki, jęczmienia jarego, owsa i żyta. Najwyższą zawartość związków fenolowych stwierdzono w glebie z mulczem z owsa (o 20% więcej, niż w glebie kontrolnej). Najwyższą zawartość wybranych kwasów fenolowych stwierdzono w glebie z mulczami z owsa i żyta. W glebie z mulczami najczęściej stwierdzano kwas ferulowy. Jednocześnie w żadnej glebie (kontrolnej i z mulczami) nie stwierdzono występowania kwasów fenolowych: protokatechowego i chlorogenowego. Równocześnie na tych samych poletkach z mulczami stwierdzono zmniejszenie zachwaszczenia pierwotnego kukurydzy, wyraźne zwłaszcza z mulczem z owsa, jęczmienia jarego i gorczycy białej. Zastosowane mulcze bardziej ograniczały wzrost chwastów jednoliściennych. Wyraźne zmniejszenie ilości związków fenolowych w glebie oraz zwiększenie zachwaszczenia pierwotnego kukurydzy obserwowano w okresie zwiększania ilości opadów i wilgotnej pogody.

Genus Serratula is known for the presence of steroid compounds which belong to the group of ecdysones. Former phytochemical works from the late sixties and early seventies indicate, in some Serratula species, the occurrence of a phenolic glycoside-arbutin. This has been confirmed in the present work through finding an α,β-arbutin anomer in Serratula quinquefolia M.B. New botanical data concerning the classification of genus Serratula suggest that the species in question should be regarded as belonging to genus Klasea (currently a section of genus Serratula). There has been an attempt to find a key to the chemical division within the taxons.

Hericium erinaceum (Bull.: Fr.) Pers. is an edible fungus of great significance in medicine. It is rarely found in Europe, in contrast, it is common in Japan and North America. Its fruitbodies have been well-known for hundreds of years in traditional Chinese medicine and cuisine. A cradle of H. erinaceum cultivation is Asia. In Eastern Europe is rare in natural habitats, but can be successfully cultivated. Both fruitbodies and mycelia are rich in active, health promoting substances. Tests of substances extracted from this mushroom carried out on animals and in vitro have given good results. They can be used in the treatment of cancer, hepatic disorders, Alzheimer’s and Parkinson’s diseases, wound healing. They improve cognitive abilities, support the nervous and immune systems. Promising results have been reported in clinical trials and case reports about the human treatment (e.g., recovery from schizophrenia, an improvement of the quality of sleep, alleviation of the menopause symptoms). The subject of this paper is to summarize information about the development of mycelium, the best conditions for cultivation of fruitbodies, bioactive substances and their use in medicine.

Abiotic stress factors are among the major causes of lower crop yields. It is known, that in response to cold and/or osmotic stress, crops activate various defense mechanisms, including morphological, physiological and metabolic adaptations. Secondary metabolism, especially phenolic compounds, seem to be an important factor of stress-induced metabolic re-engineering as their levels are alternated by abiotic stress in plants. Despite the fact, that the nature and function of phenolic compounds was already studied in various plant species, it is important to define tissue-specific changes induced by two most potent abiotic stressors – low temperature and decreased water potential. Moreover, in fields, the appearance of single stress is rather rare. Usually two or more factors are acting in parallel, which may potentially result in different effects. Therefore, the aim of this study was to analyze selected elements of secondary metabolism in roots of germinating soybean seeds under cold stress, osmotic stress and both stresses combined. In addition the effects of constant and persistent stress were compared to those induced by sudden and brief stress appearance, as well as after the post-stress recovery process. In the presented study standard methods for identification and quantification of phenolic acids and isoflavones were used and the antioxidant capacity of the radicle extracts was measured. The phenolic metabolism in plants was greatly intensified in response to cold and osmotic stress and remained at high level during the post-stress recovery. The amount and composition of both phenolic acids and identified isoflavones also changed in stress- and duration-dependent manner. This proves an important role of phenolic compounds in abiotic stress response of germinating soybean seeds and opens up new perspectives for further investigations.

Liquid shoot culture of Salvia officinalis L. in MS medium containing IAA (0.1 mg l-1) and BAP (0.45 mg l-1) was developed and evaluated in relation to shoot multiplication and antioxidant compound (carnosic acid, carnosol and rosmarinic acid) accumulation. In the liquid medium, on average, 3 new shoots per explant (shoot tip) were obtained within 3 weeks. The shoots produced 8.2±0.02 mg of diterpenoids and 31.2±0.29 mg of rosmarinic acid per gram of dry weight. Shoot proliferation and diterpenoid content increased when triacontanol (5, 10 or 20 pg l-1) was added to the liquid medium. In optimum conditions (at 20 pg l-1 TRIA) almost 7 shoots were formed per explant after 3 weeks. An increase in diterpenoid production (expressed as the sum of carnosol and carnosic acid) ranged from 30% to 50% and dependended on triacontanol concentration tested. The level of diterpenoids in triacontanol-treated shoots was similar to the content of compounds in commercial herbal product (dried leaves of S. officinalis) (10-12 mg g-1 dry wt). Triacontanol did not increase rosmarinic acid production, but the content of the phenolic as compound in shoots grown in liquid culture (31 mg g-1 dry wt) was even 24 times higher compared to samples of dried leaves of S. officinalis plants. We also demonstrated that the highest amounts of CA, Car and RA were accumulated in young, top parts of sage shoots. This observation could be useful for improving the selection of material for the extraction of natural antioxidants from S. officinalis.

The concentrations of carnosic acid, carnosol and rosmarinic acid in different materials from differentiated (multiple shoot cultures and regenerated plants) and undifferentiated (callus and cell suspension) in vitro cultures of Salvia officinalis were determined by HPLC. The results suggested that diterpenoid (carnosic acid and carnosol) production is closely related to shoot differentiation. The highest diterpenoid yield (11.4 mg g-1 for carnosic acid and 1.1 mg g-1 for carnosol) was achieved in shoots of 10-week-old micropropagated plants. The levels were comparable to those found in shoots of naturally growing plants. Undifferentiated callus and cell suspension cultures produced only very low amounts of carnosol (ca. 0.05 mg g-1 of dry weight). In contrast, content of rosmarinic acid in callus and suspension cultures as well as shoots growing in vitro and in vivo was similar and ranged between 11.2 and 18.6 mg g-1 of dry weight.

This report describes the effect of triacontanol on shoot multiplication and production of antioxidant compounds (carnosic acid, carnosol and rosmarinic acid) in S. officinalis cultures grown on MS basal medium (agar solidified medium supplemented with 0.1 mg l-1 IAA, 0.45 mg l-1 BAP). It was found that shoot proliferation significantly increased when triacontanol at concentrations of 5, 10 or 20 µg l-1 was added to the medium. HPLC analysis of acetone and methanolic extracts of sage shoots showed that the production of diterpenoids, carnosic acid/carnosol ratio, as well as, contents of rosmarinic acid were also affected by the treatment with triacontanol. The highest stimulation effect of triacontanol was observed on the production of carnosol, where the treatment with 20 µg l l-1 increased the content of this diterpenoid 4.5-fold compared to that in the control (sage shoots growing on MS basal medium, only).

The study focused on the production of compounds with antioxidant activity in hairy root and shoot cultures of Salvia officinalis grown in laboratory-scale sprinkle nutrient bioreactors. HPLC analysis showed that production of rosmarinic acid in transformed roots (34.65±1.07 mg l-1) was higher that in shoot culture (26.24±0.48 mg l-1). In the latter diterpenoids: carnosic acid (1.74±0.02 mg l-1) and carnosol (1.34±0.01 mg l-1) were also found. Biomass accumulation after a growth period in the bioreactor was also studied. An 18-fold increase in hairy root biomass was recorded after 40 days of culture. In sage shoot culture, biomass increased 43 times after 21 days of bioreactor run. The current operating conditions of the bioreactor were not suitable for the propagation of Salvia officinalis mainly due to the hyperhydricity problem of leaves and stems.

The aim of the research was to determine changes in the chemical composition in lupin seeds during seed germination in a solution containing selected nitrogen compounds. Blue lupin seeds of the Neptun cultivar were germinated for 3 or 4 days in darkness, at 24°C in 1% solutions containing the following nitrogen compounds: urea, nitric acid, ammonium sulphate, methionine or yeast extract. The control consisted of seeds germinated in water. The presence of all nitrogen sources has resulted in a poorer growth of the sprouts as compared to seeds germinated in water. There was a significant increase in the true protein content as well as a significant decrease in the content of crude ash and alkaloids in the germinated seeds. The seeds germinated for 4 days had significantly higher total protein content and significantly lower content of nitrogen-free extract compounds and oligosaccharides as compared to the seeds germinated for 3 days. A significant influence of the nitrogen source on the concentration of some amino acids was found. Cystine was found to be absent in seeds germinated in the nitric acid solution, while there occurred a twofold increase in the cystine content and a fivefold increase in the methionine content in seeds germinated in the aqueous solution of methionine and an 50% increase in the cystine concentration was observed in seeds germinated in the yeast extract and ammonium sulphate solution. Methionine and cystine were the limiting amino acids in all the samples, accept of seeds germinated in the methionine solution were it was valine. Aqueous solutions of ammonium sulphate and yeast extract were found to have the most advantageous influence on the chemical composition of lupin germination products.

The effect of methyl jasmonate (MJ) vapors on content of phenolic compounds: free phenolic acids, total quercetin, and total phenolics in etiolated buckwheat seedlings were studied. The data presented show that low concentration of MJ (10-8 M) had no influence on trans-cinnamic acid (CA), but stimulated the accumulation of chlorogenic acid in hypocotyls and cotyledons of buckwheat seedlings. A moderate dose of MJ (10-6 M) did not change the level of chlorogenic acid in the hypocotyls and cotyledons, but CA synthesis was promoted in cotyledons, whereas in hypocotyls no significant effect was found. Highest concentration of MJ (10-4 M) caused small decline of CA in hypocotyls, but large stimulation of the acid production in cotyledons was noted. MJ had stimulatory effect on caffeic acid forming, but inhibited synthesis of vanillic acid in hypocotyls and cotyledons. Lowest concentration of MJ (10-8 M) elicited accumulation of quercetin glycosides in both studied tissues of buckwheat seedlings, however at higher doses (10-6 and 10-4 M) did not affect the flavonol level. The obtained results suggest that nonequivalent influence of methyl jasmonate on the phenolics composition can be a result of various mechanisms of MJ uptake, transforming and/or its translocation in buckwheat hypocotyls and cotyledons. Decline of anthocyanins level in buckwheat hypocotyls caused by MJ cannot be explained by enhanced accumulation of quercetin glycosides or free phenolic acids, but probably by synthesis of other unknown phenolic compounds.

The aim of the presented study was to determine the effect of foliar application of CaCl2 on the level of β-carotene, lutein, tocopherols and phenolic compounds in the leaves of lettuce cv. Omega (Lactuca sativa L.). The experiments were conducted in the greenhouse of the Faculty of Cultivation and Fertilisation of Horticultural Plants, University of Life Sciences in Lublin. During the growing period, 7 and 14 days before harvest, the plants were treated with a solution of CaCl2 with concentrations of 0.1 M and 0.2 M. Plants sprayed with water were the control treatment in the experiment. Analyses were performed on whole leaves and the blade without midrib. The obtained results indicate that the leaves of lettuce cv. Omega are a good source of phenolic compounds, carotenoids and tocopherols. The dominant carotenoid in lettuce cv. Omega was β-carotene. Foliar application of CaCl2 on the plants did not have any significant effect on the level of β-carotene and lutein in the whole leaves. The contents of tocopherols, total phenolic compounds and chlorogenic acid were dependent on the concentration of CaCl2. The application of 0.1 M CaCl2 solution in the plants resulted in a decrease in the level of total phenolic compounds and chlorogenic acid, and an increase in tocopherol content. However, the treatment of the plants with 0.2M CaCl2 solution caused a lowering of the concentration of tocopherols and an increase in the content of chlorogenic acid and total phenolic compounds.

The research has been performed on roots of Vitis vinifera, cv. Himrod, obtained from seedlings grown under chill stress conditions (+10oC in the day and +7oC at night), under optimum conditions (+25oC in the day and +18oC at night) and from seedling which underwent a recover period after the chill stress treatment. The purpose of the study has been to determine quantitative and qualitative changes in phenolic compounds as well as to demonstrate changes in antiradical properties of extracts from grapevine roots, which appeared as a result of chill stress and during recovery under the optimum conditions following the stress. Phenolic compounds from grapevine roots were extracted using 80% acetone. The total content of phenolics was determined by colorimetry. The content of tannins was tested by precipitation with bovine serum albumin. The reducing power as well as DPPH• free radical and ABTS+• cation radical scavenging activity of the extracts were also tested. In order to identify phenolic compounds present in the extracts the RP-HPLC technique was employed. The tested material was found to contain tannins and three identified phenolic acids: ferulic, caffeic and p-coumaric ones. The latter occurred in the highest concentrations (from 4.46 to 6.28 µg/g fresh matter). Ferulic acid appeared in smaller amounts (from 1.68 to 2.65 µg/g fresh matter), followed by caffeic acid (from 0.87 to 1.55 µg/g fresh matter). Significantly less total phenolic compounds occurred in roots of seedlings subjected to chill stress. However, the total content of these compounds increased significantly in roots of plants which underwent recovery after chill stress. Concentration of tannins was determined by two methods. The content of condensed tannins was depressed in roots as a result of low temperature stress, whereas the content of condensed and hydrolysing tannins (determined via the BSA method) rose under chill stress conditions. A significant increase in tannins in root extracts (determined with both methods) was found during the recovery process after the stress. The three identified phenolic acids appeared in grapevine roots as ester-bound compounds. It has been demonstrated that the content of phenolic acids significantly fell as a result of low temperatures, but increased during recovery after chill stress. The weakest ability to scavenge DPPH• and ABTS+• free radicals as well as the reducing power were shown by the extract obtained from grapevine roots from the seedlings subjected to chill stress. Both free radical scavenging activity and reducing power were observed to increase considerably during recovery after stress. This seems to prove that during the recovery process following chill stress the synthesis of antioxidative compounds in grapevine roots is much more intensive.

Calocera viscosa (Pers.: Fr.) Fr. (Basidiomycota) from Dacrymycetaceae family is a widespread species of mushroom in Poland. The aim of this study was to investigate the content of indole compounds in fruiting bodies and in mycelium cultured in vitro on solid and liquid medium of this species. Fruiting bodies of Calocera viscosa were collected in coniferous forests in south Poland and were used to derive in vitro cultures. The optimal medium composition for cultures was determined. Fresh material: fruiting bodies and mycelium from culture in vitro was frozen and then dried by lyophilization. The crushed dry biomass was extracted with petroleum ether to remove oil fraction, material was dried and extracted with methanol. Analysis of indole compounds was performed in methanol extracts using chromatographic methods: TLC, UV Vis, EIMS and HPLC. This analysis presented in all three extracts the following indole compounds: L-tryptophan, 5-hydroxytryptophan, 5-methyltryptophan, melatonin and indole (contents fluctuated in the range: 0.37 to 11.88 mg/100 g d.w.). 5-hydroxytryptophan contents in all extracts were significant and amounted to 11.88 mg/100 g d.w. in fruiting bodies, and 11.42 in mycelium from liquid cultures and and 10.59 in mycelium from solid cultures. In addition, the fruiting bodies and mycelium from cultures on liquid medium revealed the presence of serotonin (0.39 and 3.19 mg/100 g d.w. respectively).
Od tysięcy lat owocniki grzybów wyższych były wykorzystywane jako źródło pożywienia. Obecnie mogą być pozyskiwane nie tylko ze stanu naturalnego i hodowli komercyjnych lecz również z kultur in vitro prowadzonych w odpowiednich warunkach. Grzyby jadalne są coraz intensywniej badane ze względu na produkcję biologicznie aktywnych metabolitów wtórnych. Biologicznie i leczniczo aktywne metabolity grzybów są używane w terapii tak poważnych schorzeń jak np.: choroby krążenia, cukrzyca, miażdżyca oraz choroby nowotworowe. Niektóre z metabolitów wykazują działanie: przeciwwirusowe, przeciwbakteryjne, przeciwrobacze. W konwencjonalnej medycynie, w leczeniu onkologicznym najdłużej stosowane są polisacharydy grzybowe, stąd też są ich najlepiej poznanymi metabolitami. Innymi grupami szerzej badanych związków są również liczne związki fenolowe, terpenowe, indolowe, witaminy, biopierwiastki (np. selen) o działaniu antyoksydacyjnym. Obiektem badań w ramach niniejszej pracy był gatunek grzyba wielkoowocnikowego Calocera viscosa (Pers.: Fr.) Fr. (Basidiomycota). Owocniki tego gatunku występują pospolicie w lasach iglastych Polski południowej. Materiał do badań stanowiły owocniki zebrane w Nowej Białej (powiat Nowy Targ) oraz mycelium otrzymane z kultur in vitro, które wyprowadzono z owocników tego gatunku. Kultury prowadzono na pożywce stałej i płynnej wytrząsanej wg Oddoux w temperaturze 25 ± 2°C przez 14 dni w warunkach sztucznego oświetlenia o intensywności 900 lx. Owocniki oraz zebrane biomasy z kultur in vitro wysuszono metodą liofilizacji, ekstrahowano eterem naftowym w celu usunięcia frakcji lipidowej, a następnie prowadzono ekstrakcję metanolem. Uzyskane wyciągi metanolowe po zagęszczeniu, oczyszczono metodą preparatywnej chromatografii TLC na płytkach (DC Alufolien Kiesel gel F-254), a następnie analizowano na zawartość związków indolowych metodą HPLC (identyfikacje związków indolowych dokonano metodami spektralnymi UV-VIS i EIMS). Na podstawie przeprowadzonych analiz stwierdzono obecność we wszystkich trzech ekstraktach następujących związków indolowych: tryptofanu, 5-hydroksytryptofanu, 5-metylotryptofanu, melatoniny i indolu (zawartości wahały się w przedziale: 0.37 do 11.88 mg/100g s.m.). Związkiem indolowym występującym w największych ilościach we wszystkich ekstraktach był 5-hydroksytryptofan. Dodatkowo w owocnikach i w mycelium z kultur na podłożu płynnym stwierdzono obecność serotoniny (odpowiednio: 0.39 i 3.19 mg/100g s.m).

The effect of copper stress on betacyanin accumulation and guaiacol peroxidase (GPOD) activity in leaves of different age was evaluated in red beet (Beta vulgaris L. var. Crosby Egyptian) plants. In hydroponic culture, plants were treated with 0.3 μM (control), 50 μM, 100 μM, and 250 μM of CuSO4 for 6 days. Copper was taken up and accumulated in old roots but was not translocated to leaves. However in young leaves, the increase of lipid peroxidation and reduction of growth were evident from day 3 of copper exposure; whereas in old leaves, the lipid peroxidation and growth were the same from either copper-treated or control plants. In response to copper exposure, the betacyanin accumulation was evident in young leaves by day 3, and continued to increase until day 6. Betacyanin only were accumulated in old leaves until day 6, but the contents were from 4 to 5 times lower than those observed in young leaves at the same copper concentrations. GPOD activity increased 3.3- and 1.4-fold in young and old leaves from day 3 of copper treatment respectively, but only in the young leaves was sustained at the same level until day 6. Old roots shown betacyanin in the control plants, but the betacyanin level and growth were reduced with the copper exposure. In contrast, young roots emerged by copper effect also accumulated copper and showed the highest betacyanin content of all plant parts assayed. These results indicate that betacyanin accumulation and GPOD activity are defense responses to copper stress in actively growing organs.

Grapevine seedlings Vitis vinifera were grown in a greenhouse under optimum conditions (soil moisture ca 70%) and under drought stress (soil moisture ca 35%). In addition, some of the plants subjected to drought underwent subsequent regeneration under optimum conditions. Drought stress caused accumulation of total phenolic compounds in grapevine roots, which may indicate that these compounds play an important role in the adaptation of roots to growth under stress conditions. Phenolic acids found in the roots occurred in the ester-bound form only. p-coumaric acid was present in the highest concentrations (6.2 to 10.5 µg/g fresh matter). The content of ferulic acid was lower, ranging from 2.4 to 4.6 µg/g fresh matter. The lowest concentration in grapevine roots was achieved by caffeic acid (2.4 to 2.9 µg/g fresh matter). The levels of p-coumaric and ferulic acids in roots rose significantly under the drought stress, while the concentration of caffeic acid increased during the post-drought recovery period. This may suggest that some of the phenolic acids protect plants under stress conditions. All the extracts from grapevine roots had antioxidative properties, but the antiradical activity of the extracts obtained from roots subjected to drought stress was inferior to the control. The same extracts were also characterised by depressed reducing power. The results imply that tolerance of grapevine to soil drought may be associated with the value of antioxidative potential in root tissues of these plants.
