The objective of the present study was to demonstrate the effect of dietary resistant dextrins, as potential prebiotics, on the intestinal microflora of young rats. Enzyme-resistant dextrin, prepared by heating of potato starch in the presence of hydrochloric (0.1% dsb) and tartaric (40% dsb) acid at 130ºC for 2 h (CA-dextrin). The experiment was performed on 24 Wistar male rats at 3-wk of age, divided by analogues in three experimental groups (control, starch and dextrin). Analyses determined the overall bacterial counts and the counts of Lactobacillus, Bifidobacterium, Bacteroides and Clostridium strains within the feces and cecal contents of rats using fluorescence in situ hybridization method. CA-dextrin had no effect on primary growth indicators (body weight, body weight gain, dietary consumption) or the mass of the small intestine and the cecum, but dextrins caused a reduction in pH and the concentration of ammonia within the cecal contents. That supplementation of diet with resistant dextrins had a positive effect on composition of intestinal microflora in rats. It increased the counts of Bifidobacterium and Lactobacillus strains both in the feces and in the cecum. Moreover, it reduced the counts of Clostridium and Bacteroides strains. These results may suggest that resistant dextrins exerted a prebiotic-like effect in the large intestine.

In the present study, enzyme-resistant dextrin, prepared by heating of potato starch in the presence of hydrochloric (0.1% dsb) and citric (0.1% dsb) acid at 130ºC for 3 h (CA-dextrin), was tested as a source of carbon for probiotic lactobacilli and bifidobacteria cultured with intestinal bacteria isolated from feces of three healthy 70-year old volunteers. The dynamics of growth of bacterial monocultures in broth containing citric acid (CA)-modified dextrin were estimated. It was also investigated whether lactobacilli and bifidobacteria cultured with intestinal bacteria in the presence of resistant dextrin would be able to dominate the intestinal isolates. Prebiotic fermentation of resistant dextrin was analyzed using prebiotic index (PI). In co-cultures of intestinal and probiotic bacteria, the environment was found to be dominated by the probiotic strains of Bifidobacterium and Lactobacillus, which is a beneficial effect.

Opis:
Schinus areira L. is a native plant from South America used for centuries in traditional medicine. Here, we investigate the antimicrobial activity of four essential oils extracted from leaves and fruits of S. areira exhibiting different chemical profiles. The antibacterial activity against the human pathogenic bacteria Staphylococcus aureus susceptible as well as methicillin resistant strain was assessed by the broth microdilution assay. The results showed that the limonene-rich oil extracted from the leaves and fruits have potent antibacterial effect on S. aureus ATCC 25923, while the α-phellandrene-rich fruit oil having a lower content of limonene showed the lowest antibacterial efficacy. In this work, for the first time, we demonstrated the bactericidal activity of essential oils isolated from fruits and leaves of S. areira against susceptible and methicillin resistant S. aureus strains. All results point out the potential use of the S. areira oils as antimicrobial agents to be used, at least against Staphylococcal infections.

Opis:
Resistant starches are type of dietary fibers. However, their physiological effects depend on the way they resist digestion in the gastrointestinal tract. The objective of this study was to examine the hypothesis that new type of RS4 preparations, of in vitro digestibility of about 50%, obtained by cross-linking and acetylation, acts as a prebiotic by increasing short chain fatty acids content in cecum digesta. The rats were fed with diet containing pregelatinized, cross-linked and acetylated starches as a main carbohydrate source. Pregelatinized, but not chemically modified, potato starch was used in the composition of the control diet. After two weeks of experiment the increase of short chain fatty acids contents in ceceum digesta was observed. The intake of starch A, cross-linked only with adipic acid, resulted in increase of about 40% of short chain fatty acids content, whereas starch PA cross-linked with sodium trimetaphosphate and adipic acid of about 50%. The utmost twofold increase was observed in the case of the production of propionic acid. In contrast, the content of butyric acid increased (12%) only as an effect of consumption of starch PA and even decreased (about 30%) in case of starch A. Both RS4 starches caused an increase of the production of acetic acid by more than 40%. No changes in serum biochemistry, liver cholesterol and organ weights of rats were stated.

We aimed to characterize the role of NAD(P)H:quinone oxidoreductase (NQO1) in apoptosis induction by antitumour quinones RH1 (2,5-diaziridinyl-3-hydroxymethyl-6-methyl-1,4-benzoquinone) and MeDZQ (2,5-dimethyl-3,6-diaziridinyl-1,4-benzoquinone). Digitonin-permeabilized FLK cells catalyzed NADPH-dependent single- and two-electron reduction of RH1 and MeDZQ. At equitoxic concentrations, RH1 and MeDZQ induced apoptosis more efficiently than the nonalkylating duroquinone or H2O2. The antioxidant N,N'-diphenyl-p-phenylene diamine, desferrioxamine, and the inhibitor of NQO1 dicumarol, protected against apoptosis induction by all compounds investigated, but to a different extent. The results of multiparameter regression analysis indicate that RH1 and MeDZQ most likely induce apoptosis via NQO1-linked formation of alkylating species but not via NQO1-linked redox cycling.

Multidrug resistance (MDR) of tumour cells is related to the overexpression of ATP-dependent pumps responsible for the active efflux of antitumour agents out of resistant cells. Benzoperimidine and anthrapyridone compounds exhibit comparable cytotoxic activity against sensitive and MDR tumour cells. They diffuse extremely rapidly across the plasma membrane and render the ATP-dependent efflux inefficient. Such uptake could disturb an energy metabolism of normal cells possessing an elevated level of ATP-dependent proteins, especially erythrocytes having a high level of the MRP1, MRP4 and MRP5 proteins. In this study the effect of five antitumour agents: benzoperimidine (BP1), anthrapyridones (CO1, CO7) and reference drugs used in the clinic: doxorubicin (DOX) and pirarubicin (PIRA), on the energetic state in human erythrocytes has been examined. These compounds have various types of structure and kinetics of cellular uptake (slow - DOX, CO7, moderate - PIRA, fast - BP1, CO1) resulting in their different ability to saturate ATP-dependent transporters. The energetic state of erythrocytes was examined by determination of purine nucleotide contents (ATP, ADP, AMP), NAD+ and values of adenylate energy charge (AEC) using an HPLC method. It was found that the level of nucleotides as well as the AEC value of erythrocytes were not changed during 24 h of incubation with these agents independently of their structure and ability to saturate ATP-dependent pumps. This is a very promising result in view of their potential use in the clinic as antitumour drugs against multidrug resistant cancers.
