A total of 94 pectolytic and 60 nonpectolytic Pseudomonas isolates were obtained from 250 samples of rotted vegetable specimens representing various economically important vegetables. The isolates were identified on the basis of standard biochemical tests. Pseudomonas fluorescens biovar V and II and Pseudomonas putida were the most abundant species among pectolytic isolates and Pseudomonas fluorescens biovar I among nonpectolytic ones. Only 3 Pseudomonas viridiflava isolates were identified and all of them were obtained from potato. Isolates of pectolytic phenotype were scattered among nonpectolytic ones irrespective of their taxonomical status. Isolates identified biochemically, as Pseudomonas marginalis were also present in nonpectolytic group. PCR method is unsuitable for identification and differentiation of bacteria belonging to pectolytic fluorescens Pseudomonas group due to great diversity of species. However, the results of PCR amplification of the genes encoding pectate lyase suggest that genes responsible for production of this enzyme may also be present in isolates of nonpectolytic phenotype.
Z 250 prób, z różnych gatunków roślin warzywnych z objawami mokrej zgnilizny wyodrębniono 94 izolaty pektynolitycznych i 60 izolatów nie pektynolitycznych bakterii z rodzaju Pseudomonas. Identyfikację i różnicowanie izolatów przeprowadzono przy zastosowaniu standardowych testów fizjologiczno-biochemicznych. Spośród izolatów z grupy pektolitycznych Pseudomonas najliczniej występował gatunek P. fluorescens, który był reprezentowany przez 4 biowary (oprócz czwartego), przy czym dominowały biowary II i V. Mniej licznie występował gatunek P. putida, a P. viridiflava był reprezentowany jedynie przez 3 izolaty, pochodzące z ziemniaka. Podobna była struktura populacji izolatów z grupy niepektolitycznych Pseudomonas. Metoda PCR okazała się być mało przydatna do wykrywania i różnicowania izolatów z grupy pektolitycznych Pseudomonas ze względu na duże ich zróżnicowanie. Amplifikacja DNA metodą PCR wykazała, że geny kodujące enzym liazy pektynowej występują także w izolatach niepektolitycznych rodzaju Pseudomonas.

Due to the importance of the biological control of plant diseases, testing and introducing new biocontrol-active microorganisms is a major concern among plant pathologists. The causal agent of cotton seedling damping-off disease is Rhizoctonia solani. In this regard, we tried to investigate the antagonistic activities of Pseudomonas aureofaciens (chlororaphis) 30–84 (phenazine producing wild type and non-phenazine producing mutant) strains on R. solani, in comparison with some isolates of P. fluorescent under both in vitro (laboratory) and in vivo (greenhouse) conditions. In the laboratory experiment, the inhibitory effects of all the bacteria, on the growth of R. solani, were evaluated using the dual culture procedure. Results showed that five isolates of P. fluorescent along with both strains of P. aureofaciens significantly inhibited the growth of R. solani. Effective bacterial antagonists were then evaluated in a greenhouse experiment where cotton seeds were coated with their suspensions and were sown in pasteurised field-soil. The soil had been pre-inoculated with a virulent isolate of R. solani. The efficacy of the bacterial antagonists was evaluated by counting the number of surviving seedlings in different treatments, at 15 and 60 days after sowing, for determining pre- and post-emergence damping-off incidence. According to the results of the greenhouse experiment, at both intervals, two isolates of P. fluorescens along with both strains of P. aureofaciens caused significant increases in the number of healthy seedlings, in comparison with the untreated control, and a commonly used fungicide (carboxin-thiram). The efficacy of phenazine producing a wild type strain of P. aureofaciens was higher than its non-phenazine producing mutant, indicating that phenazine plays an important role in the antagonistic activity of P. aureofaciens. Effective bacterial antagonists were then studied for their antagonistic mechanisms. The results showed that all four bacteria employed different mechanisms. The bacteria produced siderophore, and volatile metabolites and non-volatile metabolites, in their antagonistic activities. The results of this study suggest that P. auerofaciens may be a new biocontrol agent for controlling cotton seedling mortality disease.

Opis:
The suitability of low-cost impedance sensors for microbiological purposes and biofilm growth monitoring was evaluated. The sensors with interdigitated electrodes were fabricated in PCB and LTCC technologies. The electrodes were golden (LTCC) or gold-plated (PCB) to provide surface stability. The sensors were used for monitoring growth and degradation of the reference ATCC 15442 Pseudomonas aeruginosa strain biofilm in invitro setting. During the experiment, the impedance spectra of the sensors were measured and analysed using electrical equivalent circuit (EEC) modelling. Additionally, the process of adhesion and growth of bacteria on a sensor’s surface was assessed by means of the optical and SEM microscopy. EEC and SEM microscopic analysis revealed that the gold layer on copper electrodes was not tight, making the PCB sensors susceptible to corrosion while the LTCC sensors had good surface stability. It turned out that the LTCC sensors are suitable for monitoring pseudomonal biofilm and the PCB sensors are good detectors of ongoing stages of biofilm formation.

Bioremediation is based on microorganisms able to use pollutants either as a source of carbon or in co-metabolism, and is a promising strategy in cleaning the environment. Using soil contaminated with petroleum products from an industrial area in Saudi Arabia (Jubail), and after enrichment with the polycyclic aromatic hydrocarbon (PAH) naphthalene, a Methylobacterium radiotolerans strain (N7A0) was isolated that can grow in the presence of naphthalene as the sole source of carbon. M. radiotolerans is known to be resistant to gamma radiation, and this is the first documented report of a strain of this bacterium using a PAH as the sole source of carbon. The commonly reported Pseudomonas aeruginosa (strain N7B1) that biodegrades naphthalene was also identified, and gas chromatography analyses have shown that the biodegradation of naphthalene by M. radiotolerans and P. aeruginosa did follow both the salicylate and phthalate pathways.

Plant growth-promoting rhizobacteria (PGPR) are a collection of microorganisms often used to support and promote plant development and combat plant infectious diseases with various biological control methods. The most significant restricting factors for agricultural productivity worldwide are abiotic constraints. In the present study, seven bacterial isolates from the rice rhizosphere were selected for detailed tests based on results obtained in experiments determining the ACC deaminase synthesis and drought tolerance at !0.30 MPa PEG level. Screening results of the stress tolerance analysis of the seven isolates for elevated temperature (50°C), alkalinity (10% NaCl), and drought (-1.2 MPa) showed that abiotic stress resistance was less prevalent in DRO2, DRO13, and DRO43 isolates than in DRO17, DRO28, DRO35, and DRO51 isolates. During the study, it was observed that DRO17, DRO28, and DRO51 tended to maintain similar cell density at -0.73 MPa PEG level, as observed at -0.30 MPa stress condition. No bacterial growth was observed at higher PEG level (-1.12 MPa) for any bacterial isolate. Four strains of Pseudomonas (DRO17, DRO28, DRO35, and DRO51) exhibited salinity and temperature tolerance. Antifungal screening using the bangle method showed that DRO35 was highly antagonistic towards Rhizoctonia solani 4633, followed by Fusarium moniliforme 4223, with an inhibition of 64.3% and 48%, respectively. The DRO28 isolate exhibited 72.5% growth inhibition for Fusarium moniliforme 4223, while the DRO51 isolate showed 38.2% growth inhibition for Bipolaris hawaiiensis 2445. DRO17 reduced the growth of Rhizoctonia solani 4633, and Curvularia lunata 350 by 36% and 31%, respectively. In conclusion, the screening of bacterial strains with promising stress tolerance and antifungal characteristics could support farmers to achieve the required positive outcomes in the agriculture field.

Endophytes are an important constituent of sustainable agriculture because of their ability to produce a large number of agriculturally important metabolites. A salt-tolerant fluorescent green pigment-producing endophytic bacterium was isolated on 2.5% NaCl-supplemented nutrient agar from the leaf samples of Zanthoxylum alatum Roxb. The isolate Z1B4 was identified as Pseudomonas fluorescens based on morphological features, fatty acid methyl ester analysis, biochemical tests, and 16S rRNA gene sequencing. P. fluorescens Z1B4 showed positive results for tricalcium phosphate solubilization; 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity; and production of auxins, siderophores, hydrogen cyanide, and ammonia. P. fluorescens Z1B4 also showed strong antagonistic activity against Curvularia lunata (MTCC 283), Fusarium verticillioides (MTCC 3322), and Alternaria alternata (MTCC 1362) and exhibited stress tolerance to a wide range of temperature and pH and concentrations of NaCl and calcium salts. Under natural conditions, following inoculation with the isolate Z1B4, a significant increase in the growth of pea and maize test plants in pots was observed compared to that of uninoculated control plants. The rifampicin-resistant mutant Z1B4Rif was recovered from the roots, shoots, and leaves of the test plants, indicating that the isolated endophytic bacterium can grow well within different plant tissues. The present study indicated that the endophytic bacterium P. fluorescens Z1B4 can be used as a bacterial inoculant in stressed environments for sustainable agriculture.

Background . Melanin finds enormous applications in different industries for its unique photoprotective and anti-oxidant properties. Due to its emerging demand, scientific researchers are putting efforts to unravel more microorganisms with a potential of producing melanin on large scale. Hence, the present study was aimed at the isolation of extracellular melanin producing microorganisms from lime quarries of Karnataka, India. Besides this, the tyrosinase gene governing melanin synthesis in different organisms were compared in silico to understand its evolutionary aspects. Material and methods . Melanin producing microorganisms were screened on tyrosine gelatin beef extract agar medium. Potential isolate was explored for submerged production of melanin in broth containing L-tyrosine. Melanin was characterized by UV-Vis spectroscopy, thin layer and high performance liquid chromatographic techniques. Antibacterial activity of melanin was performed by agar well assay. Comparative tyrosinase gene sequence analysis was performed by using Geneious 2021.1 trial version software. Results . Pseudomonas otitidis DDB2 was found to be potential for melanin production. No antibacterial activity was exerted by the melanin against tested pathogens. The in silico studies showed that the common central domain of tyrosinase protein sequence of selected Pseudomonas sps. exhibited 100% identity with the common central domain of Homo sapiens tyrosinase (NP_000363.1). Conclusions . Our study shows the production of melanin in good quantities by the isolate Pseudomonas otitidis DDB2 which can be explored for scale-up process. Since the melanin formed is of eumelanin type and the tyrosinase gene sequence of several Pseudomonas sp. showed relatedness to humans, this molecule may be further developed for sunscreen formulations.

Twenty-six cabbage heads stored under typical conditions in a storage hall in Moravia, Czech Republic, were tested for the presence of bacteria by the method of isolation from three different parts of the cabbage heads. Isolations were carried out from stalks, inner and superficial leaves. Two samplings were done; in November 2015 and February 2016. Bacterial cultures were sequenced in the part of 16S rRNA region; bacteria were identified according to the sequences obtained. The most prevalent bacteria were of the genus Pseudomonas. Genera: Klebsiella, Erwinia, Pantoea, Bacillus were also identified. The results provided an interesting insight into the bacterial spectrum in stored cabbage heads and their dynamics during storage. Th e nucleotide sequences which were found were saved in GenBank/NCBI under accession numbers KX160104-KX160145.

Fluorescent Pseudomonas (FP) is a major group of plant growth promoting rhizobacteria and a well-known synthesizer of siderophores, which imparts a selective advantage on rhizosphere competence and their biocontrol traits. The present study was aimed at examining the factors affecting the production of siderophores and their potential biocontrol traits. Sixteen FP isolates were shortlisted based on their siderophore-producing ability in chrome azural S medium. The isolates were checked for variations in siderophore production under varying incubation times, temperatures, pH, iron (Fe3+) concentrations and mutagens. In addition, the iron binding affinity of siderophores, mycelial inhibition assay and plant growth promotion traits were assessed. Results showed that the siderophore production was highly influenced by the time of incubation, changes in pH, temperature and iron concentration. Chemical characterization showed that the produced siderophores were hydroxamates. Maximum siderophore production was observed at pH 7 whereas UV and EtBr exposure invariably suppressed siderophore production drastically in all isolates. All FPs from maize rhizosphere showed excellent siderophore production which could be due to the competence in strategy-II of the plant rhizosphere and significant growth inhibition on Fusarium oxysporum. Our results suggest the inclination of siderophores to iron, in terms of various criteria affecting production and the possible role of environmental mutations that affect the natural iron harvesting mechanism.

Pseudomonas syringae pv. syringae (Pss) constitutes a diverse group of bacterial strains that cause canker of stone fruits, blight of cereals and red streak of sugarcane. The purpose of this study was to determine how diverse Iranian strains of Pss are when they come from different hosts. We compared a total of 32 Pss strains isolated from stone fruits, barley, wheat and sugarcane from different geographical regions of Iran based on their phenotypic and molecular properties. Strains showed some variation regarding carbon and nitrogen utilization. Pss strains were similar in their protein banding patterns. Additional bands were found in sugarcane strains. Most strains showed one indigenous plasmid DNA and a few had two and some none. The genes of syrB and syrD encoding syringomycin synthesis and secretion, respectively, were amplified using specific primers in polymerase chain reaction. Syringomycin, producing strains amplified two DNA fragments of 752 and 446 bp representing syrB and syrD genes, respectively. Primer specificity was shown for Pss using various genera. Based on the results of this study, it is suggested that Pss strains from different hosts and geographical regions show diversity in phenotypic and molecular characters. It is thought that phenotypic variation is due to adaptation to specific hosts and niches for survival and pathogenicity.
