Opis:
Animal models based on N-methyl-d-aspartate (NMDA) receptor blockade have been widely reported. Ketamine and MK-801, the two noncompetitive antagonists of NMDA receptors, produce behaviors related to schizophrenia and exacerbated symptoms in patients with schizophrenia. The study presented here investigated the effect of subchronic dosing (once-daily, 7 day) of histamine H3 receptor (H3R) antagonists, ciproxifan (CPX) (3 mg/kg, i.p.) and clobenpropit (CBP) (15 mg/kg, i.p) including clozapine (CLZ) (3.0 mg/kg, i.p.) and chlorpromazine (CPZ) (3.0 mg/kg, i.p.), the atypical and typical antipsychotic, respectively, on MK-801(0.2 mg/kg, i.p.)-induced locomotor activity, and dopamine and histamine levels in rats. Atypical and typical antipsychotic was used to serve as clinically relevant reference agents to compare the effects of the H3R antagonists. MK-801 significantly increased horizontal activity which was reduced with CPX and CBP. The attenuation of MK-801-induced locomotor hyperactivity produced CPX and CBP were comparable to CLZ and CPZ. Dopamine and histamine levels were measured in striatum and hypothalamus, respectively, of rat brain. The MK-801 induced increase of the striatal dopamine level was reduced in rats pretreated with CPX and CBP including CLZ. CPZ also significantly lowered striatal dopamine levels, though the decrease was less robust compared to CLZ, CPX and CBP. MK-801 increased histamine content although to a lesser degree. Subchronic treatment with CPX and CBP exhibited further increased histamine levels in the hypothalamus compared to MK-801 treatment alone. Histamine H3 receptor agonist, R-α methylhistamine (10 mg/kg, i.p.) counteracted the effect of CPX and CBP. The findings of the present study support our previous work showing positive effects of CPX and CBP on MK-801-induced schizophrenia like behaviors in rodents. However, clinical studies have reported no antipsychotic effects with histamine H3 receptor antagonists.

Opis:
Introduction and objective. Young green barley is the most valuable source of nutrients and bioactive substances. It has a broad spectrum of health-promoting properties such as antioxidant, anti-inflammatory, hypoglycaemic, anti-depressant, anti-atherosclerotic and anticancer. The presented study is an attempt to extend this knowledge with particular emphasis on the possibility of using green barley in colon cancer prevention. Materials and method. Extracts were prepared on the basis of two commercial products: ground dried barley grass (YGB INT) and powder of young green barley juice (YGB GW). Their influence on colon epithelial cells (CCD841 CoN) viability and proliferation were analyzed by LDH and MTT assays. Anticancer properties of extracts were screened on colon cancer cell lines (LS180, HT-29) by MTT and BrdU assays. Changes in cells morphology induced by extracts were investigated after May-Grünwald-Giemsa staining. Results. Tested extracts were not toxic against CCD841 CoN and did not affected their proliferation or morphology (LDH test, MTT test, microscopy observation). The MTT revealed that extracts significantly inhibited proliferation of colon cancer cells in a dose-dependent manner. Results of BrdU test confirmed antiproliferative properties of extracts, but opposite to MTT test, indicated YGB GW as a better anticancer agent. Light microscopy observation proved the data obtained from both MTT and BrdU tests and additionally suggested the ability of the extracts to induce necrosis in LS180 and HT-29 cells. Conclusions. The study demonstrated that YGB extracts specifically inhibit proliferation of colon cancer cells without any undesirable effect on colon epithelial cells. Obtained results will provide a rationele for the future development of dietary supplements which could be beneficial in colon cancer chemoprevention.

Opis:
Introduction and objective. Since antiquity, C. babylonica (L.) L. extracts has been used as a remedy for primary health care in traditional medicine. In this study, a total of seven different crude extracts (acetone, chloroform, hexane, ethylacetate, methanol, ethanol and water) from branches and leaves of C. babylonica (L.) L. were prepared to determine antimicrobial and antiproliferative activity against cancer cell lines. Materials and method. MIC assay was used for antimicrobial activity against gram positive and gram negative bacteria, and one yeast. MTT assay was applied to screen the antiproliferative activity of seven extracts, and to determine dose- and time-dependent effects of the aceton extract on A549, PC-3, MCF-7, and HeLa cell lines. Results. The aceton extract of C.babylonica (L.) L. showed the best antibacterial activity against Bacillus cereus, P. aeruginosa and C. albicans (MIC: 1.6 mg/mL). GC-MS analyses allowed six compounds to be determined; the main constituents of acetone extract from C. babylonica (L.) L. were diacetone alcohol (53.47 %), 1-dexadecene (10.19 %) and 1-tetradecene (8.67 %). In addition, seven different solvent extracts at 500 μg/mL caused antiproliferative activity between 84% – 88%, compared to control. Dose-dependent effects of the extracts on A549 cells indicated that chloroform, ethyl acetate, and aceton extract were the most effective extracts with the IC50 values of 9, 33, and 36 μg/mL, respectively. Conclusions. The results clearly demonstrate that C. babylonica (L.) L. exhibited a strong antimicrobial effect and antiproliferative activity against cancer cells in vitro. Further studies are required to isolate and characterize the active pure compounds responsible for the antimicrobial and antiproliferative activities.
