Opis:
The micromorphological studies of in vitro and field transferred plants of Duranta erecta were conducted in the present investigation. The nodal shoot segments were used as explants for the establishment of cultures. The shoots were multiplied on MS medium (Murashige and Skoog) supplemented with 6-benzylaminopurine (BAP), kinetin (Kin) and Naphthalene acetic acid (NAA). In vitro multiplied shoots were rooted on half strength MS medium augmented with indole-3 butyric acid (IBA). The rooted plantlets were successfully hardened and field transferred. A comparative foliar micromorphological studies were made of in vitro grown and field transferred plants to understand the developmental changes of micro-external structures of plants towards environment. The differences in leaf constants such as stomata, vein-islets and termination numbers and trichomes across the environments imply the developmental changes of micropropagated plants of Duranta erecta.

Foliar micromorphological study of plants describes the distinct structure and functions of foliar tissues. The structural analysis of G. superba epidermal tissues and leaf architecture revealed the stomatal type, morphology and their orientation, cuticular wax deposition, crystal arrangement and venation pattern. The stomatal density was observed maximum on abaxial epidermis. The epidermis contained ordered stomatal pattern with anomocytic stomata. The stomatal density and stomatal index was 12.0 and 29.0 respectively. The parallel lower order veins and perpendicular higher order veins form a grid-like network of venation pattern in G. superba. Con-vallaria type of cuticular wax was observed. The results of the foliar features could be applied in systematic studies and also to predict the environmental factors responsible for the development of certain leaf parameters.

The synthesis of zinc oxide nanoparticles (ZnO) using herbal extract is always environmentally benign. The present investigation focuses on the synthesis of ZnO nanoparticles using aqueous extracts of various parts of Passiflora edulis Sims. f. flavicarpa Deg. (Passion fruit). The plant is rich in alkaloids, glycosides, flavonoids and triterpenes. UV-Visible double beam spectrophotometric analysis was used for characterization of ZnO nanoparticles with different time intervals which were synthesized by plant extracts. The bio-reduction completes in two hr incubation of reaction mixtures with strong broad peaks at 332 nm from leaves, 296 nm from stem and 326 nm from flower extracts. Hence the biosynthesis of ZnO nanoparticles using aqueous extracts of P. edulis could be an alternative to chemical methods and exploited in various biofields.

The direct organogenesis from nodal meristems of Peperomia pellucida L. was reported in this study. The explants were sterilized using 0.1% mercuric chloride. Since, the plant tissues are very soft and delicate 3 min treatment is optimum for surface sterilization of the explants. The adventitious shoots were induced on MS medium augmented with 2.0 mg/L BAP. Maximum 96% explants responded on this medium combination with 12.6 ±0.74 shoots per explant (4.2 ±0.44 cm average lengths). The shoots were further multiplied on MS medium + 0.5 mg/L each of BAP and Kin, resulted in maximum shoots formation (127.2 ±0.30 shoots per explant with 7.7 ±0.30 cm in length) after 4 week. The effects of auxins were tested for induction of in vitro roots from the cut ends of the shoots. The in vitro multiplied shoots were rooted on half strength MS medium fortified with 1.5 mg/L IBA. Maximum number of roots per shoot (10.8 ±0.39 roots with 6.8 ±0.33 cm length) was induced with IBA as compared to IAA. The rooted plantlets were carefully removed from the medium, washed properly and transferred to the paper cups contained soilrite. These were kept in the greenhouse for 4-6 weeks and moistened with aqueous solution of one-fourth strength MS salts. The hardened plantlets were finally transplanted in the field under natural conditions with 100% survival rate. The developed protocol can be used for mass multiplication of this medicinal plant which can help to fulfill the gap in the demand and supply of this epiphytic medicinal plant.
