Opis:
Porcine circovirus type 2 (PCV2) is an economically important swine pathogen and, although small, it has the highest evolution rate among DNA viruses. Commercial PCV2 commercial vaccines are inactivated PCV2 isolates or a subunit vaccine based on the Cap protein of PCV2. Currently, PCV2 VLPs of individual subtype vaccines are available. Although the main prevalent genotype worldwide is PCV2b, the emerging subtype PCV2d subtype is also increasingly associated with PCV disease. The aim of the study was to evaluate the protective efficacy of VLP based on the PCV2b and 2d subtypes against the mixed challenge of two hypotype PCV2 in mice. Thirty-six female SPV Kunming mice were immunized twice with PCV2b and 2d VLPs, then challenged with PCV2b and PCV2d, to assess the immunogenicity and effectiveness of the VLPs. Vaccination of the mice with PCV2b and 2d VLPs elicited a robust antibody response specific for the PCV2. The virus load detected in the 2b and 2d spleen vaccine group was the lowest compared to other groups. Furthermore, there was no pathological damage in the HE stained sections of the 2b and 2d spleen vaccine, and no virus was detected in the immunohistochemical sections. Our data suggest that the mixed PCV2b and 2d VLP vaccine could protect mice from challenge with the mixed infection of PCV2b and PCV2d.

Opis:
Porcine circovirus type 2 (PCV2) has been known as a causative agent of reproductive failure in the sow. In the present study mouse model was used to investigate PCV2 infection. In order to investigate whether PCV2 can induce lesions of spermatocytes and oocytes, 6 male and 6 female mice were each inoculated intraperitoneally with PCV2b, and 3 male and 3 female mice mock-infected with cell culture supernatant served as controls. Samples of testes and ovaries from PCV2b-inoculated and mock-infected mice were investigated using PCR, histopathological, ultrastructural and immunofluorescent histochemical methods at 14 and 21 day post infection (dpi). The study revealed that in the virus-inoculated mice leydig cells in testes and granulosa cells in ovaries were degenerated, and a small number of spermatocytes and oocytes showed apoptosis. Positive PCV2b antigen signals were also observed in these apoptotic cells. It can be suggested that PCV2 can cause lesions of spermatocyte and oocyte prior to zygotes formation in its host.

Opis:
In order to investigate whether PCV2b can be transmitted across the placenta in sows thereby causing vertical infection of fetuses, six sows in 75 day of pregnancy were either intramuscularly inoculated with a PCV2b culture supernatant (n = 4) or mock infected with cell culture supernatant (n = 2). At parturition, 3 newborn piglets from each litter were randomly selected and euthanatized prior to suckling. Samples of the mesenteric lymph nodes, spleens, kidneys, hearts and lungs were collected for PCR, histopathological and immunohistochemical assays. The results showed that the newborn piglets from PCV2b-infected sows had histopathological lesions in the tested tissues. Moreover, PCV2b antigen and DNA were detected as well. These findings therefore suggested that porcine circovirus type 2b can be transmitted across the placenta of sows, thereby leading to PCV2b vertical infection of the fetuses.

Opis:
Serology plays an important role in laboratory diagnosis of leptospirosis. Apart from the most often used microscopic agglutination test (MAT), enzyme-linked immunosorbent assay (ELISA) seems to be useful especially in screenings of animal herds. The ELISA used for detection of antibodies against selected Leptospira serogroups in swine serum samples was investigated during the study. An essential element of this test is heat-stable antigenic preparation from cultures of Leptospira interrogans serovars Icterohaemorrhagiae, Pomona and L. borgpetersenii serovar Sejroe. The aim of the present study was to identify and analyze ELISA heat-stable antigen fractions playing a role in the reaction with leptospiral antibodies indicated in swine serum. Reactivity of the three-component antigenic preparation was compared in immunoblotting with reactivity of six heat-stable antigenic preparations made from the following single serovars: L. interrogans serovars Icterohaemorrhagiae, Pomona, Canicola, L. borgpetersenii serovars Sejroe, Tarassovi and L. kirshneri serovar Grippotyphosa. All antigenic preparations were submitted to SDS-PAGE and transferred to a nitrocellulose membrane using a semidry system. After the transfer, the membrane was incubated with diluted swine serum containing antibodies specific for one of the six above mentioned Leptospira serovars. For the three-component antigenic preparation and antigens prepared from single serovars the immunoblot revealed reaction of sera with fractions of the 20-26 kDa region and around the 14.5 kDa region. The investigated heat-stable Leptospira antigenic preparation contains fractions demonstrating serogroup- and species-specificity. Fraction 20-26 kDa showed serogroup-specific activity, whereas the fraction around 14.5 kDa showed species-specific activity.

Opis:
Swine are one of significant reservoirs and sources of Leptospira infections for man. Serological screenings help to effectively control the epidemiological situation in swine herds and to prevent transmission of Leptospira from animals to man. The purpose of this study was to investigate, by the use of serological methods, the prevalence of infections caused by selected Leptospira serogroups in swine population in Poland. A total of 7112 swine serum samples were examined. The samples were collected from January to October 2008 and came from 280 counties situated in all 16 provinces of Poland. All sera were examined preliminary by enzyme-linked immunosorbent assay (ELISA) using heat-stable antigenic preparation. The samples positive or doubtful in ELISA were investigated by microscopic agglutination test (MAT) with use of serovars Icterohaemorrhagiae, Pomona, Canicola, Sejroe, Tarassovi and Grippotyphosa. Of the collected sera examined by ELISA 73 (1.02%) samples were positive, 85 (1.20%) – doubtful and 6954 – negative. Among ELISA-positive and doubtful sera 64 samples (coming from 14 provinces) were recognized in MAT as positive. Among MAT positive samples 42.19% of sera demonstrated titres with serovar Pomona, 32.81% – with Sejroe, 14.06% – with Icterohaemorrhagiae, 6.25% – with Tarassovi, 3.13% – with Grippotyphosa and 1.56% with Canicola.

The purpose of this study was to analyze the acute effect of whole body vibration exercise (WBVE) on clinical parameters and blood values in horses. Seven horses were exposed to a 10 min WBVE at a frequency of 15-21 Hz. Clinical parameters and venous blood samples were taken before and directly after WBVE. Acute short-term WBVE produced a decrease in serum cortisol (p=0.02) and creatine-kinase (p=0.02) values. Clinical parameters, hematology, fibrinogen, lactate, IGF-I, GGT, creatinine, myeloperoxidase activity and bone marker values were not significantly changed by WBVE. In adult sound horses WBVE was well tolerated and did not cause any sign of measured discomfort.

The effect of pregnancy on bone metabolism was investigated in healthy mares. Venous blood samples were collected 7 times from 19 multiparous mares starting at 20-weeks pre-parturition, continuing 6 times in 4-week intervals, including the week of parturition and one week after parturition. Serum concentrations of osteocalcin (OC) and carboxy-terminal cross-linking telopeptide of type I collagen (CTX-I) were determined. Measurement cycles and age had a significant (p < 0.01) influence on OC and CTX-I values. Pregnancy influenced bone metabolism with peak bone formation and resorption values around the time of parturition.

This study aimed to determine the effects of dexamethasone and minocycline alone and combined treatment with N-acetylcysteine (NAC) and vitamin E on serum coenzyme Q10 (CoQ10) and matrix metalloproteinase-9 (MMP-9) levels in rats administered aflatoxin B1 (AFB1). The study was carried out on 66 male Wistar rats. Following the intraperitoneal (IP) administration of AFB1 at dose of 2 mg/kg, minocycline (45 and 90 mg/kg, IP) and dexamethasone (5 and 20 mg/kg, IP) were administered alone and combined with NAC (200 mg/kg, IP) and vitamin E (600 mg/kg, IP). CoQ10 and MMP-9 levels were analyzed using the HPLC-UV method and a commercial kit by ELISA, respectively. AFB1 increased MMP-9 level and decreased CoQ10 level compared to the control group. After dexamethasone and minocycline administration, there is no increase in CoQ10 level, which is caused by AFB1. However, dexamethasone and minocycline combined with NAC+vitamin E caused significant increases in CoQ10 levels. Dexamethasone and minocycline alone and combined with NAC+vitamin E decreased MMP-9 levels compared to the single AFB1 treated group. The use of MMPs inhibitors and oxidative stress-reducing agents is anticipated to be beneficial in the poisoning with AFB1.

Opis:
The purpose of the present review was a comparison of the abortions caused by EAV and EHV-1 viruses over the 34 years. A total of 452 tissues samples from aborted fetuses (347) or foals (105) stillborn or newborn that died within 72 hours were investigated. The material for the examinations came from different farms located throughout Poland. The tissue homogenates were examined by using virus isolation test in RK-13 and Vero cell lines and the cytopathic agent was confirmed as EHV-1 by the direct fluorescent antibody test or as EAV by the indirect fluorescent antibody test. The study indicated that EAV was isolated (104 cases, 23%) almost as equally often as EHV-1 (116 cases, 25.6%). Both, equid herpesvirus-associated abortion and the abortion induced by EAV were characterized by cyclicity. The percentage of EAV and EHV-1 isolation alternately reduced and increased, but the increase of isolation of one virus was accompanied by the decrease of the other. The domination of one virus over the other occurred in cycles of a few years.

Opis:
The aim of this study was to evaluate the acute phase response of dairy cows to repeated liver biopsy in order to estimate the safety of this procedure during the transition period. Liver biopsies (up to 1000 mg of liver tissue) were conducted twice a day, 7 days before expected parturition and 3 days after calving. The number of needle insertions for each biopsy was recorded and was dependent on the amount of obtained tissue. Blood samples were taken on day 7 before expected parturition, then on days 3, 4, 7 and 14 after calving. Body temperature was measured daily in all 30 cows from day 3 until day 14 after calving. The concentrations of haptoglobin, serum amyloid A, fibrinogen and interleukin-6 were determined in serum and plasma. In 16.7% of cows, the rectal body temperature rose by ≥ 0.5oC on the day after liver biopsy. Although the concentrations of haptoglobin, serum amyloid A and fibrinogen increased significantly after calving (p<0.01), there was no influence of the number of biopsies on the acute phase reaction and repeated biopsy during the transition period had no effect on body temperature. Therefore, the procedure may be regarded as safe for cows during the transition period.
