The environment of animals is inhabited by enormous fungal species, but only a few hundreds are pathogenic for mammals. Most of potentially pathogenic fungal species, excluding dermatophytes, seldom cause the disease in immunocompetent hosts. Data from literature indicate, that an immune system and endothermy are foundations for this mammalian relative resistance to fungal systemic infections. Stable and high temperature of the body restricts invasion and growth of potentially pathogenic fungi. Together with elevated metabolism it supports the effectiveness of mammalian immunity. The innate immunity is assigned to prevent the invasion of various microbes (including fungi) to the hosts’ organism. It consists of cellular receptors and several humoral factors as the Antimicrobial Peptides. If the physical barriers fail in stopping the invader, it is recognized as “alien” by multiple Pattern Recognition Receptors (PRRs) like Toll Like Receptors (TLRs) expressed by cells of innate immunity and/ or C-type lectins. At the same time innate inflammation begins and the complement cascade is activated. These mechanisms are able to stop and clear some fungal infections. During existing infection the adaptive immunity is induced. This review aims to show the role of mammalian endothermy and to point the most important elements of innate antifungal immunity.

Prevalence of mecA, blaZ, tetO/K/M, ermA/B/C, aph, and vanA/B/C/D genes conferring resistance to oxacillin, penicillin, tetracycline, erythromycin, gentamicin, and vancomycin was investigated in 65 staphylococcal isolates belonging to twelve species obtained from ready-to-eat porcine, bovine, and chicken products. All coagulase negative staphylococci (CNS) and S. aureus isolates harbored at least one antibiotic resistance gene. None of the S. aureus possessed more than three genes, while 25% of the CNS isolates harbored at least four genes encoding resistance to clinically used antibiotics. In 15 CNS isolates the mecA gene was detected, while all S. aureus isolates were mecA-negative. We demonstrate that in ready-to-eat food the frequency of CNS harboring multiple antibiotic resistance genes is higher than that of multiple resistant S. aureus, meaning that food can be considered a reservoir of bacteria containing genes potentially contributing to the evolution of antibiotic resistance in staphylococci.

Opis:
The efficiency of somatic cell cloning (somatic cell nuclear transfer; SCNT) as well as in vitro fertilization/in vitro embryo production (IVF/IVP) in mammals stay at relatively same level for over a decade. Despite plenty of different approaches none satisfactory break-through took place. In this article, we briefly summarize the implementation of mesenchymal stem cells (MSCs) for experimental embryology. The advantages of using MSCs as nuclear donors in somatic cell cloning and in vitro embryo culture are described. The description of results obtained with these cells in mammalian embryo genomic engineering is presented.

Polychlorinated biphenyls (PCBs) are a group of persistent environmental pollutants that impair cattle reproduction. Among other effects, PCBs can disturb the intracellular mobilization of Ca²⁺ in several cell types. Hence, it is possible that they disrupt the transduction of intracellular signals generated from gonadotropin (FSH/LH) receptors. In steroidogenic ovarian cells, a defect in Ca²⁺ mobilization may have a detrimental influence on two important processes: the secretion of steroids (E2 or/and P4) and their morphological and functional differentiation. The aim of this study was to determine the influence of PCBs: 126 (dioxin-like) 77 (ambivalent) and 153 (estrogen-like) and a mixture of PCBs (Aroclor 1248) on these processes. Bovine granulosa and luteal cells were incubated for 72 hrs with PCBs (100 ng/ml), followed by Fura 2AM dye, and the fluctuations in intracellular Ca²⁺ mobilization after FSH/LH treatment were determined using an inverted microscope coupled with a CCD camera. The intensity and area of fluorescence excited by UV light were detected in the green spectrum of visible light. Aroclor 1248 and PCBs 153 and 77 significantly decreased (P < 0.01-0.001) the effect of FSH on intracellular Ca²⁺ mobilization in granulosa cells. In luteal cells, the most effective PCB on this process was PCB 77. The results revealed adverse effects of PCBs on the mobilization of intracellular Ca²⁺. Moreover, the estrogen-like congeners were found to more effectively disturb this process than the dioxin-like PCB 126. Hence, it is possible for PCBs to have a negative influence on reproductive processes by affecting calcium mobilization.

The presence of the flaA, cadF, cdtB and iam genes of Campylobacter spp. was determined with the PCR method. The materials to investigate were 56 C. jejuni and 23 C. coli strains isolated from clinical samples (children and domestic animals). It was found that all of the Campylobacter spp. isolates from children with diarrhoea and domestic animals had cadF gene, responsible for adherence. The flaA gene was present in all Campylobacter spp. isolates derived from children and cats. Occurrence of flaA gene was confirmed in 100% of C. jejuni strains obtained from dogs. The high prevalence of the cdtB gene associated with toxin production was observed in this study (100%-Campylobacter spp. isolates obtained from dogs and cats, 97.9%-Campylobacter spp. isolates from children). The isolates showed a wide variation for the presence of iam gene. The lowest prevalence (23.5%) was detected in Campylobacter spp. obtained from dogs. The highest rates of iam detection (91.6%) were revealed in C. coli isolates from children.

Lymphocytic infiltrations located in the extracellular matrix often accompany canine skin cancer. They can be characterised as an inflammatory infiltration and/or a second tumour – lymphoma. The aim of this study was an immunohistochemical analysis of a lymphocytic infiltration which accompanies spontaneous skin cancer. Twenty basal cell carcinoma, 20 non-keratinizing squamous cell carcinoma, 20 keratinizing squamous cell carcinoma and 8 sebaceous gland carcinoma samples which were accompanied by a lymphocytic infiltration and/or secondary lymphatic follicles were verified histopathologically. The expression of bcl-2, CD3, CD79α, Ki-67, MCM-3 and MCM-7 in the lymphocytic infiltration was evaluated. Four types of lymphocytic infiltrations were found: I – diffuse bcl-2⁺, II – diffuse bcl-2⁻, III – follicular bcl-2⁺/⁻ where the centre was bcl-2⁻, and the marginal zone of the follicles and the extrafollicular area were bcl-2⁺ and IV – aggregated bcl-2⁺, where the centre and periphery were bcl-2⁺. The I and IV type corresponds to lymphoma, II type is non-neoplastic immune response and III type suggest reactive follicular hyperplasia. The proliferation of lymphocytes which demonstrated the expression of neoplastic markers (I and IV), suggests preneoplastic phase (pseudolymphoma) or lymphoma – the second independent tumour. A high proliferative index of the follicular blc-2⁺/⁻ follicular infiltration indicates an increased immunological response of the host against skin cancer.

Although prostaglandin E₂ (PGE₂) is a pro-inflammatory mediator, it also produces some effect which is anti-inflammatory in character. It is suggested that one of the mechanisms responsible for the latter effect is the increased synthesis of IL-10. The aim of this study has been to determine the influence of PGE₂ on IL-10 production by bovine CD⁴⁺ and CD⁸⁺ T cells and NK cells. With this aim, peripheral blood mononuclear cells collected from 12-month-old heifers (n = 10) were treated without or with PGE₂ (10⁻⁶ M). Flow cytometric analysis showed that PGE₂ caused a reduction in the percentage of IL-10 producing CD⁴⁺ T cells (P < 0.001), while leaving the secretion of this cytokine by CD⁸⁺ T cells and NK cells unaffected. This seems to indicate that PGE₂ in cattle does not produce an anti-inflammatory effect by increasing the synthesis of IL-10; contrary to this, it may aggravate an inflammatory response by inhibiting the secretion of this cytokine by CD⁴⁺ T cells.

The present study describes the distribution of CD4+CD8+ double-positive (DP) T cells in various immune compartments of mice with ovalbumin (OVA)-induced allergic asthma. It was found that the absolute number of DP T cells was considerably increased in the mediastinal lymph nodes and lungs of asthmatic mice as compared with that determined in the healthy subjects. On the contrary, the absolute counts of DP T cells was significantly decreased in the head and neck lymph nodes, and in peripheral blood of OVA-immunized mice. These results suggest that DP T cells may be involved in the pathogenesis of allergic asthma.

The aim of this study was to investigate the role of cats and dogs as a potential reservoir of Campylobacter spp. Rectal swabs from 83 dogs and 71 cats were examined. Samples were obtained from the animals aged between 2 weeks and 24 months living in shelters, private households, farms and from veterinary clinics located in Bydgoszcz region during routine check-up. Campylobacter spp. were isolated from 4.81% dogs and 9.86% cats, respectively. C. jejuni was predominant in this study. All strains were isolated in autumn and winter from the animals living in farms and private houses. All the animals positive for Campylobacter prevalence had access to small water basins, accidental source of food and had contact with wild birds, poultry or their feaces. Isolates characterization revealed high prevalence of Campylobacter virulence genes-flaA, cadF and cdtB. 91% of isolated strains were susceptible to erythromycin. 81% among isolated strains were susceptible to azithromycin, 64% to tetracycline and 36% to ciprofloxacin. For 2 C. jejuni strains isolated from cats Random Amplified Polymorphic DNA (RAPD) profiling indicated 80% homology between them.
