Opis:
This paper discusses 28 canine patients subjected to low-field magnetic resonance imaging (MRI) of the spinal cord for neurological indications. The authors describe and compare the used MRI sequences with an indication of the most effective sequences in MRI examinations that require short scanning time. The most effective sequences supporting a quick diagnosis of spinal diseases in dogs were SE (spin echo), FSE (fast spin echo) and 3D HYCE (hybrid contrast enhancement).

Opis:
The aim of this study was to develop and to optimize an immunohistochemistry (IHC) method for PCV2 identification and to compare it with an in situ hybridization (ISH) technique. The results demonstrated that both ISH and IHC successfully detected PCV2 viral antigens or nucleic acid in the examined tissues. Most of the slides identified previously in ISH as PCV2-positive were also positive in IHC. In the case of nearly half of the slides the results of IHC examination revealed an increase in the intensity of staining. IHC presented higher sensitivity and specificity than ISH. No negative impact of the time of paraffin block storage on ISH detection results was observed. In addition, IHC results were easier to interpret due to better image quality after staining. Overall results confirmed IHC was a reliable and useful technique for PMWS diagnosis.

Opis:
The aim of the present study was to determine the efficacy of ivermectin against Cyathostominae infections and to describe the drug's pharmacokinetic parameters during two seasonal deworming treatments in horses. The study was performed on warm-blooded mares aged 3-12 years weighing 450-550 kg. A single bolus of an oral paste formulation of ivermectin was administered at a dose of 0.2 mg/kg BW in spring and autumn. Fecal samples were tested before treatment and 1, 2, 3, 4, 6, 10, 20, 30, 40, 50, 60, 75 days after treatment. Ivermectin concentrations in blood samples collected before treatment, 0.5,1, 2, 3, 4, 6, 12, 24, 36 and 48 hours after treatment, and 3, 4, 6, 8,10, 15, 20, 25, 30, 40, 50, 60 and 75 days after drug administration were determined by high pressure liquid chromatogra-phy. Drug absorption was significantly (p<0.05) slower (tmax: 21.89±11.43 h) in autumn than in spring (tmax: 9.78±8.97 h). Maximum concentrations (Cmax) of ivermectin in the blood plasma of individual horses (8.40-43.08 ng/ml) were observed 2-24 h after drug administration during the spring treatment and 2-36 h (6.43-24.86 ng/ml) after administration during the autumn treatment. Significantly higher (p<0.05) ivermectin concentrations were found during the first 4 hours after administration in spring in comparison with those determined after the autumn treatment. The administration of the recommended dose of ivermectin resulted in 100% elimination of parasitic eggs from feces in spring and autumn treatment.

Opis:
The objective of this study was to compare the effectiveness of the multiplex PCR method and traditional light microscopy in identifying and discriminating the species of Nosema spp. spores in worker bees from winter hive debris in the Province of Warmia and Mazury (NE Poland). A total of 1000 beesdead after from the bottom of the hive from bee colonies were analyzed. Spores were identified with the use of a light microscope (400-600x magnification). Spores were assigned to species by the multiplex PCR method. The microscopic evaluation revealed the presence of Nosema spp. spores in 803 samples (80.3%). Nosema ceranae spores were observed in 353 positive samples (43.96%), Nosema apis spores were found in 300 samples (37.35%), while 150 samples (19.67%) showed signs of a mixed infection. A multiplex PCR analysis revealed that 806 samples were infested with Nosema spp., of which 206 were affected only by Nosema ceranae, 600 showed signs of mixed invasion, while no samples were infected solely by Nosema apis parasites. In two cases, the presence of spores detected under a light microscope was not confirmed by the PCR analysis. The results of the study indicate that Nosema ceranae is the predominant parasitic species found in post-winter worker bees from the bottom of the hive in the region of Warmia and Mazury.

Opis:
The most common cause of hindlimb lameness in dogs is cranial cruciate ligament rupture. In 48-77.3% of the population this trauma leads to secondary damage of the meniscus. Depending on the magnetic strength of the used device, different diagnostic accuracy can be achieved. The examination sensitivity of magnetic resonance imaging is affected by many factors which are independent of diagnostic strength, such as correct positioning of the patient, size of the stifle joint examined, or selection of the right protocol of sequences. Sensitivity of meniscus damage detection was 100% and 90%, respectively, in high- and low-field magnetic resonance. The best results were reported during examination of the stifle in dogs above 10 kg b.w. at a flexion angle of 145°, and in sagittal and dorsal planes. Regardless of the magnetic strength applied, imaging of the whole cranial cruciate ligament is difficult. Moreover, MRI allows the detection of the first signs of osteoarthritis, which were observed 4 and 6 weeks after rupture of the cranial cruciate ligament using high and low-field MRI. This also applies to lesions in the subchondral bone or a bone marrow which occurred in association with insufficiency of the stifle joint, and were mainly localized in the epiphysis of the femur and tibia. The present article provides a comparison of different examination protocols and images of damaged stifle structures, such as menisci, ligaments and bones of the stifle joint visualized with low-field and high-field magnetic resonance. Magnetic resonance arthrography is also discussed.

Opis:
Atopic dermatitis is one of the most frequent allergic diseases in dogs. There are many methods of treating its symptoms but specific immunotherapy has recently gained high popularity. Before the application of specific immunotherapy, it is necessary to identify the allergens provoking the reaction of hypersensitivity in the selected animal. This raises a question about the method of allergen identification the medical practitioner decides to use in order to obtain the most credible result. The authors of the present study decided to compare the results of intradermal allergic tests and the results of an IgE screening test carried out using the FceRIa receptor method. The aim of the study was to compare the results of both tests directed to dust and storage mites. The study proves that in case of the IgE screening tests (for a group of allergens), the sensitivity is quite high (85 to 90.69%) but the specificity of these tests is insufficient (25 to 50%). In case of antibodies for the selected mites the sensitivity and specificity was too low (65.1 to 89.4% for the sensitivity, with only 14.2 to 33.3% for the specificity). Only in case of D. petronyssinus the results were higher with the sensitivity calculated at 65.1% and the specificity at 80%. The IgE screening test carried out using the FceRIa receptor method is reliable only in case of screening test for mites and the intradermal allergic test remains the gold standard for allergy testing.

Opis:
Antibiotics are used for postsurgical wound healing purposes but unfortunately, resistance against them demands some alternatives for quick recovery. Sepsis of wounds is a challenge for medical as well as veterinary professionals. Nanoparticles have significant advantages in wound treatment and drug resistance reversal. This study was conducted to appreciate emerging alternates of antibiotics like zinc oxide nanoparticles and plant extracts in topical application. Zinc oxide is considered a good wound healer and its nanoparticles are easy to access. So, the efficacies of zinc oxide nanoparticles and sweet flag plant extract ointments were tested to compare modern and traditional therapeutics as sweet flag is considered a pure medicinal plant. Rabbits were selected for this study due to the healing properties of their skin. Wounds were inflicted on the thoracolumbar region and treated for 29 days post-surgically daily with normal saline and the ointment of zinc oxide nanoparticles and sweet flag extract ointment, prepared in a hydrophilic solvent. Wound shrinkage was observed daily and histopathological analysis was made and results were compared. Zinc oxide nanoparticles ointment showed the most satisfactory results for every parameter included in the study. No side effects of its topical application were observed. Healing was normal without any complications. The preparations of zinc oxide nanoparticles may help in the era of antibiotic resistance as topical drugs in the future.

Avian encephalomyelitis (AE) is a viral disease of poultry. Although the disease has a milder clinical course in turkeys than in chickens, reproductive flocks of turkeys are vaccinated against AE. Commercial AE ELISA kits are specifically designed for chickens, which makes it difficult to implement these tests in serological monitoring of turkey flocks. The aim of the study was to compare the AE serological results provided by two ELISA kits from different producers when testing an AE-vaccinated flock of turkey hens and their progeny. We detected differences in the sensitivity of the ELISAs for testing specific anti-AE antibody levels in turkey serum samples.

Opis:
In the present study, we used next-generation sequencing to investigate the impacts of two commercially available prescription diet regimens on the fecal microbiomes of eleven client-owned healthy pet dogs. We tested an anallergenic diet on 6 dogs and a low-fat diet on 5 dogs. Before starting the study, each dog was fed a different commercial diet over 5 weeks. After collecting pre-diet fecal samples, the anallergenic or low-fat diet was administered for 5 weeks. We then collected fecal samples and compared the pre- and post-diet fecal microbiomes. In the dogs on the anallergenic diet, we found significantly decreased proportions of Bacteroides, Ruminococcaceae, and Fusobacteriaceae, belonging to the phyla Bacteroidetes, Firmicutes, and Fusobacteria, respectively. The proportion of the genus Streptococcus belonging to the phylum Firmicutes was significantly increased upon administering the anallergenic diet. In the dogs on the low-fat diet, although the phyla Actinobacteria and Bacteroidetes tended to increase (p=0.116) and decrease (p=0.147) relative to the pre-diet levels, respectively, there were no significant differences in the proportions of any phylum between the pre- and post-diet fecal microbiomes. The anallergenic diet induced a significantly lower diversity index value than that found in the pre-diet period. Principal coordinate analysis based on unweighted UniFrac distance matrices revealed separation between the pre- and post-diet microbiomes in the dogs on the anallergenic diet. These results suggest that, even in pet dogs kept indoors in different living environments, unification of the diet induces apparent changes in the fecal microbiome.
