The Dermacentor reticulatus tick is a main vector of Babesia canis in Europe. The risk of canine babesiosis is unpredictable, due to significant differences in the prevalence of B. canis between ticks originating from closely situated regions. This phenomenon may be explained by vertical transmission of the pathogen in a vector population. Thus, molecular techniques were applied to investigate the occurrence of transovarial transmission in D. reticulatus ticks. DNA of B. canis was detected in 20.7% (6/29) of engorged female ticks collected from dogs, in every pool of eggs laid by positive females (100%, 6/6) and in larvae hatched from these eggs. In the pools of eggs collected from two positive females (2/6; 33.3%), no larvae hatched and no embryos were observed inside the eggs. Conclusions. Transovarial transmission of B. canis can be an important mechanism supporting maintenance of the pathogen in the environment without the presence of a reservoir vertebrate host. However, the efficiency of transovarial transmission in the maintenance of B. canis in natural conditions requires further field research.

Introduction and objective. It is easier and non-invasive to obtain faecal samples compared with blood samples. Molecular techniques may enable detection of parasites even in tiny amounts of blood-containing faeces. We aimed to compare the sensitivity of detection of three Babesia species and Hepatozoon canis in blood and faecal samples, including samples derived from naturally infected hosts. Materials and method. Three groups were involved: 1) Nine BALB/c mice infected with Babesia microti sampled during acute (n=3), post-acute (n=3) and chronic phases of infection (n=3); 2) Eight dogs with symptoms of babesiosis; 3) Six red foxes infected with B. vulpes, one fox infected with B. canis, four foxes infected with H. canis. Genomic DNA was extracted from blood and faeces by use of commercial kits and amplified with genus-specific primers in one-step or nested PCR reactions. Selected PCR products were sequenced. Results. No positive results for faecal samples were obtained from H. canis-positive foxes in contrast to Babesia spp. infections. Positive results from PCRs were obtained for all BALB/c mice (100%), five dogs (62.5%) and four of seven foxes (57.1%). Successful sequencing was obtained for six selected murine samples (B. microti), four canine samples (B. canis) and for one fox sample (B. vulpes). The success of B. microti detection in murine faecal samples from acute, post-acute and chronic phases was identical (100%). Conclusions. Detectability of Babesia spp. infections was lower in naturally infected dogs and foxes, compared to experimentally infected mice. Detection of DNA in faecal samples can be useful in the detection of Babesia infection in populations from which blood samples are hard to obtain, but due regard must be given to the possibility that prevalence of infection may be severely underestimated.

Di-(2-ethylhexyl)phthalate (DEHP) is widely present in the human environment. The study aimed at the investigation of potential genotoxic effects induced by subchronic exposure to DEHP in germ cells of male mice in the first period of puberty, and to check if the transmission of mutation to the next generation via the sperm is possible. 8-weeks exposure to 2,000 mg/kg and 8,000 mg/kg of DEHP diminished sperm count and quality, leading to a reduced percentage of pregnant females mated to exposed males. A slight increase in the frequency of prenatal deaths and dominant lethal mutations, as well as a significantly increased percentage of abnormal skeletons among the F1 offspring of males exposed to 8,000 mg/kg of DEHP, were observed. Exposure of the fathers did not cause a delay in the postnatal development of the offspring, except for fur development in the group of 8,000 mg/kg of DEHP. Gametes of male offspring of exposed fathers showed reduced motility. The results may suggest that diminished spermaozoa quality induced by DEHP may be coincidental with mutations leading to intrauterine deaths and skeletal abnormalities in the offspring.

Introduction and objective. Tick-borne infections constitute an increasing health problem in dogs and may lead to death, especially in young or elderly individuals. Canine babesiosis constitutes a serious health problem in dogs worldwide. The aim of the study was to verify the probability of vertical transmission of Babesia canis between the bitch and the pups. Materials and methods. In Autumn 2011, cases of babesiosis were diagnosed in a litter of 6-week-old puppies of a Central Asian Shepherd dog. Immediately following the first case of infection, blood samples were collected from all the pups in the litter (n=10) and from the female. Detection of Babesia infection was performed by molecular and microscopical techniques. Results. The presence of B. canis DNA was detected using PCR in three pups, presenting at the time or 24–48 hours later with babesiosis symptoms, and in their asymptomatic mother. The isolates derived from the pups and the female – 520 bp 18S rRNA gene fragment – were compared and analyzed. All isolates from the pups and their mother were identical and showed 100% homology with B. canis group B (EU622793), supporting the same source of infection. Additionally, the USG of the peritoneal cavity was performed in the female, presenting evidence for splenomegaly. Conclusions. On the basis of (1) the same timing of three pup cases; (2) the identical B. canis sequences derived from all positive dogs; (3) evident splenomegaly in the asymptomatic female, this provides the first evidence of the vertical transmission of this piroplasm in dogs.

Background: Studies on childhood asthma demonstrate socioeconomic disparities in medical care utilization. A lack of information for Poland prompted our investigation into this question. Its goal was to discover if the utilization of medical services by asthmatic children depends on social and family factors. Methods: Subjects were 186 children with physician-diagnosed asthma, identified through a questionnaire survey of 4,535 school children in Katowice District, Poland. Utilization of medical care was assessed by such past year events as medical visits (44.0%), any diagnostic test (35.4%) and spirometry (30.6%). Association of those events with socioeconomic variables was explored by means of logistic regression, according to the criterion p≤ 0.1. Results: After adjustment for disease severity and gender medical visits were related to younger age (p=0.009), family history of respiratory diseases (p=0.08) and rural residence (p=0.09), any diagnostic tests to younger age (p=0.08), smaller number of siblings (p=0.01) and rural residence (p=0.004); spirometry to smaller number of siblings (p=0.09) and rural residence (p=0.006). Conclusion: Clinical status and age are important determinants of utilization of medical services by asthmatic children. The effects of rural residence and family size may reflect a more attentive response to the needs of a sick child.

Objective. The aim of this study is to evaluate neurological scales, as well as biochemical and radiological parameters measured on day 10 after ischemic stroke (IS), according to their value as predictors of the long-term outcome. Materials and method. 45 patients were assessed according to the Barthel Index (BI) and National Institute of Health Stroke Scale (NIHSS) on day 10, and according to Modified Rankin Scale (mRS) 3 months after the onset of IS. On day 10 of IS, the serum level of C-reactive protein (CRP), albumin, D-dimers (DD), S100BB and Tau proteins was measured and the volume of ischemic focus assessed with the use of Computed Tomography (CT). The patients were divided into groups with good outcome (GO) and mRS 0–2, and with bad outcome (BO) and mRS 3–6. Results. NIHSS and BI scores (p<0.001), the volume of ischemic focus (p<0.01), CRP (p<0.01) and albumin level (p<0.05), but not DD, S100BB and Tau protein levels evaluated on day 10, correlated with mRS after 3 months since IS onset. Patients from the BO group were observed to have lower BI (p=0.001), higher NIHSS (p<0.01) and CRP levels (p<0.05), and bigger volume of ischemic focus (p<0.05) measured on day 10 of IS. In the GO group, there were more patients with atherosclerotic etiology (p=0.02 x2=7.856). Regression analysis showed that only the BI score assessed on day 10 of IS can predict the outcome after 3 months assessed by mRS (OR=1.102, 95%, CI:1.01–1.203; p=0.001). Conclusions. BI assessed on day 10 has a predictive value for the outcome evaluated by mRS 3 months after the onset of IS.

Introduction and objective. Infectious and parasitic diseases transmitted by ticks, such as Lyme diseases, granulocytic anaplasmosis and piroplasmosis, have been frequently reported in Europe, with increasing attention to them as an emerging zoonotic problem. The presented study was performed to assess the distribution and the density of questing ticks in three regional parks of Emilia-Romagna region of Northern Italy, and to seek molecular evidence of potential human pathogens in tick populations. Materials and Methods. In the period April-October 2010, 8,139 questing ticks were collected: 6,734 larvae, 1,344 nymphs and only a few adults – 28 females and 33 males. The abundance of Ixodes ricinus questing ticks was compared among different sampling sites and related to microclimate parameters. 1,544 out of 8,139 ticks were examined for the presence of pathogens: PCR was used to detect piroplasms DNA and Real time Taqman PCR for Anaplasma phagocytophilum and Borrelia burgdorferi s.l. Results. The predominant species was I. ricinus (overall abundance 1,075.9/100 m2); more rarely, Dermacentor marginatus (n = 37 – 0.45%), Scaphixodes frontalis (n = 13 – 0.16%), Hyalomma spp. (n = 6 – 0.07%) and Ixodes acuminatus (n = 3 – 0.04%) were also found. 28 out of 324 (8.6%) samples of ticks were PCR-positive for piroplasm DNA. 11 amplicons of 18S rRNA gene were identical to each other and had 100% identity with Babesia EU1 (Babesia venatorum) using BLAST analysis. Real time Taqman PCR gave positive results for A. phagocytophilum in 23 out of 292 samples (7.9%), and for B. burgdorferi s.l. in 78 out of 292 samples (26.7%). I. ricinus was the only species found positive for pathogens by molecular analysis; 16 tick samples were co-infected with at least 2 pathogens. Discussion. The peak of nymph presence was in May, and the higher prevalence of pathogens occurred in April-June, most often in nymphs; therefore, spring season could represent the higher risk period for the transmission of pathogens. These data could provide guidelines for the preventions of tick-trasmitted diseases in this region.

The study of tree-hole breeding mosquitoes was carried out in the tropical rainforest of Imo State Nigeria (two rural areas and two forest reserves in some parts of Orlu Senatorial Zone) between May – October 2002. Using standard entomological procedures, two macrohabitats (natural tree-holes and bamboo traps) and two microhabitats (leaf axils of cocoyams/pineapples and leaf axils of plantain/banana) were sampled for various mosquito species. Mosquitoes were recovered from all the various biotypes sampled. Types of mosquitoes species encountered, their relative abundance, as well as genera varied signifi cantly during the study (p<0.05). Four genera of mosquitoes: Aedes, Culex, Anopheles and Toxorhynchites were recovered while 16 species of mosquitoes encountered include: Aedes aegypti, Ae. africanus, Ae. simpsoni, Ae. albopictus, Ae. stokesi, Ae. taylori, Ae. apicoargenteus, Culex quinquefasciatus, Cx. nebulosus, Cx. trigripes, Cx. decens, Anopheles gambiae, An. funiestus, An. coustani and Toxorhynchites viridibasis. Most of the mosquitoes showed oviposition preferences for one or more habitats. The presence of Ae. africanus, Ae. simpsoni and Ae. aegypti indicate that the study areas were at risk of yellow fever epidemic. The presence of Anopheles and Culex species ensured endemicity of malaria and fi lariasis, while the recovery of Ae. albopictus in this region suggests a possible outbreak of dengue fever in future if not properly controlled.

Introduction: Moulds are the one of the known biological factors that have a negative impact on human health. Moulds are commonly present in residential and work environments. Materials plentiful in organic compounds, such as building materials or paints, are a splendid substrate for the development of moulds. The first documented mention of a study describing the harmful effects caused by moulds in buildings emerged in the early nineteenth century. In Copenhagen and then in Padua, moulds of the genus Penicillium, Cladosporium and Mucor were found in buildings. Objective: To present the current state of the allergic properties and other negative health effects caused by moulds isolated from buildings. Brief description of the state of knowledge: The literature and own research clearly shows that moulds and their secondary metabolites can evoke toxic effects on human and animal health, and cause symptoms similar to allergic diseases. These allergens have been noted in spores as well as other fungal fragments; however, most allergens are located in germinating spores, in the hyphal tips and in mycelia. Fungal allergy can express in different ways: asthma, rhinitis, conjunctivitis, urticaria and atopic dermatitis. Fungal allergy antigen is bound to IgE-dependent reactions but also to reactions independent of IgE. Conclusions: Moulds are a significant but difficult to detect etiologic agent of different allergic diseases. Prevention of this diseases is important for patients with suspected connection between common allergic symptoms and affinity with moulds.

Introduction. The highest risk of trichinellosis for human is considered in eating meat products containing live larvae, mostly from wild boars or pigs. Spreading of Trichinella spp. may occur in various ways, one of which is transmission by vectors. The rat is considered to be the most common vector for Trichinella parasite. The population of rats living on pig farms can play an important role in maintaining or spreading the parasite to other animals. Objective. The aim of presented survey was to investigate the occurrence of Trichinella spp. in rats on farms with pigs infected with this parasite. Materials and method. From pig farms selected for study, the muscles of collected rats were investigated by magnetic stirrer digestion method to assess occurrence of Trichinella in the rat population. Isolated Trichinella parasites were identified under stereomicroscope and multiplex PCR were performed for species identification. Results. Rats infected with Trichinella spp. were discovered on three of five investigated pig farms. The mean extent of invasion in rats from the studied farms was 23.33%. The calculated medium intensity of invasion was 4.09 lpg (larvae per gram) (SD 5.41). All larvae of Trichinella discovered from rats were identified as T.spiralis. Conclusions. The results obtained indicate that in farms with a high prevalence of Trichinella invasion in pigs there are very likely to be found rats infected by this nematode. This suggests possibility to maintain the invasion in herd and spread into neighborhood farms.

“Legal highs” known also as “smarts”, “legal drugs” or “boosters” contain in their composition psychoactive substances the production, sale and possession of which are not prohibited in legislation. They are offered for sale under the cover of collectors’ items, a salt bath, aroma sticks, or plant fertilizer. Marketing was the reason for such high sales of these “highs” in Poland. The phenomenon became of concern when information became available about the first cases of tragic health consequences. Raising the awareness of youth about the level of imminent danger and serious consequences associated with the use of legal highs may be a very effective way to develop appropriate attitudes of young people, and in the light of their own health can help them take proper life decisions. “Legal highs” are substances of natural or synthetic origin having psychostimulating properties. These compounds may differ in chemical structure, potency, half-life, metabolism and severity of side effects. Their pharmacological activity is associated with changes in the neurotransmitter system. After higher doses of “legal highs” psychotic symptoms may occur: visual and auditory hallucinations resembling schizophrenic endogenous psychoses. An alarming fact is the underestimation of the adverse effect of these substances on human health. Any actions aimed at improving this situation are extremely important; therefore, in one of the key projects undertaken at the Institute of Rural Health in Lublin, in cooperation with other scientific institutes, concerned the analysis of a selected series of “legal highs”.
