Opis:
Throughout the world, charcoal rot, caused by Macrophomina phaseolina, is one of the most destructive and widespread diseases of crop plants such as soybean. In this study, the biological control capability of 11 Trichoderma spp. isolates against M. phaseolina was investigated using screening tests. Among all the tested Trichoderma spp. isolates, inhibition varied from 20.22 to 58.67% in dual culture tests. Dual culture, volatile and non-volatile tests revealed that two isolates of Trichoderma harzianum (including the isolates T7 and T14) best inhibited the growth of M. phaseolina in vitro. Therefore, these isolates were selected for biocontrol of M. phaseolina in vivo. The results of greenhouse experiments revealed that disease severity in the seed treatment with T. harzianum isolates was significantly lower than that of the soil treatment. In most of the cases, though, soil treatment with T. harzianum resulted in higher plant growth parameters, such as root and shoot weight. The effects of T. harzianum isolates on the activity of peroxidase enzyme and phenolic contents of the soybean root in the presence and absence of M. phaseolina were determined in greenhouse conditions. Our results suggested that a part of the inhibitory effect of T. harzianum isolates on soybean charcoal rot might be related to the indirect influence on M. phaseolina. Plant defense responses were activated as an elicitor in addition to the direct effect on the pathogen growth.

The current study aimed to evaluate the biodiversity and antagonistic potential of 22 endophytic fungal species belonging to 10 endophytic genera, namely Acremonium, Alternaria, Aspergillus, Fusarium, Gliocladium, Nigrospora, Penicillium, Pochonia, Scopulariopsis, and Stachybotrys, along with 2 morphospecies of mycelia, namely a yellow sterile septate mycelium (YSSM) and a dark sterile septate mycelium (DSSM), living within healthy Triticum aestivum L. against nine wheat pathogenic fungi identified as Alternaria alternata, Alternaria cichorii, Alternaria dianthi, Alternaria sonchi, Aspergillus petrakii, Curvularia penniseti, Fusarium oxysporum, Fusarium semitectum, and Fusarium solani obtained from naturally infected plants. The highest count and frequency in T. aestivum were recorded for Fusarium subglutinans (n = 81 and Rf = 34.91%) followed by DSSM (n = 32 and Rf = 13.79%). In vitro DSSM studies showed the highest antagonistic activity of 56.52% reduction against A. petrakii, while F. subglutinans inhibited F. semitectum by 72.97%. These two endophytic isolates were then selected as fungal bioagents to control root rot disease in wheat caused by A. petrakii and F. semitectum under greenhouse conditions. Wheat plants growing from seeds inoculated with DSSM were more resistant to root rot disease in soil infected with A. petrakii, and they showed reduced root rot severity percentage (15.56%) and disease index (0.27) compared to the values of 40% and 1.13, respectively, for these parameters in untreated plants. Furthermore, treatment of seeds with F. subglutinans before planting in soil infected with F. semitectum reduced the disease severity percentage and the disease index from 50% and 2.20 to 20% and 0.47, respectively. These endophytic fungi also markedly increased the growth parameters (length, fresh weight, and dry weight of shoots and roots) of wheat plants growing in soils infected with these phytopathogens. The present study showed that wheat is a promising ecological niche for different fungal endophytes that assist their host plants to survive and resist the harmful effects of phytopathogenic fungi, improve wheat growth, and develop fungal strains as biological control solutions against wheat diseases.

Opis:
Phenylalanine ammonia lyase (PAL) is a specific branch point enzyme of primary and secondary metabolism. It plays a key role in plant development and defense mechanisms. Phenylalanine ammonia lyase from Hevea brasiliensis (HbPAL) presented a complete open reading frame (ORF) of 2,145 bp with 721 encoded amino acids. The sequence alignment indicated that the amino acid sequence of HbPAL shared a high identity with PAL genes found in other plants. Phylogenetic tree analysis indicated that HbPAL was more closely related to PALs in Manihot esculenta and Jatropha curcas than to those from other plants. Transcription pattern analysis indicated that HbPAL was constitutively expressed in all tissues examined, most highly in young leaves. The HbPAL gene was evaluated by quantitative real-time PCR (qRT-PCR) after infection with Rigidoporus microporus at 0, 12, 24, 48, 72 and 96 hours post inoculation. The expression patterns of the PAL gene differed among the three rubber clones used in the study. The transcription level of the white root rot disease tolerant clone, PB5/51 increased sharply during the latter stages of infection, while it was relatively subdued in the white root rot disease susceptible clones, RRIM600 and BPM24. These results suggest that the HbPAL gene may play a role in the molecular defense response of H. brasiliensis to pathogen attack and could be used as a selection criterion for disease tolerance.

Opis:
Black alder Alnus glutinosa (L.) Gaertn. is an important forest−forming species in Poland, with more than 5% participation in the species structure. It is a typical lowland species, especially in the river valleys, on the banks of the lakes and in the land depressions. It is one of the main species in the following habitats: boggy mixed broadleaved forest, alder and alder−ash forest, riparian forest and moist broadleaved forest. As a valuable admixture alder is introduced in the habitat of boggy coniferous forest and boggy broadleaved forest. Until recently black alder was considered in Poland as a species with a very low level of danger from the pests and diseases. The situation changed at the turn of the centuries, when the deterioration of the health status of the alder stands began, which led locally to the mass dieback of the trees, mainly in the stands which are over 20 years old. In the following years, the phenomenon was intensified. The largest number of such stands was recorded in 2006. The strongest deterioration of health condition of the trees was observed in five regional directorates of the State Forests: Białystok, Lublin, Torun, Wrocław and Olsztyn. The study gives information on lignicolous fungi found on black alder in 62 alder stands, which exhibited the most severe signs of the dieback. The research was conducted in 14 forest districts and in Poleski National Park (fig.). The surveys revealed the presence of 49 taxa of the lignicolous fungi (44 Basidiomycota and 5 Ascomycota). The most common species were Xanthoporia radiata, Stereum hirsutum, Daedaleopsis confragosa, Armillaria spp. as well as S. subtomentosum (tab. 2). The disease contributing to the local dieback of black alder in the stands (mainly in the Lublin Regional Directorate of the State Forests) is the root rot caused by Armillaria species. Many species of lignicolous fungi (e.g. Fomes fomentarius, Fomitopsis pinicola, Ganoderma applanatum) penetrate into previously weakened trees, causing decay of wood, which can accelerate the dying of single trees.

Actinomycetes are considered to be the biggest producer of bioactive compounds which are expected to have antifungal activity for controlling many fungi such as Rhizoctonia solani. The objective of this study was to obtain potential soybean rhizosphere actinomyc- etes as a biocontrol agent for R. solani which cause damping-off disease both in vitro and in vivo, including their ability to produce siderophore, chitinase, and HCN. Out of 26 isolates, 18 (56%) showed diverse antifungal activities against R. solani with percentages of inhibi- tion radial growth (PIRG) from 18.9 to 64.8%, as evaluated by a dual culture method. Ten isolates with the strongest antifungal activity were numbered for further characterization. All the tested isolates were not antagonistic towards Bradyrhizobium japonicum. These iso- lates were able to suppress damping-off disease caused by R. solani in the greenhouse exper- iment. Isolate ASR53 showed the highest disease suppression, 68% and 91% in sterile and non-sterile soil, respectively. Based on 16S rRNA sequence analysis this isolate belonged to Streptomyces violaceorubidus LMG 20319 (similarity 98.8%) according to GenBank data base available at www.ncbi.nlm.gov.nih. Furthermore, isolate ASR53 had significantly longer roots and shoots, as well as greater fresh and dry weights of seedlings than the con- trol. Crude extract derived from ASR53 isolates contained 10 dominant compounds that were biologically active against fungal pathogens. Thus, this study suggests that the applica- tion of potential actinomycetes of the soybean rhizosphere can act as a promising biocon- trol agent against damping-off disease caused by R. solani.

In 1997, supplying Plant Pathogenic Microorganism Collection, microflora of diseased root crops from field and storage was analyzed. Samples from Districts lnspectors of PIOR from area of Poland were received, and 139 isolates of fungi from 369 samples of sugar and forage beat, forage cabbage, forage carrot and potatoes were obtained. The most often fungi from genus Fusarium occurred, and 23.5% of isolates from beet, 14.2% from cabbage, 29.5% from carrot and 48,2% from potatoes were received. The second dominant was species Alternaria alternata, isolated from diseased plants in 28.9%, 50%, 18.5% and 20% respectively. Among saprophytic fungi, species Penicillium and Aspergillus were represented in 9,7% of obtained isolates. Received results suggcsted that Fusarium spp. and Alternaria alternata could be potentially dangerous for root crops as a pathogens or weak pathogens.
Wyosobniono łącznie 139 izolatów grzybów z 369 prób roślin buraka cukrowego i pastewnego, kapusty pastewnej, marchwi pastewnej i ziemniaka.

The response of the Mi-1 gene to different densities of Meloidogyne incognita race 2 was investigated under controlled conditions. Susceptible and resistant tomato seedlings were inoculated with 25, 50, 100, 200, 400, 1000, 2000, 5000 and 10 000 second-stage juveniles of M. incognita. Plants were uprooted 8 weeks after inoculation and the numbers of egg masses and galls on the roots, and second-stage juveniles in 100 g soil per pot were counted. In susceptible plants, there was a correlation between the number of egg masses on roots until 2000 J2 inoculum densities. In resistant plants, when inoculum densities increased, the number of egg masses and galls also increased. The reproduction factor ratio was >1 in the susceptible plant and <1 in the resistant plant. The data showed that the 5000 J2 inoculum was a critical limit, and 10 000 J2s were above threshold for resistant plants. The data indicate that densities of M. incognita can seriously affect the performance of the Mi-1 gene.

HIV-1 or Human Immuno Deficiency Virus-1 is the main causative agent of Acquired Immuno Deficiency Syndrome (AIDS). Human host infected with HIV - 1 extensively harbours many viral variants but very little is known about the difference in pattern[17] of evolution of phylogenetic lineages of HIV-1 non recombinant, normal inter subtype recombinant and main two specific recombinant forms of HIV-1 i.e., Circulating Recombinant Forms (CRFs) and Unique Recombinant Forms (URFs). This study is mainly concerned with study of the difference in evolutionary lineages of non-recombinant and recombinant sequences of HIV-1 genome sequences and identification of geographically rich areas which has reported high degree of HIV-1 occurrence and variety. Total 1550 HIV-1 genome sequences were obtained from HIV Los Alamos Database. The sequences were aligned using MAFFT (Multiple Alignment using Fast Fourier Transform) web server tool. Alignment was carried out using 10 different set of alignment parameter values. After alignment the aligned file was used for constructing N-J phylogenetic tree using Clustal X2 tool. Phylogenetic analysis was performed keeping in mind the category to which the sequence belongs. Upon analysis it was observed that the clade containing the probable ancestor belongs remained constant in all cases of different alignment values. Non recombinant isolates, inter subtype recombinants, CRFs, URFs all followed different patterns of evolution. Non recombinant sequences were found geographically specific and subtype specific to some extent whereas, normal recombinants were subtype specific and less geographically specific. CRFs showed variation among the pattern of their evolution. At some instances the sequences occurred as sister taxa of non-recombinant or normal inter subtype recombinant sequences, while at some instances as sister taxa of other CRFs where they were geographically specific. Three CRFs existed as completely diverged sequences. URFs were four in number; two of them were Indian isolates of while other two were Japanese isolates. URFs were found to be totally geographically specific. Geography wise high rate of variation was observed in India and Japan as these two countries had sequences belonging to all of the above categories. Cameroon and South Africa have very large number isolates and a considerable amount of genetic variation among isolates but they lack URFs.

Roots of Gladiolus x hybridus Van Houtte plants infected with aster yellows phytoplasma were examined. The infected plants had a reduced root system in comparison to control plants. Their roots were thinner and the stele organisation was changed. Phytoplasmas were present in sieve tubes, companion cells and phloem parenchyma cells of the infected plant roots. Free calcium ions were localized in the cells of infected plants. Cells of the stele of infected roots, especially these infected with phytoplasmas, showed an increase of calcium antimonite deposits in theirs protoplasts. Also the number of calcium antimonite deposits increased in sieve tubes of infected roots. The deposits were present on plasma membrane, around the sieve tube plate and also in the lumen of the sieve tube. The increase of free calcium ions in sieve tubes did not cause the occlusion of sieve tube pores. Companion cells and some parenchyma cells with phytoplasmas did not react to phytoplasma infection with an increase of Ca2+ ions in protoplast. The parenchyma cells showing signs of degeneration reacted with high increase of calcium ions. The Ca2+ ions were present mainly in cytoplasm of infected parenchyma cells. There were calcium antimonite deposits in infected plant roots xylem elements and in intracellular spaces of cortex parenchyma. Such deposits were not present in control plants.

Introduction. Periodontitis is a disease of multifactorial etiology and complex clinical manifestation. Removal of subgingival bacterial biofilm is a fundamental procedure in the treatment of periodontitis. Scaling and root planing (SRP) is currently the gold standard in the treatment of periodontitis, but in some cases such a procedure does not provide satisfactory results. The use of lasers in the treatment of periodontitis is one of the alternative adjunctive methods that has recently become more popular. Objective. The aim of the study was to evaluate the effectiveness of diode lasers in the treatment of periodontitis, based on the literature found in the Medline, Pubmed and Scopus databases published from June 2013 – June 2020. Results were based on a comparison of the effectiveness of SRP treatment in combination with photodynamic therapy or SRP in combination with laser irradiation of periodontal pockets. Results. The majority of studies confirmed the effectiveness of periodontal treatment combining SRP with photodynamic therapy or SRP with laser irradiation, compared to an SRP alone. Conclusions. Based on the recent literature, it can be concluded that SRP combined with photodynamic therapy or SRP in combination with a diode laser irradiation are effective methods to improve the outcomes of non-surgical periodontal treatment.

Opis:
Buxus sempervirens (L.) is an evergreen shrub often used in urban plantings intended for forming the trimmed hedges, as well as for creating geometrical shapes. Its decorative qualities are often diminished by the effects of pathogens colonizing the roots, shoots and leaves. The study was conducted in three consecutive growing seasons. The experimental material originated from a nursery located in southern Poland. The subjects under study were diseased leaves, stems and roots of the boxwood variety ‘Suffruticosa’. Samples consisting of 720 diseased fragments of the shrub were taken for mycological examination. Mycological isolations were performed according to the standard methods used in phytopathology. A total of 1059 colonies of fungi and fungus-like organisms were isolated from the diseased boxwood plants. The fungus Alternaria alternata dominated among all the isolates. The pathogens frequently isolated from the leaves included Macrophoma candollei, Volutella buxi and Fusarium buxicola. Those frequently isolated from the shoots included: Pestalotiopsis sydowiana, Volutella buxi and Fusarium avenaceum. The root system, in turn, was colonized in large numbers by: Fusarium oxysporum, F. avenaceum, Rhizoctonia solani, Botrytis cinerea and Phytophthora cinnamomi. Results of the three-year study show a comprehensive participation of phytopathogens in the disease process affecting the boxwood variety ‘Suffruticosa’.

Opis:
Lupine diseases caused by pathogenic fungi constitute a serious problem in agriculture. They lead to partial yield loss and deterioration of crop quality through the changes in biochemical composition of seeds or their contamination with mycotoxins. Some of common lupine diseases are fusarioses caused by Fusarium oxyspomm f. sp. lupini. Morphometric and metabolic changes were investigated in yellow lupine seedlings infected with F. oxyspomm f. sp. lupini. It was found that infection caused temporary inhibition of seedling growth, overcome at later development, and activation of ß-glycosidase and peroxidases. The changes in enzymes activity indicate the induction of defense mechanism against F. oxyspomm f. sp. lupini and inhibition of pathogen spread.

Opis:
The experiments were carried out in the years 2002 and 2003 on parsley seeds of 6 cultivars: Alba, Berlińska, Cukrowa, Kinga, Lenka, and Vistula. Mycological analysis of parsley seeds showed that the most common inhabitans were fungi from genus Alternaria (mainly A. alternata and A. radicina) and Fusarium, especially F. avenaceum and F. oxysporum. During the glasshouse investigations fungi Alternaria radicina, A. alternata and Fusarium avenaceum were the main reason for parsley damping-off. The highest number of infected seedlings was observed for Berlińska and Kinga, because in both years of experiments these cultivars had the lowest number of healthy seedlings. The highest number of healthy seedlings had cultivars Alba and Lenka, especially in the second year of experiments. In the field experiments not only fungi from genus Alternaria and Fusarium were the most often isolated from diseased parsley seedlings. Fusarium oxysporum was more often isolated from diseased field seedlings than from glasshouse parsley seedlings. Other fungies isolated often from parsley seedlings cultivated in the field were: Pythium spp., Rhizoctonia solani, Cylindrocarpon destructans and Stemphylium botryosum.
Najsilniej porażone były siewki odmian Berlińska i Kinga. Najwięcej siewek zdrowych, zwłaszcza w drugim roku badań było w przypadku odmian Alba i Lenka. W doświadczeniach polowych z zamierających siewek izolowano obok grzybów z rodzajów Alternaria i Fusarium, także inne gatunki grzybów patogenicznych. Z siewek pobranych z pola uzyskano większą liczbę izolatów Fusarium oxysporum niż z siewek ze szklarni. Także dość licznie wystąpiły grzyby: Pythium spp., Rhizoctonia solani, Cylindrocarpon destructans i Stemphylium botryosum, które mogą być sprawcami zamierania siewek pietruszki.
