This article summarizes current knowledge on the genetics and possible molecular mechanisms of human pathologies resulted from mutations within the genes encoding several myosin isoforms. Mutations within the genes encoding some myosin isoforms have been found to be responsible for blindness (myosins III and VIIA), deafness (myosins I, IIA, IIIA, VI, VIIA and XV) and familial hypertrophic cardiomyopathy (β cardiac myosin heavy chain and both the regulatory and essential light chains). Myosin III localizes predominantly to photoreceptor cells and is proved to be engaged in the vision process in Drosophila. In the inner ear, myosin I is postulated to play a role as an adaptive motor in the tip links of stereocilia of hair cells, myosin IIA seems to be responsible for stabilizing the contacts between adjacent inner ear hair cells, myosin VI plays a role as an intracellular motor transporting membrane structures within the hair cells while myosin VIIA most probably participates in forming links between neighbouring stereocilia and myosin XV probably stabilizes the stereocilia structure. About 30% of patients with familial hypertrophic cardiomyopathy have mutations within the genes encoding the β cardiac myosin heavy chain and both light chains that are grouped within the regions of myosin head crucial for its functions. The alterations lead to the destabilization of sarcomeres and to a decrease of the myosin ATPase activity and its ability to move actin filaments.

In this document, the authors present the results of the long-term monitoring of the condition of the eyes of victims of the Chernobyl catastrophe. The patient examinations were conducted using our system of standard inspection and description of the condition of the eye. We have summarized the development of this system in another paper. Ocular effects in patients as a result of the Chernobyl accident have been classified either as specific irradiation injuries, attributable only to radiation exposure, or ophthalmopathology that is observed under normal conditions, but for which radiation exposure is an important risk factor. This division is particularly apparent when examining the clean-up workers in Chernobyl.

Opis:
Regulation of gene expression in eukaryotes involves many complex processes, in which chromatin structure plays an important role. In addition to the epigenetic effects, such as DNA methylation and phosphorylation or histone modifications, gene expression is also controlled by the spatial organization of chromatin. For example, distant regulatory elements (enhancers, insulators) may come into direct physical interaction with target genes or other regulatory elements located in genomic regions of up to several hundred kilobases in size. Such long-range interactions result in the formation of chromatin loops. In the last several years, there has been a rapid increase in our knowledge of the spatial organization of chromatin in the nucleus through the chromosome conformation capture (3C) technology. Here we review and compare the original 3C and 3C-based methods including chromosome conformation capture-on-chip (4C), chromosome conformation capture carbon copy (5C), hi-resolution chromosome confomation capture (HiC). In this article, we discuss different aspects of how the nuclear organization of chromatin is associated with gene expression regulation and how this knowledge is useful in translational medicine and clinical applications. We demonstrate that the knowledge of the chromatin 3D organization may help understand the mechanisms of gene expression regulation of genes involved in the development of human diseases, such as CFTR (responsible for cystic fibrosis) or IGFBP3 (associated with breast cancer pathogenesis). Additionally, 3C-derivative methods have been also useful in the diagnosis of some leukemia subtypes.

Opis:
Voice pathology assessment using sustained vowels has proven to be effective and reliable. However, only a few studies regarding detection of pathological speech based on continuous speech are available. In this study we evaluate the usefulness of various regression models trained on continuous speech recordings from Saarbruecken Voice Database in the detection of voice pathologies. The recordings were used for extraction of speaker embeddings called x-vectors based on mel-frequency cepstral coefficients and gammatone frequency cepstral coefficients. Since the dataset used in this study is imbalanced, various over- and undersampling techniques were applied to the training set to ensure robustness of models’ decision boundaries. The models were trained on both imbalanced and resampled training sets using 5-fold cross-validation. The best results were obtained for Multi Layer Perceptron trained on GFCC-based x-vectors, achieving accuracy of 0.8184, F1-score of 0.8212, and ROC AUC score of 0.8810 for the testing set.

Opis:
Automatic detection of voice pathologies enables non-invasive, low cost and objective assessments of the presence of disorders, as well as accelerating and improving the process of diagnosis and clinical treatment given to patients. In this work, a vector made up of 28 acoustic parameters is evaluated using principal component analysis (PCA), kernel principal component analysis (kPCA) and an auto-associative neural network (NLPCA) in four kinds of pathology detection (hyperfunctional dysphonia, functional dysphonia, laryngitis, vocal cord paralysis) using the a, i and u vowels, spoken at a high, low and normal pitch. The results indicate that the kPCA and NLPCA methods can be considered a step towards pathology detection of the vocal folds. The results show that such an approach provides acceptable results for this purpose, with the best efficiency levels of around 100%. The study brings the most commonly used approaches to speech signal processing together and leads to a comparison of the machine learning methods determining the health status of the patient.

During the study authors stated that the main determinant of demoralisation of youth, joining scarfers subcultures, where they act with aggression and brutal violence, are disturbances in the process of their education and socialization. They have presented, in their opinion, the most important factors determining the processes leading to deviant and pathological behaviour of youth, whose negative final is criminal activity. These determinants, in their opinion, are: family environment, school, peer group and negative social background. According to the researchers, occurrence of dysfunction in these areas results in criminalisation of the youth, which takes place in an informal group, which are the scarfers. The authors conclude that it is necessary to start comprehensive programs aimed at improving situation in family, schools, which in addition to implementation of educational process, should focus their attention on pedagogical work. Without taking into account, in a comprehensive way, all of the indicated factors, we cannot achieve the goal of normalisation of environments associated with fans.
