In a pot experiment conducted in a greenhouse, the effect of CCC Bercema growth inhibitor to reduce vegetative shoot growth of perennial ryegrass and cocksfoot was studded. The experiment was set up in triplicate. Growth regulator was applied once, in a form of an aqueous solution. The following experimental objects were specified: control object, Bercema with a concentration of 10%, 20% and 30%. Ten measurements were performed every 3 days. The study was conducted under conditions of 12-hour artificial light daily. The results were statistically analyzed using two-factorial variance analysis . It was found that the greatest shoot growth reduction of ryegrass due to the highest inhibitor concentration (B- 30%), but for cocksfoot under the influence of the minimal concentration (B- 10%), compared to the control object. It has been noted that cocksfoot is more sensitive to the applied growth regulator, resulted in significantly greater inhibition of shoot regrowth than for ryegrass. In the experiment with the perennial ryegrass significant differences in the growth of shoots were found under the influence of inhibitor application at a concentration of 30%, compared to the control object. In the case of cocksfoot, all the concentration increases of Bercema inhibitor significantly differentiated the shoots regrowth.

Field experiments were conducted in two localities: Prusy (50°07’ N; 20°04’ E – one experiment) and Wierzbica (50°29’ N; 19°45’ E – two experiments) in 2003. The objective of this study was to analyse the influence of agronomic factors on the grain yield and on the content of macroelements in grain of different forms of naked oat. In Wierzbica the grain yield was determined statistically by the genotype, the phosphorus and potassium fertilization and the application of the plants growth regulator Moddus. In Prusy grain yield was determined only by the genotype (cultivars, strains). A concentration of macroelements in forms of oat was statistically different. In both localities the phosphorus and potassium fertilization and foliar application of urea, in general, had not the statistical influence on the content of the macroelements. An exception is the influence of the foliar application of urea on the content of potassium. Plant growth regulator Moddus caused changes in the content of macroelements. These changes were not always statistically significant, but always increased the concentration of macroelements. The second plant growth regulator Promalin did not cause changes in the concentration of macroelements.
Badano wpływ czynników agrotechnicznych na zawartość wybranych składników mineralnych w ziarnie nagoziarnistych form owsa uprawianych w dwóch miejscowościach. Doświadczenia polowe prowadzono na glebie brunatnej typowej (Wierzbica – 50°29’ N; 19°45’ E) i na czarnoziemie zdegradowanym (Prusy 50°07’ N; 20°04’ E). W Wierzbicy plon ziarna był różnicowany statystycznie przez genotyp, nawożenie fosforowo-potasowe oraz stosowanie regulatora wzrostu Moddus. W Prusach jedynym czynnikiem modyfikującym statystycznie plon ziarna był dobór odmiany/rodu. W obydwu siedliskach nawożenie fosforowo-potasowe i dolistne azotem na ogół nie miało statystycznie istotnego wpływu na zawartość makroskładników. Wyjątek stanowił wpływ nawożenia dolistnego azotem na zawartość potasu. Spośród regulatorów wzrostu tylko Moddus powodował tendencje do zwiększonej kumulacji makroskładników w ziarnie owsa nagoziarnistego.

The aim of the study was to investigate the influence of plant growth regulators on callus culture of common thyme (Thymus vulgaris L.) and the possibility of using such callus cultures to obtain rosmarinic acid. The explants used to initiate shoot culture were the seeds of common thyme. The seeds were individually placed into test tubes with MS culture media of macro- and micro-element content according to Murashige and Skoog (1962). For the purpose of initiation of cultures, leaves dissected along the vascular bundle were used. The leaves were placed on MS medium with the addition of BAP in combination with NAA in a concentration of 3 and 5 mg•dm-3. Propagation of callus cultures was conducted with the use of fragments of callus tissue placed on MS media supplemented with BAP used separately in a concentration of 3 mg•dm-3, and in combination with NAA (1, 2, 3 mg•dm-3) and 2,4-D in a concentration of 0.5, 1 or 1.5 mg•dm-3 respectively. In each stage of the experiment, explants placed on MS media without the addition of plant growth regulators were the control. It was found that initiation of common thyme cultures should be conducted on MS media without plant growth regulators. For the purpose of initiation of callus culture of common thyme, the optimal culture medium was MS with the addition of 3 mg•dm-3 BAP and NAA. Propagation of callus tissue of common thyme should be conducted on culture media supplemented with 3 mg•dm-3 BAP used in combination with 1 mg•dm-3 NAA. It was observed that the mass of the propagated callus tissue decreased with an increase of NAA content.

Opis:
Saponaria officinalis L. is a medicinal plant from the family Caryophyllaceae. It occurs in most European countries, as well as in North Africa, America and Western Asia. The aim of the study was to find an optimal method of sterilization S. officinalis seeds in order to obtain sterile seedlings and to multiply axillary shoots from nodal explants isolated from them, on MS medium with the addition of growth regulators (RW). In this study, 4 variants of seed sterilization were compared using NaClO at the following concentrations: 1.5% (2), 2% (3) and 2.5% (4) for a period of 11 minutes. The control was a variant in which 70% C2H5OH was used for pre-sterilization for 1 minute (1). The highest percentage of sprouted, seeds and sterile seedlings and the lowest percentage of contamination were obtained in variant (4). As a result of micropropagation of soapwort in in vitro cultures, axillary shoots were formed from nodal explants from axillary buds. The highest percentage of explants with shoots (95%) and callus tissue (48%) and the highest number of shoots from one explant (5.95) were obtained in the second passage on MS medium with the addition of 4 mg.dm-3 BAP and 0.5 mg.dm-3 NAA. Due to the wide possibilities of using soapwort, it is advisable to continue research aimed at developing an optimal and efficient plant regeneration system of this species.

Softwood cuttings of cornelian cherry were rooted in the mixture of high moss peat and perlite with pH 5.0, in a polyethylene foil tunnel with automatic mist system placed in unheated greenhouse. Two-nodded, sub-apical, 10–12 cm long soft-wood cuttings were obtained from mother shrubs of several dozen years old. They were treated or not with Rhizopon AA containing 1% auxin IBA. Bases of cuttings were wounded or not. Application of Rhizopon AA was essential for the rooting of the cornelian cherry. Cuttings not treated with auxin did not produce adventitious roots. Wounding of cuttings decreased rooting efficiency. Rooting efficiency of cornelian cherry was also influenced by genotype.

A field study was conducted to evaluate the effect of different plant growth regulators (PGRs) applied through seed priming and foliar spray on growth and yield of three okra cultivars grown under calcareous soils. The cultivars of Punjab Selection and Sabzpari produced significantly higher number of branches and leaves per plant, pod length and diameter and pod yield as compared to cv. Green ferry, while, cv. Punjab selection produced significantly greater number of flowers and pods per plant as compared to other two cultivars. Seed germination (%), plant height, and fruit set (%) were not affected by the cultivars. Among the PGR treatments, seed primed with GA3 resulted in significantly higher germination percentage and greater plant height at flowering. Seed priming and foliar spray with NAA and GA3 were effective in increasing the final plant height, number of branches, number of leaves, number of flowers and number of pods per plant and fruit set (%), fresh weight per pod and pod yield. However, pod diameter, pod moisture content and dry weight per pod were not influenced by the PGR treatments applied. These results suggested that the PGRs have great potential to improve seed germination, enhance growth and increase yield of okra cultivars under calcareous soils.

Mulberry (Morus alba L.) is an ornamental, medicinal, and fruit plant that belongs to the Moraceae family. One of the most important techniques used in plant biotechnology is tissue culture, which enables mass production of pathogen-free plants. Cotyledon has a high potential for shoot regeneration; however, to the best of our knowledge, there are no reports on plant regeneration from cotyledon explants of mulberry. Therefore, this study aimed to evaluate the potential of seedling-derived cotyledon segments to obtain shoot multiplication of mulberry. Various concentrations (0, 0.25, 0.5, 1, 2, 3, and 4 mg/l) of thidiazuron (TDZ) in combination with indole butyric acid (IBA) were used in a completely randomized design in three replications. The results showed that the highest percentage of regeneration frequency (96.67%) and the maximum number of shoots (4.43) were obtained on Murashige and Skoog (MS) medium supplemented with 0.25 mg/l TDZ and 0.025 mg/l IBA. In the rooting experiment, the maximum rooting percentage (83.33%) and the maximum number of roots per shoot (4.36) were obtained on MS medium containing 2 mg/l IBA. In vitro-raised plantlets were placed in pots and kept in room temperature for 30 days, and the plantlets showed more than 90% survival rate. On the basis of our results, the protocol described in this study has a high potential to be used in the micropropagation of this valuable plant.

Primula veris L. (Primulaceae) is a well-known medicinal herb. The callus induction response of three explant types: roots, cotyledons, and hypocotyls of four-week-old cowslip seedlings were evaluated. The highest statistically different callus induction rate was 93.6% and was obtained from root explants on MS medium supplemented with 0.1 mg/l BA and 5.0 mg/l PIC. Calli also appeared on 83.3% of cotyledon explants on MS medium supplemented with 1.0 mg/l BA and 3.5 mg/l 2,4-D and on 81.0% of root explants on MS medium supplemented with 0.1 mg/l KIN and 2.0 mg/l 2,4-D. These values were not statistically different. The average time required for callus initiation was 4 to 6 weeks, however, it depended on the explants type. The most suitable condition for callus proliferation and growth was MS medium with 0.5 mg/l TDZ and 0.1 mg/l NAA, and with 1.0 mg/l BA and 2.0 mg/l or 3.5 mg/l 2,4-D. No light conditions proved to be more favourable for cowslip calli induction and growth.
Primula veris L. (Primulaceae) to znana roślina lecznicza. Opracowano metodę otrzymywania in vitro tkanki kalusowej z trzech rodzajów eksplantatów: korzeni, liścieni i hypokotyli z czterotygodniowych siewek pierwiosnka lekarskiego. Najwyższy, istotny statystycznie i wynoszący 93,6% indukcji tkanki kalusowej uzyskano z eksplantatów korzeniowych na podłożu Murashige i Skooga (MS) zawierającego 0,1 mg/l BA i 5,0 mg/l PIC. Indukcję kalusa obserwowano także na 83,3% eksplantatów liścieniowych na pożywce MS uzupełnionej 1,0 mg/l BA i 3,5 mg/l 2,4-D oraz na 81,0% eksplantatów korzeniowych na pożywce MS zawierającej 0,1 mg/l KIN i 2,0 mg/l 2,4-D. Średni czas potrzebny do inicjacji kalusa wyniósł 4 do 6 tygodni i zależał od rodzaju eksplantatu. Podłoża MS uzupełnione 0,5 mg/l TDZ i 0,1 mg/l NAA oraz 1,0 mg/l BA i 2,0 mg/l lub 3,5 mg/l 2,4-D okazały się najodpowiedniejszymi dla dalszego rozwoju i wzrostu zregenerowanej tkanki kalusowej. Warunki bez dostępu światła były korzystniejsze dla indukcji i wzrostu tkanki kalusowej pierwiosnka lekarskiego.

Calycanthus fertilis Walt. is a shrub belonging to the family Calycanthaceae, has great potential as ornamental. In the literature there are no reports on methodpropagation of this shrub in in vitro cultures. Therefore, the aim of this study was a development of the method micropropagation of Calycanthus fertilis Walter. Shoot explants of the size 1 cm, with an apex or node with lateral meristems were placed on the media with mineral composition according to MS and WPM supplemented with BAP (from 0.5 to 2.0 mg·dm–3) and TDZ (from 0.1 to 0.5 mg·dm–3). BAP turnedout to significantly increase initiation frequency whereas TDZ inhibitedthe formation of adventitious shoots andcausedexplant death. The highest percentage of initiated explants were found in shoot fragments placed on WPM medium supplemented with 1.0 mg·dm–3 BAP. Primary explants which initiatedgrowth were transferredon the proliferation media containing WPM macroand microelement, with the addition of different cytokinin: BAP (from 0.5 to 2.5 mg·dm–3), KIN (1.0 to 5.0 mg·dm–3) or TDZ (from 0.1 to 0.5 mg·dm–3). Calycanthus multiplication in vitro shouldbe conductedon WPM media with 1.0 mg·dm–3 BAP. Proliferatedshoots were placedon the WPM rooting medium supplemented with auxins: IBA, IAA or NAA at the concentration from 0.1 to 2.0 mg·dm–3. Maximum rooting was obtained on WPM medium supplemented with 0.5 mg·dm–3 IBA. To sum up, it shouldbe statedthat an efficient method of micropropagation of Calycanthus fertilis Walt. has been developed.

Plants of ‘Catawbiense Boursault’ and ‘Eskimo’ rhododendrons or ‘Cannon’s Double’ and ‘Kilian’ azalea grown in 4 l containers were treated twice with chlormequat (2000, 4000 and 8000 mg · dm-3), trinexapac-ethyl (50, 100, 200, 400 mg · dm-3), daminozide (2500, 5000, 7500 mg · dm-3), proxeadione calcium (75, 150, 300 mg · dm-1) and once with paclobutrazol (50, 100, 200, 400 mg · dm-3). Shoot length of the subsequent growth flush following the treatments decreased with increasing rates of the growth retardants. The number of flower buds per plant increased with increasing rates of pacloburazol, chlormequat and daminozide. Prohexadione calcium was less effective in flower bud initiation and the worst results were obtained with trinexapac-ethyl.

The components crucial for fulfilling plant nutrient needs are, beside macroelements, also microelements. Microelement content in plant raw materials is often modified by various agronomic measures, therefore it does not always meet the requirements for this element. The research was conducted to determine the effect of agronomic factors on the content of zinc, copper, iron, cobalt and manganese in oat grain. Three field experiments were set up acc. to fractional factorial designs (25–1 and 34–1) in two localities (Wierzbica and Prusy) in 2003. Akt cv. revealed a lower content of zinc, copper, iron and manganese in comparison with STH 7000 and STH 4770 strains. Microelement concentrations in oat grain were determined by the selection of oat cultivar/strain and growth regulator dose. Zinc content in oat grain fulfilled the requirements for plants designed for human consumption and animal feeds. Optimal iron content but deficient contents of manganese, copper and cobalt were registered in oat grain treated as fodder.
Składnikami niezbędnymi do pokrycia potrzeb pokarmowych roślin, obok makroskładników, są również mikroelementy. Zawartość mikroelementów w surowcach roślinach jest często modyfikowana różnymi zabiegami agrotechnicznymi, dlatego też nie zawsze odpowiada zapotrzebowaniu na ten składnik. Celem przeprowadzonych badań było określenie wpływu czynników agrotechnicznych na zawartość cynku, miedzi, żelaza, kobaltu i manganu w ziarnie owsa. Trzy eksperymenty polowe założono według planów frakcyjnych (25–1 i 34–1) w dwóch miejscowościach (Wierzbica i Prusy) w 2003 r. Odmiana Akt cechowała się niższą zawartością cynku, miedzi, żelaza i manganu w porównaniu do rodów STH 7000 i STH 4770. Zawartość mikropierwiastków w ziarnie owsa była determinowana doborem odmiany/rodu owsa oraz dawką regulatora wzrostu. Zawartość cynku w ziarnie owsa spełniała wymogi stawiane roślinom przeznaczonym na cele konsumpcyjne i paszowe. Stwierdzono optymalną zawartość żelaza, a niedoborową manganu, miedzi i kobaltu w ziarnie owsa traktowanego jako pasza.

Opis:
Callus was inducted on wounded leaf explants from shoot tips of a particular Prunus domestica 'Węgierka Zwykła' clone cultivated in vitro. The improvement of Sweet Common Prune stock callus tissue parameters has been approached by experiments on culture protocols. Either for the induction or maintenance of tissue modified Murashige and Skoog medium, supplemented with different auxins and cytokinins at varying concentrations, was used. The goal was to obtain the highiest possible proliferative capacity of friable tissue without any signs of cell redifferentiation for about 10 weeks. The choice of auxin was an important factor regulating the rate and kind of tissue growth, and for the examined prune clone auxin alone brought a relatively small proportion of cells into division, so advantageous was to combine it with oxygenated cytokinin. Friable tissue was obtained on media supplemented with dicamba or with picloram, but not with 2.4-D neither alone nor combinated with IBA.
