The effects of externally applied sodium nitroprusside on superoxide dismutase (SOD), glutathione peroxidase (GPx), ascorbate peroxidase (APX), and catalase (CAT) antioxidant enzyme activities, nitric oxide (NO) levels have been investigated in salt stress resistant and sensitive sunflower plants. NaCl treatments and SNP treatments simultaneously with salt application (NaCl+SNP) were performed. SOD, GPx, APX and CAT antioxidant enzyme activities and NO levels showed differences in leaf tissues treated with 100 µM SNP, different concentrations of NaCl, and NaCl+SNP. SOD, GPx and APX enzyme activities were generally increased in sensitive variety, but decreased in tolerant variety. However, while general increase in CAT enzyme activity was determined in tolerant type, a reduction was established in sensitive type. An increase was determined in both types in NO levels. It is evident from these results that administration of NO donor SNP can cope with reactive oxygen species in both varieties. This study indicates that negative effects of salt stress on different sun flower varieties can be recovered by nitric oxide.

Introduction. The standard porcine-derived pancreatic enzyme replacement therapy (PERT) is a lifesaving treatment for patients with diseases causing exocrine pancreatic insufficiency (EPI). An attempt to replace PERT with microbial enzymes were undertaken. The aim was to highlight whether the mode of application, mixed with food or applied directly to the stomach, of pancreatic-like enzymes of microbial origin (PLEM) can affect their activity along the gastrointestinal tract. Materials and method. The activity of amylase, lipase and proteinase in the stomach, duodenum and ileum were tested in EPI pigs (n=6) after supplementation of PLEM, either orally – before and during feed consumption – or via the stomach – before and during feed consumption. Healthy pigs not treated with PLEM (n=3) served as controls. Activity of the enzymes measured in the chyme were obtained together with the digesta pH. Activity of the enzymatic residues in the stool samples was also checked. Results. The highest pancreatic enzyme activities were found in the duodenum of the healthy pigs (amylase 162,68 kU/ mL, lipase 507,34 kU/mL and protolitic (trypsin) activity 357,60 kU/mL). Nevertheless, the microbial enzymes remained also active along the entire length of the GIT – including stomach in EPI pigs, regardless of their route of administration. However, activity level was significantly lower. Discussion. Results indicate that the activity pattern of PLEM in the small intestine mimics the activity of the natural endogenous pancreatic enzymes in healthy pigs. The most physiological features of PLEM were observed when enzymes were offered orally. The magnitude of PLEM activity in the stomach of EPI pigs was essential and significantly higher than that measured in healthy pigs, thus being somewhat not physiological, and for health reasons of the patients should be further explored. Interestingly, specific trypsin-like activity was measured in all parts of the GIT after PLEM application. However, proteolytic activity of the experimental proteaze in in vitro studies did not exhibit trypsin-like activity.

Field study was performed on Haplic Luvisol soil to determine the effects of soil compaction and straw mulching on microbial parameters of soil under soybean. Treatments with different compaction were established on unmulched and mulched with straw soil. The effect of soil compaction and straw mulching on the total bacteria number and activities of dehydrogenases, protease, alkaline and acid phosphatases was studied. The results of study indicated the decrease of enzymes activities in strongly compacted soil and their increase in medium compacted soil as compared to no-compacted treatment. Mulch application caused stimulation of the bacteria total number and enzymatic activity in the soil under all compaction levels. Compaction and mulch effects were significant for all analyzed microbial parameters (P<0.001).

Opis:
One of the bioindicators most often applied to assess the quality of soil is its enzymatic activity. Undesirable changes in the activity of enzymes can imply excessive presence of substances which are harmful to soil environment, such as heavy metals. Being a heavy metal, zinc is also an element essential for maintaining proper functions of live organisms. The purpose of this study has been to determine the significance of changes occurring in moderately heavy soil under the influence of zinc. The experiment was carried out in three replicates under laboratory conditions. Sandy loams of pH 5.5 and 7.0 were used for the trials. The soils were contaminated with zinc according to the following design: control (natural content), raised content (70 mg Zn2+ kg–1), weakly polluted (200 mg Zn2+ kg–1 ), moderately polluted (500 mg Zn2+ kg–1), heavily polluted (1,500 mg Zn2+ kg–1) and very heavily polluted soil (5,000 mg and 10,000 mg Zn2+ kg–1). The soil samples prepared as above were brought to the moisture content of 50% maximum water capacity and incubated at 25oC for 120 days. On day 30, 60 and 120, the activity of dehydrogenases, β-glucosidase, urease, acid phosphatase and arylsulphatase was determined. Based on these determinations, the following indices were calculated: ED50, the index for resistance (RS) and the index for resilience (RL). The tests have demonstrated that as the rate of soil contamination with zinc increased, the activity of all the analyzed enzymes was significantly depressed. The negative influence of zinc contamination on the activity of particular enzymes, irrespective of the soil pH, persisted throughout the whole experiment. In respect of their sensitivity to zinc, the enzymes can be ordered as follows: arylsulphatase > dehydrogenases > acid phosphatase > urease > β-glucosidase. Zinc contamination caused lasting changes in the soil environment, but the return to the state of equilibrium was the quickest in the case of dehydrogenases (RL = 0.276), less rapid for arylsulphatase (RL = 0.173) and the slowest for acid phosphatase (RL = 0.064). In contrast, the activity of urease, instead of regenerating, was increasingly disturbed (RL = 0.350). Soil acidification was the factor that most evidently exacerbated the negative influence of zinc on the activity of β-glucosidase and arylsulphatase. Values of ED50 for the activity of particular enzymes were varied. In the soil of pH 7.0, they ranged from 3,324 mg Zn2+ kg–1 for β-glucosidase to 412 mg Zn2+ kg–1 for dehydrogenases, and in the soil of pH 5.5, they varied from 1,008 Zn2+ kg–1 for β-glucosidase to 280 mg Zn2+ kg–1 for arylsulphatase.
Aktywność enzymatyczna gleby stanowi jeden z najczęściej wykorzystywanych biowskaźników do oceny jakości gleby. Niekorzystne zmiany w aktywności enzymów mogą świadczyć o nadmiernej zawartości substancji szkodliwych dla środowiska glebowego, do których należą również metale ciężkie. Cynk jest też niezbędnym składnikiem do prawidłowego funkcjonowania organizmów żywych. Celem badań było określenie istotności zmian zachodzących w glebie o średniej kategorii agrotechnicznej ciężkości pod wpływem cynku. Doświadczenie wykonano w warunkach laboratoryjnych, w 3 powtórzeniach. Do badań wykorzystano gliny piaszczyste o pH 5,5 oraz o pH 7,0, które zanieczyszczono cynkiem wg następującego schematu: kontrola (zawartość naturalna), zawartość podwyższona (70 mg Zn2+ k g–1), słabe zanieczyszczenie (200 mg Zn2+ k g–1), średnie zanieczyszczenie (500 mg Zn2+ k g–1), silne zanieczyszczenie (1500 mg Zn2+ k g–1) i bardzo silne zanieczyszczenie (5000 mg oraz 10 000 mg Zn2+ k g–1). Tak przygotowane próbki gleby uwilgotniono do poziomu 50% maksymalnej pojemności wodnej i inkubowano w temp. 25°C przez 120 dni. W 30., 60. oraz 120. dniu określono aktywność dehydrogenaz, β-glukozydazy, ureazy, fosfatazy kwaśnej oraz arylosulfatazy. Na ich podstawie obliczono wskaźniki ED50, oporności (RS) i powrotu do równowagi (RL). W wyniku badań stwierdzono, że wraz ze zwiększeniem stopnia zanieczyszczenia gleby cynkiem aktywność wszystkich badanych enzymów ulegała istotnemu zmniejszeniu. Negatywny wpływ zanieczyszczenia cynkiem na aktywność enzymatyczną gleby, niezależnie od jej pH, utrzymywał się przez cały okres trwania badań. Enzymy pod wpływem wrażliwości na zanieczyszczenie gleby cynkiem można uszeregować następująco: arylosulfataza > dehydrogenazy > fosfataza kwaoena > ureaza > β-glukozydaza. Zanieczyszczenie cynkiem powodowało długotrwałe zmiany w środowisku glebowym, ale najszybciej do stanu równowagi powracały dehydrogenazy (RL = 0,276), nieco wolniej arylosulfataza (RL = 0,173), i najwolniej fosfataza kwaoena (RL = 0,064). Natomiast aktywność ureazy nie tyko nie regenerowała się, ale stan zaburzenia ulegał pogłębieniu (RL = -0,350). Zakwaszenie gleby w największym stopniu wzmagało negatywne działanie cynku na aktywność β-glukozydazy i arylosulfatazy. ED50 dla aktywności poszczególnych enzymów było zróżnicowane. W glebie o pH 7,0 wahało się od 3324 mg Zn2+ k g–1 dla β-glukozydazy do 412 mg Zn2+ k g–1 dla dehydrogenaz, a w glebie o pH 5,5 od 1008 mg Zn2+ k g–1 dla β-glukozydazy do 280 mg Zn2+ k g–1 dla arylosulfatazy.

Electrolyte efflux, chlorophyll, malondialdehyde content and enzymatic activities (catalase, peroxidase) in cucumber cotyledons as well as stability of model lipid membranes in the presence of some new, acyclic and cyclic, aminophosphonates were studied. They also differed in the substituents at the phosphorus and nitrogen atoms. It was found that the effectiveness of the compounds to influence mentioned parameters depended on their lipophilicity and structural features. The most active compounds were these with isopropyl substituents at the P atom unless their hydrocarbon chains at the N atom were not too long. Butyl substituents at phosphorus significantly decreased aminophosphonates efficiency. Similar effect was observed when trans-type hydrocarbon chain was replaced with a short-branched one. It also seems that cyclic compounds with hexane rings were weaker modifiers of model and biological membranes than these with pentane rings. Results show that aminophosphonates interact with plasma membranes and presumably induce oxidative stress.
Badano wpływ nowych cyklicznych i acyklicznych aminofosfonianów na zawartość chlorofilu, dialdehydu malonowego oraz aktywność enzymów (katalaza, peroksydaza) w liścieniach ogórka. Określono efekt działania związków na stabilność modelowych błon lipidowych i wypływ elektrolitów z tkanki roślinnej. Badane aminofosfoniany różniły się ponadto podstawnikami przy atomie fosforu i azotu. Stwierdzono, że efektywność działania związków zależała od ich lipofilowości i kształtu cząsteczki. Najbardziej aktywne aminofosfoniany zawierały podstawnik izopropylowy przy atomie fosforu oraz niezbyt długi łańcuch węglowodorowy przy atomie azotu. Podstawnik butylowy przy atomie fosforu istotnie obniżał efektywność działania aminofosfonianów. Wydaje się, że cykliczne związki z pierścieniem cykloheksanowym w słabszym stopniu wpływają na membrany biologiczne i modelowe w porównaniu do związków zawierających cyklopentan. Wyniki badań wskazują, że aminofosfoniany oddziałują na membrany plazmatyczne i przypuszczalnie indukują stres oksydacyjny.

The objective of this study was to investigate the effect of exogenous selenium (Se) supply (0, 2, 4, 8, 16 μM) on the growth, lipid peroxidation and antioxidative enzyme activity of 100 mM NaCl-stressed melon (Cucumis melo L.) seedlings. Salt stress significantly reduced the growth attributes including stem length, stem diameter, dry weight and increased antioxidative enzyme activity [superoxide dismutase (SOD), peroxidase (POD), catalase (CAT)]. Moreover, the plant exhibited a significant increase in electrolyte leakage and malondialdehyde (MDA) content under NaCl stress. Se supplementation not only improved the growth parameters but also successfully ameliorated the adverse effect caused by salt stress in melon seedlings. However, the mitigation of NaCl-stressed seedlings was different depending on the Se concentration. At lower concentrations (2–8 μM), Se improved growth and acted as antioxidant by inhibiting lipid peroxidation and increasing in SOD and POD enzymes activity under salt stress. At higher concentrations (16 μM), Se exerted diminished beneficial effects on growth. Whereas CAT activity was enhanced. The result indicated that Se supplementation had a positive physiological effect on the growth and development of salt-stressed melon seedlings.

Enzyme activities in soils are influenced by soil characteristics and pollutants and could be indicator of soil quality. This study was undertaken to determine the relationship between physicochemical characteristics, heavy metals contents and enzyme activities (EA) in agricultural soils in the Saiss plain (Morocco). The analysis of six agricultural soil samples (sites 1–6), collected from urban and periurban agricultural soils of Fez region, showed that soils are generally alkaline with high CaCO3 that rich 46%, EC (525–703 μS/cm), rich in organic matter (3.14–5.80%). The Cr, Cu, Ni, Pb and Zn concentrations in the studied area are generally greater than the Upper Continental Crust, with a deceasing order: Zn> Cr> Pb> Cu> Ni. Geo-accumulation index showed that soils are unpolluted to moderately polluted except site 5 and site 6 that are moderately to strongly polluted by respectively Cu, Pb, Zn and Pb. Potential ecological rіsk factors were below 40, which means low potential ecological rіsk except site 5 that has moderate potential ecological risk by Cu and Pb. Soil potential ecological risk indices were found <150 indicating low ecological risk. According to this index, Cu caused more serious pollution than the other elements. The enzyme actіvities of the sіx soil samples showed almost the similar ranges of values. These EA showed the highest values in site 5.The sensіtivity of soil enzyme to heavy metals were observed UREA>PHOS>GALA. Pearson’s correlation showed significant posіtive correlatіon between studied soil EA and between these EA and heavy metals (Cu, Pb and Zn), and significant negative correlatіon between EA and heavy metals (Cr and Ni). The results of this study enrich and provide data base of the impact of heavy metals on soil enzyme actіvities in agrіcultural soils in the Saiss plain.

The aim of this study was to assess the possibility of using biological and biochemical parameters in the evaluation of forest soil quality and changes caused by land use. The study attempted to determine a relationship between the enzymatic activity of soil, the number of earthworms and soil physico-chemical properties. The study was carried out in central Poland in adjoining Forest Districts (Przedbórz and Smardzewice). In soil samples taken from 12 research plots, basic physico-chemical properties, enzyme activity (dehydrogenase, urease) and density and biomass of earthworms were examined. Enzyme activity showed a large diversity within the forest site types studied. The correlations between the activity of the enzymes studied and C/N ratio indicated considerable importance of these enzymes in metabolism of essential elements of organic matter of forest soils. Urease and dehydrogenase activity and earthworm number showed susceptibility to soil pH, which confirmed relationships between enzyme activity and abundance of earthworms and soil pH in H2O and KCl.

Opis:
Ceruloplasmin (Cp) is the major antioxidant in plasma, a protein that carries about 95% of the total copper. Multiple biochemical activities of ceruloplasmin have been described, including copper transport or oxidation of Fe(II) to Fe(III) for subsequent uptake by transferrin and ferritin. Reduction of the pool of free Fe(II) ions by Cp prevents the generation of free radicals and reactive oxygen species by oxidation of Fe(II), thus inhibiting the Haber-Weiss reaction. The removal of both free Fe(II) and Fe(III) ions from blood plasma by polyphenols enhances the antioxidant system of the living organism. However, the mechanism of interactions between exo- and endogenous antioxidants is still under consideration. The effect of raspberry seed extract (RSE) on the ferroxidase activity of ceruloplasmin isolated from plasma of patients with chronic arterial occlusion of the lower limbs due to atherosclerosis (CpAO) was investigated. Moreover, the effect of RSE on the ferroxidase activity of Cp isolated from healthy volunteers (CpC) was also estimated. The ferroxidase activity of Cp, expressed as ΔFe(II), was determined by spectrophotometry with the use of the Fe(II) – histidine complex and ferrozine as a chromogenic reagent. The addition of RSE to samples with the same amount of both CpAO or CpC in each caused an increase in ΔFe(II). The polyphenol-rich RSE may assist Cp in the fight against free radicals and reactive oxygen species when the disease occurs due to an excessive use or reduced production of endogenous antioxidants.

Opis:
Celem badań było określenie aktywności ureazy na wybranych glebach Polski: bielicowych i brunatnych wytworzonych z piasków i glin, lessów, czarnych ziem i mad rzecznych. Badaniom poddano próbki z poziomu 0-10 cm, które pobrano z Banku Gleb Instytutu Agrofizyki PAN. Aktywność ureazy zawierała się w przedziale 0,3256 - 1,7896 pM N-NH4 kg-lh-1. Najniższą aktywność ureazową zanotowano w glebach bielicowych i wynosiła ona 0,33 pM N-NH4 kg-lh-1. W glebach brunatnych wytworzonych z piasków i lessów aktywność ureazy wahała się w granicach 0,55 - 1,00 pM N-NH4 kg-lh-1. Najwyższą aktywność stwierdzono w glebach: czarnych ziemiach, madach rzecznych, glebach murszowych i murszowatych 1,43 - 1,79 pM N-NH4 kg-lh-1. W oparciu o analizę statystyczną otrzymanych wyników można stwierdzić, że aktywność ureazowa pozostaje w ścisłym związku z zawartością materii organicznej.
The aim of the investigation was to measure activity of urease in different polish soils such as podzolic and brown developed from sands and loams, brown developed from loess, as well as in black earths and aluvials soils. The measurements were performed on samples taken from 0-10 cm depths. The urease activity was in the range of 0,3256 - 1,7896 pM N-NH4+ kg''h‘'. The lowest activity was noted in the podzolic soil and was on the level of 0,3256 pM N-NH4+ kg’'h''. In the brown and lessive soils the urease activity was in the rangę 0,55-1,00 pM N-NH4+ kg''h_l. The highest urease activity were found in black earths, alluvials soils, Eutric Histosol Soils 1,43-1,79 pM N-NH/kg1 h’’. The results obtained allow to conclude that the urease activity is tightly connected with the organie matter content of the soil.
