Opis:
Paper is one of the main materials in the manufacture of packaging for contact with food. Microbes that are present in the paper or board may penetrate into the food- health hazard for the consumers. Consistent and unified regulations have not been enacted yet in the UE to prevent health hazards which may arise from infected packaging. Examination was carried out of the microbiological contamination in various paper and paperboard materials designed for uses in food packaging. The following three methods were applied in the testing: defibering-, agar flooding- and smear method. It has been proven that the defibering method yields best results in the testing of microbes’ concentration in paper and board. In some of the commercial materials tested a high level of contamination was found raising concern whether the products can be used in contact with food. No clear criteria are presently defined to qualify the material in respect of microbiological purity.
Papier stanowi jeden z podstawowych surowców do wytwarzania opakowań mających bezpośredni kontakt z żywnością. Drobnoustroje obecne w papierze lub tekturze mogą spowodować szkodliwe następstwa dla konsumentów jeśli mają możliwość przemieszczania się z opakowania do produktu spożywczego. Przepisy prawne jedynie ogólnie określają wymagania odnośnie skażenia mikrobiologicznego podając, że papier i tektura powinny być odpowiedniej jakości mikrobiologicznej. Celem pracy było zbadanie poziomu zanieczyszczeń mikrobiologicznych w różnego rodzaju papierowych i tekturowych materiałach opakowaniowych do żywności. Przeprowadzono badania czystości mikrobiologicznej opakowań z tektury i papieru trzema metodami: rozwłókniania, zalewania agarem oraz metodą wymazową. Wykazano, że metoda rozwłókniania jest najlepsza do badania stężenia drobnoustrojów w próbkach papieru i tektury. W części badanych opakowań oznaczono wysoki poziom skażenia co, przy braku jednoznacznych kryteriów, może budzić uzasadnione wątpliwości czy taki produkt kwalifikuje się jako odpowiedni do kontaktu z żywnością. co do kwalifikacji takiego produktu jako odpowiedniego do kontaktu z żywnością.

PLA fibres with the addition of the microbicidal agent Sanitized were examined by the two quantitative standards JIS L 1902:2002 and ASTM: E2149–01. It was found that the two different methods used to assess the antibacterial activity of one sample may produce results that contrast with each other. The impact of the testing conditions upon the results obtained was deliberated. Discussed are criteria to interpret the results obtained by the use of the two methods. The antibacterial activity of lactic acid liberated from PLA in an aqueous medium was confirmed. It was also examined how the laundering of fibers affect the antibacterial activity of the samples tested.
Przedstawiono wyniki badania włókien, wykonanych z poli(kwasu mlekowego) z dodatkiem środka bójczego Sanitized, dwoma metodami ilościowymi według norm JIS L 1 902:2002 i ASTM: E2149–01:2010. Wykazano, że rozbieżność otrzymanych wyników powstała jako rezultat różnic w zastosowanych metodykach. Omówiono kryteria oceny wyników w obu normach. Potwierdzono antybakteryjne działanie kwasu mlekowego.

Opis:
Modification of commercial cellulose products commonly used in homes and hospitals was the main purpose of the research herein presented. Enhanced moisture sorption and antimicrobial properties were conferred upon regular commercial gauze. Nanoparticles of microcrystalline chitosan and complex chitosan/alginateNa/Ca were used in the modification. A method was elaborated for the preparation of polymeric materials with a particle size below 1 μm by means of an ultrasonic reactor (Hielscher UP 200S). Modified commercial dressing materials were obtained charecterised by a largely increased absorption capacity, thus easing the transportation of moisture to the outside of the dressing and providing an environment optimal for wound healing. Thanks to the internal surface developed and adequately selected composition, the modified cellulosic materials exhibit antibacterial and antifungal properties.
Głównym celem przeprowadzonych badań była modyfikacja komercyjnego materiału celulozowego w postaci gazy opatrunkowej, stosowanej w codziennym użytku jak i w szpitalach, celem nadania im zwiększonej zdolności sorpcji wilgoci jak i właściwości przeciwmikrobowych. Do modyfikacji surowca wykorzystano nano-cząsteczki mikrokrystalicznego chitozanu oraz kompleksu chitozan/alginanNa/Ca. Metodę otrzymywania polimerów o rozmiarach cząstek o wielkości poniżej 1 μm opracowano przy wykorzystaniu reaktora ultradźwiękowego. Otrzymano modyfikowane handlowe materiały opatrunkowe o kilkukrotnie wyższej zdolności sorpcyjnej, które ułatwiają odprowadzanie wilgoci na zewnątrz opatrunku i zapewniają optymalne środowisko gojenia się ran. Modyfikowane materiały celulozowe dzięki rozwiniętej powierzchni wewnętrznej i odpowiednio dobranym składzie charakteryzują się właściwościami przeciwbakteryjnymi oraz przeciwgrzybowymi.

Opis:
An impact of the decomposition products of Poly(butylene succinate adipate) (PBSA) (Bionolle 3001) in the form of polymer chips and nonwovens made of the polymer upon the qualitative and quantitative microbiological profile of the soil in which samples of the materials were incubated was investigated. The mass loss of the polymer and nonwovens as a result of their biodegradation proceeding in the soil was compared. Two kinds of the soil were applied in the biodegradation tests: garden soil and an agriculture soil taken from the experimental field plots of the Agriculture University (AU) of Cracow. The AU soil despite low moisture content is fertile and holds a great amount of microorganisms. The garden soil does not offer favourable conditions for the biodegradation process, confirmed by a low mass loss in the incubated Bionolle#3001 samples.
W ostatnich dekadach ważnym kierunkiem badań jest opracowanie takich materiałów, które będą ulegały całkowitemu rozkładowi w bardzo krótkim czasie, a produkty ich biodegradacji będą przyjazne środowisku. Gleba jest źródłem wielu mikroorganizmów charakteryzujących się różnymi właściwościami biochemicznymi. Enzymami odgrywającymi główną rolę w degradacji polimerów są esterazy i proteazy zasadowe wytwarzane przez laseczki z rodzaju Bacillus sp., grzyby z rodzaju Tritrachium sp., promieniowce z rodzaju Amycolatopsis sp., oraz Paenibacillus sp. Określono wpływ produktów rozkładu polimeru PBSA (Bionolle#3001) i włókniny otrzymanej z tego polimeru na skład jakościowy i ilościowy profilu mikrobiologicznego gleby, w której inkubowano próby polimeru i włókniny. Dokonano również porównania stopnia ubytku masy polimeru i włókniny w wyniku biorozkładu w glebie ogrodniczej i pochodzącej z pól doświadczalnych Uniwersytetu Rolniczego w Krakowie.

Introduction: Glucagonoma is a rare neuroendocrine tumor with characteristic features such as the presence of the glucagon-producing tumor, diabetes, and necrolytic migratory erythema. Case presentation: the 60-year-old patient was admitted to hospital with periodic pain in the right and middle upper abdominal appearing after eating. Laboratory tests presented a high glucose level and anemia. Tumor of the body and tail of the pancreas passing the organ pouch has been found during the surgery. Diagnosis of glucagonoma was confirmed in histopathological examination in immunohistochemical stainings: a positive reaction was observed with chromogranin, synaptophysin and CEA. The proliferative activity of Ki-67 was less than 1%. Staining for glucagon also was positive so confirmed the presence of neuroendocrine tumor - glucagonoma. Conclusions: Glucagonoma sometimes may occur without characteristic features which may cause delayed diagnosis. Early diagnosis of glucagonoma is important because it increases the chances of successful recovery.

Introduction: Whipple’s disease is a chronic systemic infectious disorder with Tropheryma whipplei as an etiologic agent, occurring rarely and affecting numerous organs and systems. The variety of symptoms and a non-typical course make it difficult to establish a proper diagnosis. Purpose: In this study, etiopathogenesis, diagnostics and treatment of Whipple’s disease were presented based on the case report of 60-year-old man diagnosed with Whipple’s disease. Case presentation: Persistent diarrhoea with weight loss, lymphadenopathy in the abdominal cavity and moderate microcytic anemia predominated in the clinical picture. Diagnosis was put based on the clinical picture and macroscopic assessment of the small intestine and the presence of macrophages filled with a PAS-positive substance in the lamina propria. To deepen diagnostics, samples collected were assessed showing macrophages with the damaged mucosa, containing numerous elongated micro-organisms whose ultrastructure corresponded to Tropheryma whipplei. The patient’s clinical conditions improved after antibiotic therapy. Conclusions: It is vital to remember about Whipple’s disease in patients with chronic diseases due to a non-specific clinical picture and difficulties in establishing a proper diagnosis. When the disease is diagnosed unequivocally, proper and effective antibiotic therapy should be instituted immediately.

Purpose: Pancreatic intraepithelial neoplasia (PanIN) is one of the most commonly occurring precancerous lesion in pancreas which leads to development of pancreatic ductal adenocarcinoma (PDAC). We assessed the presence and grade of pancreatic intraepithelial neoplasia in the course of various diseases of the pancreas and its correlations with chosen clinicopathological parameters. null Results: A total of 276 foci of PanIN were identified in 94 patients. The most common lesions were PanIN 1a and PanIN 1b which together constituted 68.2 % of all lesions, whereas the PanIN 2 was present in 21.7% and PanIN 3 in 10.1% of patients. No statistical differences were observed in gender tendency for the development of PanINs. There was correlation between age of patients and degree of PanIN (p=0.034). There is no statistical difference in PanIN frequency among patients with pancreatic ductal adenocarcinomas, neuroendocrine tumors, chronic pancreatitis and pancreatic cysts (p=0.592). Conclusions: Our study showed that important factor in development of pancreatic intraepithelial neoplasia is age and the presence of PanIN in nonneoplastic diseases in older people should be included to the group with increased risk of cancer development.

Introduction: Pancreatic cancer is characterized by its aggressiveness and poor prognosis. Furthermore, mortality is one of the highest among all types of cancers. It has been observed that the expression of Fascin-1 (the actin-bundling protein which enables the motility of cells) is higher in cancer cells and is correlated with invasiveness and metastasis. Purpose: To investigate the expression of Fascin-1 in histopathological specimens from patients treated for pancreatic cancer and its relationship with histopathological parameters. Materials and methods: The study was performed on a group of 52 patients diagnosed with pancreatic cancer in the Medical University of Bialystok Clinical Hospital. The expression of Fascin-1 was evaluated in tissue samples using the immunohistochemical method and was rated as “present” or “absent”. The analysis of histopathological parameters was performed in correlation with Fascin-1 expression. Results: Fascin-1 expression was observed in the cytoplasm of cancer cells in 42/52 cases (80.8%). Fascin-1 expression occurred more frequently among patients with lymph node metastases (92.6%) than without this type of metastases (68%) (p=0.024). Likewise, the expression of the investigated protein was observed more often with the presence of distant metastases (100%) than without those metastases (74.4%) (p=0.043). There were no statistically significant differences about age, sex, histological type of cancer, grade of histological differentiation, desmoplasia, inflammatory infiltration, foci of hemorrhage, necrosis, and MVD. Conclusion: The expression of Fasicn-1 is correlated with the presence of metastases and can be a useful marker of pancreatic cancer involvement.

Introduction: Despite the progress in gaining knowledge about carcinogenesis, it is still unclear what processes are directly responsible for the differentiation of gastric cancer into its intestinal or diffuse form. Dividing of these two forms is based on one of the oldest, yet still commonly used classifications – the classification of Lauren. There are many factors that may influence the formation of gastric tumors of various aggressiveness. Purpose: To evaluate the expression of proteins: fragile histidine triad (FHIT), E-cadherin, α-catenin, γ-catenin, cathepsin B, epidermal growth factor (EGF), HER-2, MMP-9, MCM-2, Bak, Bax, BID, Bcl-XL, p53, FasL, Bcl-2, caspase-8, procaspase-3 in gastric cancer cells, depending on the type of tumor by Lauren classification. Materials and methods: Study group consisted of 91 patients treated surgically for gastric cancer in the Second Department of General and Gastroenterological Surgery, Medical University of Bialystok in years between 2000 and 2006. Results: It is shown, that the expression of E-cadherin was significantly higher in the Lauren I gastric cancer cells than in Lauren II. In case of caspase-8 there has been significantly less frequent expression of this protein in Lauren I gastric cancer cells compared to Lauren II. The authors describe no statistically significant differences in the expression of other proteins taken into consideration. Conclusions: These results suggest the role of adhesion and apoptosis-related proteins in the development of two different types of gastric cancer according to Lauren’s classification.

Introduction: Colorectal cancer is one of the most common cancers occurring in Poland. Unfortunately, this cancer is most often diagnosed at the time of great advancement. This is caused by the appearance of specific symptoms only in the late stages of cancer. Also, such low detection of early stages of adenocarcinoma may be caused by disregarding of slight symptoms. Purpose: To analyse symptoms in patients with colorectal cancer and correlate with chosen clinical-pathological parameters. Materials and methods: The study group consisted of 46 patients diagnosed with colorectal cancer. Information on symptoms associated with cancer (subjective and objective) have been selected from patients' medical history. The presence of these symptoms was correlated with the age and sex of patients, tumor location, histological type of cancer, grade of histological malignancy (G), stage of tumor (T stage), presence of lymph node metastases and distant metastases. Results: It has been shown that the presence of pain complaints described by the patient are associated with the occurrence of cancer in the colon. The painfulness and pathological resistance diagnosed by the physician are more often associated with cancer located in the colon. In the case of tumors located in the rectum, faecal admixtures appear more frequently. It has also been shown that the presence of admixtures of blood and mucus in the stool is associated with more advanced local tumors, infiltrating pericolorectal tissues (T3 + T4). Conclusions: Familiarity with clinical symptoms of colorectal cancer could make patients more sensitive to more often screening for cancer. Analysis of these symptoms could indicate to the physician the location or stage of the cancer.

Opis:
Purpose: To evaluate the clinical significance of MMP-2 and TIMP-2 concentrations in the sera of patients diagnosed with colorectal cancer. Methods: The study group comprised 48 patients with colorectal carcinoma and 24 healthy controls. The serum concentration of MMP-2 and TIMP-2 proteins was evaluated by the ELISA method. Results: The mean level of MMP-2 in the sera of patients with colorectal cancer was 39.4 ng/ml and was significantly lower compared with the control (p<0.001). A decrease in TIMP-2 protein in the sera of patients with colorectal cancer was also observed where its mean level was 132.3 ng/ml. The concentration of MMP-2 and TIMP-2 did not correlated with any clinicopathological parameters, except for the patients’ age. In addition, the concentration of MMP-2 in the sera of patients with CRC correlated negatively with the number of white blood cells in the blood and the prothrombin index. The concentration of TIMP-2 correlated positively with potassium and urea concentration in the blood. Conclusions: The results of our study indicate lack of clinical usefulness of determining the levels of MMP-2 and TIMP-2 in the sera of patients with colorectal cancer. However, these proteins play an important role in the carcinogenesis of colorectal cancer.

Opis:
Purpose: Recent literature data indicate a key role of apoptosis in the pathogenesis of inflammatory bowel disease. The aim of the study was to evaluate the expression of Bax, Bid, Bcl-2 and Bcl-xl in non-dysplastic and dysplastic epithelium in inflamated mucosa of patients with ulcerative colitis. Methods: The study consists of 18 patients with diagnosed ulcerative colitis. The expression of proteins was determined immunohistochemically. Results: Lack of Bax expression in normal epithelium of the inflamed intestinal mucosa (94.4%) and a weak expression of this protein were found in dysplastic glandular cells (67%). The Bax expression of dysplastic epithelium correlates with reduced severity of chronic inflammation (p<0.005). Bid expression in non-dysplastic glands was found in 67% of cases vs. 16% in dysplastic epithelium that was associated with the occurrence of epithelial erosions or ulcers (p<0.05). Moderate cytoplasmic expression of Bcl-xl was noted in 27.7% of patients in normal epithelium and in 66.1% within dysplastic lesions. Bcl-xl expression in dysplastic glandular cells correlated with the presence of neutrophils in the lamina propria (p <0.05). Conclusions: The immunohistochemical expressions of Bax, Bcl-2 and Bcl-xl increase and Bid protein expression decreases in dysplastic glandular tubes as compared to non-dysplastic intestinal epithelium in inflamed mucosa, which may suggest an imbalance of controlled cell death in ulcerative colitis.

Opis:
Purpose: Growth differentiation factor-15 (GDF- 15) protein belongs to a transforming growth factor-β family which determines the growth and differentiation of cells. In cancers, GDF-15 influences on the proliferation, differentiation, viability, migration and invasiveness of cancer cells. The aim of our study was to evaluate the expression of GDF-15 in the tissue and its levels in sera of patients with colorectal cancer. Materials and methods: The level of GDF-15 in the sera of 55 patients diagnosed with colorectal cancer was determined using the ELISA method whereas expression of this protein was performed by immunohistochemical method. Results: The mean value of GDF-15 levels in the sera of patients with colorectal cancer was significantly higher than in healthy control group (p<0.001). The expression of GDF-15 in the tissue was weak, moderate and strong in 23.6%, 15.7% and 60.7% cases, respectively. Statistical analysis showed that the expression of GDF-15 correlated with patients’ age (p<0.005) and non-mucinous type of cancer (p<0.001). The high GDF-15 levels in the serum was associated with tumor size (p<0.01) and distant metastases (p<0.05). Conclusions: According to our results, we postulate that the level of GDF-15 in serum can be use to assess the metastatic behavior of colorectal cancer
