The aim of the study was to optimize conditions for producing Salmonella Enteritidis recombinant heat shock protein 60 (rHsp60). Seven Escherichia coli host strains (Rosetta, Turner, C41, C43, Origami, BL21pLys, Rosetta pLys) were transformed by a recombinant plasmid containing Hsp60 gene from Salmonella Enteritidis, and then cultured and induced by isopropyl-β-D-thiogalactopyranoside (IPTG). The highest S. Enteritidis rHsp60 yield was obtained using E. coli strain C41. Induction of this strain using IPTG allowed the yield 400 μg of S. Enteritidis Hsp60 protein/2L of culture, but by autoinduction the yield exceeded 800 μg/2L.

Opis:
Introduction. Laryngeal squamous cell carcinoma (LSCC) is a common type of head and neck malignancy. Because of unsatisfactory results of therapy, development of new strategies for LSCC treatment is needed. It is believed that heat shock protein 70 (HSP70) is involved in pathogenesis of LSCC. Thus, targeting HSP70 seems to be promising strategy for laryngeal cancer treatment. Aim. The aim of the study was to assess the HSP70 concentration in laryngeal squamous cell carcinoma specimens and its correlation with tumor volume and TNM staging. Material and methods. An ELISA method and a Bradford protein assay were used to evaluate the HSP70 concentration in peripheral blood cells, tumor tissue and lymph nodes from the patients suffering from LSCC. Results. We demonstrated that the HSP70 concentration is significantly different between examined compartments. The highest level was observed in peripheral blood, while the lowest was in the lymph nodes. The HSP70 expression was correlated to tumor volume. Conclusion. Our results showed varied expression of HSP70 in tissue from patients with LSCC, but there was no association between HSP70 concentration and TNM staging. Currently, application of HSP70 inhibition as a LSCC treatment could be rather associated with systemic blocking of this molecule than target inhibition in tumor tissue. However, further analysis on a larger group of patients is needed.

Opis:
Two genes of the extreme thermophilic archaeon Thermococcus litoralis homologous to those that code for Cpn60 chaperonins were cloned and expressed in Escherichia coli. Each of the Cpn60 subunits as well as the entire Cpn60 complex crystallize in a variety of morphological forms. The best crystals diffract to 3.6 Å resolution at room temperature and belong to the space group I422 with unit cell parameters a = b = 193.5 Å, c = 204.2 Å.

Introduction. Some studies have shown the influence of Qigong on gene expression in different cells, but there is little data associated with the influence of this kind of therapy on genes expression in pheripheral monocellucar blood cells. Objective. The aim of this study was to evaluate changes in the expression of genes associated with cellular stress response in peripheral mononuclear blood cells (PMBC) in healthy women. Materials and method. The experiment took place at the Japanese Martial Arts Centre “DOJO” in Stara Wieś, Poland, conducted over the course of a 4-day qigong training session. To evaluate the genes effect of this training, blood samples were taken before and after the training period. This experiment involved 20 healthy women (aged 56.2±9.01, body height 164.8±6.5 and mass 65.5±8.2). To determine the expression of HSF-1, HSPA1A, NF-kB, IL10 and CCL2 mRNA, 3 ml of venous blood was collected. The blood samples were placed in tubes allowing for separation (BD Vacutainer CPTTM) before and after the 4 days of qigong training. Isolated PMBC were used to determine gene expression using real-time qRT-PCR (quantitative reverse transcription polymerase chain reaction). Results. Significant decreases in NF-kB and CCL2 mRNA and increases in IL10, HSF1 and HSPA1A m-RNA were detected after 4 days of qigong training. The obtained findings suggest that qigong caused a reduction in the inflammatory and intensified anti-inflammatory gene expression, as well as a higher expression of HSF-1 and HSPA1A. Conclusion. The adaptive response to qigong training was similar to the adaptive response to physical activity and was detected through gene expression in PMBC. Furthermore, this kind of training is especially indicated for women because of their higher susceptibility to psychosocial stress when compared to men.

Opis:
Tomato is one of the most consumed vegetable crops worldwide. In the break stage, it is perishable, and it has a postharvest life that does not exceed three weeks at 12 to 15°C. The aim of this study was to evaluate the effect of immersion in water and in calcium salt solutions on the metabolism and quality of tomato of traditional Peumo Pink cultivar stored for 28 days at 10ºC plus a simulated trading period of 2 days at 20ºC. Fruits were treated in water at 5, 45 and 60ºC at two immersion times of 1 and 4 min or in 2% solutions of calcium chloride, lactate and propionate at 10 and 45ºC for 4 min. The respiration rate, ethylene production and heat shock protein gene expression as well as firmness, total phenolic content and antioxidant capacity of tomato were determined. Dipping in water at 60°C reduced the loss of firmness and respiratory rate of tomatoes up to 21 days at 10°C + 2 days at 20°C. Treatment in water at 45°C for 4 min and at 60°C for 1 or 4 min stimulated heat shock protein gene expression. However, fruit treated at 60°C for 1 or 4 min showed uneven ripening; hence, the immersion in water at 45°C for 4 min was the most recommended to extend the postharvest life of tomatoes. In turn, the immersion in calcium lactate and propionate solutions at 45°C increased total calcium content and maintained firmness after 28 days at 10°C + 2 days at 20°C. The use of high temperature and calcium salts for dipping would be beneficial to provide the vegetal tissues with calcium and reduce the softening of the tomato after prolonged cold storage.

The Hsp70.2/Hst70 gene, which belongs to the 70 kDa heat-shock protein (HSP) family, is expressed specifically in primary spermatocytes and spermatids. The regulatory elements required for a high level of testis-specific expression of the gene are placed between the two major transcription start sites T1 and T2 (approximately 350 and 115 bp upstream of the starting ATG codon). Here we have shown that sequences proximal to the exon1/intron splicing site in the 5' untranslated region of the Hsp70.2/Hst70 gene, which include a highly conserved element called box B, are required for efficient expression of the chloramphenicol acetyltransferase reporter gene in testes of transgenic mice. However, in spite of the drastically reduced overall activity, the stage-specific expression pattern of the transgene was preserved after removal of these sequences. We have also shown that GC-box located downstream of the box B (approximately 210 bp upstream of the starting ATG codon) is indispensable for efficient expression of the Hsp70.2/Hst70 gene promoter in spermatogenic cells. The GC-box specifically binds proteins present in nuclear extracts from testes (putatively Sp1-like factors). A change in the pattern of such GC-box-interacting factors corresponds to activation of the Hsp70.2/Hst70 gene, confirming the importance of this regulatory element.

The aim of this study was to investigate the impact of heat stress on production performance and oxidative stress in different plumage colors of Japanese quail. For this purpose, a total of 100 birds were used in this study. The 25 birds belonged to Wild-type (n=25, grey), Tuxedo (n=25, black), Golden (n=25, yellow) and Recessive white (n=25). The birds were reared for 42 days in an environmentally controlled room at 39°C and relative humidity of 60-65%. The body weight, body weight gain (g/bird/day), and feed conversion ratio were not different between the groups (p>0.05). However, the feed intake (g/bird/day) of the Wild-type had a higher value than the Tuxedo (black) group counterparts between 15 and 21 days different (p<0.05). There was no significant effect of heat stress on the carcass traits (p>0.05). Spleen weights were different between the groups (p<0.05). The yellow group had the highest spleen weight. The highest MDA level was found in the Recessive White variety, followed by Wild-type (grey), Golden (yellow) and Tuxedo (black), respectively. However, there were no statistical differences amongst the groups (p>0.05). There was also no statistical significance in glutathione (GSH) and superoxide dismutase (SOD) levels (p>0.05). The heat shock protein 70 kDa (HSP70) level was significantly different between the groups (p<0.001). The highest percentage was observed in the Golden (5.06%) and the lowest in the White (1.43%) variety. There was no superior color variety of Japanese quail regarding fattening performance and carcass traits. It is conceivable that when considering the stress response of the different colors, the Golden group is more sensitive to stress due to the hepatic and cellular level of HSP70.

We and others have recently obtained data suggesting that cytokine-STAT signaling in many different cell-types is a chaperoned pathway initiated at the level of specialized plasma membrane microdomains called "rafts" (the "raft-STAT signaling hypothesis"). These findings are of broad significance in that all cytokines and growth factors initiate signaling in target cells by interacting with respective cell-surface receptors. The new data suggest that raft microdomains represent the units of function at the cell-surface through which ligand-stimulated STAT signaling is initiated. Moreover, recent evidence shows the involvement of chaperone proteins in regulating the STAT signaling pathway. These chaperones include the human homolog of the tumorous imaginal disc 1 protein (hTid1) which associates with Janus kinase 2 (JAK2) at the level of the plasma membrane, heat shock protein 90 (HSP90) which associates with STAT3 and STAT1 proteins in caveolin-1-containing raft and cytoplasmic complexes, and glucose regulated protein 58 (GRP58/ER-60/ERp57), a thiol dependent protein-disulfide isomerase, found in association with STAT3 "statosome" complexes in the cytosol and in the raft fraction. We suggest a function of the HSP90 chaperone system in preserving IL-6/STAT3 signaling in liver cells in the context of fever. The identification and function of protein partners associated with specific STAT species in rafts and in cytosolic complexes, and in the efficient departure of cytokine-activated STATs from the cytosolic face of rafts towards the cell nucleus are now areas of active investigation.
