Stem cells have captured considerable scientific and clinic interest because of their potential to renew themselves and to differentiate into one or more adult cell types. Thus stem cells have been recognized as a potential tool for the development of innovative therapeutic strategies in different disease disorders. Stem cells can be discriminating based on their differentiated potential as totipotent, pluripotent, multipotent or unipotent cells. There are in general three types of stem cells: embryonic, fetal and adult stem cells. While embryonic stem cell therapy has a lot of ethical concerns due to their obtaining but also unlimited proliferation and uncontrolled differentiation, fetal and adult stem cells have been used in the treatment of different diseases. The bone marrow, peripheral blood and umbilical cord blood are ideal sources of adult stem cells because there are easily accessible and contain two types of stem cells: hematopoietic stem cells giving rise to all blood cell types and mesenchymal stem cells differentiating into cells of mesodermal lineage. This review describes the general characteristics of these stem cell populations and their current applications in regenerative medicine. Additionally induced pluripotent stem cells generated through the reprogramming of differentiated adult cells are described.
Możliwość wspomagania regeneracji nieodwracalnie uszkodzonych tkanek i narządów za pomocą transplantacji komórek macierzystych skupia uwagę środowisk medycznych i opinii publicznej. Komórki macierzyste mają zdolność do samo-odnowy oraz do różnicowania się w dojrzałe komórki struktur całego organizmu. Na określonych etapach rozwoju osobniczego wykazują różny stopień ograniczenia potencjału do dalszego różnicowania, od komórek totipotencjalnych (zygota), poprzez pluripotencjalne (komórki zarodkowe) do multipotencjalnych lub unipotencjalnych (komórki somatyczne). Zarodkowe komórki macierzyste, z uwagi na nieograniczone zdolności namnażania i pluripotencjalność wydają się najbardziej optymalne do zastosowania w terapiach regeneracyjnych, lecz zastrzeżenia natury etyczno-moralnej pozyskiwania tych komórek, a także możliwość ich niekontrolowanej proliferacji po przeszczepieniu w organizmie biorcy skłoniły badaczy do poszukiwania alternatywnych źródeł komórek macierzystych, jakimi są tkanki dojrzałego organizmu. Badania ostatnich lat wykazały, iż w dojrzałych tkankach somatycznych znajdują się pluripotencjalne komórki macierzyste, co więcej odkrycia Takahashi i Yamanaka w roku 2006 r udokumentowały możliwość przekształcenia zróżnicowanych komórek izolowanych od osobników dorosłych w komórki pluripotencjalne. Zainicjowanie nowatorskich metod badawczych, których celem będzie opracowanie skutecznych sposobów przeszczepiania somatycznych komórek macierzystych w celach regeneracyjnych jest nadzieją w terapii wielu nieuleczalnych schorzeń.

Opis:
Cell walls are at the basis of a structural, four-dimensional framework of plant form and growth time. Recent rapid progress of cell wall research has led to the situation where the old, long-lasting juxtaposition: "living" protoplast - "dead" cell wall, had to be dropped. Various attempts of re-interpretation cast, however, some doubts over the very nature of plant cell and the status of the walls within such a cell. Following a comparison of exocellular matrices of plants and animals, their position in relation to cells and organisms is analysed. A multitude of perspectives of the biological organisation of living beings is presented with particular attention paid to the cellular and organismal theories. Basic tenets and resulting corollaries of both theories are compared, and evolutionary and developmental implications are considered. Based on these data, "The Plant Body" - an organismal concept of plants and plant cells is described.

The effect of ketotifen on inflammation in the intestine during T. spiralis infection in BALB/c mice was examined. The lack of enteropathy in treated mice was accompanied to affected the size of worms and the number of worms, but did not prevent the retardation of nematodes in the intestine. Fecundity of females in vitro and the number of muscle larvae were not statistically changed after treatment. Ketotifen reduced the number of mast cells and IgA plasma cells in the intestine. As inflammatory influx, epithelial damage and IgA response are related to the action of T cells, ketotifen affected T dependent reactions in the gut of T. spiralis infected mice.

Manipulating particles in the blood pool with noninvasive methods has been of great interest in therapeutic delivery. Recently, it was demonstrated experimentally that red blood cells can be forced to translate and accumulate in an ultrasound field. This acoustic response of the red blood cells has been attributed to sonophores, gas pockets that are formed under the influence of a sound field in the inner-membrane leaflets of biological cells. In this paper, we propose a simpler model: that of the compressible membrane. We derive the spatio-temporal cell dynamics for a spherically symmetric single cell, whilst regarding the cell bilayer membrane as two monolayer Newtonian viscous liquids, separated by a thin gas void. When applying the newly-derived equations to a red blood cell, it is observed that the void inside the bilayer expands to multiples of its original thickness, even at clinically safe acoustic pressure amplitudes. For causing permanent cell rupture during expansion, however, the acoustic pressure amplitudes needed would have to surpass the inertial cavitation threshold by a factor 10. Given the incompressibility of the inner monolayer, the radial oscillations of a cell are governed by the same set of equations as those of a forced antibubble. Evidently, these equations must hold for liposomes under sonication, as well.

Mammalian somatic cells and also cells of the yeast Saccharomyces cerevisiae are capable of undergoing a limited number of divisions. Reaching the division limit is referred to, apparently not very fortunately, as replicative aging. A common feature of S. cerevisiae cells and fibroblasts approaching the limit of cell divisions in vitro is attaining giant volumes. In yeast cells this phenomenon is an inevitable consequence of budding so it is not causally related to aging. Therefore, reaching a critically large cell volume may underlie the limit of cell divisions. A similar phenomenon may limit the number of cell divisions of cultured mammalian cells. The term replicative (generative) aging may be therefore illegitimate.

Some studies show that cells are able to penetrate through pores that are smaller than cell size. It concerns especially Red Blood Cells but it also may concern different types of biological cells. Such penetration of small pores is a very significant problem in the filtration process, for example in micro or ultrafiltration. Deformability of cells allows them to go through the porous membrane and contaminate permeate. This paper shows how cells can penetrate small cylindrical holes and tries to assess mechanical stress in a cell during this process. A new mathematical approach to this phenomenon was presented, based on assumptions that were made during the microscopic observation of Red Blood Cell aspiration into a small capillary. The computational model concerns Red Blood Cell geometry. The mathematical model allows to obtain geometrical relation as well as mechanical stress relations.

Opis:
Chronic exposure to environmental ultraviolet radiation (UVR) plays a key role both in photocarcinogenesis and induction of accelerated skin ageing. Sailors commonly experience a significant number of cutaneous problems, related to the exposure to environmental factors due to their working conditions. Among these factors, sun exposure is able to determine both acute and chronic skin damage, mostly linked to the effects of the ultraviolet (UV) radiation on epidermal and dermal structures. We report a case of sailor with cutis rhomboidalis nuchae, solar elastosis, actinic keratosis (AK), numerous squamous cell carcinoma (SCC) and basal cell carcinoma (BCC) of the head and trunk.

Opis:
In vitro study at the cellular level is an important step in evaluation of biological properties of newly developed drugs. In addition, thanks to the use of cell cultures, it is possible to study the function of the cells of a specific tissue and to mimic the conditions of cell growth or disease state. Standard (conventional) cell cultures are most often based on monolayer (two-dimensional, 2D) culture. In such culture, the cells adhere to the surface of the bottle/plate in which they grow. Nevertheless, spatial (three-dimensional, 3D) cultures are also increasingly used in culture research. However, conventional cell cultures still have a limited ability to mimic the natural environment of cell growth. Lab-on-a-chip systems (called Cell-on-a-chip) are new miniature constructional and methodical solutions useful for in vitro cell cultures. Microsystems allow the development of more advanced cell models than standard in vitro cultures so far used in biological laboratories. Thanks to this, it is possible to study cell functions under conditions that mimic the natural environment of cell growth. It should be emphasized, that the microsystems cannot completely replace animal testing. Cell-on-a-chip systems can provide alternative or complementary solutions / tools for in vivo tests, while ensuring high reliability of the obtained results. So far, Lab-on-a-chip systems have been used to assess the cytotoxicity of compounds, model intercellular interactions and mimic the function of various tissues. In this article we describe Cell-on-a-chip systems which can be used as advanced models of cell cultures. At the beginning of the manuscript, Cell-on-a- chip systems and materials used for fabrication microsystems are characterized. Next, the microsystems used as platforms for cytotoxicity study are presented. A special attention has been paid to Cell-on-a-chip for spatial cell cultures using: spheroids, hydrogels, multilayers and nanoscaffolds. Finally, Organ-on-a-chip systems are shortly discussed. Schemes of Cell-on-a-chip system, types of cell cultures obtained in microscale are presented in the article. A table showing a construction materials is also added. At the end, the article is summarized.

One of the main parameters that affect the solar cell performance is cell temperature; the solar cell output decreases with the increase of temperature. Therefore, it is important to select the proper solar cell technology that performs better at a specified location considering its average temperatures. In addition, the solar cell performance is directly reflected on the overall economics of the project. This paper is proposed to evaluate the variations in the performance of different solar cell technologies related to the temperature in Amman, Jordan. Field data of weather station and three PV systems (Poly-crystalline, Mono-crystalline and Thin-film) of identical design parameters were collected from Test Field Project at Applied Science Private University, Shafa Badran, Amman, Jordan. These data were analysed in the following way. estimated specific energy yield (kWh/kWp) for the three different PV systems was calculated depending on the measured value of solar irradiance and technical specifications of the installed solar panels and inverters, then the actual energy yield at different temperatures over one year was compared with the estimated value, so the deviations could be determined and actual temperature coefficients for energy yield could be calculated, knowing that the three PV Systems have identical design parameters (tilt angle, azimuth angle, type and dimensions of mounting structure and inverter size) and same cleaning method and schedule. It was found that the thin-film solar panels are less affected by temperature with temperature coefficient of -0.0984%, and -0.109%, -0.124% for Mono-crystalline and Poly-crystalline respectively. These results can be implemented in the preliminary design steps, specifically in the selection of the solar cell technology to be installed in a specific location.

Cancer cells are characterized by an aberrant increase in protein N-glycosylation and by disruption of E-cadherin-mediated adherens junctions. However, the relationship between alterations in N-glycosylation process and loss of E-cadherin adhesion in cancer remains unclear. The mechanisms of altered expression of adhesive glycoproteins in cancer cells have not been fully elucidated. Thus, the aim of this study was to examine the expression of E-cadherin and sialyl Lewisa/x, NeuAcα2-3Gal, NeuAcα2-6Gal/GalNAc structures in the normal renal tissue and intermediate and cancerous tissues from patients with clear cell RCC. Moreover, we attempted to correlate the E-cadherin expression with some specific sugar residues of renal cancer tissue glycoproteins. The expression of E-cadherin was analysed using ELISA test and immunoblotting. Oligosaccharide structures and sialylation level were detected with ELISA test using specific biotinylated lectins or antibodies. A significant decrease of E-cadherin expression as well as a significant increase in sialylated oligosaccharides level in intermediate zone and renal cancer tissue in comparison to normal renal tissue are reported. Significant decrease in expression of cadherins and increase in sialylation of oligosaccharide structures in renal cancer tissue in comparison to normal renal tissue, and in renal cancer tissue in comparison to intermediate zone of renal tissue, are important for the future research concerning detection and quantification of cadherins and sialylated oligosaccharide structures in urine and cells of urinary sediment as possible non-invasive marker of early RCC.

Existing global models of interaction between the solar wind (SW) and the local interstellar medium (LISM) describe the heliosphere that arises as a result of this interaction. There is a strong motivation to develop a kinetic model using the Particle-in-Cell (PIC) method to describe phenomena which appear in the heliosphere. This is however a long term scientific goal. This paper describes an electrostatic Particle-in-Cell numerical model developed in the Institute of Aviation in Warsaw, winch includes mechanical and charge exchange collisions between particles in the probabilistic manner using Direct Simulation Monte Carlo method. This is the first step into developing simulations of the heliosphere incorporating kinetic effects in collisionless plasmas. In this paper we focus only on presenting the work, which have been done on the numerical PIC algorithm.
Istniejące globalne modele oddziaływania wiatru słonecznego (SW) z lokalną materią międzygwiazdową (LISM) opisują heliosferę, która powstaje w wyniku interakcji tych dwóch ośrodków. Istnieje silna motywacja do opracowania modelu kinetycznego wykorzystującego metodę Particle-in-Cell (PIC) w celu opisu zjawisk, które zachodzą w heliosferze. Jednakże jest to długoterrninowy cel naukowy. W artykule przedstawiono elektrostatyczny model numeryczny PIC. opracowany w Instytucie Lotnictwa w Warszawie, który obejmuj e kolizje mechaniczne i rezonansową wymianę ładunków pomiędzy cząstkami w sposób probabilistyczny metodą Direct Simulation Monte Carlo. Jest to pierwszy krok w opracowywaniu symulacji heliosfery zawierającej efekty kinetyczne w plazmach bezzderzeniowych. W tym artykule koncentrujemy się tylko na prezentowaniu prac, które zostały wykonane z wykorzystaniem algorytmu numerycznego PIC.

The protozoan Toxoplasma gondii, described by Nicolle and Manceaux in 1908, is a ubiquitous and cosmopolitan parasite that infects a wide range of mammal and bird species with high prevalence. The biological success of T. gondii is associated with the formation of a specific relationship between the parasite and host cells leading to the establishment of a latent, chronic infection. During primary infection, acquired mostly by the oral route, the quickly multiplying tachyzoites disseminate through the body crossing several structural-functional barriers as bloodbrain or blood-retina, then they transform into dormant bradyzoites which, enclosed in tissue cysts, occupy preferentially the brain, skeletal muscle and eye. Although T. gondii is able to infect all kinds of nucleated cells, it uses strictly defined host cells, dependent on the life-cycle phase and infection stage. The article discusses selected aspects of the parasite passing via the host body barriers as well as particular role of dendritic cells and skeletal muscle cells, used by the parasite as an very effective vehicle to disseminate throughout the host body or the site of long-term T. gondii persistence, respectively.

It owes its popularity to the great variety of roof structures it offers, with respect to both, its architecture and construction. The most important factors in creating cell vaults are the number of divisions and the height of joint level of a given structure. In this paper we would like to present the geometrical methods of creating cell vaults.
Sklepienia kryształowe występujące w architekturze od połowy XV wieku w ostatnim okresie zaczynają mieć swój renesans, gdyż oferują niezwykle dużą różnorodność przekryć, tak pod względem konstrukcyjnym jak i architektonicznym. Ich kształtowanie jest oparte o ilość podziałów oraz ustalenie wysokości poziomów węzłów struktury. W tej publikacji staramy się pokazać geometryczne sposoby ich kształtowania.
