The liver as a vital body organ is adversely affected by hazardous chemicals and drugs. Paracetamol widely used as analgesic and antipyretic drug produces severe hepatotoxicity at high doses. Present study was designed to investigate the hepatoprotective activity of Polygonum perfoliatum L. used on folklore basis. Aqueous methanolic extract of the plant was prepared. Preliminary phytochemical and HPLC analyses were carried out to identify and quantify chemical constituents respectively. For hepatoprotective activity, Wistar rats were divided into six groups as normal control, standard (silymarin) control, negative control and extract treated groups i.e., 125, 250 and 500 mg/kg/day per oral. Paracetamol was administered orally, following seven days of previously stated therapy. Biochemical parameters of hepatotoxicity such as serum glutamic pyruvate transaminase (SGPT), serum glutamic oxaloacetate transaminase (SGOT), alkaline phosphatase (ALP) and total bilirubin were measured in all groups. Histopathological evaluation of liver was also carried out. Benzoic acid, chlorogenic acid, gallic acid, m-coumaric acid, quercetin and vitamin E were detected in the plant extract through HPLC. The hepatoprotective effect of 500 mg/kg/day therapy was more pronounced than 125 and 250 mg/kg dose. However, the effect of plant extract was less pronounced than standard silymarin therapy. It can be concluded that the plant extract possessed significant hepatoprotective activity that may be attributed to quercetin, benzoic acid, gallic acid and vitamin E present in it.

In recent years there have been remarkable developments in the prevention of diseases, especially with regards to the role of free radicals and antioxidants. Ethanol-induced oxidative stress appears to be one mechanism by which ethanol causes liver injury. The protective effect of aqueous plant extract of Aframomum melegueta on ethanol-induced toxicity was investigated in male Wistar rats. The rats were treated with 45 % ethanol (4.8 g/kg b.w.t.) for 16 days to induce alcoholic diseases in the liver. The activities of alanine aminotransferase, aspartate aminotransferase and triglyceride were monitored and the histological changes in liver examined in order to evaluate the protective effects of the plant extract. Hepatic malondialdehyde and reduced glutathione, as well as superoxide dismutase and glutathione-S-transferase activities were determined for the antioxidant status. Chronic ethanol administration resulted in a statistically significant elevation of serum alanine aminotransferases and triglyceride levels, as well as a decrease in reduced glutathione and superoxide dismutase which was dramatically attenuated by the co-administration of the plant extract. Histological changes were related to these indices. Co-administration of the plant extract suppressed the elevation of lipid peroxidation, restored the reduced glutathion, and enhanced the superoxide dismutase activity. These results highlight the ability of Aframomum melegueta to ameliorate oxidative damage in the liver and the observed effects are associated with its antioxidant activities.

Hepatoprotective and antioxidant activity of Lannea coromandelica bark extract (LCBE) was investigated on thioacetamide induced hepatotoxicity in rats. Hepatotoxicity was induced by thioacetamide (TAA) administration (100 mg/kg. s.c). LCBE at different doses (400 and 200 mg/kg) were administered orally to male wistar rats. Thioacetamide caused elevation of serum concentration of AST, ALT, ALP, serum bilirubin and also reduced serum concentration of total protein, albumin, sodium, potassium in animals as compared to control (p < 0.05) but LCBE treated rats showed maximum reduction of AST [(138±5.1) IU/L], ALT [(71 ±2.7) IU/L], ALP [(140 ±1.9) IU/L] with the high dose (400 mg/kg bw) of combined aqueous and alcoholic bark extract. Whereas, serum bilurubin, cholesterol, sugar and LDH content were varied with the treatments but showed higher with the only ethanolic extract at dose of 400 mg/kg. The IC50 value was observed as (83.28 ±2.12) μg/mL, for DPPH radical scavenging activity. Result concluded that ethanolic extract and combined aqueous and alcoholic bark extract of L. coromandelica showed a potential hepatoprotective and antioxidant activities might be due to the presence of phenolic groups, terpenoids and alkaloids.

Oxidative stress has been recognized as a critical pathogenetic mechanism for the initiation and the progression of hepatic injury in a variety of liver disorders. Antioxidants, including many natural compounds or extracts, have been used to cope with liver disorders. The present study was designed to investigate the hepatoprotective effects of cassia seed ethanol extract (CSE) in carbon tetrachloride (CCl4)-induced liver injury in mice. The animals were pre-treated with different doses of CSE (0.5, 1.0, 2.0 g/kg body weight) or distilled water for 5 days, then were injected intraperitoneally with CCl4 (0.1% in corn oil, v/v, 20 ml/kg body weight), and sacrificed at 16 hours after CCl4 exposure. The serum aminotransferase activities, histopathological changes, hepatic and mitochondrial antioxidant indexes, and cytochrome P450 2E1 (CYP2E1) activities were examined. Consistent with previous studies, acute CCl4 administration caused great lesion to the liver, shown by the elevation of the serum aminotransferase activities, mitochondria membrane permeability transition (MPT), and the ballooning degeneration of hepatocytes. However, these adverse effects were all significantly inhibited by CSE pretreatment. CCl4-induced decrease of the CYP2E1 activity was dose-dependently inhibited by CSE pretreatment. Furthermore, CSE dramatically decreased the hepatic and mitochondrial malondialdehyde (MDA) levels, increased the hepatic and mitochondrial glutathione (GSH) levels, and restored the activities of superoxide dismutase (SOD), glutathione reductase (GR), and glutathione S-transferase (GST). These results suggested that CSE could protect mice against CCl4-induced liver injury via enhancement of the antioxidant capacity.

The rising number of patients with liver dysfunction due to overwhelming usage of drugs and alcohol has paved the path for researchers in an interest in herbal medicine. This is because there are only a few universally effective and available options for the treatment of common liver diseases, such as cirrhosis, fatty liver and chronic hepatitis. Herbal treatment has been used to alleviate disorders related to liver and other internal organs for many centuries in the eastern world, and have currently become a favourable therapy internationally for pathological liver conditions. In recent years, researchers have used scientific methods to evaluate the effects of plants for the treatment of liver ailments, although in many cases the mechanisms and modes of action of these plants, as well as their therapeutic effectiveness, have not been confirmed. Several hundred plants have been examined to-date, but only a handful has been studied thoroughly. Among these, the current study discusses a compilation of herbal medicines used for liver protection, such as Silymarin (milk thistle), Liv-52, Camellia sinensis (green tea), Glycyrrhizaglabra (licorice) and FuzhengHuayu. The increasing use of herbal medicines reflects their perceived effectiveness in the treatment and prevention of disease, and the belief that these treatments are safe because they are ‘natural’. The presented study evaluates the effects of herbal extracts in the treatment of liver diseases, provides a general understanding of the actions of herbal medicines, a background for understanding the hepatoprotectiveness of herbs, and the challenges that are faced by the scientific community in researching thoroughly of each and every compounds of the herbs.
