Opis:
Using ultrastructural cytochemical techniques we have found differences in the distribution of surface coat components between the invasive protozoan parasite Entamoeba histolytica and the non-invasive Entamoeba dispar. Carbohydrate-containing components and anionic sites in the cell surface of both species were detected by staining with ruthenium red and cationized ferritin, respectively. Ruthenium red staining revealed a thicker surface coat in E. histolytica trophozoites, whereas trophozoites of E. dispar showed a higher concentration of cationized ferritin particles on its surface. Mannose or glucose residues were found at the plasma membrane of both parasites treated with Concanavalin A (Con A)-peroxidase; the surface reaction product was more evident in E. dispar, compared with E. histolytica. Con A rapidly produced surface caps in E. histolytica trophozoites, whereas E. dispar showed a much less efficient mobilization of surface Con A receptors. Agglutination with Con A produced much larger clumps in E. histolytica in comparison with E. dispar. In turn, biotinylation assays revealed striking differences in the composition of surface membrane proteins in both amebic species. Overall, these results further emphasize the phenotypic differences between these two common parasites of the human intestinal tract, once considered to be the same protozoan.

Opis:
The effect of insulin, wheat germ agglutinin (WGA), peanut agglutinin (PNA) and concanavalin A (ConA) on [3H]glucosamine incorporation into pericellular glycosaminoglycans (GAGs) was investigated in two lines of cultured human dermal fibroblasts. Insulin and WGA stimulated [3H]glucosamine incorporation into hyaluronic acid (HA) and heparan sulphate (HS) without any alteration of chondroitin sulphate (CS) and dermatan sulphate (DS) contents. ConA increased [3H]glucosamine incorporation into HS, CS and DS, but had no effect on [3H]glucosamine incorporation into HA. PNA affected neither the content, nor the composition of GAGs. In contrast to PNA, ConA and WGA stimulated glycolysis and demonstrated an evident antiproliferative effect on dermal fibroblasts. Thus, both the insulin-like action of WGA and ConA on cultured dermal fibroblasts and the differences between the effects of lectins on modulation of GAGs synthesis appear to be determined by their chemical structure.

Opis:
In this study, Aspergillus oryzae Β galactosidase was immobilized on concanavalin A layered calcium alginate-cellulose beads as a bioaffi nity support. Immobilized enzyme showed a remarkable broadening in temperature-activity profi les as compared to the native enzyme and exhibited 65% activity in the presence of 5% galactose. Michaelis constant (Km) was 2.57 mM and 5.38 mM for the free and the immobilized Β galactosidase, respectively. Crosslinked Β galactosidase showed greater catalytic activity in the presence of Mg2+ and was more stable during storage at 4°C for 6 weeks. Immobilized enzyme hydrolyzed 67% lactose in milk in 8 h and 85% lactose in whey in 9 h in the stirred batch process at 50oC. The continuous hydrolysis of lactose by crosslinked Β galactosidase in spiral bed reactor exhibited 93% and 88% hydrolysis of lactose at flow rate of 20 ml/h and 30 ml/h, after 1 month operation, respectively.
