Opis:
Ultrafiltration (UF) is one of the membrane processes which is mostly used in the dairy industry for the separation and concentration of whey components or fermentation broth. Fouling of UF membranes in the food industry is primarily due to a deposition of microorganisms, proteins, fats and minerals on the membrane surface. Thus, cleaning of the membranes is an essential step of UF separation. The results from investigations of chemical cleaning of a ceramic UF membrane fouled by precipitation of whey components and yeast contained in the fermentation broth are presented. The effect of cleaning procedure on the degree of permeability restoration by the fouled membrane was studied. The results demonstrated that a combination of sodium hydroxide, phosphoric acid and sodium hypochlorite as a disinfectant could be successfully used to achieve an optimum recovery of the membrane properties.

Opis:
Green synthesis of metal (zinc) oxide nanoparticles is a fascinating subject in the field of nanobiotechnology. Nanoparticles using green plants are suitable for large scale biosynthesis at room temperature with affordable price and eco-friendly nature. The stable nanoparticles were synthesized using leaves, stem, roots, flowers, immature and mature seeds extracts of Crotalaria verrucosa. Zinc Nitrate hexahydrate [Zn(NO3)2•6H2O] was used as precursor material in this experiment. The formation of nanoparticles was monitored by visual color changes and UV-Visible spectrophotometric analysis. UV-Visible absorption peaks between 299-317 nm confirmed the presence of ZnO at nano-scale. The present technique confirmed that this plant species could be explored in the field of nanotechnology for human welfare.

This paper applies the determined suitability of nanofi ltration (NF) membrane separation for selective isolation and concentration of succinic acid from aqueous solutions which are post-fermentation multicomponent fl uids. The study analyzed the infl uence of concentration and the pH of the separated solutions on the effi ciency and selectivity of NF process that runs in a module equipped with a ceramic membrane. Moreover, the effect of applied transmembrane pressure on the retention of succinic acid and sodium succinate has been studied. The investigations have shown that in the used NF module the retention of succinic acid salt is equal almost 50% in the case of a three-component model solution, although the degree of retention depends on both the transmembrane pressure and the initial concentration of separated salt.

Membrane separation is a widely used technique whose main obstacle is membrane fouling, during which a solution or particles accumulate on the membrane surface or inside its pores. This phenomenon causes both reversible and irreversible flow resistances that can be minimized with complementary techniques such as sonication and/or stirring. The application of ultrasonic energy can increase the flux by breaking the concentration polarization and cake layer on the membrane surface without affecting the intrinsic permeability of the membrane. Our work focused on cellulase and β-glucosidase enzyme recovery through sonicated/stirred ultrafiltration from the real fermentation broth, at which the fermentation liquid was produced from tobacco plants. Membrane filtrations were carried out in a laboratory-grade batch ultrafiltration cell, extended with stirrer and ultrasound processor. The effects of stirring speed and ultrasound treatment were investigated on the membrane’s permeate flux efficiency and on the quality of recycled enzymes. Resistance values were calculated by the resistance-in-series model, as well as by Hermia’s analysis, to reveal deeper, process-related information. Our results revealed that enzymes could be efficiently separated from real fermentation broth. Furthermore, the enzyme activity test showed that neither stirred nor sonicated processes were detrimental to the enzyme activity.

Introduction. Microtiter plate assay (MPA) remains one of workhorses of in vitro biofilm research but it requires optimization of experimental conditions to fulfill the biofilm formation requirements of different bacterial pathogens. Aim. The aim was to determine the effect of TSB and RPMI1640 culture media and selected culture variables (O2 vs. 5% CO2 , extended incubation time) on the biofilm production by bacteria commonly involved in biofilm-related infections: Enterococcus faecalis (EF), Escherichia coli (EC), Staphylococcus aureus (SA), Pseudomonas aeruginosa (PA), Klebsiella pneumoniae (KP). Material and methods. The investigation was performed using the MPA with crystal violet. Results. Statistically significant (p<0.05) increase in biofilm production between 24h and 72h time points was observed for EF (TSB o2, RPMIo2 and RPMIco2 ), EC (TSBo2 ) , SA (TSBo2 , TSBco2 ), KP (TSBo2 , TSBco2 ), PA (RPMIco2, TSBco2 ). The TSB caused a significantly greater stimulation of biofilm production compared to RPM1640. It outcompeted RPMI1640 irrespective of the atmospheric conditions for SA and KP and under aerobic conditions for EF. Conclusion. Although the TSB provided the most optimal conditions for biofilm production, the process was influenced by the strain type, atmospheric conditions and period of cultivation which limits the ability to design a single universal model of the in vitro biofilm investigation.

Mackerel of Scomberomorus commerson (Lacepède, 1800) is a fish that has thick meat of a distinctive taste and is a favorite of Indonesian people. It is made into various types of processed seafood. The amount of edible flesh of the fish is 65%, meaning that the waste from the fish is 35%, including the head and bones. The purpose of this study was to determine the type and class of volatile compound components that can be drawn from mackerel head and fish bone, by making these into a broth. This study uses an experimental method by testing the composition of volatile flavor compounds and then undertaking proximate testing. The method of extracting volatile flavor compounds that was applied is Solid Phase Microextraction (SPME), while Gas Chromatography / Mass Spectrometry (GC / MS) was harnessed to identify the volatile flavor. Accordingly, 150 compounds were detected in the sample of mackerel head broth, as compared with 133 compounds in mackerel bone broth. The compounds that were detected are hydrocarbons, aldehydes, alcohols, ketones, organic compounds and others. That which has the largest proportion is pentadecane from the hydrocarbon group, with a value of 18,545%. The proximate analysis results showed that mackerel head broth samples had a 96.08% water content, 1.55% ash content, 0.28% fat content and 2.78% protein content, while samples of mackerel bone broth had a 96.69% water content, 1.54% ash content, 0.44% fat content and 1.84% protein content.

Alkohole (etanol, butanol, 1,3-propanodiol, 2,3-butanodiol, glicerol), kwasy organiczne (kw. cytrynowy, kw. bursztynowy, kw. mlekowy, kw. propionowy, kw. masłowy), cukry (glukoza), a także acetoinę rozdzielano na kolumnie Aminex HPX-87H przy zastosowaniu dwóch detektorów - refraktometrycznego i UV-DAD pracujących w układzie szeregowym. Badano wpływ poszczególnych parametrów chromatograficznych na jakość rozdziału. Wyznaczono niektóre parametry walidacyjne dla zaproponowanej metody. Opracowana metoda pozwala na jakościową analizę wielu substancji w brzeczkach fermentacyjnych w szczególności, gdy niektóre z nich koeluują ze sobą.
Alcohols (ethanol, butanol, 1,3-propanediol, 2,3-butanediol, and glycerol), organic acids (citric acid, succinic acid, lactic acid, propionic acid, and butyric acid), sugar (glucose) and acetoin were separated on an Aminex HPX-87H column using two detectors: refractive index detector and UV-DAD detector. The influence of chromatographic parameters on chromatographic separation was analyzed. Some validation parameters for proposed method were determined.

W pracy przedstawiono możliwość zastosowania ekskluzji jonów w procesie oczyszczania 1,3-propanodiolu z brzeczki fermentacyjnej. Badania przeprowadzono na kationicie w formie sodowej. Wstępnie odsoloną frakcję produktu uzyskano z ok. 50% wydajnością separacji 1,3-propanodiolu. Stopień odsolenia brzeczki wyniósł 80÷90%, a końcowe oczyszczenie i odbarwienie 1,3-propanodiolu przeprowadzono stosując wymianę jonową. W wyniku procesu otrzymano oczyszczony roztwór 1,3-propanodiolu o stężeniu 16,5 g/L.
Application of ion exclusion for the separation of 1,3-propanediol from fermentation broth was examined. The study was carried out on a cation exchange resin in the sodium form. The product fraction with the desalination degree of 80-90% and 50% separation yield of 1,3-propanediol we obtained. The final purification of 1,3-propanediol was carried out using the ion exchange. As a result of this process the purified solution of 1,3-propanediol (16.5 g/L) was obtained.

The aim of this study was to determine the effect of fish-based feed materials, as a source of readily available protein contained in fish broth and essential polyunsaturated fatty acids (PUFAs) found in fish oil, on the health of piglets and rearing results. The experiment was conducted on a commercial pig fattening farm. The study involved a total of 80 weaners with an approximate body weight of 15 kg. The experiment was carried out over a period of 40 days. Feed samples were subjected to laboratory analyses. Blood samples were collected from experimental group animals to determine serum biochemical and immunological parameters. The body weight gains of weaners, mortality rates and average feed intake per animal were calculated for the entire experimental period. The addition of fish broth and fish oil significantly improved the n3:n6 fatty acid ratio in diets. The presence of EPA and DHA in the experimental diet could have had a positive health effect on piglets, comparable with that exerted by therapeutic doses of zinc often administered to pigs of this age group. During the experiment, feed conversion ratio (FCR) gain was considerably reduced in the experimental group, with similar daily gains in the control and experimental group.

The aim of this study was to determine the sensitivity of Aspergillus niger strains isolated from birds to available antifungal drugs using different in vitro assays - classical disk diffusion, Etest® and broth microdilution NCCLS/CLSI M 38-A. The study material consisted of about 2.000 swabs and samples from different species of birds. A. niger (n=10) was accounted for 6.81% of the total pool of strains isolated. Determinations were made for 13 antifungal drugs using the disk diffusion method. The A. niger exhibited high susceptibility to enilconazole, terbinafine, voriconazole, tioconazole and ketoconazole, low susceptibility to clotrimazole, miconazole and nystatin, and resistance to amphotericin B, itraconazole, pimaricin, fluconazole and 5-fluorocytosine. Minimum inhibitory concentration (MIC) was determined for 9 antifungal drugs using the micromethod of duplicate serial dilutions in a liquid medium. A. niger strains were most susceptible to enilconazole and voriconazole. MIC ranged from 0.0625 to 0.5 μg/ml for enilconazole, with MIC90-0.5 μg/ml and MIC50-0.125 μg/ml. The corresponding values for voriconazole were 0.25-1 μg/ml, 1 μg/ml and 0.5 μg/ml. MIC for amphotericin B and terbinafine ranged from 0.5 to 4 μg/ml, while the values for the remaining drugs were highly varied. MIC was measured by the gradient diffusion method using Etest® for 5 antifungal drugs: amphotericin B, fluconazole, itraconazole, ketoconazole and voriconazole. By far the highest susceptibility was obtained in the case of voriconazole, with MIC ranging from 0.0625 to 1 μg/ml. MIC for amphotericin B ranged from 0.25 to 4 μg/ml, for itraconazole and ketoconazole ranging from 0.5 to 16 μg/ml. Methods available for this purpose are not always applicable in field conditions. The present results indicate that the Etest® technique, due to its high percentage of agreement with the M 38-A microdilution method, should find application in medical and veterinary practice.
