Opis:
Nephroprotective effects (NPE) of simple Potentilla reptans - aqueous leaf extract (Pr-ALE) leaves on Paracetamol induced kidney poisonousness in wistar rats. Adult male wistar rats (weight range (WR): 200-220g) were divided into 6 groups (n=6). Paracetamol (PA) and Silymarin (SY) stayed managed intraperitoneally arranged the 5th day to rats in all groups but the normal control. Furthermore, a significant nephroprotective (NP) of the aqueous leaf extract (ALE) and oral dose of PA and SY. Pr-ALE did not mortality or significant changes in the body weight. Progression of nephrotoxicity (NT) induced by PA in rats was interfered by Pr-ALE managed, and these effects were correspond to those managed with SY. This is the first record on NPE of Pr against PA-induced NT.

The inadequacy of antiviral drugs in the treatment of viral diseases, has led to herbal medicine. It was aimed to determine the antiviral and antioxidant activities of Silymarin and Panax ginseng against Bovine Ephemeral Fever Virus (BEFV) and Bluetongue virus (BTV) in cell culture. Silymarin and Panax ginseng were dissolved at the concentration of 400 µg/ml within distillated water. The cell proliferation test was used to evaluate the cytotoxic activity of the Silymarin and Panax ginseng. They were cytotoxic over 50 μg / ml dose in Vero cells. Hence, antiviral activities of subjects were investigated against BEFV at the 25 and 50 µg/ ml doses. However, they did not show antiviral activity at any dose level against BTV. Effects of Silymarin and Panax ginseng were evaluated on the total antioxidant capacity (TAC) and thiobarbituric acid reactive substances (TBARS), oxidative stress marker, levels in Vero cells infected with BEFV and BTV. Silymarin (25 and 50 µg/ ml) affected TAC levels in Vero cells infected with BEFV, but it did no effect the TBARS levels in Vero cells infected with BEFV and BTV. Panax ginseng decreased TBARS levels in both diseases, although it did not change TAC levels at same doses on Vero cells infected with BEFV and BTV. In conclusion, it is referred that Silymarin and Panax ginseng may have antiviral some viruses and they have antioxidant, cell protective and inhibitory effects of virus replication.

Silymarin, a flavonolignan from the seeds of 'milk thistle' (Silybum marianum (L.) Gaertn.)), has been widely used from ancient times because of its excellent hepato-protective action. It is a mixture of mainly three flavonolignans, which are, silybin, silidianin, and silychristin, with silybin being the most active. Silymarin has been used medicinally to treat liver disorders, including acute and chronic viral hepatitis, toxin/drug-induced hepatitis, and cirrhosis and alcoholic liver diseases. It has also been reported to be effective in certain cancers. Its mechanism of action includes inhibition of hepatotoxin binding to receptor sites on the hepatocyte membrane; reduction of glutathione oxidation to enhance its level in the liver and intestine; antioxidant activity; and stimulation of ribosomal RNA polymerase and subsequent protein synthesis, leading to enhanced hepatocyte regeneration. It is orally absorbed but has very poor bioavailability due to its poor water solubility. This review focuses on the various pharmacological activities of silymarin.

Opis:
Silybum marianum L. (Milk thistle) extracts are the main source of silymarin that is a mixture of various flavonolignan (silybin (silibinin), silydianin and silychristin). Silymarin of milk thistle has a hepatoprotective activity for liver cirrhosis and chronic inflammatory. Silybum marianum regeneration from hypocotyl explants and evaluation of their callogenesis, growth and total flavolignan (silymarin) upon copper sulphate (as abiotic elicitor) elicitation was targeted. Copper sulphate (CuSO4) was applied in concentrations of 0, 3, 5, 7 and 9 µM to elicit the silymarin production in cultures. The elicitation periods used in this study were 2, 4, 7, 14 and 28 days. Half-strength MS medium recorded better results relative to full-strength MS one and seed incubation in the darkness at room temperature resulted in rapid germination and reached to the gar lid after 10 days. Callus fresh and dry weights as well as growth index were gradually increased with increasing the copper sulphate concentration till 5 µM while decreased thereafter at any elicitation period. With the increase of the elicitation period, the increase of the previous parameters was observed. Flavonolignan (silymarin) was positively correlated with CuSO4 levels since all levels of copper sulphate significantly enhanced its content in relative to the control. Additionally, more silymarin was accumulated after 4 or 7 days and the accumulation significantly decreased when the elicitation period reached 14 days more. The highest silymarin (flavolignan) content (11.79 and 11.67 mg g–1 DW) was obtained when 5 or 7 µM copper sulphate levels were combined with 4 days elicitation period, being about five-fold of the control.
