Several years ago it was discovered that plant transformation with a transcribed sense transgene could shut down the expression of a homologous endogenous gene. Moreover, it was shown that the introduction into the cell of dsRNA (double-stranded RNA) containing nucleotide sequence complementary to an mRNA sequence causes selective degradation of the latter and thus silencing of a specific gene. This phenomenon, called RNA interference (RNAi) was demonstrated to be present in almost all eukaryotic organisms. RNAi is also capable of silencing transposons in germ line cells and fighting RNA virus infection. Enzymes involved in this process exhibit high homology across species. Some of these enzymes are involved in other cellular processes, for instance developmental timing, suggesting strong interconnections between RNAi and other metabolic pathways. RNAi is probably an ancient mechanism that evolved to protect eukaryotic cells against invasive forms of nucleic acids.

The full scope of regulatory RNA evolution and function in epigenetic processes is still not well understood. The development of planarian flatworms to be used as a simple model organism for research has shown a great potential to address gaps in the knowledge in this field of study. The genomes of planarians encode a wide array of regulatory RNAs that function in gene regulation. Here, we review planarians as a suitable model organism for the identification and function of regulatory RNAs.

W ciągu ostatnich lat zjawisko interferencji RNA (RNAi) stało się powszechnie wykorzystywane jako eksperymentalne narządzie do analizy genów oraz pełnionych przez nie funkcji. Wraz ze wzrostem wiedzy na temat molekularnych mechanizmów działania endogennego interferującego RNA, małe interferujące RNA (siRNA), mogą pojawić się jako grupa innowacyjnych bioleków stosowanych do leczenia wielu chorób m.in. chorób nowotworowych. Poznanie i zrozumienie szlaków molekularnych istotnych w procesie nowotworzenia stwarza możliwości dla terapii nowotworowej wykorzystującej mechanizm RNAi. Nowe terapie w leczeniu nowotworów są niezbędne, a wykorzystanie małych interferujących RNA może stanowić realną strategię.
In the last few years, RNA interference (RNAi) has become widely used as an experimental tool for the analyses of genes and their functions. With increasing knowledge about the molecular mechanisms of function of endogenous RNA interference, small interfering RNA (siRNA), may occur as innovative bio-drugs for treatment of diseases such as cancer. Knowledge and understanding of the molecular pathways important for carcinogenesis create opportunities for cancer therapy using RNAi mechanism. New therapies are essential for tumors treatment, and small interfering RNAs may provide a viable strategy.

MicroRNAs (miRNAs) are small non-coding RNAs that have been found in most of the eukaryotic organisms. They are involved in the regulation of gene expression at the post-transcriptional level in a sequence specific manner. MiRNAs are produced from their precursors by Dicer-dependent small RNA biogenesis pathway. Involvement of miRNAs in a wide range of biological processes makes them excellent candidates for studying gene function or for therapeutic applications. For this purpose, different RNA-based gene silencing techniques have been developed. Artificially transformed miRNAs (amiRNAs) targeting one or several genes of interest represent one of such techniques being a potential tool in functional genomics. Here, we present a new approach to amiRNA*design, implemented as AmiRNA Designer software. Our method is based on the thermodynamic analysis of the native miRNA/miRNA* and miRNA/target duplexes. In contrast to the available automated tools, our program allows the user to perform analysis of natural miRNAs for the organism of interest and to create customized constraints for the design stage. It also provides filtering of the amiRNA candidates for the potential off-targets. AmiRNA Designer is freely available at http://www.cs.put.poznan.pl/arybarczyk/AmiRNA/.

Rab7 GTPases are involved in membrane trafficking in the late endosomal/lysosomal pathway. In Paramecium octaurelia Rab7a and Rab7b are encoded by paralogous genes. Antipeptide antibodies generated against divergent C-termini recognize Rab7a of 22.5 kDa and Rab7b of 25 kDa, respectively. In 2D gel electrophoresis two immunoreactive spots were identified for Rab7b at pI about 6.34 and about 6.18 and only one spot for Rab7a of pI about 6.34 suggesting post-translational modification of Rab7b. Mass spectrometry revealed eight identical phosphorylated residues in the both proteins. ProQ Emerald staining and ConA overlay of immunoprecipitated Rab7b indicated its putative glycosylation that was further supported by a faster electrophoretic mobility of this protein upon deglycosylation. Such a post-translational modification and substitution of Ala140 in Rab7a for Ser140 in Rab7b may result in distinct targeting to the oral apparatus where Rab7b associates with the microtubular structures as revealed by STED confocal and electron microscopy. Rab7a was mapped to phagosomal compartment. Absolute qReal-Time PCR analysis revealed that expression of Rab7a was 2.6-fold higher than that of Rab7b. Upon latex internalization it was further 2-fold increased for Rab7a and only slightly for Rab7b. Post-transcriptional gene silencing of rab7a suppressed phagosome formation by 70 % and impaired their acidification. Ultrastructural analysis with double immunogold labeling revealed that this effect was due to the lack of V-ATPase recruitment to phagolysosomes. No significant phenotype changes were noticed in cells upon rab7b silencing. In conclusion, Rab7b acquired a new function, whereas Rab7a can be assigned to the phagolysosomal pathway.

Opis:
The future of food security in Africa is being severely threatened due to an exponential increase in population, which is almost three times the increase of food production. Maize production is constrained by stem borers which cause significant yield losses. Yield losses can be further compounded by higher temperatures due to climate changes, which are expected to increase the population of maize stem borers. While several methods have been employed in stem borer management, there is still significant damage caused by maize stem borers. This necessitates better control methods including the adoption of recent biotechnological advancement in RNA interference (RNAi) technology. This review highlights evidence of an increase in the stem borer population as well as the foreseen decline in maize production worldwide due to the effects of climatic changes. Furthermore, we have drawn attention to improved methods that have been used to control stem borers in maize production as well as a reluctant acceptance of traditional biotechnology in Africa. Finally, we suggest the application of alternative RNA interference techniques to breed maize for efficient pest control in order to achieve food security, improve nutrition and promote sustainable maize production.
