Opis:
Colorectal cancer has a high incidence rate with significant mortality and morbidity. The 15-lipoxygenase-1 (15-LOX-1) is a crucial enzyme as well as a tumor suppressor, especially in colon cancer. Bee venom, by the induction of apoptosis, is a promising new anti-cancer agent. Thus, the effects of bee venom on the expression of 15-LOX-1 m-RNA and its activities in the HT-29 cell line have been investigated in the current study. Human HT-29 colon cancer cells were treated with increasing concentrations of bee venom (1.87-30 µg/mL), and cisplatin (5 µg/mL) as the positive control for 24- and 48-hours. Then, cell viability and apoptosis were measured using the MMT assay and flow cytometry, respectively. Enzyme activity and expression was assayed using the kinetic method and real-time PCR, respectively. The results showed that the main content of bee venom was melittin with a retention time of 60 minutes. The IC50 values of bee venom at 24- and 48-hours were 6.01 µg/mL and 4.44 µg/mL, respectively. Expression of 15-LOX-1 in cancer cells treated with bee venom increased (p < 0.0005), as well as the activity of the enzyme in the presence of bee venom (p < 0.01). The current study revealed the apoptotic and cytotoxic effects of bee venom against the human colon cancer HT-29 cell line that was not seen in fibroblast cells. Findings suggest that bee venom may have therapeutic effects against the HT-29 colon cancer cell through the induction of the15-LOX-1 pathway. However, further studies are needed in this regard.

The aim of this study was to analyze the molecular mechanism of inositol hexaphosphate (InsP6) action through which it may inhibit proliferation of colon cancer cells and cell cycle progression. A kinetic study of p53 and p21WAF1 mRNA increase was performed on human colon cancer HT-29 cells after treatment with 1, 5 and 10 mM InsP6 for 6, 12, 24 and 48 h. Real-time-QPCR based on TaqMan methodology was applied to analyze quantitatively the transcript levels of these genes. The transcription of β-actin and GAPDH genes was assessed in parallel to select the control gene with least variability. The 2-ΔΔCt method was used to analyze the relative changes in gene transcription. InsP6 stimulated p53 and p21WAF1 expression at the mRNA level, with the highest increase in p21WAF1 mRNA occurring at 24 h, i.e., following the highest increase in p53 mRNA observed at 12 h. Based on these studies it may be concluded that the ability of InsP6 to arrest the cell cycle may be mediated by the transcriptional up-regulation of the p53-responsive p21WAF1 gene.

Opis:
Microfluidic systems are used in a wide range of applications, including medical diagnostics, cell engineering and bioanalytics. In this work we focused on “Lab-on-a-chip” microsystems for cell cultivation. A troublesome problem of gas bubbles entering microdevices causing signal interferences and cells damage was emphasized. A novel, integrated debubbler in the form of cylindrical traps covered with thin PDMS membrane was designed and manufactured. Demonstrated debubbler was successfully applied in a long-lasting culture of HT-29 cell aggregates.

Opis:
The aim of this work was the evaluation of ion-beam induced luminescence for the characterization of luminescent oxide materials containing rare earth elements. The yttrium aluminium garnet epilayers doped with Nd, Pr, Ho, and Tm atoms were used. The ion-beam induced luminescence spectra were excited using 100 keV $H_2^{+}$ ion beam and were recorded in the wavelengths ranging from 300 nm up to 1000 nm. The separate parts of the surface of the same samples were used for ion-beam induced luminescence and cathodoluminescence experiments. Cathodoluminescence spectra have been recorded in the range from 370 nm up to 850 nm at 20 keV e-beam in scanning electron microscope equipped with a grating spectrometer coupled with a photomultiplier. The observed narrow ion-beam induced luminescence lines can be ascribed to the well known radiative transitions in the rare-earth ions in the YAG crystals. The cathodoluminescence spectra reveal essentially the same emission lines as ion-beam induced luminescence. The decrease of the ion-beam induced luminescence lines intensity has been observed under the increasing ion fluences. The ion-beam induced luminescence may be used for characterization of transparent luminescent materials as an alternative method for cathodoluminescence and can be especially useful for observation of ion-beam damage formation in crystals.
