The aim of the study was to determine the effect of the additive on NAA and BAP on the formation of callus tissue on leaf chamomile explants, cultivar, and to determine the optimal content of said growth regulators for the initiation of callus tissue in vitro. During the experiment fragments of chamomile from the collection of the Department of Genetics, Breeding and Biotechnology of Plant were laid on a medium with mineral composition according to Murashige and Skoog - MS (1962) enriched with auxin NAA in concentrations of 3 and 5 mg·dm-3 and cytokinin BAP in a concentration of 1, 2 , 5, 7 mg·dm-3. Based on the conducted experiment it was found that the initiation of callus chamomile tissue is possible in the case of using leaf explants lined with MS in the combination of auxin NAA in concentration 3 or 5 mg·dm-3 and BAP from 1 to 7 mg·dm-3. The propagation of chamomile callus tissue should be carried out on MS proliferating medium enriched with NAA at a rate of 3 mg·dm-3 and BAP in an amount of 7 mg·dm-3. Explants laid on a medium with this composition develop a large mass of callus tissue.

The aim of the study was to determine an optimal composition of growth media for Drosera rotundifolia L. cultured in vitro. Adventitious shoots of Drosera rotundifolia originating from an established in vitro culture were placed on complete Murashige and Skoog (MS) (1962) medium, MS medium with nutrient content reduced to ¾ MS, ½ MS, ¼ MS, 1/6 MS and MS medium with selected macronutrient (KNO3, KNO3 + NH4NO3, KH2PO4 or CaCl2) content reduced by ½. The medium containing ¼ of standard MS nutrients was found the most effective, as plants in this variant had the greatest weight and developed the greatest number of adventitious shoots (37.1). It was also conducive to root growth and development. Flowering was only observed on complete MS, ¾ MS and ½ MS media and MS with reduced content of KNO3 or KNO3 + NH4NO3. Reducing mineral content to ¼ MS or more resulted in red coloration of glandular tentacles.

The aim of the study was to investigate the influence of plant growth regulators on callus culture of common thyme (Thymus vulgaris L.) and the possibility of using such callus cultures to obtain rosmarinic acid. The explants used to initiate shoot culture were the seeds of common thyme. The seeds were individually placed into test tubes with MS culture media of macro- and micro-element content according to Murashige and Skoog (1962). For the purpose of initiation of cultures, leaves dissected along the vascular bundle were used. The leaves were placed on MS medium with the addition of BAP in combination with NAA in a concentration of 3 and 5 mg•dm-3. Propagation of callus cultures was conducted with the use of fragments of callus tissue placed on MS media supplemented with BAP used separately in a concentration of 3 mg•dm-3, and in combination with NAA (1, 2, 3 mg•dm-3) and 2,4-D in a concentration of 0.5, 1 or 1.5 mg•dm-3 respectively. In each stage of the experiment, explants placed on MS media without the addition of plant growth regulators were the control. It was found that initiation of common thyme cultures should be conducted on MS media without plant growth regulators. For the purpose of initiation of callus culture of common thyme, the optimal culture medium was MS with the addition of 3 mg•dm-3 BAP and NAA. Propagation of callus tissue of common thyme should be conducted on culture media supplemented with 3 mg•dm-3 BAP used in combination with 1 mg•dm-3 NAA. It was observed that the mass of the propagated callus tissue decreased with an increase of NAA content.
