Hairy root cultures obtained after Agrobacterium rhizogenes-mediated genetic transformation can serve as a model system for studying plant metabolism and physiology, or can be utilized for the production of secondary metabolites. So far no efficient protocol of hairy root development in sugar beet has been publically released. In this work, two A. rhizogenes strains (A4T and LBA1334) carrying a binary vector pBIN-m-gfp5-ER or pCAMBIA1301 possessing gfp and uidA reporter genes were used to transform petiole explants of haploid and diploid sugar beet genotypes. Five treatment combinations of sonicated-assisted Agrobacterium-mediated transformation were compared. Hairy roots appeared on 0% to 54% of explants depending on the treatment combination used. The highest frequency was achieved when explants of a diploid genotype were sonicated for 15 s in the inoculum containing A. rhizogenes of OD600=0.5 and then co-cultured for three days. Using the same treatment combinations the explants of haploid genotypes developed hairy roots with the frequency ranging from 10% to 36%. Transformation efficiency was independent on the bacterial strain used. The results indicate that haploid sugar beet explants are amenable to transformation using A. rhizogenes, and that the efficiency of that process can be increased by applying short ultrasound treatment.

Opis:
The potential effect of genetic modification on nutritional properties of potatoes transformed to improve resistance to a necrotic strain of Potato virus Y was determined in a rat experiment. Autoclaved tubers from four transgenic lines were included to a diet in the amount of 40% and compared with the conventional cv. Irga. The experiment lasted 3 weeks and special attention was paid to nutritional properties of diets, caecal metabolism and serum indices. Genetic modification of potato had no negative effect on the chemical composition and nutritional properties of tubers, ecosystem of the caecum, activity of serum enzymes and non-specific defence mechanism of the rats. Obtained results indicate that transgenic potato with improved resistance to PVYN: line R1F (truncated gene coding for PVYN polymerase in sense orientation), R2P (truncated gene coding for PVYN polymerase in antisense orientation), and NTR1.16 (non-translated regions of PVYN genome in sense orientation) are substantial and nutritional equivalence to the non-transgenic cultivar. Tubers of transgenic line NTR2.27 (non-translated regions of PVYN genome in antisense orientation) increased the bulk of caecal digesta and the production of SCFA as compared to tubers of the conventional cultivar and the other transgenic clones. Taking into account some deviations, it seems reasonable to undertake a long-term feeding study to confirm the nutritional properties of tubers of transgenic lines.

Before discovering methods of genetic transformation, people had to improve the quality of plants by artificial selection and crossing. Nowadays, these actions have been taken up by genetic engineering which requires significantly less time to achieve the expected features. Genetically modified are mostly plants of great economic value. Changing the genome is expected to result in obtaining a plant with desired features. The most common modifications are: 1. Resistance to crop protection chemicals. Giving the plant the resistance to the herbicide allows for its farming without the risk of damaging the crop. The modified plants have either completely new or additional copies of an already present gene responsible for synthesizing enzymes decomposing herbicides. A plant able to decompose herbicides becomes resistant to them. 2. Resistance to diseases caused by fungi, viruses, and bacteria. It may be acquired by introducing a transgene encoding the enzymes destroying the pathogens’ cell wall. Another transformed gene encodes osmotin – a protein associating with the pathogen’s cell membrane, causing its destruction. Resistance to viruses is acquired by introducing genes which encode the proteins of the virus’ capsid and its enzymes. The presence of these proteins results in a milder infection caused by this virus or significantly delayed onset of the disease’s symptoms. 3. Resistance to pests – insects. The Bt gene responsible for such resistance is obtained from a soil bacteria Bacillus thuringensis. The gene encodes a specific protein – Cry – which is toxic to insects. The pest dies after consuming the plant’s cells. Currently, contrary to popular opinion, the range of genetically modified crops is rather small. In Europe, only two genetically modified plants have been allowed for use: MON 810 corn (Monsanto) and Amflora potato (BASF). However, genetic modifications of plant aimed at improving their protection against pathogens or their tolerance to plant protection chemicals give various positive results. Resistance to herbicides results in, among others: less strict rules concerning time of application, lower number of uses needed since both monocotyledonous and dicotyledonous weeds are destroyed, environment protection by reduced use of the active agent, reduction of production cost by limited expenses on herbicides. Also, implanting the plants with resistance to fungi, viruses and bacteria, as well as to pests, result in economic, environmental and health profits. Thus, there is need for continued research in the use and protection of genetically modified plants.

Opis:
The objective of this discussion is an attempt to recall that already in Polish modernism, in the work of Tadeusz Miciński (1873-1918), the idea of modifying the human phenotype was signaled. Contemporary times offer the possibility of its partial (for the time being) realization. That is why it is worth confronting this idea with the foundations of religious ethics represented by the teaching of the Roman Catholic Church. It is also worth recalling the concerns that Jürgen Habermas (b. 1929) raises about the possibility of the unrestrained development of genetic engineering. Being a secular thinker, the German philosopher analyzes the ethical dangers of liberal eugenics and, in this way, opens up the search for a consensus with religious ethics.
