Opis:
Hypericum perforatum is well known for its antidepressant and anti-inflammatory activities, for which hypericin and its derivatives are indicated to be the most active compounds. Hypericin synthase (Hyp-1) is the only protein proven to catalyze the synthesis of hypericin. In this study, the full-length cDNA of Hyp-1 was chemically synthesized according to the Hyp-1 sequence in GenBank (accession no. AY148090) and then cloned into the plasmid pET22b. Hyp-1 was expressed in Escherichia coli BL21 (DE3) and purified with a Ni-NTA column. The purified protein was used to immunize New Zealand white rabbits, from which an antiserum was purified by protein G affinity chromatography. The polyclonal antibody against Hyp-1 provides a valuable tool for the study of hypericin biosynthesis in H. perforatum. Expression of Hyp-1 and the cellular distribution of hypericin were analyzed in different organs of red-pigmented H. perforatum plantlets. The black glands were not the only site of hypericin accumulation and the results indicated that hypericin might be synthesized in mesophyll cells or in tissues of the root and/or stem and then transported to the glands. This work provides a foundation for further investigation of the regulatory mechanism of hypericin synthesis during the development of H. perforatum.

Opis:
Due to space limitations during installation, reducing low-frequency noise has always been a challenging area. Sub-wavelength structures are typically favored in such scenarios for noise reduction. This paper explores the potential of micro-slit panels (MSP) for low-frequency sound absorption. To further optimize the panel thickness, coupled MSPs (CMSP) with a distance between two MSPs of less than 1 mm are proposed. Firstly, the low-frequency absorption performances of a single MSP based on two optimized schemes – the cavity-depth optimal scheme (COS) and the panel thickness optimal scheme (TOS) – are examined and compared with those of existing ultrathin metamaterials. The results demonstrate that MSP has significant potential for low frequency sound absorption, and COS allows for a smaller overall structural thickness but a larger panel thickness than TOS. Secondly, to reduce the panel thickness, the CMSP is developed and the theoretical model of its acoustic impedance is established and validated by experiments. Then, based on the theoretical model, the low-frequency absorption potential of CMSP is optimized using COS. The results show that both the overall thickness and the panel thickness of the CMSP absorber are reduced while maintaining better performance. Furthermore, the proposed absorber achieves a subwavelength scale since its total thickness can be as small as 0.138λ.

The graphene-like two dimensional (2D) inorganic materials have been been shown great interest for a variety of applications. In this work, polymer composite nanofibres containing molybdenum disulfide (MoS2) nanosheets were obtained by electrospinning. The MoS2 nanosheets were well dispersed inside the fibres, and the nanofibres maintained the fibre morphology well with the MoS2 nanosheets embedded. The incorporation of MoS2 nanosheets changes polymer nanofibre morphology from round to ribbon-like. Moreover, through thermogravimetric (TG) analysis and dynamic mechanical thermal analysis (DMTA) measurements, it was found that the MoS2 nanosheets as an additive material led to an increase in thermal stability and in the storage modulus. This work comprises an extensive approach to producing a novel 2D inorganic-organic composite structure, which should be applicable for membrane engineering with enhanced thermal and mechanical stability.
Dwuwymiarowe nieorganiczne materiały podobne do grafenu wywołały liczne zainteresowanie w różnych zastosowaniach. W pracy otrzymano metodą elektroprzędzenia kompozytowe nanowłókna polimerowe zawierające disiarczek molibdenu (MoS2). Nanoskładniki MoS2 były dobrze rozproszone we włóknach, a ich morfologia była na zadowalającym poziomie. Włączenie nanoskładników MoS2 zmienia nanowłókna polimerowe z morfologii okrągłej na wstążkową. Co więcej, dzięki analizie termograwimetrycznej (TG) i pomiarom dynamicznej mechanicznej analizy termicznej (DMTA) stwierdzono, że dodatek MoS2 może zapewnić wzrost stabilności termicznej i zwiększyć moduł przechowywania. Praca prezentuje rozszerzone podejście do produkcji nowej dwuwymiarowo nieorganiczno-organicznej struktury kompozytowej, która może mieć zastosowanie w wytwarzaniu membran o podwyższonej stabilności termicznej i mechanicznej.

The research focused on such packing media as ceramsite, polypropylene balls, and elastic fillers, and analyzed the main characteristics of extracellular polymeric substances (EPS) in their filter biofilms. The EPS were categorized as soluble EPS (S-EPS) and bound EPS (B-EPS). The component characteristics of stratified EPS were investigated via UV-Vis spectroscopy and a three-dimensional excitation-emission matrix (3D-EEM). The results showed that the EPS content of ceramsite biofilm was 245.2 mg/g VSS, which was higher than those of elastic filler material and polypropylene ball by 1.26 and 1.51 times, respectively. The protein (PN) and polysaccharide (PS) ratio of EPS in the ceramsite filter material was highest in S-EPS and B-EPS, indicating that the EPS have a stable structure. More than 67.58% of EPS formed by ceramsite was tightly bound EPS (TB-EPS), which was beneficial to maintaining the stability of biofilms. The levels of proteins (PN) and humic substances formed by ceramsite contained in the TB-EPS were higher than those of elastic filter material and polypropylene balls. 3D-EEM fluorescence spectra revealed that TB-EPS formed by a ceramsite contained high concentrations of tryptophan, tyrosine, and humic substances. The dehydrogenase activity of biofilm formed by ceramsite filler was higher than that formed by elastic fillers and polypropylene balls.
