Two Chlamydomonas reinhardtii CW15 populations modified by an excess of copper in growth medium were obtained: a “Cu” population that was continuously grown under the selection pressure of 5 μM Cu²⁺ (for at least 48 weeks) and the “Re” population, where a relatively short (9 week) exposure to elevated copper, necessary for acquiring tolerance, was followed by a prolonged period (at least 39 weeks) of cultivation at a normal (0.25 μM) copper concentration. Cells of the Cu population were able to multiply at a Cu²⁺ concentration 16 times higher than that of the control population at a normal light intensity and at a Cu²⁺ concentration 64 times higher when cultivated in dim light. The potential quantum yield of photosystem II (FV/FM ratio) under copper stress was also significantly higher for the Cu population than for Re and control populations. The Re population showed only residual tolerance towards the elevated concentration of copper, which is revealed by an FV/FM ratio slightly higher than in the control population under Cu²⁺ stress in dim light or in darkness. We postulate that in the Chlamydomonas populations studied in this paper, at least two mechanisms of copper tolerance operate. The first mechanism is maintained during cultivation at a standard copper concentration and seems to be connected with photosynthetic apparatus. This mechanism, however, has only low adaptive value under excess of copper. The other mechanism, with a much higher adaptive value, is probably connected with Cu²⁺ homeostasis at the cellular level, but is lost during cultivation at a normal copper concentration.

The optical coherence tomography is an optical measurement technique used for investigation of wide range of scattering or semitransparent materials. The optical coherence tomography enables surface and subsurface examination of different types of materials to be performed in non-contact and non-destructive way. Our research have been concentrated on optical coherence tomography systems with polarization sensitive analysis. Such a combination of optical measurements methods improves optical coherence tomography visualization contrast and also delivers some extra information about investigated devices. However, applying polarization sensitive analysis is associated with problems related to the use of ultra broadband optical sources. We have managed studies on the improvement of signal processing method. According to obtained results the problem of signal processing method in polarization sensitive optical coherence tomography can be solved by the use of the spectroscopic analysis. In this paper we present a brief discussion about the polarization sensitive optical coherence tomography measurements supported by spectroscopic analysis of backscattered light from investigated device.

Opis:
Optical low-coherence tomography is a measurement technique for non-contact and non-destructive investigation of materials inner structure. Nowadays, this method is highly applied in medical treatment especially in dermatology and ophthalmology. During our research we have developed an optical low-coherence tomography system with polarization state analysis for structure examination of a broad range of technical materials. In this paper we present our recent measurements including polarization sensitive analysis. Those measurements were taken for semitransparent and highly scattering materials like polymer layers and polymer optical retarders, anticorrosion protective coatings or abrasive paper. We proved that with the aid of developed polarization sensitive optical coherence tomography system it is possible to investigate birefringence principles of materials as well as to improve visualization contrast of cross-sectional images.

Opis:
Optical Coherence Tomography (OCT) is one of the most rapidly advancing techniques. This method is capable of non-contact and non-destructive investigation of the inner structure of a broad range of materials. Compared with other methods which belong to the NDE/NDT group (Non-Destructive Evaluation/Non-Destructive Testing methods), OCT is capable of a broad range of scattering material structure visualization. Such a non-invasive and versatile method is very demanded by the industry. The authors applied the OCT method to examine the corrosion process in metal samples coated by polymer films. The main aim of the research was the evaluation of the anti-corrosion protective coatings using the OCT method. The tested samples were exposed to a harsh environment. The OCT measurements have been taken at different stages of the samples degradation. The research and tests results have been presented, as well as a brief discussion has been carried out.
